[Ab] Resumo:A strictly aerobic, Gram-stain-negative, rod-shaped, motile by gliding and yellow-orange pigmented flavobacterium, designated strain 9Alg 151 , was isolated from the Pacific red alga Tichocarpus crinitus. Phylogenetic analysis based on 16S rRNA gene sequences showed that the novel strain fell into the genus Aquimarina of the family Flavobacteriaceae with a 16S rRNA gene sequence similarity range of 94.2-98.2 % to the recognized species of the genus. Strain 9Alg 151 grew in the presence of 0.5-5 % NaCl and at 5-34 °C, and hydrolysed aesculin, agar, gelatin, starch, Tween 40, DNA and chitin. The predominant fatty acids were iso-C17 : 0 3-OH, iso-C15 : 0, iso-C15 : 1 G, iso-C15 : 0 3-OH, iso-C16 : 0, iso-C17 : 1ω8c and summed feature 3. The polar lipid profile comprised phosphatidylethanolamine, three unidentified aminolipids and three unidentified lipids. The major respiratory quinone was MK-6. The genomic DNA G+C content was 32.6 mol%. On the basis of 16S rRNA gene sequence data, and chemotaxonomic and phenotypic characteristics, strain 9Alg 151 represents a novel species of the genus Aquimarina, for which the name Aquimarina algiphila sp. nov. is proposed. The type strain is 9Alg 151 (=KCTC 23622 =KMM 6462 ).

[Ab] Resumo:The multifunctional lipoxygenase PhLOX cloned from Pyropia haitanensis was expressed in Escherichia coli with 24.4 mg·L yield. PhLOX could catalyze the one-step bioconversion of C18-C22 fatty acids into C8-C9 volatile organic compounds (VOCs), displaying higher catalytic efficiency for eicosenoic and docosenoic acids than for octadecenoic acids. C20:5 was the most suitable substrate among the tested fatty acids. The C8-C9 VOCs were generated in good yields from fatty acids, e.g., 2E-nonenal from C20:4, and 2E,6Z-nonadienal from C20:5. Hydrolyzed oils were also tested as substrates. The reactions mainly generated 2E,4E-pentadienal, 2E-octenal, and 2E,4E-octadienal from hydrolyzed sunflower seed oil, corn oil, and fish oil, respectively. PhLOX showed good stability after storage at 4 °C for 2 weeks and broad tolerance to pH and temperature. These desirable properties of PhLOX make it a promising novel biocatalyst for the industrial production of volatile aroma compounds.

[Ab] Resumo:While searching for transcriptional regulators that respond to changes in light regimes, we identified a MYB domain-containing protein, MYB2, that accumulates under dark and other conditions in the unicellular red alga Cyanidioschyzon merolae. The isolation and analysis of a MYB2 mutant revealed that MYB2 represses the expression of the nuclear-encoded chloroplast RNA polymerase sigma factor gene SIG2, which results in the repression of the chloroplast-encoded phycobilisome genes that are under its control. Since nuclear-encoded phycobilisome and other light-harvesting protein genes are also repressed by MYB2, we conclude that MYB2 has a role in repressing the expression of light-harvesting genes. The MYB2 mutant is sensitive to a prolonged dark incubation, indicating the importance of MYB2 for cell viability in the dark.

[Ab] Resumo:A Gram-stain-negative, rod-shaped, motile by gliding and yellow-pigmented bacterium, designated strain 10Alg 139 , was isolated from the Pacific red alga Ahnfeltiato buchiensis. The phylogenetic analysis based on 16S rRNA gene sequences showed that the novel strain belonged to the genus Polaribacter, a member of the family Flavobacteriaceae, the phylum Bacteroidetes, with highest sequence similarity to Polaribacter butkevichii KMM 3938 (99.3 %) and 93.3-98.6 % to other recognized Polaribacter species. The prevalent fatty acids of strain 10Alg 139 were iso-C15 : 0 3-OH, C15 : 0 3-OH, iso-C15:0, iso-C13 : 0, C15 : 0 and C15 : 1ω6c. The polar lipid profile consisted of the major lipids phosphatidylethanolamine, two unidentified aminolipids and four unidentified lipids. The main respiratory quinone was menaquinone 6. The DNA G+C content of the type strain is 31.8 mol%. The new isolate and the type strains of recognized species of the genus Polaribacter were readily distinguished based on a number of phenotypic characteristics. A combination of the genotypic and phenotypic data showed that the isolate from alga represents a novel species of the genus Polaribacter, for which the name Polaribacterstaleyi sp. nov. is proposed. The type strain is 10Alg 139 (=KCTC 52773 =KMM 6729 ).

[Ab] Resumo:KEY MESSAGE: We have successfully produced single-cell colonies of C. merolae mutants, lacking the PsbQ' subunit in its PSII complex by application of DTA-aided mutant selection. We have investigated the physiological changes in PSII function and structure and proposed a tentative explanation of the function of PsbQ' subunit in the PSII complex. We have improved the selectivity of the Cyanidioschyzon merolae nuclear transformation method by the introduction of diphtheria toxin genes into the transformation vector as an auxiliary selectable marker. The revised method allowed us to obtained single-cell colonies of C. merolae, lacking the gene of the PsbQ' extrinsic protein. The efficiency of gene replacement was extraordinarily high, allowing for a complete deletion of the gene of interest, without undesirable illegitimate integration events. We have confirmed the absence of PsbQ' protein at genetic and protein level. We have characterized the physiology of mutant cells and isolated PSII protein complex and concluded that PsbQ' is involved in nuclear regulation of PSII activity, by influencing several parameters of PSII function. Among these: oxygen evolving activity, partial dissociation of PsbV, regulation of dimerization, downsizing of phycobilisomes rods and regulation of zeaxanthin abundance. The adaptation of cellular physiology appeared to favorite upregulation of PSII and concurrent downregulation of PSI, resulting in an imbalance of energy distribution, decrease of photosynthesis and inhibition of cell proliferation.

[Ab] Resumo:The geological, biological and geochemical features of a particular field of hydrothermal vents, discovered in the Panarea Volcanic Complex during a research survey carried out in 2015, are described for the first time. The site, located at 70-80 m depth off the South-western coast of the islet of Basiluzzo, was named Smoking Land for the presence of a large number of wide and high active chimneys and was characterized in terms of dissolved benthic fluxes, associated macrofauna and megafauna communities and preliminary mineralogy and geochemistry of chimney structures. On the whole field, a total of 39 chimneys, different in size and shape, were closely observed and described; 14 of them showed emission of low temperature hydrothermal fluids of marine origin characterized by acidified chemical conditions. The CTD and benthic chamber measurements highlighted that the Smoking Land is able to form a sea water bottom layer characterized by variable acidity and high DIC and trace elements concentrations; these characteristics weaken moving away from the chimney mouths. The SEM-EDS analysis of the collected solid samples revealed a chimney structure principally composed by amorphous and low crystalline Fe-oxyhydroxides of hydrothermal origins. The ROV explorations revealed a wide coverage of red algae (Peyssonnelia spp.) colonized by the green algae Flabiella petiolata and by suspension feeders, mainly sponges, but also bryozoans, and tubicolous polychaetes. Although novent-exclusive species were identified, the benthic communities found in association to the chimneys included more taxa than those observed in the surrounding no-vent rocky areas. These first findings evidence a submarine dynamic habitat where geological, chemical and biological processes are intimately connected, making the Smoking Land an important site in terms of marine heritage that should be safeguarded and protected.

[Ab] Resumo:Ten stigmastane-type steroids bearing unusual Δ -24-hydroxy side chains, dictyopterisins A-J, including three pairs of C-24 epimers, dictyopterisins B/C, F/G, and I/J, were isolated from the brown alga Dictyopteris undulata Holmes, together with two previously reported analogues, (24S)- and (24R)-saringosterol. Their structures were elucidated on the basis of extensive spectroscopic analysis, with their absolute configurations at the stereogenic center C-24 of the side chain being assigned by a direct comparison of H NMR data with those of related known compounds. The absolute configurations of the steroidal nuclei of dictyopterisins A, B, and H were determined using the modified Mosher's method. The mixture of dictyopterisins D and E and dictyopterisin I exhibited promising PTP1B inhibitory activities with IC values of 1.88 and 3.47 µM, respectively, comparable to the positive control oleanolic acid (IC , 2.78 µM). In addition, the mixture of dictyopterisins D and E and dictyopterisins F-J displayed significant cytotoxicities against the human cancer cell lines HL-60 (IC from 1.02 to 2.70 µM) and A-549 (IC from 1.35 to 2.85 µM).

[Ab] Resumo:With increasing coastal infrastructure and use of novel materials there is a need to investigate the colonisation of assemblages associated with new structures, how these differ to natural and other artificial habitats and their potential impact on regional biodiversity. The colonisation of Europe's first artificial surf reef (ASR) was investigated at Boscombe on the south coast of England (2009-2014) and compared with assemblages on existing natural and artificial habitats. The ASR consists of geotextile bags filled with sand located 220m offshore on a sandy sea bed at a depth of 0-5m. Successional changes in epibiota were recorded annually on differently orientated surfaces and depths using SCUBA diving and photography. Mobile faunal assemblages were sampled using Baited Remote Underwater Video (BRUV). Distinct stages in colonisation were observed, commencing with bryozoans and green algae which were replaced by red algae, hydroids and ascidians, however there were significant differences in assemblage structure with depth and orientation. The reef is being utilised by migratory, spawning and juvenile life-history stages of fish and invertebrates. The number of non-native species was larger than on natural reefs and other artificial habitats and some occupied a significant proportion of the structure. The accumulation of 180 benthic and mobile taxa, recorded to date, appears to have arisen from a locally rich and mixed pool of native and non-native species. Provided no negative invasive impacts are detected on nearby protected reefs the creation of novel yet diverse habitats may be considered a beneficial outcome.

[Ab] Resumo:An aerobic, Gram-stain-negative, yellow-pigmented bacterium, designated strain ATAX6-5 , was isolated from a marine red alga, Asparagopsistaxiformis, in South Korea. Cells were non-motile rods showing catalase- and oxidase-positive reactions. Growth of strain ATAX6-5 was observed at 5-35 °C (optimum, 30 °C), at pH 6.0-9.5 (optimum, pH 7.0) and in the presence of 0-6.0 % (w/v) NaCl (optimum, 2 %). Ubiquinone-10 was detected as the sole isoprenoid quinone and C18 : 1ω7c, C16 : 0 and C17 : 1ω6c were identified as the major cellular fatty acids. Diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, sphingoglycolipid, phosphatidylcholine, an unknown phospholipid and four unknown glycolipids were detected as polar lipids. The G+C content of the genomic DNA was 60.4 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain ATAX6-5 formed a tight phylogenic lineage with Parasphingopyxis lamellibrachiae JAMH 0132 with a 100 % bootstrap value. Strain ATAX6-5 was most closely related to P. lamellibrachiae JAMH 0132 with a 96.9 % 16S rRNA gene sequence similarity. Based on phenotypic, chemotaxonomic and molecular features, strain ATAX6-5 clearly represents a novel species of the genus Parasphingopyxis, for which the name Parasphingopyxis algicola sp. nov. is proposed. The type strain is ATAX6-5 (=KACC 18993 =JCM 31719 ). An emended description of the genus Parasphingopyxis is also proposed.

[Ab] Resumo:An aerobic Gram-stain-negative prosthecate bacterium, designated RMAR8-3T, was isolated from a marine red alga Rhodosorus marinus in the Republic of Korea. Cells were dimorphic rods with a single polar prostheca (non-motile) or flagellum (motile) showing catalase- and oxidase-positive reactions. Growth of strain RMAR8-3T was observed at 15-45 °C (optimum, 40 °C), at pH 6.0-9.0 (optimum, pH 7.0) and in the presence of 0-10 % (w/v) NaCl (optimum, 2 %). Ubiquinone-10 was detected as the sole isoprenoid quinone and C18 : 0, summed feature 8 (comprising C18 : 1 ω7c and/or C18 : 1ω6c), C17 : 0, C12 : 0 3-OH and C16 : 0 were identified as the major cellular fatty acids. The major polar lipids were sulfo-quinovosyldiacylglycerol, glucuronopyranosyldiglyceride and monoglycosyldiglyceride. The G+C content of the genomic DNA was 66.3 mol%. Strain RMAR8-3T was most closely related to Marinicauda pacifica P-1 km-3T with a 97.6 % 16S rRNA gene sequence similarity. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain RMAR8-3T formed a tight phylogenic lineage with M. pacifica P-1 km-3T within the family Hyphomonadaceae. On the basis of phenotypic, chemotaxonomic and molecular features, strain RMAR8-3T clearly represents a novel species of the genus Marinicauda, for which the name Marinicauda algicola sp. nov. is proposed. The type strain is RMAR8-3T (=KACC 18990T=JCM 31718T).
