[Ab] Abstract:OBJECTIVE: To determine the medicinal potential of various plants and their parts extracted with different solvents. METHODS: The total phenolic content of acetonitrile/water (60%-40%) (ACN/W) and aqueous (W) extract fractions was determined by high-performance liquid chromatography (HPLC), and terpenic compounds were detected by gas chromatography/mass spectrometry (GC/MS). Antioxidant activity of the samples was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and ß-carotene bleaching method. Cell viability was investigated by thiazolyl blue tetrazolium bromide [3-(4,5-dimethylthiazol)-2-yl 2,5-diphenyltetrazolium bromide] (MTT) assay. The mechanisms involved in cytotoxic activity were investigated in a murine macrophage cell line (RAW 264.7) and cancer lines. RESULTS: Our findings show that 11 plant species exhibited biological activity. In addition, moderate antibacterial activity was reported against one or more of the tested bacterial strains at two concentrations: 300 µg and 3 mg/disc. Furthermore, our data reveal that among all plants investigated, some extract and hydrophobic fractions were potent scavengers of the DPPH radical (6.78 µg/mL < EC50 < 8.55 µg/mL). Taken together, our results show that Nerium oleander (NOACN/W) and Pituranthos tortuosus (PTACN/W) were highly cytotoxic against RAW 264.7 cells with IC80 values of 0.36, and 1.55 µg/mL, respectively. In contrast, murine macrophage cell lines had low growth and were significantly sensitive to water extracts of Thymus hirtus sp. algeriensis (THW), Lavandula multifida (LMW), and ACN/W extract of Erica multiflora (EMACN/W) at doses > 400, 47.20, and 116.74 µg/mL, respectively. The current work demonstrates that RAW 264.7 cell proliferation was inhibited by samples in a dose-dependent manner. CONCLUSION: Our findings, validated through free radical scavenging activity, agar diffusion assay, and cytotoxicity of essential oils towards cancer cells, show that ethnomedicinal plants used in this work have a novel application as a tumor suppressor.

[Ab] Abstract:Herbs and plants are mostly used as antimicrobials and antioxidants owing to the harmfulness and linked side-effects of synthetic chemical constituents. Plants and spices produce various metabolites with antibacterial and antioxidant potential. These metabolites are principally revealed as encouraging healing components or mediators which control ailments in human beings. The present study was aimed to characterize the extracts from selected medicinal plants through in-vitro activities. Winged prickly ash, green tea and thyme were selected and extracted through ethanol and methanol solutions. The extracts were assessed for antibacterial and antioxidant activities. The antibacterial potential of extracts showed the significant extent of the activity against Bacillus subtilis and E. coli. The maximum activity was noted in 80% methanolic fraction of Thymus vulgaris (15.20±0.64 mm) against Bacillus subtilis. Antioxidant potential exhibited the highest phenolic and flavonoid content in Camellia sinensis . The total phenolic content was significantly higher (1456.26±12.05 mg gallic acid) in 80% ethanolic fraction of Camellia sinensis. The flavonoid content in different plant extracts ranged from 8.17±2.02 to 376.29±7.11 mg/g. The radical scavenging DPPH assay also showed the significant antioxidant capacity of selected plants with the methanolic (50%) extract of Camellia sinensis found to be the most potent (78.95±7.12%). It was concluded that the alcoholic extracts of selected medicinal plants revealed the effective antibacterial and antioxidant activity, showing protective prospective against oxidative injury.

[Ab] Abstract:Gynodioecy is a sexual dimorphism where females coexist with hermaphrodite individuals. In most cases, this dimorphism involves the interaction of cytoplasmic male sterility (CMS) genes and nuclear restorer genes. Two scenarios can account for how these interactions maintain gynodioecy. Either CMS genes recurrently enter populations at low frequency via mutation or migration and go to fixation unimpeded (successive sweeps), or CMS genes maintain polymorphism over evolutionary time through interactions with a nuclear restorer allele (balanced polymorphism). To distinguish between these scenarios, we used transcriptome sequencing in gynodioecious Thymus vulgaris and surveyed genome-wide diversity in 18 naturally occurring individuals sampled from populations at a local geographic scale. We contrast the amount and patterns of nucleotide diversity in the nuclear and cytoplasmic genome, and find ample diversity at the nuclear level (π = 0.019 at synonymous sites) but reduced genetic diversity and an excess of rare polymorphisms in the cytoplasmic genome relative to the nuclear genome. Our finding is incompatible with the maintenance of gynodioecy via scenarios invoking long-term balancing selection, and instead suggests the recent fixation of CMS lineages in the populations studied.

[Ab] Abstract:The compositions of essential oils (EOs) from Spanish marjoram (Thymus mastichina L.) grown in several bioclimatic zones of Murcia (SE Spain) were studied to determine their absolute and relative concentrations using gas chromatography-mass spectrometry. 1,8-Cineole and linalool were the main components, followed by α-pinene, ß-pinene and α-terpineol. (-)-Linalool, (+)-α-terpineol and (+)-α-pinene were the most abundant enantiomers. When the antioxidant capacities of T. mastichina EOs and their compounds were measured by five methods, EOs and linalool, linalyl acetate, α-terpinene and γ-terpinene, among others, showed antioxidant activities. All four T. mastichina EOs inhibited both lipoxygenase and acetylcholinesterase activities, and they might be useful for further research into inflammatory and Alzheimer diseases. Bornyl acetate and limonene showed the highest lipoxygenase inhibition and 1,8-cineole was the best acetylcholinesterase inhibitor. Moreover, these EOs inhibited the growth of Escherichia coli, Staphylococcus aureus and Candida albicans due to the contribution of their individual compounds. The results underline the potential use of these EOs in manufactured products, such as foodstuff, cosmetics and pharmaceuticals.

[Ab] Abstract:The use of chemical insecticides and non-selective natural products authorized for use in organic farming may reduce the effectiveness of egg parasitoids. The side-effects of ten plant essential oils on immature stages of Trichogramma pretiosum were evaluated. Carapa guianensis, Origanum vulgare and Zingiber officinalle during the F generation, and Azadirachtin and Mentha piperita in the F generation were slightly harmful (class II: 30-79%) to the emergence of this parasitoid. All essential oils affected the longevity of females of the F and F generations. Thymus vulgaris and Z. officinalle were the oils most harmful to female longevity. Carapa guianensis proved slightly harmful (class II: 30-79%) to parasitism in the F generation when applied during the egg-larval and pre-pupal stages and O. vulgare in the F generation in the pre-pupal stage alone, of this parasitoid. The sex ratio was lower than 0.5 during the pre-pupal stage of the F generation with Azadirachtin, C. guianensis, O. vulgare, Piper nigrum and Syzigium aromaticum, but this parameter was not affected for the other biological stages of T. pretiosum in the F and F generations. The Azadirachtin, C. guianensis, M. piperita, O. vulgare, T. vulgaris and Z. officinalle oils revealed a mild toxic effect to the immature stages of T. pretiosum and, therefore, it should be used according to patterns of ecological selectivity. Allium sativum and Citrus sinensis essential oils were not harmful to T. pretiosum, and can be used in Integrated Pest Management.

[Ab] Abstract:Phenolic compounds have been studied elaborately for their efficacy to improve health and to protect against a wide variety of diseases. Herein this study, different analysis methods were implemented to evaluate the antioxidant properties of catechin and cyanidin using their standard substances and as they found in the grape seeds extracts. Total phenol contents were 107.39±8.94mg GAE/g dw of grape seeds for grape seed extract (GSE) and 218.32±10.66mg GAE/g dw of grape seeds for acid-hydrolyzed grape seed extract (AcGSE). The extracts were analyzed by HPLC-DAD system and the results showed the presence of catechin, gallic acid, chlorogenic acid and ellagic acid in the processed methanolic extract and cyanidin, gallic acid and ellagic acid in the processed acidified methanolic extract. The protective abilities of catechin and cyanidin were tested against the oxidation of DNA. The results showed that cyanidin has better protection of DNA against oxidation than catechin. GSE and AcGSE were revealed to inhibit the oxidatively induced DNA damage. GSE decreased about 57% of damage caused by the Fenton control sample. This study could show new aspects of the antioxidant profiles of cyanidin and catechin.

[Ab] Abstract:This study was performed to evaluate the metabolite recovery from different extraction methods applied to aerial parts. A high-performance liquid chromatographic method using photodiode array detector with gradient elution has been developed and validated for the simultaneous estimation of different phenolic compounds in the extracts and in their corresponding purified fractions. The experimental results show that microwave-assisted aqueous extraction for 15 min at 100 °C gave the most phenolics-enriched extract, reducing extraction time without degradation effects on bioactives. Sixteen compounds were identified in this extract, 11 phenolic compounds and five flavonoids, all known for their biological activities. Color analysis and determination of chlorophylls and carotenoids implemented the knowledge of the chemical profile of this plant.

[Ab] Abstract:The article presents the research materials of composition and the properties of biologically active compounds of aqueous ethanolic extracts of wild plants. To obtain extracts, we used raw plants containing phenolic compounds and aromatic wild plants: the herb St. John's wort (Hypericum), thyme herba (Thymus vulgaris), yarrow (Achillea millefolium), oregano (Origanum vulgaris); leaves of sage (Salviae folium); rose hips (Rosae), hawthorn fructus (Crataegus) and fruits of mountain ash (Sorbus). The optimum composition of the mixtures used and time of extraction has been established: the ratio of alcohol and water in extracting mixtures 1:1 by volume; ratio raw material:extractant - 1:7 by weight. The total content of fenolic substances in extracts of herbaceous plants varied from to 15.5 to 24.4 mg/g, and in fruit extracts from 24.2 to 29.7 mg/g. Substances of phenolic nature, including gallic and ferulic acid, rutin, hesperidin, quercetin and apigenin were identified in the studied extracts using the HPLC. The analysis of flavonoid composition showed that rutin content in the investigated extracts varied from 0.56 mg/g up to 13,80 mg/g, of quercetin - from 0.52 to 1.36 mg/g; apigenin - from 0.44 to 1.44 mg/g; hesperidin from 2.44 to 32,72 mg/g. The content of phenolic acids varied from 0.16 to 1.44 mg/g (ferulic acid) and from 0.16 to 3.12 mg/g (chlorogenic acid). Total antioxidant activity of the studied phytoextracts (dilution 1:10) ranged from 142 to 230 µg/ml (in terms of ascorbic acid), which is consistent with the results of the quantitative analysis of flavonoids. The results of the studies of antimicrobial properties of phytoextracts showed that for E. coli the most active extracts were from thyme and yarrow, and against S. aureus - from St. John's wort. Extracts of St. John's wort and yarrow were effective against Rhizopus stolonifer.

[Ab] Abstract:The present study is devoted to prepare a new antibacterial and antifungal agent based on in situ-synthesized silver nanoparticles at room temperature using Rosmarinus officinalis (R. officinalis) leaf extract. The Ag-NPs characterization by UV-visible, SEM, TEM and XRD revealed that the particles sizes were in the range of 10-33 nm. In this study, hydroalcoholic extracts were used with ultrasonic method. Ultrasonication has recently received attention as a novel bioprocessing tool for process intensification in many areas of downstream processing. The antimicrobial activities of T. daenensis and S. marianum extracts with and without the presence of Ag-NPs were investigated at concentrations from 12.5 to 50 mg/mL against Staphylococcus aureus (S. aureus, Gram-positive organism) and Escherichia coli (E. coli, Gram-negative organism), and fungal strains were Aspergillus oryzae (A. oryzae) and Candida albicans (C. albicans). Antimicrobial activity determined using agar disc diffusion method revealed that the activities of Ag-NPs/T. daenensis were superior to Ag-NPs/S. marianum and extracts (T. daenensis and S. marianum). The medicinal plant extract can be used to synthesize the Ag-NPs as an eco-friendly and inexpensive method in large scale. The results showed that the prepared Ag-NPs/extracts as good antibacterial and antifungal agents can be potentially applied against rapidly increasing of antibiotic resistance.

[Ab] Abstract:Polyphenolic compounds are considered valuable secondary plant metabolites owing to the myriad of biological activities they exert. This study aimed to investigate the effect of applying various concentrations of the plant growth regulator, salicylic acid (SA), on Thymus vulgaris L. while subjecting the plant to decreasing amounts of irrigation water. The following parameters were monitored; total polyphenolic and flavonoid content, yield and composition of the essential oil, and antioxidant activity of the alcoholic extracts. Drought alone significantly (P < 0.05) increased the polyphenolic and flavonoid content, yield of the essential oil and antioxidant activity. The total flavonoid content in control plants was 6.1 ± 0.3 mg/gm dry weight calculated in terms of rutin equivalent. However, in drought stressed plants, (irrigated at 25% of the field capacity) sprayed with 3 mM SA, the flavonoid content increased to 32.1 ± 0.1 mg/gm dry weight calculated in terms of rutin equivalent. Moreover, the total phenolic content increased from 8.5 ± 0.3 to 68.5 ± 1.2 mg/gm dry weight calculated in terms of gallic acid in the same test plants. Radical scavenging activity, using DPPH assay, was measured for the different plant treatments. A decrease from 74.4 ± 0.4 µg/ml to 36.6 ± 0.9 µg/ml of IC was recorded in the drought stressed plants (25% FC) sprayed with 3 mM SA compared with the control plants. The variability in polyphenolic composition between the control plants and plants with the highest total polyphenolic content was investigated by UPLC-ESI-MS/MS. Rosmarinic acid was detected as the major component in samples from both treatments, with a higher percentage observed upon subjecting the plant to the test conditions (25% FC and sprayed with 3 mM SA). The highest yield of the essential oil (1 ± 0.06 %v/w) was obtained from drought stressed plants (25% FC) sprayed with 2 mM SA. GC/MS analysis of oil samples revealed that the Thymol content increased with drought stress, while that of p-cymene decreased. However, an increase of p-cymene was witnessed as a result of SA spraying.
