Título: LncRNA-599554 sponges miR-15a-5p to contribute inductive ability of dermal papilla cells through positive regulation of the expression of Wnt3a in cashmere goat

Resumo: BACKGROUND: Long non-coding RNAs (lncRNAs), as post-transcriptional regulators, were thought to function in the inductive property of dermal papilla cells (DPCs) in cashmere goat. Previously, lncRNA-599554 was identified in secondary hair follicle (SHF) of cashmere goat, but its functional significance is unknown. RESULTS: In the present investigation, we verified that lncRNA-599554 had significantly higher expression at the anagen dermal papilla of cashmere goat SHF than that at telogen. Based on overexpression and knockdown techniques, we found that lncRNA-599554 contributes the inductive property of DPCs of cashmere goat, which was assessed by detecting the changes in the expression of several typical indictor genes in DPCs including ET-1, SCF, Versican, ALP, Lef1 and Ptc-1. Based on RNA pull-down assay, we verified that lncRNA-599554 directly interacted with chi-miR-15a-5p. Also, we showed that lncRNA-599554 positively regulated the Wnt3a expression in DPCs but which did not appear to involve its modulating of promoter methylation. Based on the use of Dual-luciferase reporter assays, our data indicated that lncRNA-599554 regulated the Wnt3a expression through chi-miR-15a-5p-mediated post-transcriptional level. CONCLUSIONS: We showed that lncRNA-599554 contributes the inductive property of DPCs in cashmere goat which might be achieved through sponging chi-miR-15b-5p to promote the Wnt3a expression. The results from the present investigation provided a novel insight into the functional mechanism of lncRNA-599554 in the SHF regeneration of cashmere goat along with the formation and growth of cashmere fiber.

Título: Bullous hemorrhagic dermatosis at distant sites / Dermatosis ampollosa hemorrágica a distancia

Resumo: Un hombre de 72 años, es hospitalizado por falla cardiaca aguda. Tiene historia de hipertensión arterial en manejo con enalapril e hidroclorotiazida. Desde su ingreso, se inició trom-boprofilaxis con enoxaparina 40 mg diarios por vía subcutánea en abdomen. Tres días después aparecen lesiones cutáneas ampollosas, tensas, de contenido hemático y diámetro entre 5-15 mm. Las lesiones se localizan en miembro superior derecho (Figura 1) y pie izquierdo (Figura 2), tienen haloeritematoso y no son dolorosas. Las pruebas de coagulación, proteína C reactiva, VSG y hemograma son normales. Se realizó biopsia de piel que mostró lesión subepidérmica con material hemorrágico en dermis papilar y reticular con neutrófilos escasos, sin vasculitis o trombosis. Estos hallazgos, confirman el diagnóstico de dermatosis ampo-llosa hemorrágica a distancia asociada a enoxaparina. Este constituye un efecto adverso infrecuente y autorresolutivo de las heparinas, no relacionado con la dosificación y en sitios distantes a su aplicación. Este paciente se trató con el retiro del medicamento, resolviendo las lesiones a los sietedías y sin recurrencia de las mismas luego de tres meses de seguimiento

A 72-year-old man was hospitalized for acute heart failure. He had a history of arterial hypertension being treated with enalapril and hydrochlorothiazide. Thromboprophylaxis was begun at admission with 40 mg of subcutaneous enoxaparin daily, in the abdomen. Three days later, tense, blood-filled bullous skin lesions with a 5-15 mm diameter appeared. The lesions were on the right arm (Figure 1) and the left foot (Fig-ure 2); they had an erythematous halo and were not painful Clotting, C-reactive protein, ESR and complete blood count were normal. A skin biopsy was performed, which showed a subepidermal lesion with hemorrhagic material in the papillary and reticular dermis with scant neutrophils and no vasculitis or thrombosis. These findings confirm the diagnosis of bullous hemorrhagic dermatosis at distant sites related to enoxaparin. This is an uncommon and self-resolving adverse effect of heparins, unrelated to the dose and at sites distant from their application. This patient was treated by discontinu-ing the medication, and the lesions resolved seven days later with no recurrence after three months of follow-up.

Resumo: Skin lesions of patients affected by non-ulcerated cutaneous leishmaniasis (NUCL) caused by L. (L.) infantum chagasi are characterized by lymphohistiocytic inflammatory infiltrate associated with epithelioid granuloma and scarce parasitism. However, the in situ cellular immune response of these patients is unclear. Therefore, the aim of the present study was to characterize the cellular immune response in the skin lesions of patients affected by NUCL. Methods Twenty biopsies were processed by immunohistochemistry using primary antibodies to T lymphocytes (CD4, CD8), NK cells, B lymphocytes, macrophages, nitric oxide synthase and interferon-gamma. Results Immunohistochemistry revealed higher expression of all cellular types and molecules (IFN-γ, iNOS) in the dermis of diseased skin compared to the skin of healthy individuals (p < 0.05). Morphometric analysis performed in the skin lesions sections showed the predominance of CD8+ T lymphocytes in the mononuclear infiltrate, followed by macrophages, mostly iNOS+, a response that could be mediated by IFN-γ. Conclusion Our study improves knowledge of the cellular immune response in non-ulcerated or atypical cutaneous leishmaniasis caused by L. (L.) infantum chagasi in Central America and pointed to the pivotal participation of CD8+ T lymphocytes in the host defense mechanisms against the parasite in patients with NUCL.(AU)

Resumo: OBJECTIVES: The aim of this study was to evaluate the histomorphometry of the skin of women during the reproductive period according to the Fitzpatrick classification. METHODS: Thirty women aged 30 to 45 years were included in this study. We studied the surgical sites of extracted nevi. The material was processed for routine histology and then stained with haematoxylin and eosin as well as Picrosirius red. Four-micrometre histological sections were analysed according the Fitzpatrick criteria (skin pigmentation). The skin thickness and collagen concentration were determined for the reticular dermal skin. The data were statistically analysed with ANOVA. RESULTS: Fitzpatrick skin types I and II were thicker than the other skin types. CONCLUSIONS: Our data suggest that white skin may be less thick than dark skin.

Resumo: BACKGROUND: Accumulating evidence indicates that reactive oxygen species (ROS) are an important etiological factor for the induction of dermal papilla cell senescence and hair loss, which is also known alopecia. Arctiin is an active lignin isolated from Arctium lappa and has anti-inflammation, anti-microbial, and anti-carcinogenic effects. In the present study, we found that arctiin exerts anti-oxidative effects on human hair dermal papilla cells (HHDPCs). RESULTS: To better understand the mechanism, we analyzed the level of hydrogen peroxide (H2O2)-induced cytotoxicity, cell death, ROS production and senescence after arctiin pretreatment of HHDPCs. The results showed that arctiin pretreatment significantly inhibited the H2O2-induced reduction in cell viability. Moreover, H2O2-induced sub-G1 phase accumulation and G2 cell cycle arrest were also downregulated by arctiin pretreatment. Interestingly, the increase in intracellular ROS mediated by H2O2 was drastically decreased in HHDPCs cultured in the presence of arctiin. This effect was confirmed by senescence associated-beta galactosidase (SA-ß-gal) assay results; we found that arctiin pretreatment impaired H2O2-induced senescence in HHDPCs. Using microRNA (miRNA) microarray and bioinformatic analysis, we showed that this anti-oxidative effect of arctiin in HHDPCs was related with mitogen-activated protein kinase (MAPK) and Wnt signaling pathways. CONCLUSIONS: Taken together, our data suggest that arctiin has a protective effect on ROS-induced cell dysfunction in HHDPCs and may therefore be useful for alopecia prevention and treatment strategies.

Resumo: BACKGROUND: Various health benefits have been attributed to Er-Miao-San (EMS), a traditional Chinese herbal formulation that contains equal amounts of cortex phellodendri (Phellodendron amurense Ruprecht) and rhizoma atractylodis (Atractylodes lancea D.C). However, its effect on the anti-inflammatory activity in human dermal fibroblasts (HDFs) and the mechanism underlying this effect are unknown. RESULTS: This study investigated the effects of EMS on TNF-α-induced MMP-1 expression in HDFs. Our data show that EMS inhibited TNF-α-induced MMP-1 expression in a concentration-dependent manner. Interestingly, EMS maintained IkB content without inhibiting the phosphorylation of MAPKs, which are well-established upstream kinases of NF-kB. Moreover, EMS reduced the level of nuclear p65 protein in HDFs. Luciferase assay revealed that EMS inhibits the transcriptional activity of NF-kBbystabilizing IkB. Our results show that EMS exerts its anti-inflammatory effect by inhibiting NF-kB-regulated genes such as IL-1ß and IL-8. Moreover, EMS effectively inhibited TNF-α-induced expression of MMP-1 via the NF-kBpathway. CONCLUSIONS: Taken together, our data suggest that EMS could potentially be used as an anti-inflammatory and anti-aging treatment.

Título: Knockout of p16INK4a promotes aggregative growth of dermal papilla cells

Resumo: Summary Objective: Dermal papilla cells (DPCs) are located in the hair follicles and play an important role in hair growth. These cells have the ability to induce hair follicle formation when they display aggregative behavior. DPCs derived from the androgenetic alopecia (AGA) area undergo premature senescence in vitro, associated with p16INK4a expression. The aim of the current study was to investigate the expression of p16INK4a in aggregative and non-aggregative DPCs and the effect of p16INK4a down-regulation in these cells by adenovirus-mediated RNA interference (RNAi). Method: DPCs were isolated and cultured from healthy human scalp. p16INK4a gene and protein were detected in aggregative and non-aggregative cells. Expression of p16INK4a in DPCs was silenced by infection with rAd5-CDKN1A-1p2shRNA. Cell fate was monitored after infection. The growth of cells was measured by MTT assay. Cell cycle was evaluated by flow cytometry (FCM). Results: DPCs were isolated by digestion and showed aggregative behavior for six passages. The expression of p16INK4a showed a clear upward trend in non-aggregative cells when compared with aggregative group. p16INK4a expression was silenced by rAd5-CDKN1A-1p2shRNA (p<0.05). The p16INK4a-silenced cells grew more rapidly and exhibited a trend towards aggregative growth. There was an increase in the proportion of cells in G1 phase, while those in S phase were reduced after p16INK4a gene silencing (p<0.05). Conclusion: Our results suggest that p16INK4a plays an important role in the premature senescence and aggregative behavior of DPCs. These observations can lead to novel therapeutic strategies for treatment of AGA.

Resumo: La piel tiene una función primordial en el control de la integridad del medio interno. En consecuencia, su pérdida en proporciones importantes como aquellas que ocurren en las perdidas traumáticas, cirugías reconstructivas, cirugías oncológicas y grandes quemados, es factor predisponente a complicaciones. En las últimas décadas se han desarrollado diversos métodos que permiten remplazar los principales componentes de la piel. Es así como el tejido dérmico ha sido homologado mediante sustitutos a base de biopolímeros de origen animal. Los equivalentes dérmicos le confieren resistencia mecánica a los injertos y gradualmente facilitan su colonización y reemplazo por fibroblastos y células endoteliales del paciente. La matriz dérmica es un biomaterial que ha sido utilizado por la cirugía plástica, con el propósito de reconstruir en superficie o volumen un tejido que ha sido afectado y que presenta alteraciones en su morfología. Se presenta una revisión y análisis de un ensayo clínico multicéntrico de los pacientes donde se efectuaron 336 biopsias en serie de 131 pacientes participantes, en un lapso de 7 días a 2 años post-aplicación del producto. Se obtiene como resultado que la matriz de regeneración dérmica fue bien tolerada, no hubo notificaciones de respuestas inmunológicas o histológicas con importancia clínica a la implantación del producto. En las evaluaciones clínicas no hubo notificaciones de rechazo a la matriz. Además se recopilaron datos sobre la colonización o infección de las heridas. Las consecuencias de una infección en los lugares tratados con la matriz incluyen pérdida parcial o total de absorción del producto(AU)

The skin has a major role in controlling the integrity of the internal environment. Consequently, their loss in major proportions as those occurring in traumatic losses, reconstructive surgery, cancer surgery and severe burns, is a predisposing factor for complications. In the last decades various methods have been developed that allow replacing the main components in the skin. Thus the dermal tissue has been approved by biopolymers based substitutes animal. Dermal equivalents confer mechanical strength to the grafts and gradually facilitate colonization and replacement by fibroblasts and endothelial cells of the patient. Dermal matrix is a biomaterial that has been used for plastic surgery, for the purpose of reconstructing or surface tissue that volume has been affected and having alterations in morphology. A review and analysis of a multicenter clinical trial of 336 patients where biopsies were performed in series of 131 patients participating in a span of 7 days to 2 years post- spraying. As result the dermal regeneration matrix was well tolerated, no notifications immune responses or clinically significant histological to implantation of the device. In no clinical assessments of rejection notices to the matrix. Also collected data on the colonization or infection of wounds. The consequences of infection sites treated with the matrix include partial or total loss of absorption(AU)

Resumo: Introducción: Las especies vegetales constituyen el remedio primero a los problemas de salud que aquejan a las personas. La granada es rica en ácido ascórbico, mejora la barrera epidérmica y reduce la contracción de las heridas. La flor de agua posee actividad antibacteriana. Objetivo: Evaluar el efecto cicatrizante del polvo carbonizado de Punica granatum Linn (granada) y de Eichhornia crassipes (flor de agua), en un modelo experimental en ratas. Métodos: Estudio analítico experimental con el empleo 30 ratas macho distribuidas en 3 grupos (n= 10). Grupo I y II; tratadas con el polvo carbonizado de la granada y flor de agua respectivamente. Grupo III: Tratadas con cloruro de sodio al 0,9 por ciento. Se realizó un modelo de herida por escisión en el dorso. La evolución de la cicatrización fue seguida por la velocidad de contracción de la herida en milímetros. Se realizó histología sobre biopsias de tejido cicatrizado. Para el análisis estadístico se empleó la prueba no paramétrica de Mann- Whitney. El nivel de significación se fijó en p < 0,05. Resultados: Se comprobó el efecto cicatrizante de los polvos carbonizados de la granada y flor de agua; disminuyó el área de las heridas de manera significativa respecto al grupo control. El estudio histológico mostró dermis madura grado III en los grupos I y II. Conclusiones: La aplicación tópica del polvo carbonizado de granada y de flor de agua influyó sobre el cierre de las heridas y en la maduración de la dermis, por lo cual favoreció la cicatrización(AU)

ABSTRACT Introduction: Plant species are the first remedy to the people's health problems. The pomegranate is rich in ascorbic acid, improves the epidermal barrier and reduces the contraction of wounds. The water hyacinth has antibacterial activity. Objective: To evaluate the healing effect of the charred powder of Punica granatum Linn (pomegranate) and Eichhornia crassipes (water hyacinth), in an experimental model in rats. Methods: Experimental analytical study with 30 male rats distributed in 3 groups (n = 10). Group I and II; treated with the charred powder of the pomegranate and water hyacinth respectively. Group III: treated with 0.9 percent sodium chloride. An incision wound model was made on the back. The evolution of healing was followed by the speed of contraction of the wound in millimeters. Histology was performed on biopsies of scar tissue. For the statistical analysis, the nonparametric Mann-Whitney test was used. The level of significance was set at p < 0.05. Results: The healing effect of the charred powders of the pomegranate and water hyacinth was verified; decreased the wound area significantly compared to the control group. The histological study showed mature grade III dermis in groups I and II. Conclusions: The topical application of the charred powder of pomegranate and water hyacinth influenced the closure of the wounds and the maturation of the dermis, which favored healing(AU)

Resumo: ABSTRACT Purpose: To evaluate the outcomes of dermis-fat graft procedure for orbital volume replacement in anophthalmic socket. Methods: A retrospective chart review was performed studying all dermis-fat graft surgeries done at King Khlaed Eye Specialist Hospital in the last 10-year period. Sixty-two anophthalmic socket carriers underwent dermis-fat graft during the study period. Data were collected on characteristics of the patients, postoperative complications, cosmesis, and functional results such as the ability to hold an external prosthesis. Results: Mean age of the patients included in the study was 34.2 ± 9.7 years. There were 38.7% of patients with grade 3 anophthalmic socket, 64.5% of patients had an acquired anophthalmic socket, and dermis-fat graft was performed as a secondary procedure in 61.3% of patients. Postoperative complications included lagophthalmos (22.6%), graft necrosis (17.7%), pyogenic granuloma (12.9%), decreased graft size (12.9%), malpositioned lids (3.2%), and volume deficiency (3.2%). The prosthesis was held in place in 49 patients (79%) preoperatively and in 55 patients (88.7%) postoperatively. Conclusion: Dermis-fat graft is an excellent option for congenital or acquired as well as primary or secondary anophthalmic sockets, with or without contraction. The outcomes are favorable, and complications are rare.

RESUMO Objetivo: Avaliar os resultados obtidos com o uso do enxerto dermo-adiposo para reposição de volume em cavidade anoftálmica. Métodos: Estudo retrospectivo baseado em revisão de prontuários incluindo todas as cirurgias de enxerto dermo-adiposo realizadas nos últimos 10 anos no King Khaled Eye Specialist Hospital, Saudi Arabia. O enxerto dermo-adiposo foi realizado em 62 pacientes no periodo do estudo. Os dados analisados incluíram características dos pacientes, as complicações pós- operatórias e os resultados cosméticos e funcionais, tais como a habilidade de usar prótese externa. Resultados: A média de idade dos participantes foi de 34,2 ± 9,7 anos. Segundo a classificação das cavidades, 38,7% possuíam cavidade grau 3; 64,5% possuíam cavidade anoftálmica adquirida e o enxerto dermo-adiposo foi realizado como procedimento secundário em 61,3% dos pacientes. Após o procedimento 22,6% dos pacientes permaneceram com lagoftalmo, 17,7% tiveram necrose do enxerto, 12,9% desenvolveram granuloma piogênico, 12,9% tiveram redução do tamanho do enxerto, 3,2% permaneceram com as alterações no posicionamento palpebral e 3,2% continuaram com déficit de volume na órbita. Quarenta e nove pacientes (79%) eram capazes de usar prótese externa antes da cirurgia e depois do enxerto dermo-adiposo 55 (88,7%) puderam utilizar prótese externa. Conclusão: O enxerto dermo-adiposo é uma ótima opção para tratamento de cavidades anoftálmicas congênitas ou adquiridas, assim como realizado como procedimento primário ou secundário, em cavidades com ou sem contração tecidual. Os resultados são encorajadores e as complicações são pouco frequentes.
