[Ab] Resumo:Dioscorins, the major storage proteins of yam tubers, exert immunomodulatory activities. To improve oral bioavailability of dioscorins in the intestine, recombinant dioscorin (rDioscorin) was coated with N,N,N-trimethyl chitosan (TMC) and tripolyphosphate (TPP), resulting in the formation of TMC-rDio-TPP nanoparticles (NPs). The loading capacity and entrapment efficiency of rDioscorin in the NPs were 26 ± 0.7% and 61 ± 1.4%, respectively. The NPs demonstrated a substantial release profile in the pH environment of the jejunum. The rDioscorin released from the NPs stimulated proliferation and phagocytosis of the macrophage RAW264.7 and activated the gene expression of IL-1ß and IL-6. Incubation of the NPs in the Caco-2 cell monolayer led to a 5.2-fold increase of P compared with rDioscorin alone, suggesting that rDioscorin, with the assistance of TMC, can be promptly transported across the intestinal epithelia. These results demonstrate that the TMC-rDio-TPP NPs can be utilized for elucidating the immunopharmacological effects of dioscorins through oral delivery.

[Ab] Resumo:The C57BL/6J mice were fed a 135-day normal diet or a high-fat diet (HFD) without, or concurrent with, a single yam dioscorin (80 mg/kg) or dipeptide NW (40 mg/kg) intervention every day. The final body weights (g) of mice were 26.1 ± 1.4, 34.97 ± 2.1, 31.75 ± 2.6, and 31.66 ± 3.1, respectively, for normal diet-fed, HFD-fed, dioscorin-intervened, and NW-intervened group. The mice in both intervened groups showed similar less weight gains and had significant differences (P < 0.05) compared to those in the HFD group under the same cumulative HFD intakes. The blood biochemical index of mice with dioscorin interventions showed significantly lower contents in total cholesterol and low-density lipoprotein, and NW interventions showed significantly lower total triglyceride contents compared to those of the HFD group (P < 0.05). Both intervened mice exhibited similar reductions in total visceral lipid contents and have significant differences compared to those of the HFD group (P < 0.05). The dioscorin intervention was better than NW interventions in lowering blood glucose levels by oral glucose tolerance tests and both showed significant differences (P < 0.05) compared to those in the HFD group. Yam dioscorin or dipeptide NW will potentially be used for preventive functional foods of less body weight gains and impaired glucose tolerance controls, which require further clinical trial investigations.

[Ab] Resumo:There is dearth of documented information on nutrient retention of Dioscorea cayenensis products. This study was carried out to evaluate effects of processing methods on nutrient and antinutrient retention of yellow yam products. Fresh Dioscorea cayenensis tubers were purchased from Bodija market in Ibadan, peeled, cut into small pieces, divided into nine portions. One portion was treated as raw sample, and others processed into local delicacies. All nine samples were analysed for proximate, mineral, vitamin and antinutrient composition using AOAC methods. Data were analysed using ANOVA at p=0.05. Raw yam contained 66.79g moisture, 2.62g crude protein, 0.27g lipid, 0.17g fibre, 0.63g ash, 29.69g carbohydrates, 262.30mg potassium, 61.53mg magnesium, 0.79mg iron, 0.39mg zinc, and yielded 108.26kcal energy with insignificant vitamin content/100g edible portion. Processing significantly improved macronutrients and energy content with significant reduction in all antinutrients of products (p<0.05). The yam products can serve as staple source of energy to consumers.

[Ab] Resumo:It is vital to monitor the holistic toxicokinetics of toxic Chinese herbal medicines (CHMs) for safety. Although an integrated strategy based on the area under the curve (AUC) has been proposed to characterize the pharmacokinetic/toxicokinetic properties of CHMs, improvement is still needed. This study attempted to use 50% inhibitory concentration (IC ) as weighting coefficient to investigate holistic toxicokinetics of the major diosbulbins i.e. diosbulbin A (DA), diosbulbin B (DB), and diosbulbin C (DC) after oral administration of Dioscorea bulbifera rhizome (DBR) extract. Firstly, the cytotoxicities of the three diosbulbins on human hepatic L02 cells were evaluated and the IC values were calculated. Then, integrated toxicokinetics of multiple diosbulbins based on AUC and IC were determined. Finally, correlations between integrated plasma concentrations and hepatic injury biomarkers including alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and total bile acid (TBA) were analyzed. As a result, integrated plasma concentrations were correlated well with TBA and the correlation between TBA and IC -weighting integrated plasma concentrations was better than that of AUC-weighting integrated plasma concentrations. In conclusion, the newly developed IC -weighting method is expected to generate more reasonable integrated toxicokinetic parameters, which will help to guide the safe usage of DBR in clinical settings.

[Ab] Resumo:The use of herbal medicines continues to expand globally, meanwhile, herb-associated hepatotoxicity is becoming a safety issue. As a conventional Chinese medicinal herb, Dioscorea bulbifera rhizome (DBR) has been documented to cause hepatic toxicity. However, the exact underlying mechanism remains largely unexplored. In the present study, we aimed to profile entire endogenous metabolites in a biological system using a multisample integrated metabolomics strategy. Our findings offered additional insights into the molecular mechanism of the DBR-induced hepatotoxicity. We identified different metabolites from rat plasma, urine, and feces by employing gas chromatography-mass spectrometry in combination with multivariate analysis. In total, 55 metabolites distributed in 33 metabolic pathways were identified as being significantly altered in DBR-treated rats. Correlation network analysis revealed that the hub metabolites of hepatotoxicity were mainly associated with amino acid, bile acid, purine, pyrimidine, lipid, and energy metabolism. As such, DBR affected the physiological and biological functions of liver via the regulation of multiple metabolic pathways to an abnormal state. Notably, our findings also demonstrated that the multisample integrated metabolomics strategy has a great potential to identify more biomarkers and pathways in order to elucidate the mechanistic complexity of toxicity of traditional Chinese medicine.

[Ab] Resumo:Airpotato yam (the rhizome of Dioscorea bulbifera L.) is traditionally used to treat thyroid disease and various cancers in China. However, it was found to cause hepatotoxicity during clinical practice. This study aims to identify candidate serum microRNAs (miRNAs) as diagnostic biomarkers for the liver injury induced by Airpotato yam. The results of serum alanine/aspartate aminotransferase (ALT/AST) showed the remarkable hepatotoxicity induced by ethyl acetate fraction of Airpotato yam (EF) (450mg/kg) and diosbulbin B (DB) (300mg/kg) in mice. The results of miRNAs chip analysis showed that the expression of 28 and 37 serum miRNAs was obviously altered in EF- and DB-treated mice, respectively. Among these miRNAs, miRNA-122-3p, miR-194-3p and miR-5099 have passed the further validation in serum from both EF- and DB-treated mice. Moreover, the expression of miRNA-122-3p and miRNA-194-5p was significantly increased in EF (375mg/kg)-treated mice with no significant elevation of serum ALT/AST activity. Only the expression of serum miRNA-5099 was not altered in the liver injury induced by acetaminophen (APAP), monocrotaline (MCT) or toosendanin (TSN). In conclusion, this study demonstrated that miR-122-3p and miRNA-194-5p were two sensitive biomarkers, and miR-5099 might be a specific biomarker for reflecting the liver injury induced by Airpotato yam.

[Ab] Resumo:The investigation of the constituents in the rhizomes of Dioscorea nipponica Makino subsp. rosthornii (Prain et Burkill) C. T. Ting afforded one new phenanthrene 2,2',7,7'-tetramethoxy-[1,1'-biphenanthrene]-4,4',6,6'-tetrol (7) and one new isocoumarin diorosthornoumarin (8), together with 16 known compounds (1-6 and 9-18). Their structures were established on the basis of extensive spectroscopic evidences (IR, HR-ESI-MS, NMR and optical rotation), as well as comparison with literature values. All the compounds 1-18 were firstly isolated from Dioscorea nipponica Makino subsp. Rosthornii (Prain et Burkill) C. T. Ting, and compound 9 was firstly obtained as a natural product from plants, while the compounds 11 and 14 were obtained from both the genus Dioscorea and the family Dioscoreaceae for the first time. Moreover, the antitumor activities of the compounds were tested against lung carcinoma NCI-H460 cell line. Compound 12, 13, 15 and 16 showed significant cytotoxic activities, whereas 7 displayed moderate cytotoxicity.

[Ab] Resumo:The economics of bio-solvent production are largely dependent on the cost of the fermentation substrate. Dioscorea zingiberensis C.H. Wright (DZW), the main raw material used to produce saponin, contains 13-18% starch which can be directly saccharified to a saccharification liquid of DZW starch (SLDS) that contains abundant nutrients. In this study, the water-soluble micromolecule compounds in SLDS were quantified through H NMR. Using SLDS as the substrate to conduct ABE fermentation by Clostridium beijerinckii, the fermentation cycle was shortened 24h, the maximum biomass and consumption rate of the glucose significantly increased, and the productivity of total solvents were increased by 0.244±0.010g/L/h compared to standard P medium. Expression analysis of genes encoding key enzymes involved in acetone and butanol production and glucose consumption showed that they were induced by SLDS. Taken together, SLDS is a useful and renewable glycosylated solution for ABE fermentation.

[Ab] Resumo:Numerous furanoids have been reported to be toxic, and many of them were found in herbal medicines. Toxicities of furanoids are suggested to result from the generation of cis-enedials via biotransformation. The detection of the electrophilic metabolic intermediates is a challenge. Earlier, we developed a selective approach to screen potential toxic furanoids, through which we found two major furanoids, diosbulbin B and 8-epidiosbulbin E acetate, in Dioscorea bulbifera L., a known furanoid-containing and hepatotoxic herbal medicine. In the present study, we improved the approach to analyze furanoids in D. bulbifera L., which allowed us to detect additional six potential furanoids, including diosbulbin A, diosbulbin D, diosbulbin E, diosbulbin F, diosbulbin M, and diosbulbin D glycoside. The achievements of this study enhanced the sensitivity to screen potential toxic furanoids through elevating S/N values by approximately 3 times. This will facilitate the understanding of mechanisms of toxic actions of D. bulbifera L. and other furanoid-containing toxic herbal medicines.

[Ab] Resumo:Yams (Dioscorea sp.) are staple food crops for millions of people in tropical and subtropical regions. Dioscorea alata, also known as greater yam, is one of the major cultivated species and most widely distributed throughout the tropics. Despite its economic and cultural importance, very little is known about its origin, diversity and genetics. As a consequence, breeding efforts for resistance to its main disease, anthracnose, have been fairly limited. The objective of this study was to contribute to the understanding of D. alata genetic diversity by genotyping 384 accessions from different geographical regions (South Pacific, Asia, Africa and the Caribbean), using 24 microsatellite markers. Diversity structuration was assessed via Principal Coordinate Analysis, UPGMA analysis and the Bayesian approach implemented in STRUCTURE. Our results revealed the existence of a wide genetic diversity and a significant structuring associated with geographic origin, ploidy levels and morpho-agronomic characteristics. Seventeen major groups of genetically close cultivars have been identified, including eleven groups of diploid cultivars, four groups of triploids and two groups of tetraploids. STRUCTURE revealed the existence of six populations in the diploid genetic pool and a few admixed cultivars. These results will be very useful for rationalizing D. alata genetic resources in breeding programs across different regions and for improving germplasm conservation methods.
