[Ab] Resumo:Cynomoriaceae, one of the last unplaced families of flowering plants, comprise one or two species or subspecies of root parasites that occur from the Mediterranean to the Gobi Desert. Using Illumina sequencing, we assembled the mitochondrial and plastid genomes as well as some nuclear genes of a Cynomorium specimen from Italy. Selected genes were also obtained by Sanger sequencing from individuals collected in China and Iran, resulting in matrices of 33 mitochondrial, 6 nuclear, and 14 plastid genes and rDNAs enlarged to include a representative angiosperm taxon sampling based on data available in GenBank. We also compiled a new geographic map to discern possible discontinuities in the parasites' occurrence. Cynomorium has large genomes of 13.70-13.61 (Italy) to 13.95-13.76 pg (China). Its mitochondrial genome consists of up to 49 circular subgenomes and has an overall gene content similar to that of photosynthetic angiosperms, while its plastome retains only 27 of the normally 116 genes. Nuclear, plastid and mitochondrial phylogenies place Cynomoriaceae in Saxifragales, and we found evidence for several horizontal gene transfers from different hosts, as well as intracellular gene transfers.

[Ab] Resumo:As a great deal of interest is developed to study novel bioactive components with health benefit effects from natural resources, in this paper, a rat pheochromocytoma line 12 (PC12) cell is built to observe the protective effect of a Cynomorium songaricum Rupr. polysaccharide (CSP) against H2O2-induced oxidative stress. Fluorescence microscope, flow cytometry and micro-plate reader are used to assess cell viability and apoptosis. And the levels of reactive oxygen species (ROS), 8-hydroxy-2'-deoxyguanosine (8-OH-dG), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and lactate dehydrogenase (LDH) are evaluated. The results show that, the CSP can significantly protect PC12 cells against H2O2-induced oxidative stress, increase the intracellular antioxidase system load and inhibit H2O2-induced apoptosis by scavenging of ROS, regulating cell cycle, preventing DNA damage and protecting the cell membrane. This research would be benefit for preventing and curing the oxidation-related diseases in polysaccharide study.

[Ab] Resumo:The plant, Cynomorium songaricum Rupr., is used as a traditional medicine in China and Mongolia. In the present study, two new water-soluble polysaccharides isolated from C. songaricum Rupr. were purified by successive Sephadex G-75 and G-50 column chromatographies and then characterized by high resolution NMR and IR spectroscopies. The molecular weights of two polysaccharides were determined by an aqueous GPC to be [Formula: see text] = 3.7 × 10(4) and 1.0 × 10(4), respectively. In addition, it was found that the polysaccharide with the larger molecular weight was an acidic polysaccharide. It was found that the iodine-starch reaction of both isolated polysaccharides was negative and the methylation analysis gave 2, 4, 6-tri-O-methyl alditol acetate as a main product. NMR and IR measurements and sugar analysis revealed that both polysaccharides had a (1 â†’ 3)-α-d-glucopyranosidic main chain with a small number of branches. After sulfation, the sulfated C. songaricum Rupr. polysaccharides were found to have a potent inhibitory effect on HIV infection of MT-4 cells at a 50% effective concentration of 0.3-0.4 µg/ml, a concentration that has almost the same high activity as standard dextran and curdlan sulfates, EC50 = 0.35 and 0.14 µg/ml, respectively. The 50% cytotoxic concentration was low, CC50>1000 µg/ml. In addition, the interaction between the sulfated polysaccharides and poly-l-lysine as a model protein compound was investigated by a surface plasmon resonance to reveal the anti-HIV mechanism.

[Ab] Resumo:In this study, highly selective core-shell molecularly imprinted polymers on the surface of magnetic nanoparticles were prepared using protocatechuic acid as the template molecule. The resulting magnetic molecularly imprinted polymers were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and vibrating sample magnetometry. The binding performances of the prepared materials were evaluated by static and selective adsorption. The binding isotherms were obtained for protocatechuic acid and fitted by the Langmuir isotherm model and Freundlich isotherm model. Furthermore, the resulting materials were used as the solid-phase extraction materials coupled to high-performance liquid chromatography for the selective extraction and detection of protocatechuic acid from the extracts of Homalomena occulta and Cynomorium songaricum with the recoveries in the range 86.3-102.2%.

[Ab] Resumo:The present study aimed to examine the potential anticancer properties of fixed oil obtained from Maltese mushroom (Cynomorium coccineum L.), an edible, non-photosynthetic plant, used in traditional medicine of Mediterranean countries to treat various ailments and as an emergency food during the famine. We investigated the effect of the oil, obtained from dried stems by supercritical fractioned extraction with CO2, on B16F10 melanoma and colon cancer Caco-2 cell viability and lipid profile. The oil, rich in essential fatty acids (18:3n-3 and 18:2n-6), showed a significant growth inhibitory effect on melanoma and colon cancer cells. The incubation (24 h) with non-toxic oil concentrations (25 and 50 µg/mL) induced in both cancer cell lines a significant accumulation of the fatty acids 18:3n-3 and 18:2n-6 and an increase of the cellular levels of eicosapentaenoic acid (20:5n-3) with anticancer activity. Moreover, the oil exhibited the ability to potentiate the growth inhibitory effect of the antitumor drug 5-fluorouracil in Caco-2 cells and to influence the melanin content in B16F10 cells. The results qualify C. coccineum as a resource of oil, with potential benefits in cancer prevention, for nutraceutical and pharmaceutical applications.

[Ab] Resumo:Cynomorium coccineum L. is a non-photosynthetic plant, spread over Mediterranean countries, amply used in traditional medicine. The aim of this study was to evaluate for the first time the antifungal activity of its extracts. The antifungal activity was evaluated using the macrodilution method against Candida spp., Cryptococcus neoformans and dermatophyte strains. The methanolic extract was very active against C. neoformans, Candida guilliermondii and Candida krusei, with minimal inhibitory concentrations (MIC) values of 0.025 mg/mL. This extract is more active than fluconazole against C. krusei H9. The influence of methanolic extract on the dimorphic transition in Candida albicans was also studied through the germ tube inhibition assay. More than 60% of filamentation was inhibited at a concentration of 1/4 MIC. These results are preliminary and further studies are needed to an eventual use of C. coccineum methanolic extract in the treatments of candidiasis and cryptococcosis.

[Ab] Resumo:BACKGROUND: Cynomorium songaricum Rupr. (CS) has been used as a medicine to treat many diseases as well as to alleviate age-related issues, such as memory impairment, dementia, and stress. In this study, we assessed the effects of Cynomorium songaricum extract (CSE) on the novel object recognition, cell proliferation and neuroblast differentiation in the dentate gyrus of mice by using 5-bromodeoxyuridine (BrdU) and polysialylated neural cell adhesion molecule (PSA-NCAM). We also measured serum corticosterone levels to assess its correlation with neurogenesis and stress. METHODS: Male C57BL/6 J mice were divided into 3 groups: vehicle-treated, 40 mg/kg CSE-treated, and 100 mg/kg CSE-treated. The vehicle and CSE were given to mice once a day for 3 weeks. BrdU was injected twice a day for 3 days to label newly generated cells. RESULTS: Administration of CSE significantly increased the preferential exploration of new objects in these mice. In addition, administration of CSE decreased serum levels of corticosterone. BrdU-positive cells as well as brain-derived neurotrophic factor (BDNF) mRNA expression in the dentate gyrus were higher in the CSE-treated groups than in the vehicle-treated group. PSA-NCAM-positive neuroblasts and their well-developed tertiary dendrites were also significantly increased by the treatment of CSE. These effects were prominent at the higher dosage than at the lower dosage. CONCLUSION: These results suggest that administration of CSE increases cell proliferation and neuroblast differentiation in the dentate gyrus of mice by reducing serum corticosterone levels and increasing BDNF levels in this area.

[Ab] Resumo:OBJECTIVE: To identify the Cynomorii Herba and its analogues species using DNA barcoding technique. METHOD: Total genomic DNA extracted from all materials using the DNA extraction kit. The internal transcribed spacer 2 (ITS2) regions were amplified using polymerase chain reaction (PCR), and purified PCR products were sequenced bi-directionally. Sequence assembly and consensus sequence generation were performed using the CodonCode Aligner 3.7.1. The Kimura 2-Parameter (K2P) distances and GC content were computed using MEGA 5. 0. Species identification analyses were conducted through the species identification system for traditional Chinese medicine and neighbor-joining (NJ) trees. RESULT: The ITS2 sequence lengths of Cynomorii Herba were 229 bp. The average intra-specific genetic distances of Cynomorii Herba were 0.003. The average inter-specific genetic distances between Cynomorii Herba and its adulterants species were 0.760. The results showed that the minimum inter-specific divergence is larger than the maximum intra-specific divergence. The species identification system for traditional Chinese medicine and NJ trees results indicated that Cynomorii Herba and its adulterants species can be easily identification. CONCLUSION: The ITS2 region is an efficient barcode for identification of Cynomorii Herba, which provide a new technique to ensure clinical safety in utilization of traditional Chinese medicine.

[Ab] Resumo:In natural conditions, fully ripe Cynomorium songaricum seeds parasitize in Nitraria tangutorum or N. sphaerocarpa or N. sibirica or Zygophyllum xanthoxylom and Peganum harmala, were used in this study to research the morphological characteristics, embryo rate, seed viability, 1 000-grain weight, purity, water content and the seeds of different host parasitic relationship with each other. The results showed that the morphology, color and surface characteristics of the C. songaricum seeds are very similar in different hosts. According to the seed morphology can not be judged on its host. For the host to N. tangutorum or Peganum harmala or N. sibirica, we should choose the round hole screen less than 0.923 1 mm and larger than 1.066 2 mm to cleaning seeds. For the C. songaricum seeds parasitic in N. sphaerocarpa, the choice of slightly less than 0.926 1 mm and larger than 0.985 3 mm round hole screen to cleaning. For the parasitic seeds in Z. xanthoxylom, less than 0.751 3 mm and slightly larger than 1.035 3 mm round hole screen could be used. Highy significant correlation was found among the morphological indexes in C. songaricum seeds (P < 0.01). Morphological indexes and 1 000-grain weight were significantly correlated (0.01 < P < 0.05), but with the seed viability and the embryo rate were not found significant correlation. Grain weight is not related with the seed viability and the Fully mature C. songaricum seed viability is high and water content is low. The difference of the habitats and the host plants should be considered in the seed quality assessment and classification. The C. songaricum seeds on host plants are not selective, and the C. songaricum seeds from the host plants could be parasitized in other host plants.

[Ab] Resumo:Erectile dysfunction (ED) is one of the major health concerns affects the quality of life among Thai male. The treatment of ED by the first-line drugs is limited to a certain group of patients due to their side effects and costs. Alternative medicine can be beneficial for the treatment of ED. This is a randomized, double-blind, placebo-controlled, crossover study aimed to assess the efficacy and safety of Cappra(®), a traditional herbal medicine which was used in Thailand for decades, for the treatment of mild and mild to moderate ED in Thai patients. A total of 63 patients with mild or mild to moderate ED were randomized to receive Cappra(®) or placebo for two weeks in the first period, followed by one week washout period. The patients were switched to the alternative treatment in the second period. The efficacy was assessed by the International Index of Erectile Function (IIEF) questionnaire and adverse events. Sixty one patients completed the study. There was an improvement of IIEF score for all domains in Cappra(®) group compared with placebo group. The mean change of IIEF score from baseline for erectile function domain of Cappra(®) was significantly higher than placebo (4.87 vs 3.44, p = 0.032). The most common adverse events were dizziness (13.3% Cappra(®), 9.6% placebo), face numbness (1.6% Cappra(®), 0% placebo), and tachycardia (1.6% Cappra(®), 0% placebo). The results from this study demonstrated that Cappra(®) is effective and well-tolerated and can be used as alternative therapy for mild and mild to moderate ED.
