Resumo: ABSTRACT Background: Extramedullary hematopoiesis depends on complex pathophysiological mechanisms linked to hematopoietic stem cells and the proteins considered mediators of the inflammation. The identification of hematopoietic cells outside bone marrow in the adult is an occurrence that can occasionally follows the inflammatory response, was considered a secondary occurrence, but current biomolecular studies have changed that concept. Aim: Describe the presence of clusters of precursor cells of platelets (megakaryocytes), and cells of the inflammatory response in the abdominal wall and spleen of rats with experimentally induced incisional hernias and repaired with different synthetic prostheses. Methods: Twenty-five rats with incisional hernias previously performed, were divided into groups of five animals each: Group 1, repair of the hernia defect without prosthetic implant; Group 2, repair with polypropylene prosthesis; Group 3, repair using polypropylene with low weight; Group 4, the use of polypropylene and polyglecaprone prosthesis; Group 5, of polypropylene and polyglactin prosthesis. All prostheses were cut in rhombus format with area 2,625 cm². The animals were reoperated after 10 days, the abdominal walls were removed with the viscera attached to them and the material was processed for histological study. Results: Megakaryocyte niches in the abdominal wall and spleen, occasionally removed together with the adhesions produced in animals with implantation of prostheses and significant inflammatory reaction. Conclusion: The intense inflammatory reaction due to the prostheses with polypropylene in their composition was disproportionate to the expected response, indicating that further studies should be accomplished including immunophenotyping evaluation and specific panels of monoclonal antibodies to better understand the findings.

RESUMO Racional: A hematopoiese extramedular depende de mecanismos fisiopatológicos complexos, havendo relação destas células-tronco hematopoiéticas com proteínas mediadoras da inflamação. A identificação de células hematopoiéticas fora da medula óssea no adulto, situação que ocasionalmente pode acompanhar a resposta inflamatória era considerada ocorrência secundária, mas estudos biomoleculares modificaram este conceito. Objetivo: Descrever agrupamentos de células precursoras das plaquetas (megacariócitos) e células da resposta inflamatória, na parede abdominal e no baço de ratos com hérnias incisionais induzidas experimentalmente e reparadas com diferentes próteses sintéticas. Métodos: Vinte e cinco ratos com hérnias incisionais previamente realizadas foram distribuídos em grupos com cinco animais: Grupo 1, reparo do defeito herniário sem implante de prótese; Grupo 2, reparo com prótese de polipropileno; Grupo 3, reparo empregando polipropileno com baixa gramatura; Grupo 4, utilização de prótese de polipropileno e poliglecaprone; Grupo 5, prótese de polipropileno e poliglactina. Todas as próteses foram recortadas na forma de losangos com área de 2,625 cm². Os animais foram reoperados após 10 dias, as paredes abdominais foram retiradas em bloco com as vísceras a elas aderidas e o material foi processado em rotina histológica. Resultados: Foram evidenciados nichos de megacariócitos na parede abdominal e no baço coletado juntamente com as aderências em animais com implante de próteses, além de reação inflamatória significativa. Conclusão: A intensa reação inflamatória, local e sistêmica em relação às próteses com polipropileno em sua composição, foi desproporcional à resposta esperada, requerendo aprofundamento do estudo com avaliação da imunofenotipagem e painéis específicos de anticorpos monoclonais para melhor esclarecimento.

Título: Polymer blend: a new approach for eliminating curing effect of aqueous dispersion coatings

Resumo: The aim of present work was to investigate blends of Eudragit® NE 30D with Aquacoat® ECD using different ratios to eliminate curing effect associated with individual polymers. Propranolol HCl 10% w/w was layered onto sugar cores using 5% w/w HPMC as a binder. Drug-layered-cores were coated either with pure or blends of Aquacoat® ECD: Eudragit® NE 30D in a fluidized bed coater to obtain 20% w/w coating level. Talc 35% w/w was used as anti-tacking agent. The pellets were characterized for in vitro dissolution studies, morphology, water uptake-weight loss, osmolality and adhesion of coating after curing at 60 °C or 60 °C/75% RH for 24 h. The findings revealed that Aquacoat® ECD coated pellets showed curing effect due to further gradual coalescence of polymeric particles which resulted into better film formation upon curing. In contrast, the curing effect of Eudragit® NE 30D coated pellets was caused by decrease in adhesion of coatings after curing which provided entirely different swelling behavior of uncured (localized swelling) and cured (uniform swelling) pellets. The undesired curing effect of individual polymers was eliminated by using their blends in appropriate ratio.

Resumo: BACKGROUND: Super-paramagnetic iron oxide nanoparticles (SPION) contain a chemotherapeutic drug and are regarded as a promising technique for improving targeted delivery into cancer cells. RESULTS: In this study, the fabrication of 5-fluorouracil (5-FU) was investigated with loaded Dextran (DEXSPION) using the co-precipitation technique and conjugated by folate (FA). These nanoparticles (NPs) were employed as carriers and anticancer compounds against liver cancer cells in vitro. Structural, magnetic, morphological characterization, size, and drug loading activities of the obtained FA-DEX-5-FUSPION NPs were checked using FTIR, VSM, FESEM, TEM, DLS, and zeta potential techniques. The cellular toxicity effect of FA-DEX-5-FU-SPION NPs was evaluated using the MTT test on liver cancer (SNU-423) and healthy cells (LO2). Furthermore, the apoptosis measurement and the expression levels of NF-1, Her-2/neu, c-Raf-1, and Wnt-1 genes were evaluated post-treatment using flow cytometry and RT-PCR, respectively. The obtained NPs were spherical with a suitable dispersity without noticeable aggregation. The size of the NPs, polydispersity, and zeta were 74 ± 13 nm, 0.080 and 45 mV, respectively. The results of the encapsulation efficiency of the nano-compound showed highly colloidal stability and proper drug maintenance. The results indicated that FA-DEX-5-FU-SPION demonstrated a sustained release profile of 5-FU in both phosphate and citrate buffer solutions separately, with higher cytotoxicity against SNU-423 cells than against other cells types. These findings suggest that FA-DEX-SPION NPs exert synergistic effects for targeting intracellular delivery of 5-FU, apoptosis induction, and gene expression stimulation. CONCLUSIONS: The findings proved that FA-DEX-5-FU-SPION presented remarkable antitumor properties; no adverse subsequences were revealed against normal cells.

Resumo: Stents farmacológicos foram desenvolvidos para reduzir a resposta proliferativa neointimal e consequentemente a reestenose, mais frequente limitação da intervenção coronária percutânea com balão e stents não faramcológicos. O desenvolvimento destes dispositivos baseia-se no maior entendimento da biologia da reestenose, na seleção de fármacos anti-proliferativos adequados para os diversos mecanismos envolvidos nesta complicação e no uso de plataformas/polímeros adequados para entrega do fármaco. Consequentemente o desempenho destes dispositivos depende da perfeita interação de todos estes elementos. As abordagens atuais para minimizar a reestenose são revisados neste capítulo. Embora a primeira geração dos stents farmacológicos tenha sido focada na eficácia em reduzir a reestenose, questões relacionadas à sua segurança surgiram, comprometendo seu uso mais disseminado. As novas gerações de stents farmacológicos com polímeros duráveis ou bioabsorvíveis conseguiu reduzir as taxas de nova intervenção e de trombose. Embora o modelo ideal de stent farmacológico ainda esteja em investigação, é certo que esta tecnologia já se estabeleceu como primeira linha na intervenção coronária percutânea contemporânea

Título: Discoloration and force degradation of orthodontic elastomeric ligatures

Resumo: ABSTRACT OBJECTIVE: The aim of the present study was to evaluate color changes and force degradation of orthodontic elastomeric ligatures in different stretching patterns during a 8-weeks period. METHODS: Two elastomers with the minimum and two with the maximum color changing, and gray elastomers of two brands (American Orthodontics and Ortho Technology) were selected according to an opinion poll with clinicians and color changes after 4 weeks of intraoral use were evaluated. These elastomers were mounted on special jigs fabricated using a CAD-CAM technique, underwent different stretching patterns and the force was measured in 0, 24 hours, 2, 4 and 8 weeks. During in vivo part of the study, force levels of elastomers were measured after 4 weeks on a material testing machine. Data were analyzed with four-way ANOVA and Tukey post hoc tests. RESULTS: All the elastomers showed color changing but the degree of color stability was significantly different. The mean force degradation was higher in 1-mm stretch groups. After 8 weeks, the average residual force of elastomers was 1.45 ± 0.18 N and the maximum force decay was seen in the elastomers that exhibited the maximum initial force. CONCLUSION: There is significant relationship between the stretching pattern and the amount of residual force of elastomers. Elastomers with higher initial forces exhibited higher percentages of force loss after 8 weeks. It seems that there is a relationship between initial color and color changing of elastomers.

RESUMO OBJETIVO: o objetivo do presente estudo foi avaliar as alterações de cor e a degradação da força de ligaduras elásticas em diferentes padrões de estiramento, durante um período de 8 semanas. MÉTODOS: duas ligaduras elásticas com alteração mínima de cor, duas com alteração máxima de cor e ligaduras elásticas na cor cinza de duas marcas comerciais (American Orthodontics e Ortho Technology) foram selecionadas de acordo com uma pesquisa de opinião com clínicos, e mediante a avaliação das alterações de cor após quatro semanas em uso intrabucal. Essas ligaduras elásticas foram montadas em jigs fabricados por meio da técnica CAD-CAM e foram expostas a diferentes padrões de estiramento, e a força foi medida nos tempos de 0h, 24h; 2, 4 e 8 semanas. Durante a parte in vivo do estudo, os níveis de força das ligaduras elásticas foram medidos após 4 semanas, em uma máquina de teste de materiais. Os dados foram analisados com os testes ANOVA de quatro vias e post-hoc de Turkey. RESULTADOS: todas as ligaduras elásticas mostraram alterações de cor, mas houve diferenças significativas quanto ao grau de estabilidade da cor. A média de degradação da força foi mais alta nos grupos com estiramento a 1 mm. Após 8 semanas, a média da força residual nas ligaduras elásticas foi de 1,45 ± 0,18 N e a maior degradação na força foi observada nas ligaduras elásticas que apresentaram a maior força inicial. CONCLUSÃO: há uma relação significativa entre o padrão de estiramento e a quantidade de força residual nas ligaduras elásticas. As ligaduras elásticas com as maiores forças iniciais exibiram as mais altas porcentagens de degradação da força após 8 semanas. É, também, possível que haja uma relação entre a cor inicial da ligadura elástica e sua alteração de cor.

Resumo: As próteses bucomaxilofaciais têm um importante papel na devolução da estética e de algumas funções para os pacientes. Suas boas condições são primordiais, para que possam exercer suas funções adequadamente e manter o local, onde estão inseridas, livre de infecções e inflamações. Portanto, as não formações de colônias e biofilmes bacterianos em materiais eleitos para confecção dessas próteses trarão benefícios aos pacientes reabilitados. Com esse propósito, o presente artigo verificou a eficiência bactericida dos biocidas policloreto de dialildimetilamônio (PDADMAC) e poliacrilamida catiônica (PAC) quando pincelado em resina acrílica termopolimerizável e silicone de grau médico. Os resultados mostraram que o biocida PDADMAC dissolvido em água obteve boa resposta antibacteriana, quando pincelado sobre ambos os materiais. Já o biocida PAC, entretando, mostrou eficiência antibacteriana apenas quando aplicado resina acrílica termopolimerizável... (AU)

The maxillofacial prosthesis have an important role in reestablishing esthetics and some of the patients functions. The prosthesis' condition is imperative to guarantee their appropriate function and keep the receptor site free of infections or inflammatory processes. Therefore, preventing the infection of the chosen material by bacterial colonies and biofilm can be hazardous to the patient's health. To investigate such problem, the present article evaluated the efficiency of two biocides: pol(diallyldimethylammonium chloride (PDADMAC) and polyacrylamide cationic (PAC). The biocides were applied over heat-cured acrylic resin or silicone. The results shows that the PDADMAC biocide dissolved in water had positive outcome when brushed over both materials. The PAC biocide had antibacterial efficiency only when brushed over the heat-cured acrylic resin... (AU)

Título: The outcomes of two different bulking agents (dextranomer hyaluronic acid copolymer and polyacrylate-polyalcohol copolymer) in the treatment of primary vesico-ureteral reflux

Resumo: ABSTRACT Purpose Subureteral injection of bulking agents in the endoscopic treatment of vesicoureteral reflux is widely accepted therapy with high success rates. Although the grade of vesicoureteric reflux and experience of surgeon is the mainstay of this success, the characteristics of augmenting substances may have an effect particularly in the long term. In this retrospective study, we aimed to evaluate the clinical outcomes of the endoscopic treatment of vesicoureteric reflux (VUR) with two different bulking agents: Dextranomer/hyaluronic acid copolymer (Dx/HA) and Polyacrylate polyalcohol copolymer (PPC). Materials and Methods A total 80 patients (49 girls and 31 boys) aged 1-12 years (mean age 5.3 years) underwent endoscopic subureteral injection for correction of VUR last six years. The patients were assigned to two groups: subureteral injections of Dx/HA (45 patients and 57 ureters) and PPC (35 patients and 45 ureters). VUR was grade II in 27 ureters, grade III in 35, grade IV in 22 and grade V in 18 ureters. Results VUR was resolved in 38 (66.6%) of 57 ureters and this equates to VUR correction in 33 (73.3%) of the 45 patients in Dx/HA group. In PPC group, overall success rate was 88.8% (of 40 in 45 ureters). Thus, Thus, this equates to VUR correction in 31 (88.5%) of the 35 patients. Conclusions Our short term data show that two different bulking agent injections provide a high level of reflux resolution and this study revealed that success rate of PPC was significantly higher than Dx/HA with less material.

Resumo: ABSTRACT Introduction: Various bulking agents were utilized for endoscopic correction of VUR. A study reviewing multi-institutional data showed that the amount of injection material has increased over time with the purpose of improving success rates, which also resulted in costs. We noticed an opposite trend in our center since we started using a new bulking agent. The aim of this study was to evaluate evolution of our practice with different bulking agents. Patients and Methods: Records of VUR patients who underwent subureteric injection with polyacrylate polyalcohol copolymer (PPC) and dextronomere hyaluronic acide (DxHA) between 2005 and 2014 were reviewed. Variation of different parameters throughout the study period was evaluated along with the success rate. Success was defined as complete resolution of reflux. Results: A total of 260 patients with 384 refluxing units were included. The success rate was higher in PPC group compared to DxHA group. There was no statistically significant difference between years regarding distribution of VUR grade, body weight, patient height, and age in PPC group. Despite significant reduction in injection volume, success rate did not decrease through the years with PPC. Conclusion: Different bulking agents may require different injection volumes to achieve the same success rate in endoscopic treatment of vesicoureteral reflux. Habits gained with previous experience using other materials should be revised while using a new agent.

Título: Idiosyncratic reaction after injection of polyacrylate - polyalcohol copolymer

Resumo: ABSTRACT Context: Polyacrylate-polyalcohol copolymer is a synthetic product, non-biodegradable, with low rate of therapeutic failure and lower incidence of reactions at the site of injection, when compared to biodegradable agents. We report an unprecedent, exuberant and persistent inflammatory reaction following injection of that substance. Patient: a 17 years-old patient with vesico-ureteral reflux and complete pyelocaliceal right duplication was submitted to treatment with polyacrylate-polyalcohol copolymer (STING technique). In the seventh day of post-operatory, she presented intense dysuria and hypogastric pain, without laboratory exams alterations; a symptomatic treatment was started. After two months, the symptoms persisted and an ultrasound detected thickening of bladder wall close to the uretero-vesical junction. After that exam, a cystostopic biopsy showed epithelial hyperplasia with increased edema of lamina propria, suggesting an adverse reaction to the polymer. After four months, there was complete remission, but the reflux persisted with the same grade. Hypothesis: This is an unprecedent reaction following injection of this copolymer. The presence of characteristics such as absence of infection, temporal relation between treatment and beginning of symptoms, and detection of epithelial hyperplasia at the local of injection reinforce the hypothesis of association of the substance and adverse reaction. In that patient, important complains motivated early investigation of urinary tract, that confirmed those aspects. Maybe if that reaction had occurred in patients with lower capacity of expression (such as in infants) it would be unnoticed.

Resumo: Abstract Despite its potential in the production of polymers from renewable sources, D-limonene faces difficulties in its polymerization, resulting in low monomer conversion and molar mass. In order to investigate the non-ideality inherent kinetics, this work explores different modeling strategies for D-limonene radical polymerization, using benzoyl peroxide as initiator. The starting model considered the classical approach for conventional radical polymerization. This model was then corrected by including reaction orders different from the unit. After an analysis and choice of the best model, computer simulations were compared with experimental results from literature, validating the chosen approach. It was found that the process is drastically influenced by chain transfer reactions, presenting a non-ideal behavior. Finally, an analysis of distinct reaction conditions provided information on monomer conversion, molar mass and polymer dispersity, which could guide future research in the synthesis optimization. Higher molar mass poly(limonene) were obtained by simultaneously reducing the monomer and initiator concentrations.
