[Ab] Resumo:Common vetch ( subsp. L.) is a self-pollinating annual forage legume with worldwide importance. Here, we investigate the optimal number of individuals that may represent the genetic diversity of a single population, using Start Codon Targeted (SCoT) markers. Two cultivated varieties and two wild accessions were evaluated using five SCoT primers, also testing different sampling sizes: 1, 2, 3, 5, 8, 10, 20, 30, 40, 50, and 60 individuals. The results showed that the number of alleles and the Polymorphism Information Content ( ) were different among the four accessions. Cluster analysis by Unweighted Pair Group Method with Arithmetic Mean (UPGMA) and STRUCTURE placed the 240 individuals into four distinct clusters. The Expected Heterozygosity ( ) and increased along with an increase in sampling size from 1 to 10 plants but did not change significantly when the sample sizes exceeded 10 individuals. At least 90% of the genetic variation in the four germplasms was represented when the sample size was 10. Finally, we concluded that 10 individuals could effectively represent the genetic diversity of one vetch population based on the SCoT markers. This study provides theoretical support for genetic diversity, cultivar identification, evolution, and marker-assisted selection breeding in common vetch.

[Ab] Resumo:Vicia sativa (common vetch), which contains ß-cyanoalanine (BCA) and γ-glutamyl ß-cyanoalanine (γGBCA) is used for adulteration of lentil and have neuro toxic effects on people and animals. Therefore the determination of BCA and γGBCA analytes in accurate and reliable manner has high importance for human and animal health. LC-IDMS/MS method has been developed and validated for quantitative analyses of ß-cyanoalanine and C labeled BCA is used as an internal standard in γGBCA analysis. In order to validate the method, linearity, recovery, precision (repeatability), intermediate precision, limit of detection, and limit of quantification parameters were investigated. The correlation coefficient was found to be greater than 0.99 for both analytes. The recoveriess were determined as 95.8% for BCA and 97.4% for γGBCA. The relative expanded measurement uncertainties of ß-cyanoalanine and γ-glutamyl-ß-cyanoalanine were obtained as 4.6% and 5.8%, respectively with the coverage factor, k, is 2 and at 95% confidence level.

[Ab] Resumo:The effectiveness of plant growth promoting bacteria (PGPB) in improving metal phytoremediation is still limited by stunted plant growth under high soil metal concentrations. Meanwhile, mixed planting with leguminous plants is known to improve yield in nutrient deficient soils but the use of a metal tolerant legume to enhance metal tolerance of a phytoremediator has not been explored. We compared the use of Pseudomonas brassicacearum, Rhizobium leguminosarum, and the metal tolerant leguminous plant Vicia sativa to promote the growth of Brassica juncea in soil contaminated with 400 mg Zn kg(-1), and used synchrotron based microfocus X-ray absorption spectroscopy to probe Zn speciation in plant roots. B. juncea grew better when planted with V. sativa than when inoculated with PGPB. By combining PGPB with mixed planting, B. juncea recovered full growth while also achieving soil remediation efficiency of >75%, the maximum ever demonstrated for B. juncea. µXANES analysis of V. sativa suggested possible root exudation of the Zn chelates histidine and cysteine were responsible for reducing Zn toxicity. We propose the exploration of a legume-assisted-phytoremediation system as a more effective alternative to PGPB for Zn bioremediation.

[Ab] Resumo:We examined the effects of Cd on growth, lipid peroxidation, reactive oxygen species (ROS) accumulation, antioxidant enzymatic activity, and lignin content in the roots of two varieties of Vicia sativa. Treatment with Cd decreased plant growth and increased ROS and lipid peroxidation levels to a greater extent in the Cd-sensitive variety ZM than in the Cd-tolerant variety L3. Most hydrogen peroxide (H2O2) and superoxide anion (O2(•-)) were accumulated in the cell walls and extracellular spaces in response to Cd treatments. Chemical assays and experiments using inhibitors showed that larger increases in H2O2 and O2(•-) production in ZM than in L3 were probably attributed to elevated Cd-induced nicotinamide adenine dinucleotide-peroxidase (NADH-POD) activity. Cd treatment increased the accumulation of lignin and the guaiacol peroxidase (GPOD) activities in the apoplast more significantly in ZM root than in L3. Howerver, root laccase activity was higher in L3 than in ZM. Thus Cd toxicity induced significant lignification in the roots of V. sativa, and increases in H2O2 accumulation and apoplastic GPOD activity were likely responsible for this effect.

[Ab] Resumo:BACKGROUND: The objective of this study was to investigate the effect of molasses, lactic acid bacteria and propionic acid on the fermentation quality, aerobic stability and in vitro gas production of total mixed ration (TMR) silage prepared with oat-common vetch intercrop on the Tibetan plateau. TMR (436 g kg(-1) dry matter (DM)) was ensiled with six experimental treatments: (1) no additives (control); (2) molasses (M); (3) an inoculant (Lactobacillus plantarum) (L); (4) propionic acid (P); (5) molasses + propionic acid (MP); (6) inoculant + propionic acid (LP). RESULT: All silages were well preserved with low pH (< 4.19) and NH3-N contents, and high lactic acid contents after ensiling for 45 days. L and PL silages underwent a more efficient fermentation than silages without L. P and MP silages inhibited lactic acid production. Under aerobic conditions, M and L silage reduced aerobic stability for 15 and 74 h, respectively. All silages that had propionic acid in their treatments markedly (P < 0.05) improved the aerobic stability. After 72 h incubation, all additives treatments increased (P < 0.05) the 72 h cumulative gas production and in vitro DM digestibility (IVDMD) as compared with the control. L treatment decreased (P < 0.05) in vitro neutral detergent fibre degradability. CONCLUSIONS: Our findings show that TMR prepared with oat-common vetch intercrop can be well preserved. Although propionic acid is compatible with lactic acid bacteria, and when used together, they had minor effects on fermentation, aerobic stability and in vitro digestibility of TMR silage prepared with oat-common vetch intercrop.

[Ab] Resumo:Using high throughput sequencing of the nodA gene, we studied the population dynamics of Rhizobium leguminosarum (bv. viciae, bv. trifolii) in rhizospheric and nodular subpopulations associated with the leguminous plants representing different cross-inoculation groups (Vicia sativa, Lathyrus pratensis of the vetch/vetchling/pea group and Trifolium hybridum of the clover group). The "rhizosphere-nodules" transitions result in either an increase or decrease in the frequencies of 10 of the 23 operational taxonomic units (OTUs) (which were identified with 95% similarity) depending on the symbiotic specificity and phylogenetic positions of OTUs. Statistical and bioinformatical analysis of the population structures suggest that the type of natural selection responsible for these changes may be diversifying at the whole-population level and frequency-dependent at the OTU-specific level, ensuring the divergent evolution of rhizobia interacting with different host species.

[Ab] Resumo:The accumulation of nickel ions in the roots and shoots of vetch seedlings (Vicia sativa L.) at increasing concentrations of nickel chloride in the medium was studied. It was shown that the accumulation of nickel in the shoots was increased when the concentration of nickel chloride in the medium was more than 50 µM. The bioconcentration factor and sustainability index for vetch seedlings were calculated under the experimental conditions. The obtained results were similar to parameters for other plants, grown on a nutrient medium or soil substrate. First, the obtained results allowed estimate the limits of nickel chloride concentrations for four of five zones, which correspond to the theoretical concept of dose-response curves in the studies on the influence of physiologically essential heavy metals on plants (Prasad 2010). Some parameters of oxidative stress caused by the presence of nickel chloride in the medium were shown. It seems that at low nickel concentrations in the medium in vetch seedlings the increase of several biochemical parameters (catalase activity and proline) caused by the high amylase activity in seeds.

[Ab] Resumo:The temperate and herbaceous genus Vicia L. is a member of the legume tribe Fabeae of the subfamily Papilionoideae. The genus Vicia comprises 166 annual or perennial species distributed mainly in Europe, Asia, and North America, but also extending to the temperate regions of South America and tropical Africa. The use of simple sequence repeat (SSR) markers for Vicia species has not been investigated as extensively as for other crop species. In this study, we assessed the potential for cross-species amplification of cDNA microsatellite markers developed from common vetch (Vicia sativa subsp. sativa). For cross-species amplification of the SSRs, amplification was carried out with genomic DNA isolated from two to eight accessions of 22 different Vicia species. For individual species or subspecies, the transferability rates ranged from 33% for V. ervilia to 82% for V. sativa subsp. nigra with an average rate of 52.0%. Because the rate of successful SSR marker amplification generally correlates with genetic distance, these SSR markers are potentially useful for analyzing genetic relationships between or within Vicia species.

[Ab] Resumo:The objective of this study was to evaluate effects of lactic acid bacteria and propionic acid on the fermentation quality and aerobic stability of oats-common vetch mixed silage by using a small-scale fermentation system on the Tibetan plateau. (i) An inoculant (Lactobacillus plantarum) (L) or (ii) propionic acid (P) or (iii) inoculant + propionic acid (PL) were used as additives. After fermenting for 60 days, silos were opened and the aerobic stability was tested for the following 15 days. The results showed that all silages were well preserved with low pH and NH3 -N, and high lactic acid content and V-scores. L and PL silages showed higher (P < 0.05) lactic acid and crude protein content than the control silage. P silage inhibited lactic acid production. Under aerobic conditions, L silage had similar yeast counts as the control silage (> 10(5) cfu/g fresh matter (FM)); however, it numerically reduced aerobic stability for 6 h. P and PL silages showed fewer yeasts (< 10(5) cfu/g FM) (P < 0.05) and markedly improved the aerobic stability (> 360 h). The result suggested that PL is the best additive as it could not only improved fermentation quality, but also aerobic stability of oats-common vetch mixed silage on the Tibetan plateau.

[Ab] Resumo:The chemical composition of four common vetch (Vicia sativa L.) seeds was investigated to determine their nutrition value. The result shows that the seeds are low in lipid (1.55-2.74% of dry weight), and high in the unsaturated fatty acid (74.51-77.36% of total fatty acid). The ratio of essential amino acid to non-essential amino acid (0.62-0.69) is even higher than the amount (0.38) recommended by World Health Organization. Besides, the seeds are also found rich in Mg, Mn and Cu, but with a low ratio of Ca to P (0.24-0.73), which may increase the risk of the mineral element toxicity. The results indicate that the four common vetch seeds could be taken as an alternative food source, but the possible toxic effect should be taken into consideration.
