[Ab] Resumo:SNARE proteins are essential to vesicle trafficking and membrane fusion in eukaryotic cells. In addition, the SNARE-mediated secretory pathway can deliver diverse defense products to infection sites during exocytosis-associated immune responses in plants. In this study, a novel gene (CkSNAP33) encoding a synaptosomal-associated protein was isolated from Cynanchum komarovii and characterized. CkSNAP33 contains Qb- and Qc-SNARE domains in the N- and C-terminal regions, respectively, and shares high sequence identity with AtSNAP33 from Arabidopsis. CkSNAP33 expression was induced by H2O2, salicylic acid (SA), Verticillium dahliae, and wounding. Arabidopsis lines overexpressing CkSNAP33 had longer primary roots and larger seedlings than the wild type (WT). Transgenic Arabidopsis lines showed significantly enhanced resistance to V. dahliae, and displayed reductions in disease index and fungal biomass, and also showed elevated expression of PR1 and PR5. The leaves of transgenic plants infected with V. dahliae showed strong callose deposition and cell death that hindered the penetration and spread of the fungus at the infection site. Taken together, these results suggest that CkSNAP33 is involved in the defense response against V. dahliae and enhanced disease resistance in Arabidopsis.

[Ab] Resumo:BACKGROUND: Pestiviruses in general, and Bovine Viral Diarrhea (BVD) in particular, present several potential targets for directed antiviral therapy. MATERIAL AND METHODS: The antiviral effect of (Bge.) Kitag (Dog strangling vine: DS) extract on the bovine viral diarrhea (BVD) virus was tested. First, a cytotoxicity test in MDBK (Madin-Darby bovine kidney) cells was done with all organic extract concentrations. RESULTS: The cytotoxic concentration CC for the ethyl acetate (EA) extracts was 18.2 ug/ml. In the tissue culture, infectious dose (TCID ) assay, the BVD virus decreased when treated with 18.2 ug/ml of the ethyl acetate extracts. CONCLUSION: Ethyl acetate extracts and fractions of the DS extract could be used as a potential antiviral for BVD.

[Ab] Resumo:Two new sugar-free 14,15-secopregnane-type steroids, 14-O-methyl-3-epi-hirundigenin (1) and 2-deoxyamplexicogenin A (2), along with two known sugar-free pregnane-type steroids, were isolated from the 95% ethanol extract of the roots of Cynanchum stauntonii. The structures of the new compounds were characterized on the basis of extensive spectroscopic analyses, mainly 1D and 2D NMR methods, albeit the MS experiments did not display the molecular ion peaks. Compound 2 was the aglycones of stauntosides J and K, etc., previously isolated from C. stauntonii. The isolation and identification of the new compounds graced the structural diversity of pregnane-type steroids from C. stauntonii.

[Ab] Resumo:Twenty-three undescribed previously C steroidal glycosides, named saccatols D-K and cynsaccatols I-W, together with six known analogs were isolated from the roots of Cynanchum auriculatum Royle ex Wight (Apocynaceae). Their structures were elucidated by a variety of spectroscopic techniques, as well as HRESIMS and GC method. All isolated compounds were tested their neuroprotective effects on the model of H O -induced damage to PC12 cells. Except saccatols D-F, J, cynsaccatol V and wilfoside A, other twenty-three isolates produced significantly activities against H O -induced cell damage (P < 0.001). Furthermore, cynsaccatols I, N, O and S can obvious inhibition of damaged PC12 cell apoptosis at their dosages of 1 µM by Annexin V-FITC/PI double staining assay with flow cytometry.

[Ab] Resumo:Nine 14,15-secopregnane-type C -steriosides, stauntosides U, V, V -V , W and C -C , as well as two known C -steriosides, were isolated from the roots of Cynanchum stauntonii. Stauntosides U, V and V -V share the same basic structural features of 8α:14α,14:16,15:20,18:20-tetraepoxy-14,15-secopregn-6-ene-3ß,5α,9α-triol, with the numbering system following that of C -pregnanes. The aglycones of stauntosides U, V and V -V are classified into two subcategories, the 5,9-dihydroxy groups and 5α:9α-peroxy bridge, according to the oxidative states of the two hydroxy groups at the C-5 and C-9 positions. The anti-inflammatory activity of the major compounds was assessed in an in vitro inflammatory model of mouse peritoneal macrophages using IC values of the inhibition of nitric oxide (NO) production as an indicator. Stauntosides V and V exhibited target activity with IC values of 9.3 µM and 12.4 µM, respectively, compared with dexamethasone, which was used as a positive control.

[Ab] Resumo:Many microorganisms can generate pigments with different colours and structures during the growth process. In this study, an endophytic fungus producing red pigments was isolated from Cynanchum auriculatum Royle ex Wight tissue. PCR amplification sequencing was conducted, and phylogenetic analysis was performed on the ITS region sequences. Combined with morphological observation, the fungus was identified as Stemphylium lycopersici. The antioxidant activities of the pigments were evaluated in vitro and showed good antioxidant properties. Ultraviolet (UV), Raman, and Fourier transform infrared (FTIR) spectroscopy and gas chromatography-mass spectroscopy (GC-MS) were used to analyse the pigments' components, which were shown to contain phenolics and anthraquinones. Most of these components have not been previously reported in Stemphylium lycopersici, especially physcione. This study is the first report of Stemphylium lycopersici secondary metabolites and their potential use as red pigments and antioxidants. Further optimisation of the culture conditions of this fungal strain might permit its application for pigment production.

[Ab] Resumo:Effect of acid hydrolysis on the morphology, structure and digestion property of starch from Cynanchum auriculatum Royle ex Wight was investigated in this study. The hydrolysis degree of C. auriculatum starch rapidly increased to 63.69% after 4days and reached 78.67% at the end of 9days. Morphology observation showed that the starch granules remained intact during the first 4days of hydrolysis. However, serious erosion phenomenon was observed after 5days and starch granules completely fell into pieces after 7days. During acid hydrolysis process, the crystal type of hydrolyzed starch changed from original C -type to final A-type. Small-angle X-ray scattering patterns showed the semi-crystalline growth rings started to be hydrolyzed after 4days. The proportions of single helix and amorphous components as well as amylose content in starch gradually decreased, whereas the proportion of double helix components continuously increased during acid hydrolysis. However, the contents of rapidly digestible starch, slowly digestible starch and resistant starch were almost constant during acid hydrolysis process, indicating the in vitro digestion property of C. auriculatum starch was not affected by acid hydrolysis. Our results provided novel information on the inner structure of C. auriculatum starch granules.

[Ab] Resumo:The present study aims to evaluate the antiparasitic activity of active components from Cynanchum paniculatum against Ichthyophthirius multifiliis. The antiparasitic activities of two bioassay-guided fractionationated compounds from C. paniculatum identified as Cynatratoside-A and Cynanversicoside C, by comparing spectral data (NMR and ESI-MS) with literature values, were evaluated by in vitro assay. These showed that both could kill theronts of I. multifiliis at a concentration of 10·0 mg L-1, with the median effective concentration (EC50) values of 4·6 mg L-1 and 5·2 mg L-1 for Cynatratoside-A and Cynanversicoside C, respectively. Encysted tomonts were killed at concentrations of 8·0 mg L-1 with both compounds. In vivo experiments demonstrated that fish treated with both compounds at 15·0 mg L-1 carried significantly fewer parasites than controls (P < 0·05). There were no mortalities among treated fish group compared with 75% mortality of untreated fish. Cynatratoside-A and Cynanversicoside C are therefore potential candidate drugs for use against I. multifiliis.

[Ab] Resumo:The root of Cynanchum auriculatum Royle ex Wight is a traditional Chinese herbal medicine and healthy food. Although C. auriculatum has already been processed into starch for human consumption in China, the structural characterizations of C. auriculatum starch is still unknown. Therefore, the morphology, structural and physicochemical properties of C. auriculatum starch were investigated in this study. C. auriculatum starch exhibited both spherical and polygonal shapes with granule size ranging from 2 to 12µm. Some void cavities and serpentine channels were observed in the inner of starch granules. X-ray powder diffraction pattern revealed that C. auriculatum starch was a C -type with relative crystallinity of 25.19%. Small-angle X-ray scattering spectrum indicated C. auriculatum starch had a lamellar repeat distance of 9.21nm. The proportions of single helix, double helix and amorphous components in C. auriculatum starch were 3.42%, 27.11% and 69.47%, respectively. The amylose content of C. auriculatum starch was 28.0% with the gelatinization temperature ranging from 59.3 to 70.1°C. The maximum weight loss rate of C. auriculatum starch appeared at 309°C. In addition, C. auriculatum starch showed higher swelling power than other starches tested. Our results suggest C. auriculatum starch will have wide applications in food industry.
