[Ab] Resumo:Successive chromatography of EtOAc-soluble extracts of the fruiting body of Sparassis crispa (Wulf.) resulted in isolation of four new aromatic compounds, sparoside A (1) and sparalides A-C (3-5), two new naturally occurring compounds, 2 and 6, and eight known compounds, 7-14. The chemical structures were determined by interpretation of nuclear magnetic resonance and mass spectrometry spectroscopic data. Extract, solvent-soluble fractions of the extract, and all of the pure compounds isolated from the fractions were subjected to the mRNA expression assay for proprotein convertase subtilisin/kexin type 9 (PCSK9). Among them, sparoside A (1), hanabiratakelide A (8), adenosine (11), and 5α,6α-epoxy-(22E,24R)-ergosta-8(14),22-diene-3ß,7ß-diol (14) exhibited potent inhibitory activities on PCSK9 mRNA expression, with IC values of 20.07, 7.18, 18.46, and 8.23 µM, respectively (berberine, positive control, IC = 8.04 µM), suggesting that compounds 1, 8, 11, and 14 are suitable for use in supplements to the statins for hyperlipidemia treatments.

[Ab] Resumo:BACKGROUND: The aim of this study was to characterize the molecular relationship, origin and historical biogeography of the species in important brown rot fungal genus Laetiporus from East Asia, Europe, Pan-America, Hawaii and South Africa. We used six genetic markers to estimate a genus-level phylogeny including (1) the internal transcribed spacer (ITS), (2) nuclear large subunit rDNA (nrLSU), (3) nuclear small subunit rDNA (nrSSU), (4) translation elongation factor 1-α (EF-1α), (5) DNA-directed RNA polymerase II subunit 2 (RPB2), and (6) mitochondrial small subunit rDNA (mtSSU). RESULTS: Results of multi-locus phylogenetic analyses show clade support for at least seventeen species-level lineages including two new Laetiporus in China. Molecular dating using BEAST estimated the present crown group diverged approximately 20.16 million years ago (Mya) in the early Miocene. Biogeographic analyses using RASP indicated that Laetiporus most likely originated in temperate zones with East Asia and North America having the highest probability (48%) of being the ancestral area. CONCLUSIONS: Four intercontinental dispersal routes and a possible concealed dispersal route were established for the first time.

[Ab] Resumo:White rot fungi (WRF) are applicable to biodegradation of recalcitrant pollutants. However, excessive biomass growth typical for WRF cultivation can hinder their large scale applications. Therefore, immobilization of Irpex lacteus to liquid-core alginate beads restricting excessive mycelium growth and simultaneously keeping high degradation rate of pollutants was tested. Effective diffusivities of dyes to the beads varied from (2.98 ± 0.69) × 10 to (10.27 ± 2.60) × 10 m /s. Remazol Brilliant Blue R (RBBR), Reactive Orange 16 (RO16), and Naphthol Blue Black (NBB) were used as model dyes. The immobilized fungus decolorized model dyes when applied both in microwell plates and in fluidized bed reactors. Using the microwell plates, the apparent reaction rate constants ranged from (2.06 ± 0.11) × 10 to (11.06 ± 0.27) × 10 1/h, depending on the dye used and its initial concentration. High initial concentrations negatively affected the dye decolorization rate. No fungal growth outside the beads was observed in fluidized bed reactors and thus no operational problems linked to an excessive biomass growth occurred. When RBBR was decolorized in subsequent batches in the fluidized bed reactor, the apparent reaction rate constant increased from (11.63 ± 0.35) × 10 to (29.26 ± 7.19) × 10 1/h.

[Ab] Resumo:A fungus J2 producing laccase with high yield was screened in soils and identified as Abortiporus biennis. The production of laccase was induced by 0.1 mM Cu , 0.1 mM tannic acid, and 0.5 M ethanol. The laccase from Abortiporus biennis J2 was purified to electrophoretic homogeneity by a couple of steps. The N-terminal amino acid sequence of the enzyme was AIGPTADLNISNADI. The properties of the purified laccase were investigated. The result showed the laccase from Abortiporus biennis J2 is a thermo and pH stable enzyme. The laccase activity was inhibited by Hg , Cd , Fe , Ag , Cu , and Zn , while promoted by Mg , Mn at 10 mM level. Purified laccase was used to the clarification of litchi juice. After treatment with this laccase, the phenolic content of litchi juice had been found to be greatly reduced along with an increase in the clarity of the juice. The result indicated the potential of this laccase for application in juice procession.

[Ab] Resumo:Lignocellulolytic enzyme activities of selective fungi Daedalea flavida MTCC 145 (DF-2), Phlebia radiata MTCC 2791 (PR), and non-selective fungus Flavodon flavus MTCC 168 (FF) were studied for pretreatment of cotton stalks. Simultaneous productions of high LiP and laccase activities by DF-2 during early phase of growth were effective for lignin degradation 27.83 ± 1.25 % (w/w of lignin) in 20-day pretreatment. Production of high MnP activity without laccase in the early growth phase of PR was ineffective and delayed lignin degradation 24.93 ± 1.53 % in 25 days due to laccase production at later phase. With no LiP activity, low activities of MnP and laccase by FF yielded poor lignin degradation 15.09 ± 0.6 % in 20 days. Xylanase was predominant cellulolytic enzyme produced by DF-2, resulting hemicellulose as main carbon and energy source with 83 % of cellulose recovery after 40 days of pretreatment. The glucose yield improved more than two fold from 20-day DF-2 pretreated cotton stalks after enzymatic saccharification.

[Ab] Resumo:The present work constitutes a contribution to the phytochemical investigation of aiming to search for effective cytotoxic constituents against tumor cell lines in vivo. Bioassay-guided separation of the ethylacetate extract of afforded four new isocryptoporic acid (ICA) derivatives, ICA-B trimethyl ester ( ), ICA-E ( ), ICA-E pentamethyl ester ( ), and ICA-G ( ), together with nine known cryptoporic acids. These isocryptoporic acids are isomers of the cryptoporic acids with drimenol instead of albicanol as the terpenoid fragment; their structures were elucidated on the basis of spectroscopic evidences (UV, IR, HRMS, and NMR) and comparison with literature values. All isolates show certain cytotoxic activities against five tumor cell lines. Among them, compound showed an comparable activity to that of the positive control -platin, while other compounds exhibited weak cytotoxic activities.

[Ab] Resumo:This paper presents a first scale up under non-sterile conditions of the biodegradation process of 2-naphthalensulfonic acid polymers (NSAP) contained in a petrochemical wastewater by two white-rot fungi (Bjerkandera adusta and Pleurotus ostreatus). The biodegradation experiment was conducted first in flasks and then in packed-bed bioreactors filled with inert and biodegradable carriers (straw), the latter acting as both physical support and carbon source. Reactor inoculated with P. ostreatus attached on straw worked under non-sterile conditions for three months showing 30 ± 5% NSAP degradation. Respirometric tests showed that the fungal treatment was also able to significantly increase the biodegradable fraction of the wastewater COD, which rose from 9% to 40%. It was observed that the fungal degradation of the straw in the bed releases non-biodegradable by-products. Taking into account this contribution to nbCOD, the combined treatment of fungi and activated sludge could theoretically be able to reduce the original COD by up to 73%.

[Ab] Resumo:By the method of ¹H NMR prescreening and tracing the diagnostic proton signals of the methyl groups, three additional new triquinane-type sesquiterpenoids-chondrosterins K-M (1-3) and the known sesquiterpenoid anhydroarthrosporone (4)-were isolated from the marine fungus Chondrostereum sp. Their structures were elucidated on the basis of MS, 1D, and 2D NMR data. Chondrosterin K is a rare hirsutane sesquiterpenoid, in which a methyl group was migrated from C-2 to C-6 and has a double bond between C-2 and C-3. Compounds 1-3 showed significant cytotoxicities against various cancer cell lines in vitro.

[Ab] Resumo:We report for the first time the extraction, isolation, and the proposed structure of a polysaccharide from the fruiting bodies of Daedalea quercina. The monosaccharide composition of D. quercina isolate (DQW1Pa1) was mainly glucose as identified using GC-MS. FTIR-ATR spectroscopy and absolute configuration studies showed that this polysaccharide is a ß-d-glucan. Its average molecular weight obtained using size exclusion chromatography was 1.6×10 Da, consistent with glucans derived from the order Polyporaceae. MALDI-QTOF MS/MS was carried out to identify the linkage and connectivity of the glucose units. Collision Induced Dissociation (CID) of selected parent ions of different oligosaccharide lengths showed the presence of characteristic glycosidic bond cleavages B /C , the linear backbone by 1-6 linkage, and the cross-ring fragment, A . Presence of branching unit was identified from high intensity A fragment and verified from diagnostic ion of [D] and [D-H O] types. To confirm the linkage assignment obtained using MALDI-QTOF MS/MS, DQW1Pa1 was subjected to methylation analysis. Results showed the presence of 1-3, 1-6, 1- and 1-3-6 linked glucose in the order of decreasing abundance, respectively. The repeating unit of isolate DQW1Pa1 was deduced as 1-3 linked linear glucose backbone with branches composed of three 1-3 linked glucose units connected to backbone by 1-6 linkage.

[Ab] Resumo:Ceriporiopsis latemarginata and Antrodiella angulatopora are two Neotropical polypores that are very similar morphologically and are characterized by effused-reflexed basidiomes, large angular pores, small ellipsoid to ovoid basidiospores and a monomitic hyphal system with presence of thick-walled hyphae with infrequent clamps. In order to verify the evolutionary relations of C. latemarginata and to find its taxonomic placement, morphological, biological, and phylogenetic studies were carried out, as well as comparisons with Antrodiella s.s. and Ceriporiopsis s.s. Mono- and poly-sporic cultures were obtained and used for mating system and nuclear behaviour studies, as well as for DNA sequence analyses of the internal transcribed spacer and large subunit of ribosomal RNA gene, translation elongation factor 1-alpha gene and RNA polymerase II second largest subunit-like gene. Type specimens of C. latemarginata and A. angulatopora were morphologically examined and their synonymy was confirmed. The results obtained support the description of a new polypore genus Rickiopora in the residual polyporoid clade characterized by a bipolar mating system, an astatocoenocytic nuclear behaviour, and a monomitic hyphal system that display a metachromatic reaction in cresyl blue.
