[Ab] Resumo:A bacterial strain designated Tese-5T was isolated from a water convolvulus field in Taiwan and characterized using the polyphasic taxonomic approach. Strain Tese-5T was an aerobic, Gram-stain-negative, non-motile, rod-shaped bacterium and formed bright yellow coloured colonies. Strain Tese-5T grew at 15-35 °C (optimum, 30 °C), with 0-1.0 % NaCl (optimum, 0-0.5 %) and at pH 5.5-7 (optimum, pH 6). The major fatty acids (>10 %) of strain Tese-5T were C18 : 1ω7c, summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and C16 : 0. The polar lipid profile comprised phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylcholine, phosphatidylmonomethylethanolamine, phosphatidyldimethylethanolamine and sphingoglycolipid. The major polyamine was spermidine. The major isoprenoid quinone was Q-10. The DNA G+C content was 65.7 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain Tese-5T belonged to the genus Novosphingobium and showed the highest levels of sequence similarity to Novosphingobium chloroacetimidivorans BUT-14T and Novosphingobium mathurense SM117T (96.3 %). Phenotypic characteristics of the novel strain also differed from those of the closest-related species of the genus Novosphingobium. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain Tese-5T represents a novel species in the genus Novosphingobium, for which the name Novosphingobium ipomoeaesp. nov. is proposed. The type strain is Tese-5T (=BCRC 80904T=LMG 28838T=KCTC 42656T).

[Ab] Resumo:AIMS: To identify a potential pathogenic isolate of fungus on Convolvulus arvensis and to determine its phytotoxic activity, which revealed the presence of toxic metabolites responsible for the toxicity against the target weed. METHODS AND RESULTS: A high virulent isolate of the fungus, Phoma crystallifera was isolated from symptomatic field bindweed in the west of Iran and was screened for the production of phytotoxins, which promoted necrosis on the detached leaves and seedlings of field bindweed in the bioassays. The isolate was distinct from other isolates of the fungi on the basis of morphological characteristics and the combined sequence database of the ITS region, partial LSU rDNA and ß-tubulin gene. Isolate P. crystalifera P6 produced the highest amount of phytotoxins after 21 days in a shacked culture of Richard's broth. The active metabolites were isolated from a cell-free culture filtrate by ethyl-acetate and purified by thin layer chromatography. The result indicated that six out of nine spots had phytotoxic activity in the bioassays, with R values of 0·16, 0·30, 0·36, 0·43, 0·57 and 0·81. CONCLUSIONS: Phoma crystallifera P6 and its active metabolites showed significant phytotoxic effects on the detached leaves of C. arvensis. SIGNIFICANCE AND IMPACT OF THE STUDY: To date, there are no reports of possible biocontrol agent(s) on C. arvensis in Iran. Thus, P. crystallifera P6 is introduced here as a severe pathogenic fungus and which can be used as a biocontrol agent against C. arvensis.

[Ab] Resumo:In this study, a rapid green synthesis of the silver nanoparticles (AgNPs) using leaves extract of Convolvulus arvensis was investigated. UV-Visible spectra showed the SPR band at around 430nm which proved the AgNP formation. The reduction of silver ions to nanoparticles was completed within 150min. Particle size analysis verified the formation of AgNPs with average size of 28nm, while their PDI was 0.2. XRD patterns verified the crystalline nature of produced AgNPs. SEM images verified the presence of spherical nanoparticles with no evidence of aggregations. FTIR analysis verified the involvement of the phenolic compounds in AgNP formation. It was found that the biosynthesized AgNPs have the antibacterial activity against human pathogen E. coli. Also, nanoparticles exhibit biofilm degrading activity against both bacterial strains of S. aureus and P. aeruginosa. Furthermore, the synthesized AgNPs prevented the coagulation of blood samples. The accelerated reduction of methylene blue (MB) in the presence of AgNPs with rate constant of 0.108min , confirmed the catalytic potential of nanoparticles.

[Ab] Resumo:Oral administration of the total alcohol extract of Convolvulus pilosellifolius Desr. (250 and 500 md/kg) showed potent anti-ulcerogenic activity in absolute ethanol-induced ulcer model in rats; it showed percent protection of control ulcer by 69.2 and 84.6%, respectively, while standard ranitidine (100 mg/kg) exhibited 46.2%. Bio-guided work leads to isolation of two novel compounds (1 and 2), which were identified through H, C NMR, HMPC, HMQC and DEPT as: methyl 2-(hydroxymethyl) octanoate, named as amanitate, and 16-amino-9,13-dimethyl-17-(prop-1-en-2-yl)-hexadecahydro-1H-cyclopenta[a] phenanthren-3-ol, named as asmatol. Both compounds (50 mg/kg) possessed anti-ulcerogenic activity with 95.4% and 55.84% protection, respectively. Two known compounds (3 and 4) were also isolated and identified through comparison with authentic samples and confirmed through different NMR techniques as kampeferol and quercetin. These compounds also showed anti-ulcerogenic activity with 78.38% and 5.38% protection, respectively. The cytoprotective mechanism explains the potent anti-ulcerogenic activity of the total alcohol extract and the isolated compounds. The extract was highly safe as the LD was more than 5000 mg/kg. These results were well supported by the sub-chronic toxicity study, as the extract (500 mg/kg) administrated orally to rats for 35 consecutive days showed no alteration in the liver and kidney functions. [Ab] Resumo:BACKGROUND: Although switchgrass has been developed as a biofuel feedstock and its potential for bioethanol and bio-oil from fast pyrolysis reported in the literature, the use of the seeds of switchgrass as a source of triglycerides for biodiesel production has not been reported. Similarly, the potential for extracting triglycerides from coffeeweed (an invasive plant of no current economic value) needs to be investigated to ascertain its potential economic use for biodiesel production. RESULTS: The results show that coffeeweed and switchgrass seeds contain known triglycerides which are 983 and 1000 g kg(-1) respectively of the fatty acids found in edible vegetable oils such as sunflower, corn and soybean oils. In addition, the triglyceride yields of 53-67 g kg(-1) of the seed samples are in the range of commercial oil-producing seeds such as corn (42 g kg(-1) ). CONCLUSION: The results also indicate that the two non-edible oils could be used as substitutes for edible oil for biodiesel production. In addition, the use of seeds of switchgrass for non-edible oil production (as a feedstock for the production of biodiesel) further increases the total biofuel yield when switchgrass is cultivated for use as energy feedstock for pyrolysis oil and biodiesel production. © 2016 Society of Chemical Industry.

[Ab] Resumo:In recent years, use of plants for remediation of contaminated soils, especially removal of heavy metals, is considered. The current study tends to investigate the removal of lead and nickel ions by the Convolvulus tricolor (CTC), was grown for 30 days in different concentrations of lead and nickel ions. Then concentration of them in soil and different organs of the plant was measured by atomic absorption spectrometry. The highest absorbed of them occurred in concentration 0.001N, which highest Pb(2+) accumulation is in the aerial parts of the plant: leaf > stem > root and for Ni(2)+: root > stem > leaf. No ion was observed into the flowers and nickel ion absorption is more of lead ion in different plant organs of CTC. The experimental isotherm data were investigated using isotherms of Langmuir, Freundlich, BET, Temkin and Dubinin-Radushkevich (DRK). The correlation coefficient for all of them is calculated that show the best correlation coefficient for Ni(2+) adsorption is obtained BET model, whereas for Pb(2+) adsorption in root is Freundlich model but for its leaf and stem is BET model. The results show, CTC is suitable for Pb(2+) and Ni(2+) and this technique is in-situ method, simple, and low cost.

[Ab] Resumo:OBJECTIVE: To isolate the neuroprotective component from Convolvulus pluricaulis Chois. (Convolvulaceae) which can be used as a lead molecule in the treatment of Huntington's disease (HD). METHODS: The methanolic extract of whole plant was fractionated into four fractions; chloroform, ethyl acetate, n-butanol, and aqueous fraction. The chloroform and ethyl acetate fractions were pooled on the basis of antioxidant activity and TLC profile and charged into silica gel column. Four subfractions were collected from column (FrA, FrB, FrC, and FrD) and further assessed for antioxidant potential by 2,2-diphenyl-1-picrylhydrazyl in vitro assay. 3-Nitropropionic acid (3-NP) was administered to Wister rats for induction of HD. Different fractions were administered for 14 days. Different behavioral alterations were assessed in between study period. Animals were euthanized immediately following the last behavioral session, and biochemical parameters were measured. RESULT AND DISCUSSION: Systemic administration of 3-NP showed marked motor deficits and oxidative damage in rats. Only FrB showed significant antioxidant activity and on further purification gave pure compound (scopoletin). Our study showed that FrB (20 mg/kg) pre-treatment significantly attenuated the loss in body weight, improved the locomotor activity, grip strength, and gait abnormalities. It also has attenuated the increased malondialdehyde and nitrite levels, and restored superoxide dismutase and reduced GSH enzyme activity in the striatum and cortex in 3-NP-treated groups. These results suggest that C. pluricaulis Chois. exhibits a neuroprotective effect by accelerating brain antioxidant defense mechanisms in 3-NP-treated rats.

[Ab] Resumo:An acylated anthocyanin trioside was isolated from pink flowers of Convolvulus althaeoides using a combination of chromatographic techniques. On the basis of MS (MALDI-TOF) and NMR (1H NMR, 1H-1H COSY, 1D TOCSY, HSQC, HMBC, 13C CAPT) its structure was determined as cyanidin 3-O-[6-O-(4-O- (6-O-(E-caffeoyl)-ß-D-glucopyranosyl)-ß-L-rhamnopyranosyl)-ß-D-glucopyranoside]-5-O-ß-D-glucopyranoside. The absolute configuration of the aldose enantiomer moieties was determined from the separation of the corresponding thiazolidine diastereoisomer derivatives by HPLC-DAD. This is a novel trisaccharide within the flavonoids, and the first complete characterization of a linear glucosyl-rhamnosyl-glucoside (4'-glucosylrutinoside) within the anthocyanins. Whilst acylation of rhamnose moieties in position 4 is quite common, the occurrence in anthocyanins of a glycosylated rhamnose is a unique trait. Although many complex anthocyanins have been found amongst the Convolvulaceae, the genus Convolvulus had not yet been surveyed for anthocyanins and thus this account could be of significance within the current chemotaxonomy of this family.

[Ab] Resumo:Bois noir is an economically important grapevine yellows that is induced by 'Candidatus Phytoplasma solani' and principally vectored by the planthopper Hyalesthes obsoletus Signoret (Hemiptera: Cixiidae). This study explores the 'Ca. P. solani' genetic variability associated to the nettle-H. obsoletus and bindweed-H. obsoletus systems in vineyard agroecosystems of the central-eastern Italy. Molecular characterization of 'Ca. P. solani' isolates was carried out using polymerase chain reaction/restriction fragment length polymorphism to investigate the nonribosomal vmp1 gene. Seven phytoplasma vmp-types were detected among the host plants- and insect-associated field-collected samples. The vmp1 gene showed the highest polymorphism in the bindweed-H. obsoletus system, according to restriction fragment length polymorphism analysis, which is in agreement with nucleotide sequence analysis. Five vmp-types were associated with H. obsoletus from bindweed, of which one was solely restricted to planthoppers, with one genotype also in planthoppers from nettle. Type V12 was the most prevalent in both planthoppers and bindweed. H. obsoletus from nettle harbored three vmp-types, of which V3 was predominant. V3 was the only type detected for nettle. Our data demonstrate that planthoppers might have acquired some 'Ca. P. solani' profiles from other plant hosts before landing on nettle or bindweed. Overall, the different vmp1 gene rearrangements observed in these two plant hosts-H. obsoletus systems might represent different adaptations of the pathogen to the two host plants. Molecular information about the complex of vmp-types provides useful data for better understanding of Bois noir epidemiology in vineyard agroecosystem.

[Ab] Resumo:CONTEXT: Convolvulus pluricaulis Chois. (Convolvulaceae), a well-known Ayurvedic "Medhya Rasayana" (nervine tonic), is extensively used for different central nervous system (CNS) disorders. OBJECTIVE: The objective of this study was to evaluate the protective effect of standardized hydro-methanol extract of C. pluricaulis (CPE) and its fractions, namely ethyl acetate (EAE), butanol (BE), and aqueous (AE), against 3-nitropropionic acid (3-NP) induced neurotoxicity in rats. MATERIALS AND METHODS: The extract of the whole plant was standardized on the basis of scopoletin content (0.014%) using thin layer chromatography densitometric analysis. CPE (100 and 200 mg/kg) and its fractions, namely EAE (15 and 30 mg/kg), BE (25 and 50 mg/kg), and AE (50 and 100 mg/kg) were administered orally for 20 d. Their protective effect against 3-NP (10 mg/kg, i.p. for 14 d) was assessed by the effect on various behavioral parameters, namely body weight, locomotor activity, grip strength, gait pattern, and the effect on cognitive dysfunction. Biochemical parameters for oxidative damage were also assessed in the striatum and cortex regions of the brain. RESULTS: Administration of 3-NP induced HD-like symptoms that were indicated by reduced body weight, locomotor activity, memory, grip strength, and oxidative defense. CPE (200 mg/kg), EAE (30 mg/kg), and BE (50 mg/kg) significantly (p < 0.001) attenuated 3-NP induced reduction in locomotor activity, grip strength, memory, body weight, and oxidative defense in comparison with 3-NP-treated animals on 10 and 15 d. CONCLUSION: The present study suggested that CPE has a protective action against 3-NP-induced neurotoxicity and can be further explored for its efficacy against Huntington's disease.
