[Ab] Resumo:INTRODUCTION: Aralia elata leaves are known to have several biological activities, including anti-arrythmia, antitumor, anti-inflammatory, anti-fatigue, antimicrobial and antiviral effects. Our previous study found that triterpenoid saponins from the leaves of A. elata had antitumor effects. Quantification of the triterpenoids is important for the quality control of A. elata leaves. OBJECTIVE: To establish high-performance liquid chromatography coupled with evaporative light scattering detection (HPLC-ELSD) for the simultaneous determination of four major triterpenoid saponins, including Aralia-saponin IV, Aralia-saponin VI, 3-O-ß-d- glucopyranosyl-(1 â†’ 3)-ß-d-glucopyranosyl-(1 â†’ 3)-ß-d-glucopyranosyl oleanolic acid 28-O-ß-d-glucopyranoside (Aralia-saponin TTP)and Aralia-saponin V. METHODOLOGY: The separation was carried out on a Dikma Diamonsil C column (4.6 mm × 250 mm, 5 µm) efficiently with gradient elution consisting of acetonitrile and water. RESULTS: All calibration curves showed good linear regression (R  > 0.9996) within the ranges of tested concentrations. This validated method was applied to determine the contents of the four major triterpenoid saponins in 53 samples from different regions of northeast China. Hierarchical clustering analysis was first used to classify and differentiate Aralia elata leaves. CONCLUSION: The method developed was successfully applied to analyse four major triterpenoid saponins in Aralia elata leaves which is helpful for quality control of the herb. [Ab] Resumo:A phytochemical investigation was carried out to the root of Aralia echinocauis, and a new neolignan glycoside, 3-{3,5-dimethoxy-4ß-d-Glucopyranoside-2-[3-(3-ß-d-Glucopyranoside-4-methoxy-phenyl)-allyl]-phenyl}-prop-2-en-1-ol (1), together with a known saponin, araliasaponin II (2) for the first time was obtained. The chemical structure of compound 1 was identified mainly by the analysis of NMR including 1D and 2D NMR in combination with High Resolution Electrospray Ionisation Mass (HR-ESI-MS). This paper herein describes the isolation and structural elucidation of compound 1.

[Ab] Resumo:var. is widely distributed in China and used as a traditional herbal medicine for the treatment of digestive and immune system diseases. The present study aimed to search for novel oleanolic-type triterpenoids in low-polarity fractions. Six new triterpene derivatives ( - ), together with two known compounds were isolated from the barks of var. . Their structures were elucidated by 1D- and 2D-NMR spectroscopic analysis and chemical methods. They were identified as 3-oxo-oleana-11,13(18)-dien-28,30-dioic acid ( ), 30-hydroxy-3-oxo-oleana-11,13(18)-dien-28-oic acid ( ), 3ß-hydroxy-oleana-11,13(18)-dien-28-oic acid-28- -ß-d-glucopyranoside ( ), 3ß,30-dihydroxy-oleana-11,13(18)-dien-28-oic acid-28- -ß-d-glucopyranoside ( ), 3ß-hydroxy-oleana-11,13(18)-dien-28-oic acid-3- ß-d-xylopyranosyl-(1 → 2)-ß-d-glucopyranoside ( ), 3ß,29-dihydroxy-oleana-9(11),12-dien-28-oic acid-28- -ß-d-glucopyranoside ( ), namely, araliachinolic acids I and II and araliachinosides I-IV. The cytotoxicity of the isolated compounds was tested against HepG2, A549, SGC7901, and MCF7 cell lines, but no apparent activity was observed at a concentration of 50 µM.

[Ab] Resumo:PURPOSE: Aralia elata var. mandshurica (Rupr. & Maxim.) J.Wen syn. A. mandshurica Rupr. & Maxim is evaluated for its medicinal application. The aim of this study is to analyze pharmacological studies on A. elata var. mandshurica published until December 2015. METHODS: The information regarding the chemistry, safety, effectiveness, and pharmacological and clinical effects of A. elata was systematically collected from the scientific literature through library catalogs; online services such as E-library.ru, Medline/PubMed, Scopus, Web of Science, and Google Scholar. RESULTS: A. elata is often considered an example of a medicinal plant used in Chinese, Korean, and Japanese traditional medicine. However, the contemporary applications of Aralia in officinal medicine result primarily from a large number of pharmacological and clinical investigations carried out in the former USSR in the mid-20th century. Since the 1950s, medicinal preparations from radices of A. elata and radices of A. mandshurica have secured an established position within Russian/USSR medicine as evidenced by the inclusion of the drug in recent editions of the National Pharmacopoeia of the USSR and in the Register of Medicinal Preparations of Russia. Pharmacological studies on animals have shown that Aralia increases physical working capacity and affords a stress-protective effect against a broad spectrum of harmful factors including cold stress, immobilization, UV irradiation, and low air pressure. The phytoadaptogen exerts an effect on the central nervous, reproductive, immune, respiratory, and gastrointestinal systems; the metabolic syndrome including hypolipidemic and antidiabetic effects; and blood coagulation. Together with general properties of adaptogens, Aralia has its own specificity, which manifests in cardioprotective and antiarrhythmic activities. Studies on isolated organs, cells, and enzymes have revealed that Aralia preparations exhibit antioxidant activities and enhance sarcoplasmic reticulum Ca -ATPase activity, inhibit endoplasmic reticulum stress-associated apoptosis markers (GRP78, CHOP, Caspase-12, and JNK), and increase phosphorylation of STAT3 and Bcl2/Bax ratio; they also show cytotoxic activities against some tumor cell lines; affect NF-κB and PPARs activities; and regulate biosynthesis of pro-inflammatory cytokines and inflammation-related protein expression, tissue respiration, and oxygen consumption. In healthy subjects, Aralia increases mental performance, working capacity, and endurance of movement. Numerous clinical trials have shown the efficiency of Aralia preparations in patients with traumatic brain injury (accompanied with asthenic syndrome and neurotic reactions, depression, neurasthenia, and psychasthenia), neurological diseases (accompanied with astheno-depressive and astheno-hypochondriasis syndromes), myasthenia syndrome (accompanied with chronic post-influenza arachnoiditis), and arterial hypotension. Aralia tincture and "Saparal" are useful as antiviral remedies. Radioprotective properties of Aralia have been reported in pregnant women. Synergistic antiobesity effect was reported for the combination of A. mandshurica and Engelhardtia chrysolepis extracts and antidiabetic effect for the combination of Aralia and glipizide. Promising stress-relieving effects of Aralia are reported for professionals whose work requires a high level of attention. Its proposed ability to moderate stress-induced damage and dysfunction in the cardiovascular tissue might make Aralia the adaptogen of choice among patients with higher risk for cardiovascular diseases. Because Aralia extract administration appears to affect plasma glucose level and hepatic lipid accumulation and ameliorate hyperinsulinemia, it might also provide benefits and be the adaptogen of choice for patients with obesity and diabetes. CONCLUSION: This review describes the considerable diversity of pharmacological effects of A. elata reported in numerous studies carried out in the former USSR and other countries, which have been confirmed over >47 years of use of the plant as an official medicinal remedy. The knowledge discussed in this review can be applied to the expansion of the use of this high-value plant in the pharmacotherapy of European and other countries and for the further discovery of new drugs based on the secondary metabolites of this plant. Modern approaches in mechanisms of action, including a study of gene expression profiling, suggest the most up-to-date challenges for the future research of Aralia.

[Ab] Resumo:In the present study, we isolated and characterized one purified polysaccharide (AEP-1) from the leaves of Aralia elata, and investigated its effect on human osteosarcoma (OS) U-2 OS cell line and analyzed its mechanism. MTT assays showed that AEP-1 markedly inhibited the growth of U-2 OS cells in a dose- and time-dependent fashion, suggesting a cytotoxic effect. The AEP-1 also dose-dependently induced DNA fragmentation and caused apoptotic death in U-2 OS cells. The event of apoptosis was accompanied by increased ratio of Bax/Bcl-2, depolarization of mitochondrial membrane potential (Δym) and the release of cytochrome c from mitochondria into the cytoplasm. Moreover, AEP-1 treatment triggered the activation of caspase-9 and caspase-3, as well as the cleavage of poly (ADP-ribose) polymerase (PAPR) in U-2 OS cells. All these results suggests that the induction of apoptosis by AEP-1 in U-2 OS cells occurs through the mitochondria-dependent pathway and AEP-1 may be useful in treating OS and improving cancer chemotherapy.

[Ab] Resumo:Aralia elata Seem. (A. elata) is a traditional Chinese medicine to treat some diseases. This investigation aims to evaluate the pharmaceutical safety of the ethanol extract of A. elata leaves, namely ethanol leaves extract (ELE), in Beagle dogs. In sub-chronic oral toxicity study, dogs were treated with the ELE at doses of 50, 100 and 200 mg/kg for 12 weeks and followed by 4 weeks recovery period. During experimental period, clinical signs, mortality, body temperature, food consumption and body weight were recorded. Analysis of electrocardiogram, urinalysis, ophthalmoscopy, hematology, serum biochemistry, organ weights and histopathology were performed. The results showed that both food consumption and body weight significantly decreased in high-dose group. Treatment-related side effects and mortality were observed in high-dose female dogs. Some parameters showed significant alterations in electrocardiogram, urinalysis, serum biochemistry and relative organ weights. These alterations were not related to dose or consistent across gender, which were ascribed to incidental and biological variability. The findings in this study indicated that the no-observed adverse effect level (NOAEL) of the ELE was 100 mg/kg in dogs and provided a vital reference for selecting a safe application dosage for human consumption.

[Ab] Resumo:Five new diterpenoids (1-5) were isolated from the roots of Aralia melanocarpa, together with four known compounds, 7ß-hydroxy-ent-pimara-8(14),15-diene-19-oic acid (6), 18-norpimara-8(14),15-dien-4-ol (7), ent-16ßH,17-isovalerate-kauran-19-oic acid (8), and ent-16α,17-dihydroxykauran-19-oic acid (9). Based on the MS, IR, and NMR spectral analysis, the structures of the five new diterpenoids (1-5) were elucidated. The cytotoxic activities of compounds 1-9 were assayed, and compounds 1 and 2 showed cytotoxicity in four cancer cell lines with IC50s from 4.2 to 8.2 µM.

[Ab] Resumo:In traditional Asian medicine, Aralia cordata (AC) is a known as a pain reliever and anti-inflammatory drug. Although several of its biological activities have been reported, the immunomodulatory effects of a hot water extract of AC (HAC) have not yet been described. The aim of this study was to investigate whether HAC modulates the activation of macrophages, which play important roles in innate immune responses against microbial pathogens, and if so, to determine the molecular mechanisms by which HAC mediates this process. It was found that HAC activates bone marrow-derived macrophages (BMDM) and increases amounts of nitric oxide and proinflammatory cytokines in a dose-dependent manner. In addition, HAC was found to induce phosphorylation of NF-κB and mitogen-activated protein kinases (MAPKs), including c-Jun N-terminal kinases, extracellular signal-regulated kinases and p38. Interestingly, these effects were absent in BMDM prepared from myeloid differentiation protein 88-knockout mice. Polysaccharides from HAC exerted stronger immunostimulatory effects than HAC itself. Furthermore, orally administered HAC clearly enhanced clearance of the intracellular pathogen Listeria monocytogenes by boosting innate immune responses. These results demonstrate that HAC exerts immunostimulatory effects through the TLR/MyD88 and NF-κB/MAPK signal transduction pathways.

[Ab] Resumo:Two new triterpenoids, 3ß-hydroxyoleana-11,13(18)-diene-28,30-dioic acid (1) and 3-oxooleana-11,13(18)-diene-28,30-dioic acid (2), one novel triterpenoid glycoside, 3ß-O-(6'-O-methyl-ß-D-glucuronopyranosyl)oleana-11,13(18)-dien-28-oic acid (3) along with six known compounds (4 - 9) were isolated from the stem bark of Aralia armata (Wall.) Seem. Their structures were elucidated through extensive spectroscopic methods. The herbicidal activities of these compounds against Bidens pilosa L., an invasive weed in P. R. China, were evaluated. Compounds 3, 5, and 6 exhibited more significant herbicidal activities on B. pilosa than the positive-control pendimethalin. Their possible use as herbicidal chemicals or model compounds deserved more attention. The effects of compounds 1 - 9 on Spodoptera litura cultured cell line Sl-1 cell proliferation and its morphology were also evaluated. The results indicated that compounds 1 - 5 affected Sl-1 cell proliferation. Compound 3 showed more obvious proliferation inhibition activities on Sl-1 cell than the positive-control rotenone. With regard to the effect on morphology, compound 2 significantly changed Sl-1 cell, resulting in cell blebbing and vacuole forming. Triterpenoids aremedicinally and agriculturally important, and cytotoxicity of the three new compounds 1 - 3 deserved further studies.

[Ab] Resumo:Accurate identification of the plant species that are present in herbal medicines is important for quality control. Although the dried roots of Aralia continentalis (Araliae Continentalis Radix) and Angelica biserrata (Angelicae Pubescentis Radix) are used in the same traditional medicine, namely Dok-Hwal in Korean and Du-Huo in Chinese, the medicines are described differently in the national pharmacopeia. Further confusion arises from the distribution of dried Levisticum officinale and Heracleum moellendorffii roots as the same medicine. Medicinal ingredients from all four plants are morphologically similar, and discrimination is difficult using conventional methods. Molecular identification methods offer rapidity and accuracy. The internal transcribed spacer 2 (ITS2) region of the nuclear ribosomal RNA gene (rDNA) was sequenced in all four plant species, and the sequences were used to design species-specific primers. Primers for each species were then combined to allow sample analysis in a single PCR reaction. Commercial herbal medicine samples were obtained from Korea and China and analyzed using the multiplex assay. The assay successfully identified authentic medicines and also identified inauthentic or adulterated samples. The multiplex assay will be a useful tool for identification of authentic Araliae Continentalis Radix and/or Angelicae Pubescentis Radix preparations in Korea and China.
