Resumo: BACKGROUND: Hyperpigmentation disorders such as post-inflammatory hyperpigmentation are major concerns not only in light-skinned people but also in Asian populations with darker skin. The anti-tyrosinase and immunomodulatory effects of sericin have been known for decades. However, the therapeutic effects of sericin on hyperpigmentation disorders have not been well documented. METHODS: In this study, we used an in vitro model to study the anti-tyrosinase, tolerogenic, and anti-melanogenic effects of sericin on Staphylococcus aureus peptidoglycan (PEG)-stimulated melanocytes, dendritic cells (DCs), and artificial skin (MelanoDerm™). Enzyme-linked immunosorbent assay, conventional and immunolabeled electron microscopy, and histopathological studies were performed. RESULTS: The results revealed that urea-extracted sericin has strong anti-tyrosinase properties as shown by a reduction of tyrosinase activity in melanin pigments both 48 h and 10 days after allergic induction with PEG. Anti-inflammatory cytokines including interleukin (IL)-4, IL-10, and transforming growth factor-p were upregulated upon sericin treatment (10, 20, and 50 µg/mL), whereas production of allergic chemokines, CCL8 and CCL18, by DCs was diminished 48 h after allergic induction with PEG. Moreover, sericin lowered the expression of micropthalmia-associated transcription factor (MITF), a marker of melanogenesis regulation, in melanocytes and keratinocytes, which contributed to the reduction of melanin size and the magnitude of melanin deposition. However, sericin had no effect on melanin transport between melanocytes and keratinocytes, as demonstrated by a high retention of cytoskeletal components. CONCLUSION: In summary, sericin suppresses melanogenesis by inhibition of tyrosinase activity, reduction of inflammation and allergy, and modulation of MITF function.

Resumo: BACKGROUND: Pigmentation development, is a complex process regulated by many transcription factors during development. With the development of the RNA sequencing (RNA-seq), non-coding RNAs, such as miRNAs, lncRNAs, and circRNAs, are found to play an important role in the function detection of related regulation factors. In this study, we provided the expression profiles and development of ncRNAs related to melanocyte and skin development in mice with black coat color skin and mice with white coat color skin during embryonic day 15 (E15) and postnatal day 7 (P7). The expression profiles of different ncRNAs were detected via RNA-seq and also confirmed by the quantitative real-time PCR (qRT-PCR) method. GO and KEGG used to analyze the function the related target genes. RESULTS: We identified an extensive catalogue of 206 and 183 differently expressed miRNAs, 600 and 800 differently expressed lncRNAs, and 50 and 54 differently expressed circRNAs, respectively. GO terms and pathway analysis showed the target genes of differentially expressed miRNA and lncRNA. The host genes of circRNA were mainly enriched in cellular process, single organism process. The target genes of miRNAs were mainly enriched in chromatin binding and calcium ion binding in the nucleus. The function of genes related to lncRNAs are post translation modification. The competing endogenous RNA (ceRNA) network of lncRNAs and circRNAs displays a complex interaction between ncRNA and mRNA related to skin development, such as Tcf4 , Gnas , and Gpnms related to melanocyte development. CONCLUSIONS: The ceRNA network of lncRNA and circRNA displays a complex interaction between ncRNA and mRNA related to skin development and melanocyte development. The embryonic and postnatal development of skin provide a reference for further studies on the development mechanisms of ncRNA during pigmentation.

Resumo: Se reporta el caso de un paciente varón de 68 años de edad que presentó tumoración exofítica en planta de pie derecho que al estudio histopatológico fue compatible con melanoma acral amelanótico desmoplásico. El melanoma desmoplásico es una variante histológica de presentación poco frecuente, más aún el tipo amelanótico, y de localización acral, por lo que requiere de un diagnóstico precoz para el tratamiento oportuno, mejorando así el pronóstico del paciente.

We report the case of a male patient aged 68, who presented an exophytic tumor in his right foot plant that, by histopathological study, was compatible with acral amelatonic desmoplastic melanoma. Desmoplastic melanoma is a subtype of rare representation, and even more the amelatonic type of acral location, therefore it requires early diagnosis for prompt treatment and thereby improving patient prognosis.

Resumo: Clásicamente, el vitiligo ha sido definido como una enfermedad de la piel en la cual los melanocitos (MC) son erradicados de una epidermis lesionada por células T autorreactivas con la presencia de moléculas del sistema inmune y otros componentes no inmunológicos que resultan en la pérdida de pigmento y lesiones cutáneas en el paciente. Anteriormente, la ausencia y daño en los MC ha sido asociado a mayor riesgo de cáncer de piel incluyendo melanoma. Sin embargo, en vitiligo se ha identificado la presencia de melanocitos 'no pigmentados', similar a individuos albinos, que aparentemente confieren mayor resistencia para el desarrollo de melanoma en estas personas. Estos hechos aparentemente contradictorios se complican aún más cuando los antigenos de los MC reconocidos por el sistema inmune durante la respuesta autoinmune o en la inmunidad antitumoral son los mismos en ambas enfermedades. Un análisis de las similitudes y diferencias entre la respuesta inmune contra MC observada en vitiligo y su rol en la inmunidad tumoral observada en melanoma, podría llevar a entender mejor el rol de estas células y al futuro desarrollo de nuevas terapias para ambas enfermedades.

Classically, vitiligo has been defined as a skin disease in which melanocytes (MC) are eradicated from lesional epidermis by MC- reactive T cells, as well as other non-immune and immune components, resulting in disfiguring loss of pigmento Moreover, the absence 01' damage on MC has frequently been associated to a major risk to develop skin cancer including melanoma. However, patients with vitiligo have also shown 'non-pigmented' MC in epidermis similar to individuals with albinism, and these cells are apparently conferring resistance of developing melanoma. These seemingly contradictory facts are further complicated because, the MC antigens which are immunologically recognized are shared for both diseases producing fairly different results. An analysis of the similarities and differences between the autoimmunity observed in vitiligo and the tumour immunity observed in melanoma might lead to a better understanding of the MC roles and the development of new therapies for both diseases.

Resumo: RESUMEN Los melanomas mucosos son tumores poco frecuentes y de mal pronóstico. Presentan un comportamiento agresivo, y pueden tener varias localizaciones en el aparato digestivo. Este tipo de tumores es más frecuente en la región anorectal. Se presentó un caso de una paciente femenina de 59 años, con un cuadro de varios meses de evolución; caracterizado por ardor, prurito anal y sensación de masa que prolapsaba a través del ano. Se constató al examen físico masa tumoral aspecto polipoideo. Se realizó polipectomía donde la biopsia arrojó como resultados el aspecto histológico de un melanoma mucoso de canal anal.

ABSTRACT Mucous melanomas are few frequent and have a bad prognosis. They present an aggressive behavior and might have several locations in the digestive system. This kind of tumors is more frequent at the anus-rectal region. The authors presented the case of a female patient, aged 59 years, with clinical conditions of several months of evolution, characterized by burning, anal pruritus and the sensation of a mass prolapsing through the anus. A tumor mass of polypoid aspect was found at physical examination. Polypectomy was carried out and the biopsy showed the histologic aspect of a mucous melanoma of the anal canal.

Resumo: Abstract Melanoacanthoma is a rare variant of seborrheic keratosis, which is notable for dark pigmentation and fast radial growth, making it difficult to distinguish from melanoma. Histologically, it is characterized by proliferation of keratinocytes and dendritic melanocytes. The authors report a scalp lesion, fast growing, suspected by dermoscopy and confocal microscopy examination, with dendritic cells distributed throughout the lesion. Based on these findings, it was not possible to classify this lesion as clearly benign, so it was excised. Histopathologic evaluation and immunostain were consistent with melanoacanthoma.

Resumo: Abstract Divided nevus, also known as kissing nevus, is a rare variant of congenital melanocytic nevi in which there are two adjacent nevi in areas of the body that undergo embryonic cleavage. The original description of this type of lesion was on the eyelid. The location on the penis is even rarer, with only 17 case reports in the literature so far, and only one of them described the dermoscopic findings. We report the case of a patient with divided nevus of the penis and its clinical, dermoscopic and histopathological features.

Título: Blue nevus with satellitosis: case report and literature review

Resumo: Abstract: Blue nevus is a benign melanocytic lesion, typically asymptomatic and of unknown etiology. Many histological subtypes are recognized, the most commons being: common blue nevus, cellular blue nevus, and combined blue nevus. New rare variants have been described in the literature, with emphasis on eruptive blue nevus, plaque, agminate, linear, with satellitosis, disseminated, familial and targetoid. The diagnosis of blue nevus usually presents no difficulties, however, the presence of structures such as irregular edges or satellitosis, are highly suggestive of malignancy, and the differential diagnosis with malignant blue nevus and melanoma with peripheral spread should be considered. We report a case of blue nevus with satellitosis in a 15-year-old female patient.
