[Ab] Resumo:Olfaction is a key sense routing foraging behaviour in parasitoids. Preferences for food, mate and host stimuli can be innate in parasitic wasps. Alternatively, learning-mediated mechanisms play a crucial role. Females of the braconid parasitoid Psyttalia concolor exploit olfactory cues arising from tephritid hosts and related microhabitats. However, little is known on the olfactory stimuli routing males searching for mates. In this study, we focused on the attractiveness of Bactrocera oleae-induced olive volatiles towards P. concolor males. Furthermore, we evaluated learning occurrence in virgin males, when trained for selected unattractive volatile organic compounds (VOCs) associated with mate rewards. (E)-ß-Ocimene, α-pinene and limonene attracted virgin males in Y-tube bioassays. Unattractive VOCs evoked positive chemotaxis after associative learning training. P. concolor males exposed to VOCs during a successful or unsuccessful mating, showed short-term preference for these VOCs (<1 h). However, memory consolidation was strictly dependent on reward value. Indeed, males experiencing a successful mating showed a fast consolidation into protein dependent long-term memory, appearing after 24 h. On the other hand, males experiencing a less valuable training experience (i.e. unsuccessful courtship), did not show consolidated memory after 24 h. Overall, our findings suggest that P. concolor virgin males may exploit VOCs from the host microhabitat to boost their mate searching activity, thus their reproductive success. However, since learning is a costly process, P. concolor males retained durable memories just in presence of a valuable reward, thus avoiding maladaptive behaviours.

[Ab] Resumo:Integrative taxonomy has resolved the species status of the potentially invasive Ceratitis rosa Karsch into two separate species with distinct ecological requirements: C. rosa "lowland type" and the newly described species Ceratitis quilicii De Meyer, Mwatawala & Virgilio sp. nov. "highland type". Both species are tephritid pests threatening the production of horticultural crops in Africa and beyond. Studies were carried out by constructing thermal reaction norms for each life stage of both species at constant and fluctuating temperatures. Non-linear functions were fitted to continuously model species development, mortality, longevity and oviposition to establish phenology models that were stochastically simulated to estimate the life table parameters of each species. For spatial analysis of pest risk, three generic risk indices were visualized using the advanced Insect Life Cycle Modeling software. The study revealed that the highest fecundity, intrinsic rate of natural increase and net reproductive rate for C. rosa and C. quilicii was at 25 and 30°C, respectively. The resulting model successfully fits the known distribution of C. rosa and C. quilicii in Africa and the two Indian Ocean islands of La Réunion and Mauritius. Globally, the model highlights the substantial invasion risk posed by C. rosa and C. quilicii to cropping regions in the Americas, Australia, India, China, Southeast Asia, Europe, and West and Central Africa. However, the proportion of the regions predicted to be climatically suitable for both pests is narrower for C. rosa in comparison with C. quilicii, suggesting that C. quilicii will be more tolerant to a wider range of climatic conditions than C. rosa. This implies that these pests are of significant concern to biosecurity agencies in the uninvaded regions. Therefore, these findings provide important information to enhance monitoring/surveillance and designing pest management strategies to limit the spread and reduce their impact in the invaded range.

[Ab] Resumo:The tephritid fruit fly Zeugodacus tau (Walker) is a polyphagous fruit pest of economic importance in Asia. Studies based on genetic markers indicate that it forms a species complex. We report here (1) the complete mitogenome of Z. tau from Malaysia and comparison with that of China as well as the mitogenome of other congeners, and (2) the relationship of Z. tau taxa from different geographical regions based on sequences of cytochrome c oxidase subunit I gene. The complete mitogenome of Z. tau had a total length of 15631 bp for the Malaysian specimen (ZT3) and 15835 bp for the China specimen (ZT1), with similar gene order comprising 37 genes (13 protein-coding genes-PCGs, 2 rRNA genes, and 22 tRNA genes) and a non-coding A + T-rich control region (D-loop). Based on 13 PCGs and 15 mt-genes, Z. tau NC_027290 (China) and Z. tau ZT1 (China) formed a sister group in the lineage containing also Z. tau ZT3 (Malaysia). Phylogenetic analysis based on partial sequences of cox1 gene indicates that the taxa from China, Japan, Laos, Malaysia, Bangladesh, India, Sri Lanka, and Z. tau sp. A from Thailand belong to Z. tau sensu stricto. A complete cox1 gene (or 13 PCGs or 15 mt-genes) instead of partial sequence is more appropriate for determining phylogenetic relationship.

[Ab] Resumo:transformer (tra) is a switch gene of sex determination in many insects, particularly in Dipterans. However, the sex determination pathway in Bactrocera cucurbitae (Coquillett), a very destructive pest on earth, remains largely uncharacterized. In this study, we have isolated and characterized one female-specific and two male-specific transcripts of the tra gene (Bcutra) of B. cucurbitae. The genomic structure of Bcutra has been determined and the presence of multiple conserved Transformer (TRA)/TRA-2 binding sites in Bcutra has been found. BcuTRA is highly conservative with its homologues in other tephritid fruit flies. Gene expression analysis of Bcutra at different developmental stages demonstrates that the female transcript of Bcutra appears earlier than the male counterparts, indicating that the maternal TRA is inherited in eggs and might play a role in the regulation of TRA expression. The conservation of protein sequence and sex-specific splicing of Bcutra and its expression patterns during development suggest that Bcutra is probably the master gene of sex determination of B. cucurbitae. Isolation of Bcutra will facilitate the development of a genetic sexing strain for its biological control.

[Ab] Resumo:Bactrocera invadens Drew, Tsuruta & White, Bactrocera papayae Drew & Hancock, and Bactrocera philippinensis Drew & Hancock, key pest species within the Bactrocera dorsalis species complex, have been recently synonymized under the name Bactrocera dorsalis (Hendel). The closely related Bactrocera carambolae Drew & Hancock remains as a discrete taxonomic entity. Although the synonymizations have been accepted by most researchers, debate about the species limits remains. Because of the economic importance of this group of taxa, any new information available to support or deny the synonymizations is valuable. We investigated the chemical epicuticle composition of males and females of B. dorsalis, B. invadens, B. papayae, B. philippinensis, and B. carambolae by means of one- and two-dimensional gas chromatography-mass spectrometry, followed by multiple factor analyses and principal component analysis. Clear segregation of complex cuticule profiles of both B. carambolae sexes from B. dorsalis (Hendel) was observed. In addition to cuticular hydrocarbons, abundant complex mixtures of sex-specific oxygenated lipids (three fatty acids and 22 fatty acid esters) with so far unknown function were identified in epicuticle extracts from females of all species. The data obtained supports both taxonomic synonymization of B. invadens, B. papayae, and B. philippinensis with B. dorsalis, as well as the exclusion of B. carambolae from B. dorsalis.

[Ab] Resumo:Tephritid fruit flies are amongst the most damaging insect pests of horticulture globally. Some of the key fruit fly species are managed using the sterile insect technique (SIT), whereby millions of sterile males are released to suppress reproduction of pest populations. Male annihilation technique (MAT), whereby sex specific lures are used to attract and kill males, is often used to reduce wild male numbers before SIT programs commence, providing released sterile males an increased numerical advantage. Overall program efficacy might be improved if MAT could be deployed simultaneously with SIT, continuously depleting fertile males from pest populations and replacing them with sterile males. However, such 'male replacement' requires a means of suppressing attraction of released sterile males to lures used in MAT. Previous studies have found that exposure of some fruit flies to lure compounds as mature adults can suppress subsequent response to those lures, raising the possibility of pre-release treatments. However, this approach requires holding flies until after maturation for treatment and then release. The present study takes a novel approach of exposing immature adult male Queensland fruit flies (Bactrocera tryoni, or 'Qfly') to raspberry ketone (RK) mixed in food, forcing these flies to ingest RK at ages far younger than they would naturally. After feeding on RK-supplemented food for two days after emergence, male Qflies exhibited a reduction in attraction to cuelure traps that lasted more than 20 days. This approach to RK exposure is compatible with current practises, in which Qflies are released as immature adults, and also yields advantages of accelerated reproductive development and increased mating propensity at young ages.

[Ab] Resumo:Few efforts have been made in Mexico to monitor Rhagoletis pomonella (Walsh) (Diptera: Tephritidae) in commercial hawthorn (Crataegus spp.) crops. Therefore, the main objectives of this study were to evaluate infestation levels of R. pomonella in feral and commercial Mexican hawthorn and to assess the efficacy of different trap-lure combinations to monitor the pest. Wild hawthorn was more infested than commercially grown hawthorn at the sample site. No differences among four commercial baits (Biolure, ammonium carbonate, CeraTrap, and Captor + borax) were detected when used in combination with a yellow sticky gel (SG) adherent trap under field conditions. However, liquid lures elicited a slightly higher, although not statistically different, capture. Cage experiments in the laboratory revealed that flies tended to land more often on the upper and middle than lower-bottom part of polyethylene (PET) bottle traps with color circles. Among red, orange, green, and yellow circles attached to a bottle trap, only yellow circles improved fly captures compared with a colorless trap. A PET bottle trap with a red circle over a yellow background captured more flies than a similar trap with yellow circles. An SG adherent yellow panel trap baited with ammonium carbonate was superior to the improved PET bottle trap (red over a yellow background) baited with different liquid proteins, but a higher proportion of females and no differences in fly detection were measured in PET traps baited with protein lures. These trials open the door for future research into development of a conventional nonadherent trap to monitor or control R. pomonella.

[Ab] Resumo:Degradation models for multilure fruit fly trap dispensers were analyzed to determine their potential for use in large California detection programs. Solid three-component male lure TMR (trimedlure [TML], methyl eugenol [ME], raspberry ketone [RK]) dispensers impregnated with DDVP (2, 2-dichlorovinyl dimethyl phosphate) insecticide placed inside Jackson traps were weathered during summer (8 wk) and winter (12 wk) in five citrus-growing areas. Additionally, TMR wafers without DDVP, but with an insecticidal strip, were compared to TMR dispensers with DDVP. Weathered dispensers were sampled weekly and chemically analyzed. Percent loss of TML, the male lure for Ceratitis capitata (Wiedemann) Mediterranean fruit fly; ME, the male lure for Bactrocera dorsalis (Hendel), oriental fruit fly; RK, the male lure for Bactrocera cucurbitae (Coquillett), melon fly; and DDVP was measured. Based on regression analyses for the male lures, TML degraded the fastest followed by ME. Degradation of the more chemically stable RK was discontinuous, did not fit a regression model, but followed similar seasonal patterns. There were few location differences for all three male lures and DDVP. Dispensers degraded faster during summer than winter. An asymptotic regression model provided a good fit for % loss (ME, TML, and DDVP) for summer data. Degradation of DDVP in TMR dispensers was similar to degradation of DDVP in insecticidal strips. Based on these chemical analyses and prior bioassay results with wild flies, TMR dispensers could potentially be used in place of three individual male lure traps, reducing costs of fruit fly survey programs. Use of an insecticidal tape would not require TMR dispensers without DDVP to be registered with US-EPA.

[Ab] Resumo:The olive fruit fly Bactrocera oleae (Diptera: Tephritidae) is the most devastating pest of cultivated olive (Olea europaea L.). Intraspecific variation in plant resistance to B. oleae has been described only at phenotypic level. In this work, we used a transcriptomic approach to study the molecular response to the olive fruit fly in two olive cultivars with contrasting level of susceptibility. Using next-generation pyrosequencing, we first generated a catalogue of more than 80,000 sequences expressed in drupes from approximately 700k reads. The assembled sequences were used to develop a microarray layout with over 60,000 olive-specific probes. The differential gene expression analysis between infested (i.e. with II or III instar larvae) and control drupes indicated a significant intraspecific variation between the more tolerant and susceptible cultivar. Around 2500 genes were differentially regulated in infested drupes of the tolerant variety. The GO annotation of the differentially expressed genes implies that the inducible resistance to the olive fruit fly involves a number of biological functions, cellular processes and metabolic pathways, including those with a known role in defence, oxidative stress responses, cellular structure, hormone signalling, and primary and secondary metabolism. The difference in the induced transcriptional changes between the cultivars suggests a strong genetic role in the olive inducible defence, which can ultimately lead to the discovery of factors associated with a higher level of tolerance to B. oleae.

[Ab] Resumo:Interest in insect metamorphosis has primarily focused on juvenile hormone (JH) and ecdysone. Compared to ecdysone signaling, the molecular action of JH is less well established because Methoprene-tolerant (Met) as the JH receptor has been identified until recently. In vitro studies have indicated that Met forms an active JH-dependent complex with one partner protein, Taiman (Tai). However, the related studies on Tai's role in insect metamorphosis are very limited. In this study, five Tai isoforms differing in C-terminal region are identified from the oriental fruit fly Bactrocera dorsalis, an important worldwide pest infesting fruits and vegetables. The spatiotemporal expression pattern analysis indicates that BdTai-A and BdTai-B are highly expressed in early larvae while BdTai-D is more abundant in middle-late larvae. Meanwhile, in vivo methoprene stress leads to dramatic expression pattern fluctuation of BdTai isoforms. The subsequent reverse genetic study reveal that all Tai isoforms (denoted as "Tai-core") depletion in larvae stage of B. dorsalis produce precocious larvae-pupae development, i.e. shortened pupation process and miniature pupae. Further knockdown of individual Tai isoform show that silence of BdTai-E causes the same phenotype as of BdTai-core RNAi. The current data suggest that BdTai-E is involved in transducing the JH signal that represses metamorphosis. Besides, isoforms should be considered when studying Tai functions.
