[Ab] Resumo:Saussurea involucrata Kar. et Kir. is a hardy, dicotyledonous plant featured by strong tolerance to severe stress conditions. In a previous study, we created a cDNA library to characterize genetic factors associated with the cold acclimation response of S. involucrata. The full-length cDNA of one dehydrin-like gene, SiDHN, was obtained by RT-PCR and the SiDHN gene was further characterized in this study. The full-length SiDHN cDNA consists of 703 base pairs (bp) with a 333bp open reading frame encoding a protein comprising 111 amino acid residues. The alignment of the deduced amino acid sequence showed that SiDHN protein shared 36% identity with one homolog in Helianthus annuus. To evaluate its biological functions, 35S:SiDHN recombinant plasmid was introduced into tobacco using Agrobacterium tumefaciens and the resistance of transgenic plants to freezing and drought stresses were analyzed. Compared with the wild-type, transgenic tobacco plants showed greater resistance to freezing and drought stresses. In this study, we provided evidence that SiDHN can enhance plant cold and drought resistance, suggesting that SiDHN could be potentially used to genetically improve plant tolerance to abiotic stresses.

[Ab] Resumo:ETHNOPHARMACOLOGICAL RELEVANCE: The root of Saussurea costus (Aucklandia lappa Decne, Aucklandiae Radix, SC) and Thuja orientalis L. (TOL) have been traditionally used as anti-inflammatory agents in Korea. However, they have not been studied for the efficacy of atopic dermatitis (AD) treatment, a chronic inflammatory skin disease. We investigated the efficacy of topical applications with 1,3-butyleneglycol extracts of SC and TOL to alleviate the symptoms of AD. MATERIALS AND METHODS: HaCaT cells and the dorsal skin of Nc/Nga mice had a local exposure of house mite extracts and 2,4-dinitrochlorobenzene (DNCB), respectively. After lesions developed, we topically applied 1,3-butylen glycol (vehicle; control), SC (30%), TOL (30%), or SC (15%)+TOL (15%) to the skin lesions for 5 weeks. The normal-control was not exposed to DNCB. The skin thickness, mast cell infiltration, serum immunoglobulin E (IgE) and IgG1 and gene expressions of interleukin (IL)-4, IL-13, and IFN-γ in the dorsal skin and HaCaT cells were measured. RESULTS: Chlorogenic acid (129.6±10.2µg/g) for SC and catechin and apigenin (93.4±13.2 and 16.9±1.3µg/g, respectively) for TOL were used as indicator compounds for the strength of the extracts. SC+TOL decreased the expression of protease-activated receptor-2 and ICAM-1 and the release of TNF-α and IL-6 in HaCaT cells activated by 3µg/mL house mite extracts in comparison to either of SC or TOL alone. In Nc/Nga mice challenged with DNCB, SC+TOL synergistically attenuated clinical symptoms of AD such as erythema, hemorrhage, edema, excoriation and dryness in the dorsal skin better than either SC or TOL alone. Histological analysis of the dorsal skin also showed that SC+TOL treatment significantly and additively decreased the inflammatory cellular infiltrate, including mast cells and eosinophils in comparison to either of SC or TOL. SC+TOL also decreased serum IgE and IgG1 levels and the expression of IFN-γ, IL-4, and IL-13 mRNA in dorsal skin in DNCB-treated Nc/Nga mice. CONCLUSION: SC+TOL relieved the symptoms of AD by reducing pro-inflammatory activity and over-activated immune responses. These data suggest that SC+TOL may be an effective alternative intervention for the management of AD.

[Ab] Resumo:Hispidulin is a naturally occurring flavonoid isolated from a traditional Chinese medicinal herb, Saussurea involucrata. In this study, the regulating role of hispidulin on the mRNA expression level of enzymes involved in lipid metabolism was examined in vitro and in vivo. Moreover, the in vivo lipid-modulating effect of hispidulin was compared with that of fenofibrate, a classical PPARα agonist. Our results in present study demonstrated that hispidulin can directly bind to and activate PPARα as an agonist and thus modulate the downstream lipid-metabolizing genes. Moreover, hispidulin could attenuate dyslipidemia in high fat diet induced dyslipidemia rat model. Although further studies are needed, this study provided evidence for the potential use of hispidulin in dyslipidemia management.

[Ab] Resumo:Aucklandialappa Decne (ALD) is one of the traditional herbs to treat various kinds of disorders including asthma, cough, vomit, diarrhea, hepatitis and cholecystitis. However, its effects on indigestion and particularly antiulcer activity of ethanol extract have not been studied. In the study, the Aucklandia lappa Decne extract (ALDE) was investigated to see if it againstgastric injury effects through traditional pathways. Ethyl alcohol and epinephrine hydrochloride were used to induce acute gastric mucous membrane damage in adult SD rats and Kunming mice, respectively. This present study evaluated its effects on peptic ulcer of ALDE treatment in SD rats and Kunming mice. In acute gastric mucous membrane damage induced by ethyl alcohol in rats, the results indicated that three ALDE treatment groups highly significantly decreased the mucosal damage index as compared to the model group. Furthermore, this mucosal damage index of the mid-dose group significantly decreased while the high-range dose group highly significantly decreased, respectively, as compared to the SO group. The ulcer inhibition rate of low -dose, mid-dose and high-dose ALDE treatment groups reached 68.64%, 72.67% and 74.91%, respectively. In acute gastric mucous membrane damage induced by pyloric ligation in rats, the results indicated that three ALDE treatment groups highly significantly decreased the mucosal damage index as compared to the model group. The mucosal damage index of middose group significantly decreased while the high-range dose group highly significantly decreased, respectively as compared to the SO group. The ulcer inhibition rate of low-dise, mid-dose and high-dose ALDE treatment groups reached 68.64%, 72.67% and 74.91%, respectively. In acute gastric mucous membrane damage induced by pyloric ligation in rats, the results indicated that three ALDE treatment groups highly significantly decreased the mucosal damage index of, respectively, as compared to the model group. Furthermore, this mucosal damage index of the midrange dose group significantly decreased while the high-dose group highly significantly decreased, respectively, as compared to the SO group. The ulcer inhibition rate of low-dose, mid-dose and high-dose ALDE treatment groups reached 68.64%, 72.67% and 74.91%, respectively. Our results indicated that ALDE exhibits a marked effect on peptic ulcer activity in animals, which supports previous results of its use in traditional Chinese medicine.

[Ab] Resumo:ETHNOPHARMACOLOGICAL RELEVANCE: Saussurea involucrate (Kar. et Kir.) Sch. -Bip is an endangered species of the Compositae family, and this species has long been used for the treatment of rheumatoid arthritis, dysmenorrhea, stomachache, and altitude sickness in Uighur folk and Chinese medicine. AIM OF THE STUDY: This study aimed to investigate the different therapeutic efficacy of alcohol infusion (SEI) and water decoction (SWD) of S. involucrata in treating rheumatoid arthritis using complete Freund's adjuvant (CFA)-induced arthritis (AA) in a rat model. MATERIALS AND METHODS: Male Wistar rats (160-180g) were immunized by intradermal injection of 0.1mL of CFA into the left hind metatarsal footpad. Aspirin was chosen as the positive drug. Alcohol infusion (SEI, 400 and 800mg/kg) and water decoction (SWD, 443 and 886mg/kg) of S. involucrata aerial parts were orally administered from day 1 and continued for 21 days. Arthritis severity was evaluated by arthritic score, body weight loss, and paw swelling. The levels of TNF-α, IL-ß, and IL-6 in the serum of AA rats were detected by enzyme linked immunosorbent assay (ELISA). Histological changes in the ankle joint were also analyzed in the AA rats. RESULTS: Both SEI and SWD significantly ameliorated AA severity, as suggested by the modulatory effects on body weight loss, paw swelling, and arthritic score. Histopathological improvement in the joint architecture was also observed in the SEI- and SWD-treated AA rats. The overproduction of TNF-α, IL-1ß, and IL-6 was remarkably attenuated in the serum of all treated rats. Furthermore, the therapeutic effect of SWD was more potent than that of SEI in treating rheumatoid arthritis using AA in a rat model, which was reported for the first time. CONCLUSIONS: These results suggested that the extract of S. involucrata significantly attenuated adjuvant arthritis in rats by decreasing the levels of TNF-α, IL-1ß, and IL-6 in the serum. S. involucrata has the potential to be regarded as a candidate for the treatment of human arthritis. Moreover, the therapeutic effect of SWD was much better than alcohol infusion, indicting that active constituents are mainly in the water extract, which is helpful for the clinical treatments to choose the appropriate process.

[Ab] Resumo:In a screen for mammalian thioredoxin reductases inhibitors, an MeOH extract from the roots of Saussurea lappa C.B. Clarke (Compositae) inhibited the activity of cytosolic thioredoxin reductase (TrxR1). Bioassay-guided separation of the extract led to the isolation of a new TrxR1 inhibitor, dehydrocostus lactone (DHC), a guaiane-type sesquiterpene. The content of DHC in the extract was determined to be 0.4%. DHC inhibited human cervical carcinoma HeLa cells with an IC50 of ∼12.00 µM but displayed less cytotoxicity to human immortalized normal liver cells L02. We observed that DHC killed HeLa cells through induction of apoptosis. DHC inhibited the activity of TrxR1 in HeLa cells, which elicited an accumulation of reactive oxygen species (ROS) in cells and a collapse of the intracellular redox equilibrium and eventually induced apoptosis of HeLa cells.

[Ab] Resumo:As part of pharmacological-phytochemical integrated studies on medicinal plants from Indian flora, costunolide (1) and dehydrocostus lactone (2), were isolated as major phytochemicals from Saussurea lappa, a plant traditionally used in different Asian systems of medicine. A series of 1,4-disubstituted-1,2,3-triazoles conjugates were synthesized through diastereo selective Michael addition followed by regioselective Huisgen 1,3-dipolar cycloaddition reactions. All these triazolyl derivatives (5a-5j) & (7a-7j) were well characterized using modern spectroscopic techniques and evaluated for their anticancer activity against a panel of five human cancerous celllines. The results indicated that all the analogs displayed moderate cytotoxic activity.

[Ab] Resumo:Upward migration of plants to barren subnival areas is occurring worldwide due to raising ambient temperatures and glacial recession. In summer 2012, the presence of six vascular plants, growing in a single patch, was recorded at an unprecedented elevation of 6150 m.a.s.l. close to the summit of Mount Shukule II in the Western Himalayas (Ladakh, India). Whilst showing multiple signs of stress, all plants have managed to establish stable growth and persist for several years. To learn about the role of microbes in the process of plant upward migration, we analysed the root-associated microbial community of the plants (three individuals from each) using microscopy and tagged amplicon sequencing. No mycorrhizae were found on the roots, implying they are of little importance to the establishment and early growth of the plants. However, all roots were associated with a complex bacterial community, with richness and diversity estimates similar or even higher than the surrounding bare soil. Both soil and root-associated communities were dominated by members of the orders Sphingomonadales and Sphingobacteriales, which are typical for hot desert soils, but were different from communities of temperate subnival soils and typical rhizosphere communities. Despite taxonomic similarity on the order level, the plants harboured a unique set of highly dominant operational taxonomic units which were not found in the bare soil. These bacteria have been likely transported with the dispersing seeds and became part of the root-associated community following germination. The results indicate that developing soils act not only as a source of inoculation to plant roots but also possibly as a sink for plant-associated bacteria.

[Ab] Resumo:A new sesquiterpenoid, 10α-methoxyartemisinic acid (1) was isolated from Saussurea lappa roots along with two known compounds, costunolide (2) and dehydrocostus lactone (3) by high-speed countercurrent chromatography with purities of 85, 95 and 98%, respectively. Their structures were determined on the basis of spectral data including 1D and 2D NMR and HREIMS.
