Resumo: The aim this work was to evaluate the genetic variability within and among populations grown on sisal region of Bahia. The genetic diversity of 140 population of sisal was analyzed using ISSR molecular markers. Samples were collected in six counties in Bahia. Eighteen primers of ISSR were used, in which nine of them were effective in the amplification of DNA samples to yield 143 polymorphic loci. The average percentage of polymorphism found in the population was 64%. The average heterozygosis (He) and Shannon-Wiener index (I) were 0.180 and 0.279, respectively. 73% of the molecular variance can be due to intrapopulation differences. The populations were divided into two groups according to their geographical location, showing a structure of genetic variability in space. The GST 0.235, enough to avoid that there is a strong population differentiation. The genetic structure of sisal can be exploited for the creation of gene banks for conservation in situ and ex situ to obtain individuals of good commercial quality. There is genetic variability among sisal genotypes. ISSR molecular markers are efficient to analyze the divergence between sisal genotypes, assisting improvement work.

O objetivo deste trabalho foi avaliar a variabilidade genética existente entre e dentro das populações cultivadas na região sisaleira baiana. A diversidade genética de 140 indivíduos de sisal foi analisada por meio de marcadores moleculares ISSR. As amostras foram coletadas em seis municípios baianos. Utilizando-se dezoito primers de ISSR, dos quais, nove foram eficientes na amplificação do DNA das amostras analisadas, obtendo-se 143 locos polimórficos. O percentual médio de polimorfismo encontrado nas populações foi de 64%. A média de heterozigosidade (He) e do índice de Shannon-Wiener (I) foram de 0,180 e 0,279, respectivamente. 73% da variância molecular pode ser atribuída às diferenças intrapopulacionais. As populações foram divididas em dois grupos, de acordo com sua localização geográfica, evidenciando uma estruturação da variabilidade genética no espaço. O GST de 0,235 foi suficiente para evitar que exista uma forte diferenciação populacional. A estruturação genética do sisal pode ser explorada para a criação de bancos de germoplasma, visando à conservação in situ e ex situ, para a obtenção de indivíduos de boa qualidade comercial. Há presença de variabilidade genética entre os genótipos de sisal. Marcadores moleculares ISSR são eficientes para analisar a divergência entre os acessos de sisal, auxiliando trabalhos de melhoramento.

Resumo: Agave tequilana Weber var. 'Azul' is grown for the production of tequila, inulin and syrup. Diverse bacteria inhabit plant tissues and play a crucial role for plant health and growth. In this study culturable endophytic bacteria were extracted from leaf bases of 100 healthy Agave tequilana plants. In plant tissue bacteria occurred at mean population densities of 3 million CFU/g of fresh plant tissue. Three hundred endophytic strains were isolated and 16s rDNA sequences grouped the bacteria into eight different taxa that shared high homology with other known sequences. Bacterial endophytes were identified as Acinectobacter sp., A. baumanii, A. bereziniae, Cronobacter sakazakii, Enterobacter hormaechei, Bacillus sp. Klebsiella oxytoca, Pseudomonas sp., Enterococcus casseliflavus, Leuconostoc mesenteroides subsp. mesenteroides and Gluconobacter oxydans. Isolates were confirmed to be plant growth promoting bacteria (PGPB) by their capacities for nitrogen fixation, auxin production, phosphate solubilization, or antagonism against Fusarium oxysporum AC132. E. casseliflavus JM47 and K. oxytoca JM26 secreted the highest concentrations of IAA. The endophyte Acinectobacter sp. JM58 exhibited the maximum values for nitrogen fixation and phosphate solubilization index (PSI). Inhibition of fungi was found in Pseudomonas sp. JM9p and K. oxytoca JM26. Bacterial endophytes show promise for use as bio-inoculants for agave cultivation. Use of endophytes to enhance cultivation of agave may be particularly important for plants produced by micropropagation techniques, where native endophytes may have been lost.

Título: Seed germination of Agave species as influenced by substrate water potential

Resumo: BACKGROUND: Plants of Agave spp. perform Crassulacean acid metabolism (CAM) and are highly drought-tolerant, but little is known concerning seed germination under low water availability. The aim of this study was to assess the effect of substrate water potential (ΨW) on seed germination and contrast hydrotime parameters of seven valuable and commercially-important Agave species from different geographical distributions and climatic regions of Mexico. Our hypothesis was that seed germination of Agave species is not affected by low water availability independently of seed biomass and the climate of their distribution area. RESULTS: Seed germination (at 25°C and in the dark) between 85 and 100% for all species occurred within 80 - 180 h at -0.03 MPa and 250 - 430 h at -1.0 MPa. Seed germination at -1.5 MPa declined to less than 50% (p < 0.05) for A. asperrima and A. cupreata but did not change significantly for A. americana var. marginata, A. lechuguilla and A. striata, although they showed the lowest mean base water potential (-2.01 to -2.64 MPa). Seed germination of 40% Agave species, from arid and semi-arid climates in this study, was not affected by the lower ΨW. CONCLUSION: Germination of seeds of Agave species is moderately affected by low water availability, is partially dependent of their ecological distribution, and is independent of seed mass.

Resumo: The present study evaluated the anti-inflammatory and analgesic properties of Agave sisalana Perrine in classic models of inflammation and pain. The hexanic fraction of A. sisalana (HFAS) was obtained by acid hydrolysis followed by hexanic reflux. Anti-inflammatory properties were examined in three acute mouse models (xylene ear oedema, hind paw oedema and pleurisy) and a chronic mouse model (granuloma cotton pellet). The antinociceptive potential was evaluated in chemical (acetic-acid) and thermal (tail-flick and hot-plate test) models of pain. When given orally, HFAS (5, 10, 25 and 50 mg/kg) reduced ear oedema (p < 0.0001; 52%, 71%, 62% and 42%, respectively). HFAS also reduced hind paw oedema at doses of 10 mg/kg and 25 mg/kg (p < 0.05; 42% and 58%, respectively) and pleurisy at doses of 10 mg/kg and 25 mg/kg (41% and 50%, respectively). In a chronic model, HFAS reduced inflammation by 46% and 58% at doses of 10 mg/kg and 25 mg/kg, respectively. Moreover, this fraction showed analgesic properties against the abdominal writhing in an acetic acid model (at doses of 5-25 mg/kg) with inhibitory rates of 24%, 54% and 48%. The HFAS also showed an increased latency time in the hot-plate (23% and 28%) and tail-flick tests (61% and 66%) for the 25 mg/kg and 50 mg/kg doses, respectively. These results suggest that HFAS has anti-inflammatory and analgesic properties.

Resumo: Steroidal saponins from the plant Agave brittoniana with activity against the parasite Trichomona vaginalis. The genus Agave (Agavaceae), includes more than 300 species; around 16 of them show an homogeneous distribution throughout Cuba. Agave brittoniana (ssp. brachypus), is an endemic subspecies that grows in the central region of the country and its leaves are traditionally used in the treatment of parasitic diseases. The parasite Trichomonas vaginalis causes the disease known as trichomoniasis, that infects the genital tract. To test in vitro the plant against Trichomona vaginalis, the dried and powdered leaves were extracted three times with ethanol-water (7 : 3) by maceration at room temperature. The solvent was removed under reduced pressure and the extract was suspended in distilled water, defatted with n-hexane, and extracted with water-saturated n-butanol. After solvent removal, a portion of the n-butanol extract was hydrolyzed. After extraction with ethyl acetate the hydrolysis products were compared with authentic sapogenins samples using thin layer chromatography (TLC). Most of the sapogenins (yuccagenin and diosgenin) were isolated and their structures were confirmed. using nuclear magnetic resonance (NMR) experiments. The n-butanol extract was subjected to a separation process through column chromatography to obtain five fractions. After multiple separation processes by reversed phase high performance liquid chromatography (HPLC), the most active one produced one refined fraction that contained two saponins with the same aglycone (diosgenin) and one yuccagenin based saponin. Best results of the activity were obtained with the yuccagenin derived glycoside. Rev. Biol. Trop. 56 (4): 16451652. Epub 2008 December 12.

El género Agave, familia Agavaceae, tiene más de 300 especies, con aproximadamente 16 distribuidas en toda Cuba. Una de ellas, el Agave brittoniana Trel. (ssp. brachypus), es una subespecie endémica y sus hojas son tradicionalmente utilizadas en el tratamiento de enfermedades parasitarias. Se realizaron estudios "in vitro" de la actividad de productos de esta planta frente a Trichomona vaginalis. Las hojas secas y pulverizadas fueron extraídas tres veces con una mezcla de etanol-agua (7: 3) mediante maceración a temperatura ambiente. El disolvente fue evaporado a presión reducida y el extracto fue suspendido en agua destilada, desengrasado con n-hexano, y extraído con n-butanol saturado con agua. Luego de una extracción con acetato de etilo, los productos de la hidrólisis fueron comparados con patrones de sapogeninas mediante la cromatografía de capa fina (CCD). Aislamos las sapogeninas mayoritarias (yuccagenina y diosgenina) y confirmamos sus estructuras utilizando técnicas de resonancia magnética nuclear. Por otra parte, el extracto n-butanólico fue sometido a un proceso de separación biodirigido mediante cromatografía de columna, obteniéndose cinco fracciones. Después de múltiples separaciones, la más activa rindió una fracción purificada con dos sapogeninas con el mismo aglicón (diosgenina) y un glicósido de yucagenina. Los mejores resultados de esta actividad fueron obtenidos con el glicósido derivado de la yucagenina.

Resumo: It was evaluated the anthelmintic activity of Agave sisalana (sisal) juice against gastrointestinal nematodes and its potential toxic effects in goats. In vitro tests showed more than 95 percent reduction in larval counts of the genus Haemonchus spp. at concentrations between 86.5 and 146.3 mg.mL-1. In vivo the percent reduction of larvae of the fourth (L4) and fifth (L5) stages of Haemonchus, Oesophagostomum and Trichostrongylus was less than 95 percent in groups GI and GII, and between 80 and 90 percent in group GIII. A. sisalana juice at the concentrations tested in vitro was effective against gastrointestinal nematodes in goats; however, its anthelmintic efficacy was reduced when administered to animals.

Foi avaliada a atividade anti-helmíntica do suco de Agave sisalana (sisal) contra nematódeos gastrintestinais e possíveis efeitos tóxicos em caprinos. Nos testes in vitro, encontrou-se redução superior a 95 por cento na contagem de larvas do gênero Haemonchus spp. nas concentrações entre 86,5 e 146,3 mg.mL-1. In vivo, o percentual de redução de larvas de quarto (L4) e quinto (L5) estágios de Haemonchus, Oesophagostomum e Trichostrongylus foi inferior a 95 por cento para o GI e GII, e entre 80 e 90 por cento para o GIII. O suco de A. sisalana nas concentrações testadas in vitro foi efetivo contra nematódeos gastrintestinais de caprinos, apresentando, no entanto, reduzida eficácia anti-helmíntica quando administrado nos animais.

Resumo: Third instars of white grubs of six species associated to agave tequilero in Jalisco, México were described from 1,145 specimens collected from soil samples from September 2006 to August 2007, in the municipalities of Ixtlahuacán del Río, Tepatitlán de Morelos and San Juan de Escobedo, Jalisco, México. Diagnostic characters were illustrated and a key was also included. Cyclocephala comata (Bates) was the most abundant species (63.2 percent), followed by Phyllophaga ravida (Blanchard) (21.9 percent), Phyllophaga polyphylla (Bates) (9.4 percent), Phyllophaga misteca (Bates) (4.1 percent), Strategus aloeus (L.) (1.1 percent) and Anomala hoepfneri (Bates) (0.3 percent). Phyllophaga ravida and A. hoepfneri are reported for the first time on the agave plant and the latter is a new record for the State of Jalisco. All Melolonthidae species showed a marked seasonality with lower number of larvae in June 2007 and high number in August 2007, which is associated with the region's rainy season and the agave plant age, respectively.

Se describen las larvas de tercer instar de seis especies de gallinas ciegas asociadas al cultivo de agave tequilero en el estado de Jalisco, México, con base en 1,145 especimenes, de muestras de suelo, colectados de Septiembre de 2006 a Agosto de 2007, en los municipios de Ixtlahuacán del Río, Tepatitlán de Morelos y San Juan de Escobedo, Jalisco, México. Se incluyen ilustraciones diagnósticas y una clave para el reconocimiento de las especies. Cyclocephala comata (Bates) fue la especie más abundante (63.2 por ciento), seguida por Phyllophaga ravida (Blanchard) (21.9 por ciento), Phyllophaga polyphylla (Bates) (9.4 por ciento), Phyllophaga misteca (Bates) (4.1 por ciento), Strategus aloeus (L.) (1.1 por ciento) y Anomala hoepfneri (Bates) (0.3 por ciento). Phyllophaga ravida y A. hoepfneri se reportan por primera vez asociadas al agave tequilero y esta última es nuevo registro para el estado de Jalisco. Las especies de melolóntidos presentaron una marcada estacionalidad, con un más bajo número de larvas en Junio de 2007 y mayor en el mes de Agosto de 2007, lo que coincidió con el periodo de lluvias en la región y edad de las plantas, respectivamente.

Título: Effects of substrate water potential in root growth of Agave salmiana Otto ex Salm-Dyck seedlings

Resumo: The objective of this study was to test the hypothesis that root of maguey (Agave salmiana Otto ex Salm-Dyck) seedlings reacts during the first 24 h to low substrate water potential (Yw), by anatomical modifications. Three-4 cm root length seedlings were planted in vermiculite for 24 h at Yw between -0.03 and -2.35 MPa. Root dimensions, proline content and anatomy were evaluated. Substrate ψw between -0.65 and -2.35 MPa did not significantly affect longitudinal root growth. However, proline content significantly increased from 1.6 to 2.1 emoles mg-1. Significant reductions of transverse root area (41 percent), thickness of mucilage covering the epidermis (47 percent), thickness of epidermis (between 15 and 46 percent), area of the parenchyma (between 35 and 41 percent) and number of vessels (up to 28 percent) were observed with Yw of -2.35 MPa. In contrast, thickness of xylem wall, diameter of xylem vessels and the number of cells of the cortex of the differentiation root region significantly increased (64, 17, and 97 percent, respectively). The anatomical changes associated with low substrate Yw indicate a net increase of root apoplatic paths; structures involved in water conduction increased their diameter under low substrate Yw conditions and anatomical changes occurred during the first 24 h of water stress.

Resumo: Shoot apex, leaf primordia, leaf sections and roots from Mexican prickly poppy seedlings, were inoculated with Agrobacterium tumefaciens harboring the binary vector pCAMBIA2301, which contained the beta-glucuronidase (uid A) gene. Histochemical beta-glucuronidase (GUS) assay in infected explants showed transient gus gene expression between 3 and 12 days after inoculation. To our knowledge, this is the first report of A. mexicana susceptibility to A. tumefaciens-mediated genetic transformation.
