[Ab] Resumo:Based on the study of actual nutrition and availability of macroelements there was found that adult population of the city of Omsk was established to refer to the group at risk for the development of micronutrient deficiency associated with low content of a set of essential elements (copper, zinc, calcium, selenium) in the ration, being prior for correction.There was executed the hygienic substantiation developed the fermented acid milk bioproduct of a functional purpose "Bifidin" enriched with micronutrients, prior for the population of the Omsk region. Introduction of this bio-product into the ration allows to effectively correct disorders of the mineral status, that was manifested in a decline in the proportion of patients with insufficiency of calcium (from 37.9 to 24.1%; p = 0.013), copper (from 51.7 to 25.9%; p = 0.004), selenium (from 96.6 to 84.5%; p = 0.026), zinc (from 58.6 to 48.3% of the subjects; p = 0.264). Upon the completion of the prophylactic course (intake "Bifidin" of 200 ml for 60 days), there was noted the gain in the concentration in hair: calcium (by 24.4%; p=0.441), zinc (by 8.0%; p=0.0008), copper (by 8.8%; p < 0.001), selenium (by 41.5%; p < 0.001). There was established the efficacy of the use of bio-product "Bifidin" enriched with micronutrients for the improvement of the structure of nutrition and element status of the adult population of the city of Omsk. There was justified the extensive use of dairy products enriched with micronutrients both for the reduction of the prevalence of microelementoses and the improvement of the nutritional status.

[Ab] Resumo:Cannabinoids including, but not limited to Δ9-tetrahydrocannabinol, 11-hydroxytetrahydrocannabinol, and (-)-11-nor-Δ9-tetrahydrocannabinol-9-carboxylic acid are known to toxicologists and synthetic chemists as difficult compounds because they are subject to numerous degradative pathways. It is the purpose of this short review article to discuss common pathways that result in the disappearance of cannabinoids - such as conjugate formation, adsorption to surfaces, chemical reactions, microbial action, thermal decomposition, chemical bonding, photosensitivity, sample handling, analytical methodology, and micelle trapping - and to point out possible ways to avoid such degradation.

[Ab] Resumo:Brominated flame retardants (BFRs) and Dechlorane Plus (DP) were measured in both human hair and paired serum samples from a cohort of university students in South China. Segmental analysis was conducted to explore gender difference and the relationships between the hair and serum. The concentrations of total PBDEs in the hair and serum samples were in a range of 0.28-34.1ng/g dry weight (dw) and 0.16-156ng/g lipid weight (lw), respectively. Concentrations of ∑DPs (sum of the syn-DP and anti-DP isomers) in all hair samples ranged from nd-5.45ng/g dry weight. Concentrations of most PBDEs and decabromodiphenylethane (DBDPE) in distal segments (5-10cm from the scalp) were higher than those in the proximal segments (0-5cm from the scalp) (t-test, p < 0.05), which could be due to the longer exposure time of distal segments. The proximal segments exhibited a unique congener profile, more close to that in the serum rather than the distal segments of hair. An obvious gender difference was found in the levels of ∑PBDEs using integrated hair samples, while the difference disappeared when considering alone the proximal segments of hair (0-5cm from scalp) for both genders. This paper provides supplement to the current knowledge on sources of BFRs and DPs in hair and declares the importance of segmental analysis.

[Ab] Resumo:OBJECTIVES: To identify the species of mammalian hair using COⅠ gene mini-barcoding technology. METHODS: A pair of universal primers for mammalian COⅠ gene mini-barcoding were designed. The hair DNA samples of experimental animals from 11 species in 5 orders, mammalia was amplified by PCR technology with universal primers, and the PCR products were sequenced by bi-directional DNA and after the sequences splicing the results were deposited into the BOLD database to perform the homology comparison. RESULTS: The DNA of hair from all experiment animal species could be totally amplified by the mini-barcoding universal primers designed in this study. The length of amplified fragment was 147 bp. The result of homology comparison in the database showed that the closest matching species were consistent with the experiment animal species. In addition to the matching degree of (98.99%), all homology matching degree of the other experiment animals were 100%, and the intraspecific genetic distance of was 1%. The interspecific genetic distance was ten times more than the intraspecific genetic distance which could be used for species identification. CONCLUSIONS: The COⅠ gene mini-barcode technology is established and can provide fast and accurate species identification for hair of mammals.

[Ab] Resumo:Human hairs are the trace evidence most commonly encountered at many crime scenes. However, they have not been effectively utilized for actual criminal investigations because of the low accuracy of their morphological inspection, low detection rate of short tandem repeat (STR) typing, and the problem of heteroplasmy in mitochondrial DNA analysis. Here, we examined the possibility of individual discrimination by comparing profiles of bacterial flora on hair. We carried out the profiling of terminal restriction fragment length polymorphisms (T-RFLP) of the amplified bacterial 16S ribosomal RNA (rRNA) gene from hair samples. Compared with existing STR typing methods that use hair roots, this method using hair shafts allowed the detection of stable bacterial DNA. We successfully obtained the T-RFLP profile from single hair shafts of all volunteers tested. The profiles were specific to each individual, and multiple profiles obtained from the individual him/herself showed higher similarity than those from different individuals. These individual-specific profiles were stably obtained from samples from most volunteers, when collected again after 6months. Storage of the collected hair samples at -30°C was effective for obtaining reproducible T-RF profiles. When unidentified hair samples collected in the laboratory were compared with a pre-constructed database, 17 of 22 hairs were assigned to a small group of people, including the corresponding individuals. These results show that T-RFLP analysis of bacterial flora on a hair shaft found at a crime scene could provide useful information for narrowing down a suspect.

[Ab] Resumo:The Polled Intersex Syndrome (PIS) is responsible for the absence of horns in homozygous and heterozygous goats causing a female-to-male sex reversal in the homozygous polled genotypic female (XX) goats. A simple and efficient non-invasive method was elaborated to detect the genotypic sex from hair and faecal samples using a pair of primers to amplify the X- and Y-linked alleles of the amelogenin gene. The PCR products were easily distinguishable using agarose gel electrophoresis: we detected an X-specific single band in samples originating from healthy phenotypic females and double (X- and Y-) bands in samples from males. The new PCR method is applicable for diagnosing the sex of PIS-affected animals already as newborn kids, in contrast with the phenotypic findings appearing only after puberty, and thus it may replace the cumbersome chromosome investigations.

[Ab] Resumo:Wastes and tailing ponds of Tyrnyauz tungsten and molybdenum factory are the main sources of heavy metal incoming into environment in Kabardino-Balkarian Republic. The factory was closed more than 10 years ago and the recultivation of it's tailing ponds, where accumulated hundreds of millions tones of wastes, was completed. The aim of this investigation was an assessment of their possible influence on children residing in the vicinity of these tailing ponds (village Bylym). Village Verhny Baksan located about 30 km upstream of the valley of the Baksan River was chosen as reference (pure) locality. As a results of the performed investigations we revealed that in drinking water of Bylym and Verhny Baksan concentrations of molybdenum were 2.10±0.42 pg/l and 0.31±0.15 pg/l correspondingly, which is remarkably lower than maximum permitted concentrations. The concentrations of Mo, Cu and Pb in children's hair in both villages were practically the same, which indicates to the absence of their accumulation in human organism. But the quantity of cells with cytogenetic disorders in buccal epithelial cells in children from Bylym was 4.1 times higher in comparison with the corresponding index of uncontaminated area. The obtained data demonstrate that genotoxic effect of remedied tailing ponds retains.

[Ab] Resumo:The paper presents information about the content of benzo(a)pyrene in soil samples of petroleum products and the content of heavy metals (iron, copper, lead, arsenic, manganese, chromium, cadmium and mercury) in the soil and hair samples of children. Soil samples and hair were taken in October 2014 in the north-western part of the Yamal-Nenets Autonomous District. The values obtained were compared with the approximately permissible levels and maximum allowable concentrations adopted in the Russian Federation. Levels are determined compounds in the soil in general do not exceed the allowable concentrations. In hair samples there was noted marked exceeding of the recommended values for zinc, copper, chromium and iron 10%, 16.7%, 83.3% and 100% samples, respectively. The cause of the high iron content in the hair samples may be due to the quality of consumed water, which is characterized by low mineralization and high iron content. Increased chromium content of more than 80% of the hair samples can be caused, as in the case of iron, the quality of water consumed, but also a high proportion of local foods in the diet of people, rich by chromium. A conducted study on the state of the environment in the village, away from areas of hydrocarbon production, indicates to the low importance of anthropogenic impact.

[Ab] Resumo:The objective of the study was to demonstrate how mitochondrial DNA (mtDNA) can be used to determine the species origin of animal microtraces. The study included pieces of cat and dog hair without the root, a fragment of cooked chicken bone (0.1g), three goose down samples (0.028 g), a pork swab, a pork scratching (5×5×5 mm), and pork lard (0.22 g). DNA was isolated from all of these samples using the method appropriate for the particular source material. The extracts had DNA concentration exceeding 5.4 ng/µl with A purity range of 1.14-1.88. Next, the samples were subjected to PCR and real-time PCR with species-specific primers and primers complementary to mitochondrial DNA (mtDNA). Control reactions based on the amplification of eukaryotic-specific fragment (18S rRNA) were additionally performed. PCR and real-time PCR products for detection of species-specific mtDNA were obtained for all templates, whereas during the detection of eukaryote DNA no product was obtained for dog and cat hair only. The poor quality of the obtained DNA did not prevent the analysis. The results showed that mitochondrial DNA is suitable for identification of small or highly processed samples, in which genomic DNA often cannot be analyzed.

[Ab] Resumo:To assess the association between exposure to the tobacco, heavy metals and phthalate on early pregnancy and missed abortion.42 women with missed abortion and 57 matched controls (women with normal pregnancies) were recruited between March and May 2012, from the Department of Gynecology and Obstetrics, First Affiliated Hospital of Guangxi Medical University and the People Hospital of Guangxi Zhuang Autonomous Region. The questionnaire survey was carried on to learn about the basic conditions, as well as smoking history of all participants. The levels of tobacco, heavy metal, and phthalate exposure were compared between the 2 groups by measuring nicotine, cocaine, cadmium (Cd), manganese (Mn), plumbum (Pb) and dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP), butyl benzyl phthalate (BBP), di-2-ethyl hexyl phthalate (DEHP) in the hair samples.Out results showed that significant differences in age (P = .042), premarital examination (P = .041), passive smoking (P = .021), and heavy metal exposure (P = .022) were found in the case group compared to the control. In addition, the concentration of nicotine (P = .037), cotinine (P = .018), Cd (P = .01), Pb (P = .038) and DEHP (P = .001) in the hair were significantly higher in the case group. Furthermore, logistic analysis revealed that age [Odds Ratio (OR) 1.172, 95% confidence interval (CI) 1.036-1.327], Cd (OR 8.931, 95% CI 2.003-39.811), Cotinine (OR 4.376, 95% CI 1.159-16.531), DEHP (OR 1.863, 95% CI 1.103-3.146) were important factors contributing to the missed abortion (P < .05).It was demonstrated that high gestational age, passive smoking, heavy metals, and the phthalate exposure were the risk factors for missed abortion, while the premarital health examination was a protective factor. Avoiding these harmful substances before getting pregnant and during the early stages of pregnancy, might help prevent missed abortions.
