[Ab] Resumo:Studies addressing the toxicity of pesticides towards non-target organisms focus on the median lethal concentration and biochemical response of individual pesticides. However, when determining environmental risks, it is important to test the combined effects of pesticides, such as insecticides and herbicides, which are frequently used together in agricultural areas. Here we aimed to investigate the toxic effects of the combined use of the herbicide atrazine and the insecticides, endosulfan, indoxacarb, and thiamethoxam on Gammarus kischineffensis. To do this, we tested the activities of oxidative stress, detoxification, and neurotoxicity biomarkers. Compared to atrazine alone, we detected higher glutathione-S-transferase, catalase and superoxide dismutase activities (oxidative stress biomarkers) when atrazine was combined with either endosulfan or indoxacarb. However, higher IBR values were determined in organisms where pesticide mixtures were used according to individual use. Based on these results, mixtures of atrazine and other pesticides may cause synergistic effects and may be evidence of increased toxicity and oxidative stress.

[Ab] Resumo:Regeneration is a complex and dynamic process, mobilizing diverse cell types and remodelling tissues over long time periods. Tracking cell fate and behaviour during regeneration in active adult animals is especially challenging. Here, we establish continuous live imaging of leg regeneration at single-cell resolution in the crustacean . By live recordings encompassing the first 4-5 days after amputation, we capture the cellular events that contribute to wound closure and morphogenesis of regenerating legs with unprecedented resolution and temporal detail. Using these recordings we are able to track cell lineages, to generate fate maps of the blastema and to identify the progenitors of regenerated epidermis. We find that there are no specialized stem cells for the epidermis. Most epidermal cells in the distal part of the leg stump proliferate, acquire new positional values and contribute to new segments in the regenerating leg.

[Ab] Resumo:Gammarus minus, a freshwater amphipod living in the cave and surface streams in the eastern USA, is an excellent model for investigating evolutionary adaptation to the subterranean environment. RNA-Seq was conducted on one pair of morphologically distinct sister populations inhabiting surface and cave habitats to identify genes that were differentially expressed in the two populations, as well as to compare levels and patterns of genetic variation within and between populations. Of the 104,630 transcripts identified in the transcriptome assembly, 57% had higher average levels of expression in the cave population. After Benjamini-Hochberg correction for multiple tests, 1517 and 551 transcripts were significantly upregulated or downregulated, respectively, in the cave population, indicating an almost three-fold enrichment of cave-upregulated genes. The average level of nucleotide diversity across all transcripts was significantly lower in the cave population. Within the cave population, where the average nucleotide diversity of cave-downregulated transcripts was 75% that of the cave-upregulated transcripts, a highly significant difference, whereas within the spring population the nucleotide diversities of cave-downregulated and cave-upregulated transcripts was virtually identical. Three lines of evidence suggest that the reduced variation in cave downregulated transcripts is due to positive selection in the cave population: 1) the average neutrality index of cave-downregulated genes was < 1, consistent with positive selection, and significantly less than that of cave-upregulated genes; 2) Tajima's D was positively correlated with the cave:surface expression ratio, and 3) cave-downregulated transcripts were significantly more likely to be highly diverged from their surface homologs than cave-upregulated transcripts. Five transcripts had fixed premature termination codons in the cave population. The expression patterns and sequence variation in one such transcript, encoding the DNA repair protein photolyase, were examined in more detail and provide the first evidence for the relaxation of functional constraint in this light-dependent protein in a subterranean population.

[Ab] Resumo:Predators shape prey populations by elimination of individuals (consumptive effects) and by inducing modifications in prey behaviour, physiology or morphology (NCE-non-consumptive effects). Due to the resource allocation to defence, decreased feeding and higher stress, the costs of predator NCEs can be considerable. Therefore, the resistance to NCEs may be crucial for population growth and interspecific competition. We tested the resistance of Ponto-Caspian gammarids Dikerogammarus villosus and Pontogammarus robustoides to NCEs imposed by their predator, the racer goby Babka gymnotrachelus. As D. villosus is often avoided by predators in the presence of alternative food, we hypothesised that it would bear lower behavioural and physiological costs of anti-predator responses. We tested gammarid feeding in short-time experiments (2-4 h) with food (chironomid larvae) located at various distances from the stony shelter (to enforce food searching, Experiment I) or in the direct gammarid proximity (no searching needed, Experiment II). Moreover, we checked the predator effect on gammarid growth in a 2-week Experiment III. Both gammarids exposed to predators reduced feeding efficiency outside the shelter (Experiment I). Contrary to our expectations, the response of D. villosus was stronger. When food was provided in their direct proximity (Experiment II), the feeding of both species was unaffected by predators, indicating that a shelter supplied with food can reduce predator NCEs. The growth of P. robustoides was reduced in the presence of predators (Experiment III), whereas that of D. villosus was unaffected. Although D. villosus has a more effective defence strategy than P. robustoides, it bears similar or even higher behavioural costs of NCEs. However, it exhibits the higher resistance to the long-term predator presence, sustaining its growth rate under such conditions. This may be one of the factors contributing to the great invasion success of D. villosus, currently taking place in European fresh waters.

[Ab] Resumo:Understanding the effect of temperature on ecologically important species has become a major challenge in the context of global warming. However, the consequences of climate change cannot be accurately predicted without taking into consideration biotic interactions. Parasitic infection, in particular, constitutes a widespread biotic interaction, and parasites impact their hosts in multiple ways, eventually leading to consequences for communities and ecosystems. We explored the effect of temperature on the anti-predator behavior of a keystone freshwater invertebrate, the amphipod Gammarus fossarum. Gammarids regularly harbor manipulative acanthocephalan parasites that modify their anti-predator behavior in ways that potentially increase the probability of trophic transmission to their definitive hosts. We investigated the impact of temperature on gammarids infected by two acanthocephalan parasites, Pomphorhynchus tereticollis and Polymorphus minutus. Uninfected and naturally infected gammarids were acclimatized to different temperatures, and their behavior was measured. Our results showed that the effect of infection on the phototaxis of gammarids increased with increasing temperature, with a stronger effect induced by P. tereticollis. In contrast, temperature had no effect on the alteration of refuge use or geotaxis observed in infected gammarids. Our results provide the first direct evidence that temperature can affect the extent of behavioral alteration brought about by certain parasite species. However, the consequences of increased trophic transmission remain elusive; the supposedly key anti-predatory behavior was not significantly affected by exposure of gammarids to different temperatures.

[Ab] Resumo:During morphogenesis numerous morphogenetic factors ensure the production of a target phenotype. By disrupting these processes, a toxic exposure during this period could cause an increase of phenotypic defects. In the present study, embryos of the freshwater amphipod Gammarus fossarum were exposed throughout the embryogenesis to increasing concentrations of fenoxycarb (0, 0.5µgL , 5µgL and 50µgL ), a growth regulator insecticide analog of the insect juvenile hormone. In addition, to identify morphogenesis' sensitive period, embryos were exposed during either early or late embryonic development to 5µgL of fenoxycarb. In newborn individuals from exposed embryos, three phenotypes were investigated: i) eye pigmentation, ii) length of the antenna and gnathopod of both left and right sides and iii) midgut tissue state. Developmental homeostasis was assessed by measuring fluctuating asymmetry and inter-individual variance of both the antenna and gnathopod. Exposure to 5µgL and 50µgL fenoxycarb throughout the embryonic development induced a delayed hatching and altered appendages size. Moreover, exposure to 5µgL throughout the embryogenesis and during the gastrulation phase impaired eye pigmentation, while exposure to 50µgL resulted in increased tissue damages of the midgut. No significant increase of fluctuating asymmetry was observed in exposed individuals, neither for the antenna nor for the gnathopod. These results demonstrate that fenoxycarb can alter embryonic development of G. fossarum without disrupting developmental homeostasis.

The Amphipoda includes six suborders, the Pseudingolfiellidea, Hyperiidea, Colomastigidea, Hyperiopsidea, Senticaudata (described in a previous contribution (Lowry & Myers 2013)) and Amphilochidea. The suborder Ingolfiellidea is raised to order status. A cladistic tree, based on morphology, is presented illustrating the relationships of the Amphipoda at parvorder level. A tree for the families of the Physomatidira and Physocephalatidira, a tree for the Maxillipiidira, Oedicerotidira, Eusiridira and Amphilochidira and a tree for the Synopiidira, Haustoriidira and Lysianassidira, are provided. Families are listed together with their included genera. New families are diagnosed.

[Ab] Resumo:Sunamphitoe Spence Bate, 1857 is one of several ampithoid amphipod genera to be reasonably specific to its algal host. Sunamphitoe was recently shown to be a senior synonym of Peramphithoe Conlan & Bousfield, 1982 by both morphological and molecular phylogenetic analyses that included selected exemplars of both putative genera. Phylogenetic analysis considering all species of Sunamphitoe herein corroborates the validity of this synonymy and also delineates several morphological groups within the genus. Eight new species are descibed herein, Sunamphitoe angrox sp. nov., S. batavia sp. nov., S. dampierensis sp. nov., S. jonathani sp. nov., S. lehae sp. nov., S. mixtura sp. nov., S. naturaliste sp. nov., and S. stevesmithi sp. nov., all from Australia and New Zealand; the status of the problematic New Zealand species, Sunamphitoe aorangi is resolved.
