[Ab] Resumo:In Russia, the species of the genus Iris L., series Lacteae Doronkin, the taxonomic structure of which still remains controversial, are found in the south of Siberia and the Russian Far East, as well as in other regions of Asia. Sequence analysis of three chloroplast DNA regions (rps4, trnL­trnF, and trnS­trnG) shows that, in Russia and adjacent countries, there are two genetically and geographically isolated Lacteae species. I. oxypetala Bunge grows in the south of the Russian Far East, and I. lactea Pall. grows in Siberia, Mongolia, and Kazakhstan. Genetic differentiation between the populations of I. lactea is extremely low and statistically insignificant (the fixation index ΦST = 0.057, P > 0.05), pointing to the unity of the gene pool and the absence of other Lacteae species in this area.

[Ab] Resumo:A number of novel bacteria including members of rarely cultivated phyla, Acidobacteria and Verrucomicrobia, were successfully isolated from the roots of two emergent plants, Iris pseudacorus and Scirpus juncoides, by a simple culturing method. A total of 47.1% (66 strains) for I. pseudacorus and 42.1% (59 strains) for S. juncoides of all isolates (140 strains from each sample) were phylogenetically novel. Furthermore, Acidobacteria and Verrucomicrobia occupied 10.7% (15 strains) and 2.9% (4 strains) of I. pseudacorus isolates, and 2.1% (3 strains) and 3.6% (5 strains) of S. juncoides isolates, respectively, indicating that plant roots are attractive sources for isolating rarely cultivated microbes.

[Ab] Resumo:Iritectol G, a novel iridal-type triterpenoid containing an uncommon tetrahydrofuran moiety, was isolated from the rhizomes of Iris tectorum. The structure was elucidated by comprehensive spectroscopic analysis. Iritectol G inhibited spontaneous and 4-aminopyridine-evoked calcium oscillations in primary cultured neocortical neurons with IC values of 8.2µM and 12.5µM, respectively. Further electrophysiological study demonstrated that iritectol G preferred to interact with inactivated state of voltage-gated sodium channel with an IC value of 7.0µM. These data demonstrated that iritectol G was a novel sodium channel inhibitor.

[Ab] Resumo:This study focused on the effects of plant compositions on removal rates of pollutants in microcosms through investigating rhizosphere microbial populations, photosynthetic efficiency and growth characteristics. Mixed-culture groups improved the removal efficiency of TN and TP significantly but exhibited lower COD removal rates. Total plant biomasses were improved as the species richness increased, but the N/P content in the plants was mainly affected by the type of species. The mixed-culture groups showed lower photosynthesis rates and oxygen supply generated from roots under high irradiation. Microbial communities of the cultured groups in the rhizosphere exhibited significant differences. According to principal component analysis (PCA), the fungi were the typical microbes of SPA, SPAB, and SPABC, resulted in improvement in nutrient accumulation. These results demonstrated that a mixed culture strategy can represent the overyielding of biomass, promote the photo-protection mechanism, and will further increase the removal rates of pollutants in a constructed wetland.

[Ab] Resumo:Iris lactea is a perennial halophyte and is tolerant to Cd. However, the mechanisms underlying this Cd tolerance are still poorly understood. In this study, morphological, physiological and biochemical responses of I. lactea to a 21 d exposure to different concentrations of Cd (0-150mgL ) were investigated. I. lactea plants showed no toxicity symptoms except for a small reduction in growth at 100 and 150mgL Cd, along with the enhancement of H O and MDA content in comparison to the control. The activities of SOD and POD were significantly enhanced and Ca accumulated with increasing Cd concentrations. Moreover, most Cd was retained in roots and only a small amount was transported to the shoots with increasing external Cd concentrations. Cd content had a negative correlation with content of K, Fe, Zn, and Mn and a positive correlation with Mg content in shoots and roots, which had no influence on these contents of mineral nutrients in shoots and chlorophyll levels with the increase of Cd concentrations. The Cd translocation factors were always less than 1 and bioaccumulation factors ranged from 3.43 to 15.6 across all treatments, suggesting that I. lactea might be effectively used in phytostabilization of Cd contaminated soils. Overall, the findings suggest that I. lactea could reduce photoinhibition and oxidative damage and maintain metal ion homeostasis in plant tissue by limiting translocation of Cd from roots to shoots and enhancing induction of antioxidant enzyme activities, thereby improving its Cd tolerance.

[Ab] Resumo:Cancer is possibly one of the most devastating and complex disease and therefore involves chemotherapy as one of the frontline strategies in its therapy. However, expected toxicity and resistance with chemotherapeutic agents encourage us to use the plant derived natural chemotherapeutic sources at the clinical stage of cancer therapy. In view of this strategy, herein new glycosides and isoflavonoids were isolated from Iris kashmiriana Baker and subjected to structure elucidation followed by their biological evaluation. Isolated compounds and their derivatives were purified by the column chromatography and structural identification was made by a combination of various spectroscopic technique vis. UV, IR, H NMR, C NMR, DEPT, 2-D NMR and mass spectrometry coupled with chemical analysis. Furthermore, an in silico library of isolated isoflavones and its analogues were designed. NF-kappaB (transcription factor that facilitates angiogenesis, inflammation, invasion and metastasis) was selected as a target to evaluate the anticancer and antioxidant activity of isoflavones and its analogues. Designed library of isoflavones and analogues were docked into the active site of NF-kappa B and the most active 15 analogues were selected for synthesis. Finally, all compounds were evaluated for their cytotoxicity against various cell lines and antioxidant activity with different methods that demonstrate their anti-cancer and anti-oxidant potential. The cell cycle specificity of the cytotoxicity was further analyzed by corresponding analysis, using flow cytometer. Most of the compounds exhibit moderate activity, whereas the 5,7,8-trihydroxy-3-(4-methoxyphenyl)-4H-chromen-4-one, 5,7,8-trihydroxy-3-(4-hydroxyphenyl)-4H-chromen-4-one, 5,7,8-triacetoxyoxy-3-(4-methoxyphenyl)-4H-chromen-4-one and 6,7-diacetoxyoxy-3-(4-methoxyphenyl)-4H-chromen-4-one showed distinct broad-spectrum anticancer activity with IC values ranges between 3.8 and 5.6 µg/mL. Cell cycle analysis indicates that these compounds induced cell cycle arrest at G2/M phase.

Their structures were elucidated on the basis of spectroscopic data interpretation.

[Ab] Resumo:Iris species are well recognized as wealthy sources of isoflavonoids. In the present study, phytochemical investigation of the rhizomes of Iris germanica (Iridaceae) procure the isolation of two new isoflavonoids namely, 8-hydroxyirilone 5-methyl ether (2) and 8-hydroxyirilone (3), along with eight known isoflavonoids: irilone 4'-methyl ether (1), irilone (4), irisolidone (5), irigenin S (6), irigenin (7), irilone 4'-O-ß-d-glucopyranoside (8), iridin S (9), and iridin (10). The isolated flavonoids were structurally characterized with the assist of comprehensive spectroscopic analyses (UV, IR, 1D and 2D NMR, and HRMS) and comparing with the published data. They were estimated for their antioxidant and antidaibetic capacities using DPPH and α-amylase inhibition assays, respectively. Compounds 2, 3, and 4 exhibited prominent antioxidant activities with IC values of 12.92, 9.23, and 10.46µM, respectively compared to propyl gallate (IC 7.11µM). Moreover, 2-5 possessed highest α-amylase inhibitory activity with % inhibition 66.1, 78.3, 67.3, and 70.1, respectively in comparison to acarbose (reference α-amylase inhibitor). Additionally, their structure-activity relationship has been discussed.

[Ab] Resumo:Six new iridal-type triterpenoids containing an unprecedented cyclopentane ring, polycycloiridals E-J (1-6), were isolated from a large-scale re-extraction of Iris tectorum. A possible biosynthesis pathway is postulated. The known spirioiridotectal D (7) was also obtained in the current investigation, and its structure was unequivocally defined using X-ray diffraction data. Compound 7 suppressed LPS-activated NO production in the BV2 cell line with an IC value of 0.54 µM.

[Ab] Resumo:As a green remediation technology, phytoremediation is becoming one of the most promising methods for treating petroleum hydrocarbons (PHCs)-contaminated soil. Pot culture experiments were conducted in this study to investigate phytoremediation potential of two representative Iridaceae species (Iris dichotoma Pall. and Iris lactea Pall.) in remediation of petroleum hydrocarbon-contaminated saline-alkali soil from the Dagang Oilfield in Tianjin, China. The results showed that I. lactea was more endurable to extremely high concentration of PHCs (about 40,000 mg/kg), with a relatively high degradation rate of 20.68%.The degradation rate of total petroleum hydrocarbons (TPHs) in soils contaminated with 10,000 and 20,000 mg/kg of PHCs was 30.79% and 19.36% by I. dichotoma, and 25.02% and 19.35% by I. lactea, respectively, which improved by 10-60% than the unplanted controls. The presence of I. dichotoma and I. lactea promoted degradation of PHCs fractions, among which saturates were more biodegradable than aromatics. Adaptive specialization was observed within the bacterial community. In conclusion, phytoremediation by I. dichotoma should be limited to soils contaminated with ≤20,000 mg/kg of PHCs, while I. lactea could be effectively applied to phytoremediation of contaminated soils by PHCs with at least 40,000 mg/kg.
