[Ab] Resumo:Ultrasound-assisted aqueous extraction (UAAE) was used to extract oil from Clanis bilineata (CB), a traditional edible insect that can be reared on a large scale in China, and the physicochemical property and antioxidant capacity of the UAAE-derived oil (UAAEO) were investigated for the first time. UAAE conditions of CB oil was optimized using response surface methodology (RSM) and the highest oil yield (19.47%) was obtained under optimal conditions for ultrasonic power, extraction temperature, extraction time, and ultrasonic interval time at 400 W, 40°C, 50 min, and 2 s, respectively. Compared with Soxhlet extraction-derived oil (SEO), UAAEO had lower acid (AV), peroxide (PV) and p-anisidine values (PAV) as well as higher polyunsaturated fatty acids contents and thermal stability. Furthermore, UAAEO showed stronger antioxidant activities than those of SEO, according to DPPH radical scavenging and ß-carotene bleaching tests. Therefore, UAAE is a promising process for the large-scale production of CB oil and CB has a developing potential as functional oil resource.

[Ab] Resumo:Stem-borers in the genus Papaipema (Lepidoptera: Noctuidae) range from highly polyphagous agricultural pests to specialists on more than 20 families of flowering plants, many of them highly toxic. Papaipema is the largest genus of noctuids endemic to North America and provides an excellent study system for the evolution of noctuid host plant use. To improve the availability of genomic resources for such investigations, we performed de novo transcriptome sequencing and assembly for two specialist Papaipema with unusual larval hosts: P. speciosissima, which is associated with ferns, and the undescribed P. "sp. 4," which is associated with bamboo. The resulting transcriptomes were similar in terms of completeness, gene count, and gene identity, but we identified some 8,000 genes (~17% of each transcriptome) not shared between the two species. While some of these have identifiable orthologs in other Lepidoptera, ~5% of each transcriptome consists of species-specific genes. We examine the function of these genes and find that almost half have retrotransposon-related functional domains. The potential role of species-specific genes is discussed, and the expansion of certain retrotransposon families in Papaipema is examined.

[Ab] Resumo:The present study deals with glutathione-S-transferase (GST) based detoxification of pesticides in Helicoverpa armigera and its potential application in eliminating pesticides from the environment. Dietary exposure of a pesticide mixture (organophosphates - chlorpyrifos and dichlorvos, pyrethroid - cypermethrin; 2-15ppm each) to H. armigera larvae resulted in a dose dependant up-regulation of GST activity and gene expression. A variant GST from H. armigera (HaGST-8) was isolated from larvae fed with 10ppm pesticide mixture and it was recombinantly expressed in yeast (Pichia pastoris HaGST-8). HaGST-8 had a molecular mass of 29kDa and was most active at pH 9 at 30°C. GC-MS and LC-HRMS analysis validated that HaGST-8 was effective in eliminating organophosphate type of pesticides and partially reduced the cypermethrin content (53%) from aqueous solutions. Unlike the untransformed yeast, P. pastoris HaGST-8 grew efficiently in media supplemented with pesticide mixtures (200 and 400ppm each pesticide) signifying the detoxification ability of HaGST-8. The amino acid sequence of HaGST-8 and the already reported sequence of HaGST-7 had just 2 mismatches. The studies on molecular interaction strengths revealed that HaGST-8 had stronger binding affinities with organophosphate, pyrethroid, organochloride, carbamate and neonicotinoid type of pesticides. The abilities of recombinant HaGST-8 to eliminate pesticides and P. pastoris HaGST-8 to grow profusely in the presence of high level of pesticide content can be applied for removal of such residues from food, water resources and bioremediation.

[Ab] Resumo:AIM: A database based on distributional records of Eurasian Zephyrus hairstreaks (Lepidoptera: Lycaenidae: Theclini) was compiled to analyse their areas of endemism (AoEs), species richness and distribution patterns, to explore their locations of past glacial refugia and dispersal routes. METHODS: Over 2000 Zephyrus hairstreaks occurrences are analysed using the NDM/VNDM algorithm, for the recognition of AoEs. Species richness was calculated by using the option 'Number of different classes' to count the different classes of a variable presented in each 3.0°×3.0° grid cell, and GIS software was used to visualize distribution patterns of endemic species. RESULTS: Centres of species richness of Zephyrus hairstreaks are situated in the eastern Qinghai-Tibet Plateau (EQTP), Hengduan Mountain Region (HDMR) and the Qinling Mountain Region (QLMR). Latitudinal gradients in species richness show normal distribution with the peak between 25° N and 35° N in the temperate zone, gradually decreasing towards the poles. Moreover, most parts of central and southern China, especially the area of QLMR-EQTP-HDMR, were identified as AoEs that may have played a significant role as refugia during Quaternary global cooling. There are four major distributional patterns of Zephyrus hairstreaks in Eurasia: Sino-Japanese, Sino-Himalayan, high-mountain and a combined distribution covering all three patterns. CONCLUSIONS: Zephyrus hairstreaks probably originated at least 23-24 Myr ago in E. Asia between 25° N to 35° N in the temperate zone. Cenozoic orogenies caused rapid speciation of this tribe and extrusion of the Indochina block resulted in vicariance between the Sino-Japanese and the Sino-Himalayan patterns. The four distribution patterns provided two possible dispersal directions: Sino-Japanese dispersal and Sino-Himalayan dispersal.

[Ab] Resumo:Insect gut immunity plays a key role in defense against microorganism infection. The knowledge of insect gut immunity has been obtained mostly from Drosophila melanogaster. Little is known about gut immunity in the diamondback moth, Plutella xylostella (L.), a pest destroying cruciferous crops worldwide. In this study, expressions of the immune-related genes in the midgut of P. xylostella orally infected with Staphylococcus aureus, Escherichia coli and Pichia pastoris were profiled by RNA-seq and qRT-PCR approaches. The results revealed that the Toll, IMD, JNK and JAK-STAT pathways and possibly the prophenoloxidase activation system in P. xylostella could be activated by oral infections, and moricins, gloverins and lysozyme2 might act as important effectors against microorganisms. Subsequent knock-down of IMD showed that this gene was involved in regulating the expression of down-stream genes in the IMD pathway. Our work indicates that the Toll, IMD, JNK and JAK-STAT pathways may synergistically modulate immune responses in the P. xylostella midgut, implying a complex and diverse immune system in the midgut of insects.

[Ab] Resumo:PURPOSE: Glycopeptides are widely used for the treatment of meticillin-resistant Staphylococcus aureus (MRSA) infections. Although difficult to detect, isolates with reduced (GISA), hetero (hGISA) or complete (GRSA) resistance to glycopeptides are increasingly reported. Optimal therapy for such strains is unknown. We compared the in vitro and in vivo activity of tedizolid (TED), a recently licensed oxazolidonone, with vancomycin (VAN) and teicoplanin (TEIC) combined with fusidic acid (FD) or rifampicin (RIF) against S. aureus (SA) with reduced susceptibility to glycopeptides. METHODS: Susceptibility was determined for six (GISA, hGISA and GRSA) reference strains and 72 clinical MRSA isolates screened for hGISA/GISA-like phenotypes. Synergy and bactericidal activity were assessed using chequerboard and time-kill assays. The G. mellonella wax moth caterpillar model was used to measure the activity of TED and the combinations in vivo. RESULTS: Glycopeptide MICs (VAN/TEIC) ranged from 0.5-8/4 and 0.125-1 for TED. No significant synergy was noted when VAN/TEIC were combined with either RIF or FD. Time-kill assays confirmed that TED was bacteriostatic but superior to VAN and TEIC against GISA strains. In G. mellonella TED was more effective than TEIC monotherapy versus GISA strains. The combination of TEIC with RIF was the most effective combination overall, both in vitro and in vivo. CONCLUSIONS: TED had good in vitro activity versus MRSA including those with reduced susceptibility to glycopeptides. Although bacteriostatic, it was effective in the G. mellonella model and superior to TEIC in the treatment of GISA. Although this supports the use of TED for MRSA and GISA, the TEIC/RIF combination also warrants further study.

[Ab] Resumo:Experience is well known to affect sensory-guided behaviors in many herbivorous insects. Here, we investigated the effects of natural feeding experiences of Helicoverpa armigera larvae on subsequent preferences of larval approaching and feeding, as well as the effect of host-contacting experiences of mated females on subsequent ovipositional preference. The results show that the extent of experience-induced preference, expressed by statistical analysis, depended on the plant species paired with the experienced host plant. Larval feeding preference was much easier to be induced by natural feeding experience than larval approaching preference. Naïve larvae, reared on artificial diet, exhibited clear host-ranking order as follows: tobacco ≥ cotton > tomato > hot pepper. Feeding experiences on hot pepper and tobacco could always induce positive feeding preference, while those on cotton often induced negative effect, suggesting that the direction of host plant experience-induced preference is not related to innate feeding preference. Inexperienced female adults ranked tobacco as the most preferred ovipositional host plant, and this innate preference could be masked or weakened but could not be reversed by host-contacting experience after emergence.

[Ab] Resumo:Understanding interactions between herbivores and parasitoids is essential for successful biodiversity protection and monitoring and for biological pest control. Morphological identifications employ insect rearing and are complicated by insects' high diversity and crypsis. DNA barcoding has been successfully used in studies of host-parasitoid interactions as it can substantially increase the recovered real host-parasitoid diversity distorted by overlooked species complexes, or by species with slight morphological differences. However, this approach does not allow the simultaneous detection and identification of host(s) and parasitoid(s). Recently, high-throughput sequencing has shown high potential for surveying ecological communities and trophic interactions. Using mock samples comprising insect larvae and their parasitoids, we tested the potential of DNA metabarcoding for identifying individuals involved in host-parasitoid interactions to different taxonomic levels, and compared it to standard DNA barcoding and morphological approaches. For DNA metabarcoding, we targeted the standard barcoding marker cytochrome oxidase subunit I using highly degenerate primers, 2*300 bp sequencing on a MiSeq platform, and RTAX classification using paired-end reads. Additionally, using a large host-parasitoid dataset from a Central European floodplain forest, we assess the completeness and usability of a local reference library by confronting the number of Barcoding Index Numbers obtained by standard barcoding with the number of morphotypes. Overall, metabarcoding recovery was high, identifying 92.8% of the taxa present in mock samples, and identification success within individual taxonomic levels did not significantly differ among metabarcoding, standard barcoding, and morphology. Based on the current local reference library, 39.4% parasitoid and 90.7% host taxa were identified to the species level. DNA barcoding estimated higher parasitoid diversity than morphotyping, especially in groups with high level of crypsis. This study suggests the potential of metabarcoding for effectively recovering host-parasitoid diversity, together with more accurate identifications obtained from building reliable and comprehensive reference libraries, especially for parasitoids.

[Ab] Resumo:Traditionally, species of the genus Zinaida were assigned to the genus Polytremis, until molecular evidence revealed that the former is a distinct genus. Nine species in Polytremis sensu Evans have since been removed and assigned to Zinaida; however, there is still uncertainty as to the taxonomic status of an additional seven Polytremis species. Moreover, the interspecific relationships within Zinaida have remained unresolved. To further investigate the taxonomic statuses and interspecific relationships within Zinaida, a molecular phylogeny of most species of Zinaida and its allies was inferred based on regions of the mitochondrial COI-COII and 16S and nuclear EF-1α genes (3006 bp). The results revealed that Zinaida is monophyletic and consists of four intra-generic clades that correspond to morphological characteristics. Clade A (Z. suprema group) consists of P. kiraizana, Z. suprema, and P. gigantea, with the latter two as sister species. Clade B (Z. nascens group) consists of seven species, and is the sister group of Clade C (Z. pellucida group), which comprises sister species Z. pellucida and Z. zina. In Clade B, Z. caerulescens and Z. gotama, and Z. theca and Z. fukia are sister species, respectively. On the basis of our molecular evidence and morphological features, we have moved P. gigantea, P. kiraizana, P. jigongi, and P. micropunctata to the genus Zinaida as new combinations. We review morphological characteristics and discuss the distribution of each of these groups in the light of our phylogenetic hypothesis, and provide a comprehensive taxonomic checklist.

[Ab] Resumo:We analyzed the antifungal susceptibility profiles, genotypes, and virulence of clinical Aspergillus terreus isolates from six university hospitals in South Korea. Thirty one isolates of A. terreus, comprising 15 respiratory and 16 ear isolates were assessed. Microsatellite genotyping was performed, and genetic similarity was assessed by calculating the Jaccard index. Virulence was evaluated by Galleria mellonella survival assay. All 31 isolates were susceptible to itraconazole, posaconazole, and voriconazole, while 23 (74.2%) and 6 (19.4%) showed amphotericin B (AMB) minimum inhibitory concentrations (MICs) of ≤ 1 mg/L and > 4 mg/L, respectively. Notably, respiratory isolates showed significantly higher geometric mean MICs than ear isolates to AMB (2.41 vs. 0.48 mg/L), itraconazole (0.40 vs. 0.19 mg/L), posaconazole (0.16 vs. 0.08 mg/L), and voriconazole (0.76 vs. 0.31 mg/L) (all, P <0.05). Microsatellite genotyping separated the 31 isolates into 27 types, but the dendrogram demonstrated a closer genotypic relatedness among isolates from the same body site (ear or respiratory tract); in particular, the majority of ear isolates clustered together. Individual isolates varied markedly in their ability to kill infected G. mellonella after 72 h, but virulence did not show significant differences according to source (ear or respiratory tract), genotype, or antifungal susceptibility. The current study shows the marked diversity of clinical isolates of A. terreus in terms of antifungal susceptibilities, genotypes and virulence in the G. mellonella model, and ear isolates from Korean hospitals may have lower AMB or triazole MICs than respiratory isolates.
