[Ab] Resumo:Both selenium (Se) and polysaccharides from (Maxim.) Li (PFPs) ( ) have been reported to possess antioxidative and immuno-protective activities. Whether or not Se-containing polysaccharides (Se-PFPs) have synergistic effect of Se and polysaccharides on enhancing the antioxidant and immune activities remains to be determined. We previously reported that polysaccharides isolated from Se-enriched (Se-PFPs) possessed hepatoprotective effects. However, it is not clear whether or not they have anti-mutagenic effects. In the present study, we compared and evaluated anti-mutagenic effects of Se-PFPs at three concentrations (1.35, 2.7 and 5.4 g/kg body weight) with those of PFPs, Se alone or Se + PFPs in mice using micronucleus assay in bone marrow and peripheral blood as well as mitomycin C-induced chromosomal aberrations in mouse testicular cells. We also elucidated the underlying mechanism. Our results demonstrated that Se-PFPs inhibited cyclophosphamide (CP)-induced micronucleus formation in both bone marrow and peripheral blood, enhanced the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) in mouse liver, and reduced the activity and expression of cytochrome P450 1A (CYP4501A) in mouse liver in a dose-dependent manner. In addition, we found that the anti-mutagenic potential of Se-PFPs was higher than those of PFPs, Se alone or Se + PFPs at the same level. These results suggest that the anti-mutagenic potential of Se-PFPs may be mediated through the inhibition of the activity and expression of CYP4501A. This study indicates that application of Se-PFPs may provide an alternative strategy for cancer therapy by targeting CYP1A family.

[Ab] Resumo:Nontoxic natural products useful in skin care cosmetics are of considerable interest. Tyrosinase is a rate-limiting enzyme for which its inhibitor is useful in developing whitening cosmetics. Pyracantha koidzumii (Hayata) Rehder is an endemic species in Taiwan that exhibits tyrosinase-inhibitory activity. To find new active natural compounds from P. koidzumii, we performed bioguided isolation and studied the related activity in human epidermal melanocytes. In total, 13 compounds were identified from P. koidzumii in the present study, including two new compounds, 3,6-dihydroxy-2,4-dimethoxy-dibenzofuran (9) and 3,4-dihydroxy-5-methoxybiphenyl-2'-O-ß-d-glucopyranoside (13), as well as 11 known compounds. The new compound 13 exhibited maximum potency in inhibiting cellular tyrosinase activity, the protein expression of cellular tyrosinase and tyrosinase-related protein-2, as well as the mRNA expression of Paired box 3 and microphthalmia-associated transcription factor in a concentration-dependent manner. In the enzyme kinetic assay, the new compound 13 acted as an uncompetitive mixed-type inhibitor against the substrate l-3,4-dihydroxyphenylalanine and had a Km value against this substrate of 0.262 mM, as calculated using the Lineweaver-Burk plots. Taken together, our findings show compound 13 exhibits tyrosinase inhibition in human melanocytes and compound 13 may be a potential candidate for use in cosmetics.

[Ab] Resumo:The study of phenotypic and genetic variation of obligate parthenogenetic organisms contributes to an understanding of evolution in the absence of genetic variation produced by sexual reproduction. Eriosoma lanigerum Hausmann undergoes obligate parthenogenesis in Mexico City, Mexico, due to the unavailability of the host plants required for sexual reproduction. We analysed the phenotypic and genetic variation of E. lanigerum in relation to the dry and wet season and plant phenology. Aphids were collected on two occasions per season on a secondary host plant, Pyracantha koidzumii, at five different sites in the southern area of Mexico City, Mexico. Thirteen morphological characteristics were measured from 147 to 276 individuals per site and per season. A multivariate analysis of variance was performed to test the effect of the season, site and their interaction on morphological traits. Morphological variation was summarised using a principal component analysis. Genetic variation was described using six enzymatic loci, four of which were polymorphic. Our study showed that the site and season has a significant effect on morphological trait variation. The largest aphids were recorded during cold temperatures with low relative humidity and when the plant was at the end of the fruiting period. The mean genetic diversity was low (mean H e = .161), and populations were genetically structured by season and site. Morphological and genetic variations appear to be associated with environmental factors that directly affect aphid development and/or indirectly by host plant phenology.

[Ab] Resumo:BACKGROUND: The aim of this study was to investigate the influence of a combination of selenium-enriched green tea polysaccharides (Se-GTP) and Huo-ji polysaccharides (HJP) on the immune function and antioxidant activity in mice. RESULTS: The results showed that the indices of spleen and thymus were markedly increased, and the activity of natural killer (NK) cell was promoted in mice treated with the combination of Se-GTP and HJP. The combined treatment of Se-GTP and HJP also reduced the content of tumour necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in splenocytes. In addition, the activities of glutathione peroxidase (GPx) and superoxide dismutase (SOD) were remarkably enhanced, and malondialdehyde (MDA) levels were significantly reduced in mice treated with combination of Se-GTP and HJP. Furthermore, the combined treatment of Se-GTP and HJP increased nuclear factor erythroid 2-related factor (Nrf2) expression at mRNA and protein levels in splenocytes. The effects of the combination treatment of Se-GTP and HJP in mice were stronger than with Se-GTP or HJP treatment alone. CONCLUSION: Our study suggests that the combined administration of Se-GTP and HJP can synergistically improve immune function and decrease the oxidative stress by enhancing the mechanisms involved in the clearance of free radicals.

[Ab] Resumo:We have previously reported that polysaccharides extracted from Pyracantha fortuneana (Maxim.) Li (P. fortuneana) lowered the oxidative stress and inhibited the inflammatory responses in mice. Our present study aims to determine the effects of Selenium enriched P. fortuneana polysaccharides (Se-PFPs) against carbon tetrachloride (CCl4)-induced liver injury in a mouse model. Our results displayed that CCl4 remarkably elevated the levels of alanine transferase (ALT), aspartate transaminase (AST), lactic dehydrogenase (LDH), cholesterol, triglycerides in serum. However, similar to BP treatment, supplementation of mice with Se-PFPs resulted in reversal of ALT, AST, LDH, cholesterol, triglycerides in serum. Contrary to CCl4, supplementation of mice with Se-PFPs elevated the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) and levels of glutathione (GSH) in liver. Furthermore, Se-PFPs treatment increased the expression of GPx and catalase (CAT) at mRNA and protein levels in liver which were decreased in CCl4 group. Contrary to CCl4, Se-PFPs supplement decreased the levels of thiobarbituric acid reactive substances (TBAR) and H2O2, which served as lipid peroxidation biomarker. Our study indicates that Se-PFPs administration is effective in attenuating CCl4-induced liver injury. The mechanism underlying this effect may be attributed to the reduction of oxidative stress and inflammation in the liver by Se-PFPs through up-regulation of the antioxidant system. Our study suggests that Se-PFPs might be a potential dietary agent in the prevention of hepatic damage.

[Ab] Resumo:Arge pyracanthae Wei & Shinohara, n. sp. is described from Mt. Hupingshan, Hunan Province, China, including COI sequences from two specimens. Larvae are solitary external leaf feeders on Pyracantha fortuneana (Maxim.) H. L. Li (Rosaceae). Field observations and rearing experiments showed that this species has a multivoltine life cycle. This is the first record of an argid sawfly associated with Pyracantha.

[Ab] Resumo:Proanthocyanidins (PCs) with poor bioavailability were argued for their health benefits. In this study, water-soluble polymeric polyphenolic PCs fractions from Pyracanthafortuneana fruit were used to investigate whether the presence of PCs is correlated with the increased cell antioxidant activities (CAA) of quercetin (Q). The results indicated that the most decrement in the values of EC50, which Q inhibited peroxyl radical-induced DCFH oxidation effective in the HepG2 cells, was observed to be 2.91 (vs. control 5.97) in the present of the fraction with 15.8 of the average degree of polymerisation of PCs (ADP). Also, the order of efficacy was the same with the ADP of PCs. Further, this effect is associated with the improvement of the solubility and stability of Q after the addition of the PCs. Our current study suggests that the additive effects of PCs on small molecular polyphenols may be responsible for their antioxidant benefits in vivo.

[Ab] Resumo:Firethorn fruits have been used in traditional medicine owing to their diuretic, cardiac and tonic properties. In this study, the antiradical activities and phenolic, phytosterol, lipid-soluble vitamin and fatty acid contents of firethorn fruits were investigated. Antiradical activities of firethorn extracts (ethanol, water, methanol, acetone and diethyl ether) were determined by using DPPH(·), ABTS(·+) and OH(·) radical-scavenging assays. The scavenging activity of the firethorn ethanol, methanol and acetone extracts on ABTS is higher than BHT. The scavenging activity of the firethorn methanol and ethanol extracts on DPPH is higher than BHT. δ-Tocopherol, α-tocopherol, vitamin K, vitamin D, ß-sitosterol, ergosterol, stigmasterol, and the major fatty acids linolenic acid and palmitic acid were determined in firethorn. These results indicate that firethorn can be a good source of natural antioxidant owing to the presence of fatty acids, vitamins, phytosterols and phenolic compounds, and antiradical properties. Moreover, this study is the first report about antiradical and phytochemical properties of Pyracantha coccinea fruit extracts.

[Ab] Resumo:A natural biosorbent obtained from Pyracantha coccinea was modified with an anionic surfactant to facilitate its dye removal ability. Modified biosorbent was successfully employed for the decolorization of Methyl Violet (MV)-contaminated solutions. A three-variable Box-Behnken design for response surface methodology was used to examine the function of independent operating variables. Optimum pH and biosorbent amount were found to be 6.0 and 0.055 g, respectively. The effects of temperature and ionic strength on the dye removal performance of biosorbent were also investigated. A biosorption equilibrium was attained within 30 min and experimental data fitted well to the pseudo-second-order model. The Langmuir isotherm model fitted adequately to the equilibrium data. The maximum monolayer biosorption capacity of the modified biosorbent was found to be 254.88 mg g(-1). Good biosorption yields were also recorded in continuous biosorption system. Ion exchange and complexation could be suggested as possible mechanisms for the biosorption. The developed modified biosorbent was regenerated up to 80.30 % by 0.005 M HCl. At real wastewater conditions, it has 86.23 ± 0.21 and 94.51 ± 1.09 % dye removal yields in batch and column systems, respectively. Modified biomaterial can be used as an effective biosorbent for the removal of MV dye from aqueous solution with high biosorption performance.

[Ab] Resumo:This study focused on the improvement of the decolorization potential of biomass derived from Pyracantha coccinea. Alkyl benyzldimethyl ammonium chloride (ABDAC) was used as modification agent. Batch mode decolorization potential of modified biosorbent was explored at different operating conditions. ABDAC modification significantly increased the biosorption yield to 97.27%, which was 3.88 times higher than that of natural biomass. The prepared biosorbent was effectively used for the decolorization of Reactive Red 45 contaminated solutions after the optimization of biosorption conditions. The non-linear regression analysis was used to evaluate the isotherm and kinetic model parameters. Process followed the Langmuir isotherm model and the highest monolayer capacity of 152.49 mg g(-1) was obtained with a small amount of modified biosorbent. Kinetic studies indicated fast decolorization rate of the process following the pseudo-first-order model. Biosorption performance of the prepared biosorbent was tested in RR45 containing real wastewater sample. The possible dye biosorbent interactions in the biosorption process were explored by zeta potential, scanning electron microscobe and FTIR analysis.
