[Ab] Resumo:Ribosome-inactivating proteins (RIPs) are toxic enzymes that are mostly biosynthesized by plants. RIPs are N-glycosidases that cleave an essential adenine molecule from the 28S rRNA. This is followed by the irreversible inhibition of protein synthesis leading to cell death. By fusing RIPs to cancer cell specific targeting ligands RIPs have been utilized for targeted anti-tumor therapy. The anti-tumoral efficiency of such conjugates depends significantly on the N-glycosidase activity of the RIP domain. Different methods have been developed in order to determine the N-glycosidase activity of RIPs and RIP domain containing anti-tumor toxins. However the existing methods are elaborate and include radioassays, HPLC and enzymatic conversion assays. Here, a simple and cost effective N-glycosidase assay is presented, which is based on the direct determination of the released adenine by thin-layer chromatography (TLC) and TLC-densitometry. An adenine based single stranded oligonucleotide is used as substrate. Following TLC development the released adenine is quantified on silica glass plates by UV absorbance at 260nm.

[Ab] Resumo:A biological method for synthesising silver nanoparticles (AgNPs) was developed using the callus extracts from L. under sunlight at 25,000 lx. The AgNPs were characterised using transmission electron microscopy, atomic force microscope, X-ray diffraction and Fourier transform infrared spectroscopy. The AgNPs were mostly spherical with the size of 2.1 to 45.2 nm (average 10.9 nm). Pulse treatments of AgNPs at 125, 250 and 500 mg/l for 1 h extended vase life of cut carnation ( cv. Green Land) flowers. Four dominant bacteria strains sp., and were isolated from the stem-ends of cut flowers. AgNP pulse inhibited significantly bacterial growth in vase solution and cut stem ends during all of the vase period. The bacteria related blockage in the stem-ends was significantly alleviated by AgNP pulse because of its higher antibacterial efficacy against the dominant bacteria. In addition, ethylene release of cut carnation flowers was inhibited in response to AgNP pulse. This is the first time that the biologically synthesised AgNPs could be applied as a promising preservative agent for cut carnation flowers.

[Ab] Resumo:, a perennial herbaceous flower and a member of the Caryophyllaceae family, has strong resistance to cold and drought stresses. To explore the transcriptional responses of to individual and combined stresses, we performed transcriptome sequencing of seedlings under normal conditions or subjected to cold treatment (CT), simulated drought treatment (DT), or their combination (CTDT). After de novo assembly of the obtained reads, 112,015 unigenes were generated. Analysis of differentially expressed genes (DEGs) showed that 2026, 940, and 2346 genes were up-regulated and 1468, 707, and 1759 were down-regulated in CT, DT, and CTDT samples, respectively. Among all the DEGs, 182 up-regulated and 116 down-regulated genes were identified in all the treatment groups. Analysis of metabolic pathways and regulatory networks associated with the DEGs revealed overlaps and cross-talk between cold and drought stress response pathways. The expression profiles of the selected DEGs in CT, DT, and CTDT samples were characterized and confirmed by quantitative RT-PCR. These DEGs and metabolic pathways may play important roles in the response of to the combined stress. Functional characterization of these genes and pathways will provide new targets for enhancement of plant stress tolerance through genetic manipulation.

[Ab] Resumo:Host specialization has important consequences for the diversification and ecological interactions of obligate pathogens. The anther-smut disease of natural plant populations, caused by Microbotryum fungi, has been characterized by specialized host-pathogen interactions, which contribute in part to the isolation among these numerous fungal species. This study investigated the molecular variation of Microbotryum pathogens within the geographic and host-specific distributions on wild Dianthus species in southern European Alps. In contrast to prior studies on this pathogen genus, a range of overlapping host specificities was observed for four delineated Microbotryum lineages on Dianthus hosts, and their frequent co-occurrence within single-host populations was quantified at local and regional scales. In addition to potential consequences for direct pathogen competition, the sympatry of Microbotryum lineages led to hybridization between them in many populations, and these admixed genotypes suffered significant meiotic sterility. Therefore, this investigation of the anther-smut fungi reveals how variation in the degrees of host specificity can have major implications for ecological interactions and genetic integrity of differentiated pathogen lineages.

[Ab] Resumo:Recently, the investigation of bioactive constituents from natural products has been performed extensively around the world, yet many plants remain to be studied, even now. The author is especially interested in saponins, which are included in various natural products and Kampo medicines. Based on these study aims, we isolated the chemical constituents of Dianthus japonicus and Clethra barbinervis. The aerial parts of Dianthus japonicus were isolated into nine new (1-9) and seven known oleanane-type triterpene saponins. The structural elucidation of their chemical constituents was examined by several spectroscopic methods. Two new C-glycosyl flavones, two glycosidic derivative of anthranilic acid amides and a maltol glucoside were also isolated from this plant. The isolated compounds were evaluated according to cytotoxic inhibition toward A549 cell lines. Compounds 7-9 showed moderate inhibitory activity. The isolation of a MeOH extract of Clethra barbinervis leaves led to three new triterpene glucosides, i.e. an ursane (ryobunin A, 22), a seco-ursane (ryobunin B, 23) and an oleanane-type glucoside (ryobunin C, 24), along with four known compounds.

[Ab] Resumo:Zinc-lead mining wastes remain largely unvegetated and prone to erosion for many years because of phytotoxic levels of residual heavy metals, low nutrient status and poor physical structure. The optimal solution for these areas is to restore plant cover using species which spontaneously appear on the spoils. These species are adapted to the conditions of tailings, and their establishment will promote further vegetation by increasing soil organic matter and development of a soil system capable of supporting the nutrient and water requirements of plants and microoorganisms. The potential of Dianthus carthusianorum and Biscutella laevigata to stabilize mine spoils was analysed in a three-year pot experiment. Post-flotation wastes accumulated after Zn and Pb recovery from ores, were collected from tailings and used as a substrate for plant growth. Seeds for seedling production were collected from plants growing spontaneously on mine tailings. Prior to the establishment of the three-year pot experiment, the substrate was amended with fertilizer NPK or municipal sewage sludge, supplemented with K O (SS). Substrate samples were collected for chemical analyses, dehydrogenase and urease activities measurements each year at the end of the growing season. The plants were harvested three years after the amendments. Both tested plant species were equally suitable for revegetation of the tailings. The amendment including both SS and NPK resulted in an increase of C , N , available P, K, Mg contents, an increase of dehydrogenase (DHA) and urease activities and a decrease in the concentrations of the soluble forms of Zn, Pb and Cd. However, nutrient content, DHA activity and plant biomass were higher with SS than NPK addition. NPK application enhanced the substrate properties after the first growing season, while positive effects of SS use were still observed after three years. A longer-lasting positive effect of SS than NPK application was probably due to the high organic matter content in SS, which was gradually decomposing and releasing nutrients.

[Ab] Resumo:Hyperhydricity occurs frequently in plant tissue culture and can severely affect commercial micropropagation and genetic improvement of the cultured plantlets. Hyperhydric shoots are charaterzized by high water content, but how this occurs is still a subject of investigation. Silver ion (Ag ) can reduce the extent of hyperhydricity in plants, but its effect on the reversion of hyperhydric plantlets and the underlying mechanism of reversion has not been clarified. In this study, about 67% of the hyperhydric Dianthus chinensis L. plantlets were found to revert to normal condition when the plantlets were cultured in medium supplemented with 29.4µmolL AgNO . Water content and hydrogen peroxide (H O ) content in the guard cells of these plantlets were reduced, while stomatal aperture and water loss rate were increased. AgNO also reduced the content of endogenous ethylene and expression of ethylene synthesis and ethylene signal transduction-associated genes. Reduced accumulation of ethylene consequently led to an increase in stomatal aperture mediated by decreased H O content in the guard cells. These results adequately verified the role of AgNO in the reversion of hyperhydricity in D. chinensis L. and also provided clues for exploring the cause of excessive water accumulation in hyperhydric plants.

[Ab] Resumo:The aim of the study was to investigate volatile compounds from the aerial parts of Dianthus acicularis of the genus Dianthus of the family Caryophyllaceae grown wild in Northern Kazakhstan for the first time. D. acicularis is a typical Trans-Volga-Kazakhstani endemic. D. acicularis has high resistance to the bacterial wilt, a serious disease caused by Burkholderia caryophylli. The qualitative and quantitative compositions of the specimens of the essential oils were analysed by the method of GC-MS. The main constituents of D. acicularis essential oil were methyl ketones - 2-pentadecanone (26.9-32.2%) and 2-tridecanone (4.7-17.7%), identified for the first time in the Dianthus genus. The methyl ketone activity provides protection of the plants from herbivores and fungal pathogens. One can suppose that the presence of 2-pentadecanone and 2-tridecanone in the essential oil of carnation coniferous provides its resistance to different insects and pathogens, including the resistance to the bacterial wilt.

[Ab] Resumo:One new oleanolic acid triterpenoid saponin, 3-O-ß-D-glucopyranosyl olean-11, 13(18)-diene-23,28-dioic acid, (hereafter referred to as DS-1) was isolated from the traditional Chinese medicinal plant Dianthus superbus (D. superbus). DS-1 plays an important role in the bioactivity of D. superbus. Thus, a sensitive, reliable and accurate reversed-phased liquid chromatography with tandem mass spectrometry (LC-MS/MS) in negative ion mode was developed and validated for the quantification and pharmacokinetic study of DS-1 in rats plasma. The pharmacokinetic profile showed that DS-1 was rapidly absorbed and eliminated in plasma, indicating that significant accumulation of the compound in biological specimen is unlikely. In addition, poor absorption into systemic circulation was observed after oral administration of DS-1, resulting in low absolute bioavailability (0.92 %).

[Ab] Resumo:BACKGROUND: Hepatitis is a viral infection of hepatitis B virus (HBV). Limitations of drug used in the management of it opens the interest related to alternative medicine. The given study deals with the antiviral activity of L. (DSL) against HBV & . MATERIAL AND METHODS: study liver cell line HepG2.2.15 was used by transinfected it with HBV. Cytotoxicity stduy was performed by using different concentrations of DSL such as 50, 100, 200, 500 & 1000 µg/ml. Anti HBV activity of DSL was estimated by assesing the concentration of HBsAg and HbeAg in cell culture medium by using ELISA. Whereas study was performed on ducklings and antiviral activity of DSL (100, 200, 400 mg/kg) was confirmed by estimating the serum concentration of HBV DNA and histopathology study of hepatocytes in HBV infected ducklings. RESULT: Result of the study suggested that >500 µg/ml concentration of hydroalcoholic extract of DSL was found tobe cytotoxic. It was also observed that DSL significantly ( <0.05) reduces the concentration of antigenes in cell culture media as per the concentration and days of treatment dependent. Moreover study confirms the anti viral activity of DSL (200 & 400 mg/kg) as it significantly ( <0.05) decreases the serum concenetration of HBV DNA in HBV infected dukling compared to control group. Histopathology study was also reveals the hepatprotective effect of DSL in HBV infected ducklings. CONCLUSION: The given study concludes the antiviral activity DSL against HBV by and models.
