[Ab] Abstract:The extract of ethanol, methanol and DMSO of pennyroyal leaves from Mugla Region (Turkey) were tested for antimicrobial activity against eleven bacterial and one yeast strain by disc diffusion method. Among the extracts assayed, the methanol extract of pennyroyal leaves exhibited significant antimicrobial activity against Salmonella typhimurium and Staphylococcus aureus with 20 mm diameter inhibition zone. The DMSO extract of pennyroyal leaves displayed significant activity against S.aureus (19 mm) and Bacillus subtilis (21mm) showing inhibition zone of 19 mm and 21 mm diameter, while the ethanolic extract showed significant antimicrobial activity against S.aureus (17mm) and Klebsiella pneumoniae (20mm) inhibition zones.? On comparint the MIC value of ethanol, methanol and DMSO leaf extract, methanolic extract of pennyroyal presented best activity (MIC 8 mg ml-1) against S. typhimurium CCM 583 and S.aureus ATCC 6538/P. Analyses of GC/MS determined eleven compounds viz., neophytadiene (69.95%), Pulejon 7.85%, Pinane 4.81%, Bicyclo (3.1.1 Heptane 2.6.6.6 trimethyl) 4.68%. In conclusion, methanolic extracts of M. pulegium showed antimicrobial activity because of high neophytadiene content.

[Ab] Abstract:In this paper, Mentha pulegium leaves extract was used as a green reducing agent for the synthesis of silver-nanoparticles. The synthesized silver-nanoparticles were characterized by UV-VIS spectrophotometry, transmission electron microscopy, X-ray spectroscopy and used as an adsorbent for preconcentration of trace levels of cadmium (ІІ). After the desorption of cadmium (ІІ) in 5 mol L formic acid, the desorbent solution was aspirated into the flame atomic absorption spectrometry for the determination of cadmium. In order to optimize the experimental condition, a response surface methodology based on central composite design was used. The optimum conditions are: pH: 8.6, amounts of adsorbent: 30 mg, 10 min extraction time and desorption time of 2 min. Under the optimum condition, the calibration curve was linear in the range of 5-200 µg L cadmium (ІІ) ion with a correlation coefficient of 0.9995. The limit of detection was 1.1 µg L and the relative standard deviation for 25 µg L cadmium (ІІ) ion was 3.0% (n = 5). In order to check the applicability of the proposed method, different real samples were analyzed. Also, the accuracy of this method was successfully checked by the analysis of certified reference material and spike tests.

[Ab] Abstract:Mentha pulegium (Lamiaceae) tea has been used as a traditional medicine; however, the modulatory effect of M. pulegium extracts on damage to human erythrocytes associated to t-butyl hydroperoxide (t-BHP) exposure remains to be investigated. Accordingly, we perform this study in order to test the hypothesis that aqueous and ethanolic extracts of M. pulegium could modulate the hemolysis associated to t-BHP exposure, non-protein thiol (NPSH) oxidation and lipid peroxidation (measured as thiobarbituric acid reactive substances - TBARS) in human erythrocytes. Samples were co-incubated with t-BHP (4 mmol/L) and/or aqueous or ethanolic extracts (10-1000 mg/mL) during 120 min to further analysis. We found that both extracts, when associated to t-BHP, potentiate NPSH oxidation and hemolysis. Moreover, both extracts significantly prevents against t-BHP-induced TBARS production. A significant correlation among hemolysis and NPSH levels was found. Taking together, our data points that the association of M. pulegium extracts with t-BHP culminates in toxic effect to exposed erythrocytes, besides its protective effect against t-BHP-induced TBARS production. So, we infer that the use of this extract may exert negative effect during painful crisis in sickle cell anemia. However, more studies are still necessary to better investigate/understand the mechanism(s) involved in the toxic effect resultant from this association.

[Ab] Abstract:This study was planned in order to investigate effective essential oils to inhibit in-vitro growth of Methicillin resistant Staphylococcus aureus (MRSA). In this study using disc diffusion method anti MRSA activity of ten diverse essential oils extracted from traditional plants namely Thymus vulgaris L, Mentha pulegium, Ocimum sanctum, Mentha piperita, Cymbopogon citratus, Rosmarinus officinalis L., Cortex cinnamom, Citrus nobilis x Citrus deliciosa, Origanum vulgare and Mentha sp. was examined. All the essential oils inhibited growth of S. aureus to different extent, by exhibiting moderate to elevated zones of inhibitions. Essential oils of cinnamon (Cortex cinnamomi) and thyme (Thymus vulgaris L) were observed to be the most powerful against MRSA strains used in this study. At lowest concentration of 25µl/ml essential oils comprehensible zone of inhibition was found 9±0.085mm and 8±0.051mm respectively, and at elevated concentrations there was a total decline in growth of MRSA and a very clear zone of inhibition was observed. A synergistic effect of essential oils in amalgamation with amoxicillin a Penicillin group of antibiotic was also examined. Interestingly a strong synergism was observed with oregano (Origanum vulgare) and pennyroyal mint (Mentha pulegium) essential oils, which were not so effective alone driven out to be important synergistic candidate. Our results demonstrated that essential oils of cinnamon and thyme can be used as potential antimicrobial agent against the Methicillin-resistant Staphylococcus aureus infections and Amoxicillin antibacterial activity can be enhanced using active constituents present in oregano and pennyroyal mint essential oils.

[Ab] Abstract:Herpes simplex virus type 1 (HSV-1) causes serious infections particularly in immunocompromised patients. Methanolic extract of four plants were evaluated for their anti-viral effects against acyclovir resistant HSV-1 in HeLa cell line. The 50% cytotoxic concentration (CC50) as well as the effective minimal cytotoxic concentration of each plant extract were evaluated by MTT assay. Antiviral effects of the plant extracts on HSV-1 were examined at different concentrations of the extract. The effective minimal cytotoxic concentration was evaluated at different times of virus replication after infection. Virus titration was assessed by tissue culture infectious dose 50 (TCID50) method. Among the 4 plant extracts evaluated only Mentha pulegium L. extract was shown to exert the highest antiviral activity, with selectivity index (SI) 10.25. Direct treatment of HSV-1 with Mentha pulegium L. extract resulted in 1.7 log10 TCID50 reduction in virus titers after one hour. The highest reduction in HSV-1 infectivity was obtained 1 hour after the infection of the cells with virus resulting in 2.1 log10 TCID50 reduction as compared to the control. The antiviral effects of Mentha pulegium L. extract on HSV-1 after virus infection was more remarkable than the virucidal activity.

[Ab] Abstract:Chemical composition and antifungal activity of essential oils of Algerian Mentha species were studied. Chemical compositions of different Mentha species oils (Mentha rotundifolia, M. spicata, M. pulegium, and M. piperita) were investigated by capillary GC and GC/MS, and their antifungal activities were evaluated by means of paper disc diffusion method and minimum inhibitory concentration (MIC) assays. In total, 98 components from all Mentha species were identified. All oils were rich in monoterpene-oxygenated components. In addition, we reported fumigant antifungal activity of Algerian Mentha essential oils against four fungi: Botrytis cinerea, Penicillium expansum, Monilinia laxa, and M. fructigena. All oils demonstrated very good inhibition especially against B. cinerea, M. laxa, and M. fructigena. Both Monilinia fungi were extremely sensitive to all Algerian Mentha oils, which suggests that Mentha essential oils have the potential to be used as bio-pesticides to protect fruit trees, such as apple and pear trees, and provides an alternative to chemical pesticides.

[Ab] Abstract:The aim of the study was the determination of the chemical composition of Mentha pulegium L. and Rosmarinus officinalis L. essential oils and the evaluation of their antileishmanial, antibacterial and antioxidant activities. Essential oils (EOs) were isolated using steam distillation and the chemical composition was determined using GC-MS analysis. The antibacterial activity was tested against ten pathogenic strains using the diffusion method, the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) by microtitration assay. The antioxidant activity was estimated by DPPH free radical scavenging ability and ferric-reducing power. The antileishmanial activity was tested against Leishmania major, Leishmania tropica and Leishmania infantum using MTT (3-(4.5-dimethylthiazol-2yl)-2.5-diphenyltetrazolium bromide) assay. The yield of essential oils (v/w %) M. puleguim and R. officinalis based on dry weight were 5.4 and 2.7% respectively. GC/MS analysis of R. officinalis essential oil (ROEO) revealed the presence of 29 components, mainly represented by oxygenated monoterpenes (63.743%) and hydrocarbons monoterpenes (21.231%). Mentha pulegium essential oil (MPEO) revealed 21 components, mainly represented by oxygenated monoterpenes (83.865%). The major components of ROEO were α-pinene (14.076), 1,8-Cineole (23.673) and camphor (18.743), while menthone (21.164) and pulegone (40.98) were the main major components of MPEO. M. pulegium and R. officinalis EOs showed a significant antioxidant activity compared with ascorbic acid and Trolox to the IC values of 58.27 ± 2.72 and 85.74 ± 7.57 µg/mL respectively revealed by reducing power assay. As for the antibacterial effect, the highest zone diameters were shown by the MPEO against Bacillus subtilis (30 ± 1.43 mm) and Proteus mirabilis (28 ± 1.32 mm). These values are significantly important compared with those of the commercialized antibiotic (Erythromycin and Chlorophenicol). The lowest MIC and MBC values were obtained with MPEO against S. aureus MBLA (MIC = MBC = 0.25% (v/v)). While, ROEO has exhibited a bactericidal effect against Listeria monocytogenes (MIC = MBC = 0.5% (v/v)), Bacillus subtilis (MIC = MBC = 1% (v/v)) and Escherichia coli (MIC = MBC = 1% (v/v)). For the antileishmanial effect, ROEO is the most active against L. major (IC = 1.2 ± 0.36 µg/mL. While, the MPEO has the most leishmanicidal effect against L. major (IC = 1.3 ± 0.45 µg/mL). These findings show that the EOs of M. pulegium and R. officinalis are good sources of bioactive molecules that could have potential applications in the food and pharmaceutical industries.

[Ab] Abstract:Background It is important to consider the optimum conditions and processing factors (like solvent type) influencing activity of plant antioxidants for utilization in food and biological systems. Methods The antioxidant capacity and phenolic content of two Mentha species, namely, Mentha pulegium L. (MP) and Mentha rotundifolia (L.) Huds (MR), were studied and six solvent systems were used. The total antioxidant capacity of the mint species extracts was evaluated using phosphomolybdenum method and the free radical-scavenging capacity by 2,2-diphenyl-1-picrylhydrazyl radical-scavenging assay. Results The efficiency of the used solvents to extract phenols from the two species varied considerably. The highest total phenolic content was obtained from methanol extract of MP (25.3±1.3 mg GAE/gdw) and total flavonoid content from methanol extract of MR (10.1±0.1 mg QE/gdw). High phenol content was significantly correlated with high antioxidant capacity. The methanol extracts showed the highest radical scavenging activity. All the extracts showed variable antioxidant capacity by the formation of phosphomolybdenum complex. Acetone extract of MP and methanol extract of MR exhibited marked reducing power in this method. Conclusions Our findings identified the appropriate solvent for extracting MP and MR phenolics which might provide a rich source of natural antioxidants.

[Ab] Abstract:ETHNOPHARMACOLOGICAL EVIDENCE: Mentha pulegium L. leaves are used in the Iranian traditional medicine for the treatment of functional dyspepsia. AIM OF STUDY: To study the efficacy and safety of M. pulegium in the treatment of functional dyspepsia patients fulfilling the Rome III criteria. MATERIALS AND METHODS: The efficacy and safety of a standardized Mentha pulegium leaf extract (drug extract ratio: 15.9:1, extraction solvent: 70% v/v aqueous ethanol) (330mg three times daily taken for 2 months) as add-on to one famotidine 40mg tablet per day in the treatment of 50 functional dyspepsia patients were compared with those of a parallel placebo group (n =50). RESULTS: The extract significantly decreased the total dyspepsia score measured by the Hong Kong dyspepsia index compared to the placebo and baseline (P=0.011 and P<0.001 respectively). The stomach pain, upper abdominal bloating, upper abdominal dull ache, belching and total dyspepsia scores were decreased from baseline in the extract group significantly compared to the placebo (P<0.001, P<0.001, P=0.003, P<0.001 and P<0.001 respectively). However, the decreases of other dyspepsia symptoms scores from baseline in the extract group were not significant compared to the placebo (P>0.05). The extract improved the quality of life measured by the SF-36 questionnaire significantly compared to the placebo and baseline (P=0.003 and P<0.001 respectively). Moreover, the extract lowered the rate of H. pylori infection determined by the urease test significantly compared to the placebo and baseline (P=0.001 and P<0.001 respectively). The extract did not significantly affect the complete blood count and liver and kidney function tests (P>0.05). The patients did not experience any adverse drug effect. CONCLUSIONS: M. pulegium extract (genuine drug extract ratio: 19.4:1; extraction solvent: 70% v/v aqueous ethanol) 270mg three times daily taken for 2 months as adjunct to one famotidine 40mg tablet per day seems safe, improves dyspeptic symptoms and quality of life and eradicates H. pylori in functional dyspepsia patients.

[Ab] Abstract:A simple and eco-friendly method for efficient synthesis of stable colloidal silver nanoparticles (AgNPs) using extracts is described. A series of reactions was conducted using different types and concentrations of plant extract as well as metal ions to optimize the reaction conditions. AgNPs were characterized by using UV-vis spectroscopy, transmission electron microscopy, atomic force microscopy, dynamic light scattering, zetasizer, energy-dispersive X-ray spectroscopy (EDAX) and Fourier transform infrared spectroscopy (FTIR). At the optimized conditions, plate shaped AgNPs with zeta potential value of -15.7 and plasmon absorption maximum at 450 nm were obtained using high concentration of aqueous extract. Efficient adsorption of organic compounds on the nanoparticles was confirmed by FTIR and EDAX. The biogenic AgNPs displayed promising antibacterial activity on , , and . The highest antibacterial activity of 25 µg mL-1 was obtained for all the strains using aqueous extract synthesized AgNPs. The aqueous extract synthesised AgNPs also showed considerable antifungal activity against fluconazole resistant . The cytotoxicity assay revealed considerable anticancer activity of AgNPs on HeLa and MCF-7 cancer cells. Overall results indicated high potential of extract to synthesis high quality AgNPs for biomedical applications.
