[Ab] Abstract:The total flavonoids from Hemerocallis citrina baroni are regarded as a green and natural health care product with many beneficial impacts on human health. In this study, ultrasound-synergized electrostatic field extraction (UEE) of the total flavonoids (TF) from H. citrina was investigated. Significant independent variables of the extraction, including the electrostatic field, ultrasonic power, ethanol concentration and extraction time, were optimized using the Box-Behnken (BB) method, and the optimal extraction conditions were obtained by response surface methodology (RSM). The extraction yield using UEE was compared with the yields obtained using only ultrasound extraction (UE) and water bath extraction (WE), using a UV-vis spectrophotometer. The best extraction yield of 1.536% using UEE was achieved under the following optimal conditions: electrostatic field of 7kV, ultrasonic power of 500W, ethanol concentration of 70% and extraction time of 20min. The optimal solid-liquid ratio (1:25g/mL) and extraction temperature (55°C) were determined by single factor experiments. Compared to other extraction methods, UEE not only increases the extraction yield of TF but also exhibits an excellent antioxidant activity in assays of the scavenging capacity for DPPH, hydroxyl and superoxide anion radicals. The availability of the UEE method can be supported by the ultrasonic cavitation effect, which plays the most important role in the UEE method. The electrostatic field can be regarded as a random disturbance for sonication, which can strengthen the cavitation effect and increase the cavitation yield. Moreover, the amount of iodine release in potassium iodide solution well validated the synergetic effect between the ultrasound and electrostatic field.

[Ab] Abstract:Gene expression analysis using reverse transcription quantitative real-time PCR (RT-qPCR) requires the use of reference gene(s) in the target species. The long yellow daylily, Hemerocallis citrina Baroni. is rich in beneficial secondary metabolites and is considered as a functional vegetable. It is widely cultivated and consumed in East Asian countries. However, reference genes for use in RT-qPCR in H. citrina are not available. In the present study, six potential reference genes, actin (ACT), AP-4 complex subunit (AP4), tubulin (TUB), ubiquitin (UBQ), 18S and 60S ribosomal RNA, were selected and their expression stability in different developmental stages, organs and accessions was evaluated using four statistical software packages (geNorm, NormFinder, BestKeeper, and RefFinder). For commercial flower buds of different landraces, the combination of 60S, TUB, and AP4 was appropriate whereas ACT and 60S was suitable for normalization of different organs. In addition, AP4 exhibited the most stable expression in flower buds among different developmental stages. UBQ was less stable than the other reference genes under the experimental conditions except under different organs was 18S. The relative expression levels of two genes, primary-amine oxidase (HcAOC3) and tyrosine aminotransferase (HcTAT) which play important roles in alkaloid biosynthesis were also examined in different organs of the 'Datong' landrace, which further confirmed the results of selected reference genes. This is the first report to evaluate the stability of reference genes in the long yellow daylily that can serve as a foundation for RT-qPCR analysis of gene expression in this species.

[Ab] Abstract:Eight new γ-lactam alkaloids, hemerominors A-H (1-8), including two pair of epimers (1-4), together with six known compounds (9-14) were isolated from the roots of Hemerocallis minor Mill. The structures of 1-8 were established on the basis of extensive NMR studies and HR-MS measurements as well as comparison with literature data. The absolute configurations of 1-8 were determined by CD spectral analysis and modified Mosher's method. All of compounds were evaluated for their inhibitory effects against LPS-induced NO production in RAW264.7 macrophages. Among them, compound 13 exhibited moderate inhibitory activity against NO production and with IC value of 18.0 µM.

[Ab] Abstract:This study was aimed at the identification and quantification of the protein components of the pollen grains in parallel with the distal stigmatic tissue of tetraploid cultivars. Proteomes were analyzed using iTRAQ 4plex labeling, peptides separation by online RP-nano-LC and analysis by ESI-MS/MS. Protein identification and quantification were made using the Asparagales database as a reference. A total of 524,037 MS/MS spectra were produced from pollen and stigma samples. From these, a total of 8368 peptides wereidentified corresponding to 994 unique peptides and 432 protein groups. Among them, 128 differentially expressed proteins were retained for further analysis. In absence of the daylily genome availability, we exploited numerous databases and bioinformatics resources to exploring the putative biological functions of these proteins. The profile of differentially expressed proteins suggests an important representation of functions associated to the signalling and response against endogenous and environmental stresses, including several enzymes implicated in the biosynthesis of antibiotics. The abundance in stigma of several structural proteins of the ribosomal sub-units as well as of the core histones suggest that the translation processes and the regulation of gene expression in stigma is a more active mechanism than in pollen. In addition, pollen prioritizes the synthesis of fructose and glucose as opposed to sucrose in stigma as a source of energy. Finally, the modulated proteins in Hemerocallis point to several pathways that give potential clues concerning the molecular mechanisms underlying the functions of the pollen and the stigmatic fluid in daylily reproduction.

[Ab] Abstract:BACKGROUND: Daylily flowers, the flower and bud parts of Hemerocallis citrina or H. fulva, are well known as Wang-You-Cao in Chinese, meaning forget-one's sadness plant. However, the major types of active constituents responsible for the neurological effects remain unclear. This study was to examine the protective effects of hydroalcoholic extract and fractions and to identify the active fractions. METHODS: The extract of daylily flowers was separated with AB-8 resin into different fractions containing non-phenolic compounds, phenolic acid derivatives and flavonoids as determined using UPLC-DAD chromatograms. The neuroprotective activity was measured by evaluating the cell viability and lactate dehydrogenase release using PC12 cell damage models induced by corticosterone and glutamate. The neurological mechanisms were explored by determining their effect on the levels of dopamine (DA), 5-hydroxy tryptamine (5-HT), γ-aminobutyric acid (GABA), noradrenaline (NE) and acetylcholine (ACh) in the cell culture medium measured using an LC-MS/MS method. RESULTS: Pretreatment of PC12 cells with the extract and phenolic fractions of daylily flowers at concentrations ranging from 0.63 to 5 mg raw material/mL significantly reversed corticosterone- and glutamate-induced neurotoxicity in a dose-dependent manner. The fractions containing phenolic acid derivatives (0.59% w/w in the flowers) and/or flavonoids (0.60% w/w) exerted similar dose-dependent neuroprotective effect whereas the fractions with non-phenolic compounds exhibited no activity. The presence of phenolic acid derivatives in the corticosterone- and glutamate-treated PC12 cells elevated the DA level in the cell culture medium whereas flavonoids resulted in increased ACH and 5-HT levels. CONCLUSION: Phenolic acid derivatives and flavonoids were likely the active constituents of daylily flowers and they conferred a similar extent of neuroprotection, but affected the release of neurotransmitters in a different manner.

[Ab] Abstract:There are limited data concerning the role of endogenous H S in prolonging the postharvest of vegetables and fruits. Using a fluorescence microscope with a specific probe, we discovered that, during the senescence of postharvest daylily flower, endogenous H S homeostasis was impaired. The activities of two important synthetic enzymes of H S, l- and d-cysteine desulfhydrase, exhibited decreasing tendencies. However, NaHS (a H S donor) not only blocked the decreased H S production but also extended the postharvest life of daylilies. These beneficial roles were verified by the alleviation of lipid peroxidation and the increased activities of antioxidant enzymes. Meanwhile, the energy status was sustained, and the respiration rate was decreased. In contrast to NaHS, the addition of an inhibitor of H S synthesis alone aggravated lipid peroxidation and lowered energy charge. Together, the present study implies that endogenous H S alleviates senescence of postharvest daylilies via increasing antioxidant capacity and maintained energy status.

[Ab] Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Depression induce distressed emotional state and cognitive deficits simultaneously, which both should be improved in the treatment. Hemerocallis citrina Baroni (HC) is a traditional herbal medicine in Eastern-Asia areas and the total phenols extract of HC (HCPE) contains the main active ingredients. It has been reported that HC has the emotional improvement effect. But the cognitive effect of HC was seldom researched. AIM OF THE STUDY: We designed to evaluate the antidepressant and cognitive improvement effect of HCPE using a chronic unpredictable mild stress (CUMS) model, and the potential mechanisms were explored by investigating the corticosterone (CORT), monoamine neurotansmitters, brain-derived neurotropic factor (BDNF) and oxidative stress. MATERIALS AND METHODS: The depression rats were induced by CUMS procedures and treated with HCPE (10, 20, 40mg/kg/day, by gastric gavage). The antidepressant effect was evaluated by sucrose preference test, open field test and body weight, while the cognitive improvement was investigated using morris water maze test. Besides, the levels of monoamine neurotransmitters in the hippocampus and frontal cortex were measured by ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). The serum CORT and BDNF in hippocampus were test using enzyme-linked immunosorbent assay (ELISA) kits. The oxidative stress indicators in frontal cortex were also analyzed. RESULTS: HCPE (40mg/kg) improved the emotion and cognition related behaviors in depression effectively. Moreover, HCPE increased the neurotransmitters concentration (5-HT, DA and NE) in the hippocampus and frontal cortex compared with CUMS rats. Meanwhile, the CUMS induced changes of serum corticosterone level and the hippocampus BDNF level were reversed. Besides, HCPE reduced malondialdehyde (MDA) in the frontal cortex of model rats. CONCLUSION: It suggested that HCPE could improve the depression-like emotional status and associated cognitive deficits in CUMS rats, which might be mediated by regulation of neurotransmitters and BDNF levels in brain, alleviation of corticosterone level as well as the alleviation of oxidative stress.

[Ab] Abstract:The effects of a special ornamental plant Hemerocallis middendorfii Trautv. et Mey. on remediating petroleum-contaminated soil from the Dagang Oilfield in Tianjin, China, was studied by a greenhouse pot-culture experiment and the gradients of TPHs were 0, 10,000 and 40,000 mg · kg⁻¹. The results suggested that H. middendorfii had a high tolerance to TPHs (≤ 40,000 mg · kg⁻¹). And H. middendorfii significantly (P < 0.05) promoted the removal rate of TPHs (53.7% and 33.4%) compared with corresponding controls (31.8% and 12.0%) by natural degradation, respectively. The relative abundance of amino acids, organic acids and sugars and others in soil were analyzed by gas chromatography-mass spectrometry (GC-MS), and PCA and PLS-DA models were to investigate the rhizospheric mechanisms. The results suggested that H. middendorfii changed the distribution characteristics of each component in soil, and the glucopyranoside played a key role in the removal of TPHs. Furthermore, the results about comparative metabolic profile showed that some special metabolites were only found in the contaminated groups, including alanine, tetradecanoic acid, hexadecanoic acid and 9,12-octadecadienoic acid. Additionally, the exposure of TPHs changed the primary metabolic flux of roots, and caused the significant (P < 0.01) change of metabolites. In conclusion, H. middendorfii might be an enduring ornamental plant for effective remediating TPHs (≤ 40,000 mg · kg⁻¹) in soil. But the exposure of TPHs had changed the metabolic profile of H. middendorfii in roots, which might be the metabolic response of H. middendorfii to petroleum-contaminated soil.

[Ab] Abstract:Soil moisture variation in dryland sloping jujube. orchard was investigated after introducing two economic crops, i.e., feed Brassica napus (JR) and Hemerocallis fulva (JH) planted between jujube rows. Jujube tree without inter-row crop was set as control (CK). The results showed that mean soil moisture for JR and JH in the 0-180 cm soil layer increased by 6.2% and 10.1% compared with CK, respectively. Soil moisture changed mainly in the 0-60 cm soil layer in growth stage of Jujube trees. Soil moisture in JR and JH treatments significantly increased in the 0-60 cm soil layer, which could meet the demand in water resource of jujube plantation. The water consumption of jujube trees also mainly concentrated in the 0-60 cm soil layer. There was a significant decay exponential relationship between the soil moisture in the 0-20 cm layer and the drought duration after rainfall. During the 18-day dry period after rain, the soil moisture contents of JR and JH were apparently higher than that of CK. In conclusion, the jujube-crop intercropping system improved the soil moisture condition. It was an effective measure to overcome the seasonal drought in jujube orchards on the loess hilly region.

[Ab] Abstract:A previous study found that the antidepressant-like effects of ethanolic extracts from Hemerocallis citrina are predominantly related to the flavonoid, hesperidin. The study herein aimed to explore the antidepressant-like mechanism of hesperidin in mice induced by chronic mild stress (CMS). The results indicated that hesperidin reversed the reduction of sucrose preference and the elevation of immobility time in mice induced by CMS. In addition, the increase in serum corticosterone levels and decrease in hippocampal extracellular signal-regulated kinase (ERK) phosphorylation and brain-derived neurotrophic factor (BDNF) levels in CMS mice were also ameliorated by hesperidin treatment. In contrast, improvement by hesperidin was suppressed by pretreatment with ERK inhibitor SL327. Taken together, our findings confirmed the antidepressant-like effect of hesperidin and indicated that hesperidin-induced BDNF up-regulation was mediated in an ERK-dependent manner.
