Título: Intercellular mediators in bone remodeling regulation in the experimental renal pathology / Mediadores intercelulares de la regulación de la remodelación ósea en un modelo experimental de patología renal

Resumo: Bone metabolism disorders are characterized by an imbalance of bone resorption and formation in the bone remodeling process. Glucocorticoids that are used to treat kidney diseases exacerbate these disorders. P-selectin and galectin-3 are molecules involved in the sclerotic process in kidney, whereas bone resorption is regulated by the interaction between the nuclear factor activator kappa b receptor (RANK), its ligand (RANKL) and the RANKL decoy receptor osteoprotegerin (OPG). The aim of this study was to investigate the cellular and molecular mechanisms of disruption of bone remodeling regulation processes, reflected by intercellular mediators (RANKL, OPG, P-selectin and galectin-3) in chronic kidney disease experimental model treated with glucocorticoids. Rats were divided into four groups of 10 animals each. The first group, the control group, included intact animals. The second group consisted of rats with impaired bone remodeling resulting from chronic kidney disease (experimental group (CKD). The third group was a group of animals with impaired bone remodeling due to exposure to glucocorticoids (experimental group (GCs)). The fourth group consisted of rats with impaired bone remodeling in chronic kidney disease, followed by exposure to glucocorticoids (experimental group (CKD + GCs)). The effects of CKD and glucocorticoid were evaluated biochemically, histologically and by measuring bone density. An enzymelinked immunoassay was used to measure intercellular mediator levels in the serum. The bone density in the experimental groups was reduced compared to the control group. RANKL levels in animals of three experimental groups were higher than in intact animals. Serum levels of OPG were higher in CKD and GCs groups than in intact animals. At the same time, in the animals' blood serum of the CKD + GCs group, the levels of OPG were lower, than those in animals from the control group. The levels of galectin-3 in the serum of the experimental groups GCs and CKD + GCs were lower than in intact animals. The serum levels of galectin-3 in animals of the CKD group were higher than those in animals from the control group. The levels of P-selectin were lower in the serum of the GCs group than in intact animals. At the same time, the levels of P-selectin were higher in the CKD and CKD + GCs groups, than those in animals from the control group. In conclusion, the study of the complex system of bone remodeling regulation, which includes many factors and their interactions, may lead to the development of new methods for treating patients with chronic kidney disease in order to prevent osteoporosis in the future. Las enfermedades metabólicas óseas se caracterizan por un desequilibrio en el proceso de remodelación ósea en los que participan mediadores tales como receptor del activador del factor nuclear- kappa- b (RANK), su ligando (RANKL) y la osteoprotegerina (OPG). Los glucocorticoides, recuentemente empleados en el tratamiento de la enfermedad renal crónica, exacerban este desequilibrio. En la enfermedad esclerótica renal, las moléculas de adhesión celular P-selectina and galectina-3 tienen un rol fundamental. El objetivo de esta trabajo fue estudiar las alteraciones en los mediadores de la remodelación ósea (RANKL, OPG, P-selectina and galectina-3) en un modelo de enfermedad renal crónica con tratamiento glucocorticoideo. Ratas Wistar hembras fueron divididos en 4 grupos: control (C); enfermedad renal crónica con afección de la remodelación ósea (ERC); animales con afección de la remodelación ósea expuestos a glucocorticoides (GC); enfermedad renal crónica con afección de la remodelación ósea tratados con glucocorticoides (ERC+GC). Los efectos de la ERC y los GC fueron evaluados bioquímicamente, histológicamente y por medición de la densidad ósea. RANKL, OPG, Pselectina and galectina-3 se cuantificaron en muestras de sangre venosa empleando enzimoinmuno análisis. En los 3 grupos experimentales la densidad ósea se evidenció reducida y los niveles séricos de RANKL elevados respecto al grupo control. Los niveles de OPG en los grupos ERC y GC fueron superiores mientras que en el grupo ERC+GC menores respecto a los animales controles. Galectina 3 plasmática en GC y ERC+GC se encontró reducida y aumentada en los animales ERC, en comparación con los animales controles. La concentración sérica de P-selectina sérica fue mayor en los grupos ERC y ERC+GC, y menor en los animales GC respecto a los niveles plasmáticos de los animales intactos. El avance del conocimiento sobre la regulación de la remodelación ósea a través de la interacción de mediadores sistémicos, en un futuro, puede conducir al desarrollo de nuevas estrategias terapéuticas para la prevención de la osteoporosis en pacientes con enfermedad renal crónica. Título: Use of GC/MS to identify chemical constituents and cytotoxic activity of the leaves of Phoradendron mucronatum and Phoradendron microphyllum (Viscaceae)

Resumo: ABSTRACT Phoradendron mucronatum and P. microphyllum are plants that found in tropical and subtropical areas, used in traditional medicine and popularly known as mistle-thrush. The aim of this study was to identify the chemical constituents of different leaf extracts from P. mucronatum and P. microphyllum and assess cytotoxic activity against strains from a human tumour cells. Extracts obtained with hexane, dichloromethane, chloroform and ethyl acetate from the leaves were analysed by gas chromatography coupled with mass spectrometry (GC-MS) and the cytotoxicity was assessed by the MTT method (bromide (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)). The tested human tumour cells were NCI-H292 (human pulmonar mucoepidermoid carcinoma), MCF-7 (human breast adenocarcinoma) and HEp-2 (epidermoid carcinoma of the larynx). Analysis by GC/MS of the extracts from leaves of P. microphyllum and P. mucronatum detected 51 different compounds, such as alkaloids, diterpenes, triterpenes, sterols, alcohols, aldehydes, fatty acids and hydrocarbons. In the cytotoxic evaluation, hexane and ethyl acetate extracts from the leaves P. microphyllum inhibited cell growth of NCI-H292 strains (72.97%) and HEp-2 (87.53%), respectively. The extracts of P. mucronatum species showed an inhibitory effect towards NCI-H292 (83.19%/hexane), MCF-7 (88.69%/dichloromethane) and HEp-2 (93.40%/hexane). The extracts showed cytotoxic activity against the tested strains, especially the P. mucronatum, which presented the highest percentages of inhibition of cell growth.

Resumo: Chloroform extract (CE) and fractions obtained from Aldama arenaria roots were evaluated for their in vitro antiproliferative activity against 10 human tumor cell lines [leukemia (K-562), breast (MCF-7), ovary expressing a multidrug-resistant phenotype (NCI/ADR-RES), melanoma (UACC-62), lung (NCI-H460), prostate (PC-3), colon (HT29), ovary (OVCAR-3), glioma (U251), and kidney (786-0)]. CE presented weak to moderate antiproliferative activity (mean log GI50 1.07), whereas fractions 3 and 4, enriched with pimaranetype diterpenes [ent-pimara-8(14),15-dien-19-oic acid and ent-8(14),15-pimaradien-3ß-ol], presented moderate to potent activity for most cell lines, with mean log GI50 of 0.62 and 0.59, respectively. The results showed promising in vitro antiproliferative action of the samples obtained from A. arenaria, with the best results for NCI/ADR-RES, HT29, and OVCAR-3, and TGI values ranging from 5.95 to 28.71 µg.ml -1, demonstrating that compounds of this class may be potential prototypes for the discovery of new therapeutic agents.

El extracto de cloroformo (CE) y las fracciones obtenidas de las raíces de Aldama arenaria fueron evaluadas por su actividad antiproliferativa in vitro contra 10 líneas celulares tumorales humanas [leucemia (K-562), mama (MCF-7), ovario que expresa un fenotipo resistente a múltiples fármacos (NCI/ADR-RES), melanoma (UACC-62), pulmón (NCI-H460), próstata (PC-3), colon (HT29), ovario (OVCAR-3), glioma (U251) y riñón (786-0)]. CE presentó actividad antiproliferativa débil a moderada (log GI50 promedio de 1.07), mientras que las fracciones 3 y 4, enriquecidas con diterpenos de tipo pimarane [ent-pimara-8 (14), ácido 15-dien-19-oico y ent-8 (14), 15-pimaradien-3ß-ol], presentaron actividad moderada a potente para la mayoría de las líneas celulares, con un log GI50 promedio de 0.62 y 0.59, respectivamente. Los resultados mostraron una prometedora acción antiproliferativa in vitro de las muestras obtenidas de A. arenaria, con los mejores resultados para NCI/ADR-RES, HT29 y OVCAR-3, y valores de TGI que van desde 5.95 a 28.71 µg.ml -1, lo que demuestra que los compuestos de esta clase pueden ser prototipos potenciales para el descubrimiento de nuevos agentes terapéuticos.

Resumo: Knowledge of the radiochemical purity of radiopharmaceuticals is mandatory and can be evaluated by several methods and techniques. Planar chromatography is the technique normally employed in nuclear medicine since it is simple, rapid and usually of low cost. There is no standard system for the chromatographic technique, but price, separation efficiency and short time for execution must be considered. We have studied an alternative system using common chromatographic stationary phase and alcohol or alcohol:chloroform mixtures as the mobile phase, using the lipophilic radiopharmaceutical [99mTc(MIBI)6]+ as a model. Whatman 1 modified phase paper and absolute ethanol, Whatman 1 paper and methanol:chloroform (25:75), Whatman 3MM paper and ethanol:chloroform (25:75), and the more expensive ITLC-SG and 1-propanol:chloroform (10:90) were suitable systems for the direct determination of radiochemical purity of [99mTc(MIBI)6]+ since impurities such as99mTc-reduced-hydrolyzed (RH),99mTcO4- and [99mTc(cysteine)2]-complex were completely separated from the radiopharmaceutical, which moved toward the front of chromatographic systems while impurities were retained at the origin. The time required for analysis was 4 to 15 min, which is appropriate for nuclear medicine routines.

Resumo: Chloroform, methanol and aqueous extracts of Penstemon roseus and Senna crotalarioides were investigated for their anti-inflammatory and antioxidant properties. The aqueous extract had no activity on TPA-induced ear edema, but the chloroform and methanol extracts of both plants caused a significant inhibition of the edema. The chloroform extracts showed activity on carrageenan-induced paw edema and mouse ear edema induced by multiple topical applications of TPA, and only P. roseus (dose of 100 mg/kg) exhibited anti-arthritic activity. The methanol extracts of P. roseus and S. crotalarioides had the most activity in the DPPH and reducing power tests. The LD50 values in mice of both chloroform extracts were considerate as slight acute toxic and higher than the doses used in this study.

Las propiedades antiinflamatorias y antioxidantes de los extractos de cloroformo, metanol y agua de Penstemon roseus y Senna crotalarioides fueron investigadas. El extracto acuoso no tuvo actividad sobre el edema auricular inducido con TPA, pero el extracto de cloroformo y etanol de ambas plantas inhibieron significativamente el edema. El extracto de cloroformo mostró actividad sobre el edema plantar inducido con carragenina y el edema auricular en ratón inducido por múltiples aplicaciones de TPA, y solo P. roseus y S. crotalarioides tuvieron la mejor actividad en las pruebas de DPPH y poder reductor. Los valores de la DL50 en ratón de ambos extractos de cloroformo son considerados con ligera toxicidad aguda y mayores que las dosis empleadas en el estudio.

Resumo: En el año 1899, en el viejo Hospital de Clínicas de Buenos Aires Eugenio Py filmó una intervención quirúrgica que practicó el Dr. Alejandro Posadas para resecar un quiste hidatídico de pulmón. En la película se ve a Posadas operando, y administrando la anestesia al Practicante Rodolfo S. Roccatagliata que tiene en su mano derecha un frasco, del cual deja caer esporádicamente un fármaco sobre una máscara anestésica. Es probable que la anestesia se realizó con el cloroformo y la máscara de Schimmelbusch. En las conclusiones, se considera que esta película testimonia que la primera filmación de una anestesia general en el mundo se hizo en Buenos Aires.

In 1899 at the old Hospital de Clínicas in Buenos Aires, Eugenio Py filmed a surgical procedure performed by Dr. Alejandro Posadas to extirpate a lung hydatid cyst. The film shows Posadas operating and Rodolfo S. Roccatagliata administering anesthesia, holding in his right hand a bottle from which he sporadically drops a drug to an anesthetic mask. It is probable that the anesthetic was chloroform, and the mask that of Schimmelbusch. The conclusion is that that this film is proof of being the first one of general anesthesia in the world, taken in Buenos Aires.

Em 1899, no velho Hospital de Clínicas de Buenos Aires, Eugenio Py filmou uma intervenção cirúrgica realizada pelo Dr. Alejandro Posadas para resseção de um cisto hidático de pulmão. No filme se vê ao Dr. Posadas operando e ao praticante Rodolfo S. Roccatagliata administrando a anestesia, quem tem um frasco na mão direita do qual cai, esporadicamente, um fármaco a uma máscara anestésica. É provável que o anestésico fosse o clorofórmio e a máscara, a de Schimmelbusch. Nas conclusões se considera que este filme é um testemunho de que a primeira filmagem de uma anestesia geral no mundo foi feita em Buenos Aires.

Resumo: El presente trabajo tiene por objeto recordar al Dr. Ricardo Gutiérrez como el iniciador de la pediatría argentina y como uno de los poetas más destacados de la segunda generación romántica de nuestro país. Así mismo, dar a conocer su labor como anestesiólogo (en esa época cloroformista), específicamente en la humana tarea de la “supresión de los dolores del parto”, de acuerdo con su tesis de doctorado aprobada el 30 de mayo de 1868 por la Facultad de Medicina de la Universidad de Buenos Aires. En los albores de la anestesia, en nuestro país y en el extranjero, a quien la administraba se lo llamaba cloroformista o eterizador, de acuerdo al fármaco anestésico empleado. Las conclusiones de la presente investigación histórica son las siguientes: 1. A este prestigioso médico se lo debe considerar uno de los pioneros argentinos en emplear un anestésico para suprimir los dolores en las parturientas. 2. Su tesis de doctorado referida a la anestesia obstétrica fue la primera sobre ese tema aprobada por la Facultad de Medicina de la Universidad de Buenos Aires.

The purpose of this paper is to remember Dr. Ricardo Gutiérrez as pioneer of Argentine pediatrics and one of the most prominent poets of the second romantic generation in our country. Also, to make known his work as anesthesiologist (chloroformist in those times), specifically in the humane task of “suppressing delivery pains” as per his doctoral thesis approved on May 30, 1868 by the Buenos Aires University School of Medicine. In the early days of anesthesia, in our country and abroad, people who administered anesthesia were called chloroformists or etherizers according to the anesthetic drug used. Follow are the conclusions of this historical research: 1. This prestigious physician must be considered one of the Argentine pioneers in the use of an anesthetic to suppress pain in women in labor. 2. His doctoral thesis concerning anesthesia in obstetrics was the first one on this subject, approved by the Buenos Aires University School of Medicine.

Este artigo tem como objetivo lembrar o Dr. Ricardo Gutiérrez como iniciador da pediatria argentina e um dos poetas mais destacados da segunda geração romântica da Argentina. Também, divulgar seu trabalho como anestesiologista (naquela época, cloroformista), especificamente na humana tarefa da “supressão das dores de parto”, como consta na sua tese de doutorado aprovada em 30 de maio de 1868 pela Faculdade de Medicina da Universidade de Buenos Aires. Nos alvores da anestesia, tanto na Argentina como no exterior, quem administrava anestesia era chamado cloroformista ou eterizador, de acordo com o fármaco anestésico utilizado. As conclusões da presente investigação histórica são as seguintes: 1. Este destacado médico deve ser considerado um dos pioneiros argentinos em usar um anestésico para eliminar as dores nas parturientes. 2. Sua tese de doutorado referida à anestesia obstétrica foi a primeira sobre esse assunto aprovada pela Faculdade de Medicina da Universidade de Buenos Aires.

Resumo: The hypoglycemic effects of hexane, chloroform and methanol extracts from fruits of Ferocactus latispinus and Ferocactus histrix were evaluated by oral administration to normoglucemic and streptozotocin-induced severe diabetic rats (SD). The anti-diabetic effect was examined by blood glucose, triglycerides, lipid peroxidation, total cholesterol levels in the serum, glycogen content of liver and skeletal muscles, superoxide dismutase (SOD) catalase (CAT), glutathione reductase (GR) and glutathione peroxidase (GSHPx) levels. The most active extracts were obtained with chloroform. Chloroform extracts from F. latispinus and F. histrix increased activities of SOD, GR, GSHPx and CAT, hepatic glycogen content, glucose-6-phosphatase (G6Pase) and the plasma insulin levels. They also, decreased glucokinase (GK) and TBAR (thiobarbituric acid assay). Of the two plants studied F. latispinus showed better antihyperglycemic and antihyperlipidemic effects that F. histrix. In conclusion F. latispinus and F. histrix possesses significant antihyperglycemic properties after 4 h after a single oral dose. It can also improve hyperlipidemia and hypoinsulinemia in streptozotocin-induced diabetic. These results demonstrated that F. latispinus and F. histrix typically used as a health food, has strong antidiabetic effects in vivo, thus, it may have beneficial properties in the prevention of diabetes.

Los efectos hipoglucemiantes de extractos obtenidos con hexano, cloroformo y metanol a partir de frutos de Ferocactus latispinus y Ferocactus histrix fueron evaluados por la administración oral a ratas normales y con diabetes severa (SD) inducida por estreptozotocina. Los extractos más activos fueron obtenidos con cloroformo el cuál incrementa los niveles de SOD, GR, GSHPx y el CAT, el contenido de glucógeno hepático, la glucosa-6-fosfatasa (G6Pase) y los niveles de insulina plasmática. También producen disminución de la glucoquinasa (GK) y TBARS. De las dos plantas estudiadas la F. latispinus presento mayor actividad antihiperglicemiante y antihiperlipidémicos que la F. histrix. En conclusión F. latispinus y F. histrix pueden mejorar la hiperlipidemia y la hipoinsulinemia en animales diabéticos inducida por estreptozotocina. Estos resultados demostraron que F. latispinus y F. histrix utilizadas normalmente como un alimento saludable, tiene fuertes efectos antidiabéticos in vivo, por lo tanto, pueden tener propiedades beneficiosas en la prevención de la diabetes.
