[Ab] Resumo:Competition and fouling defence are important traits that may facilitate invasions by non-indigenous species. The 'novel weapons hypothesis' (NWH) predicts that the invasive success of exotic species is closely linked to the possession of chemical defence compounds that the recipient community in the new range is not adapted to. In order to assess whether chemical defence traits contribute to invasion success, anti-bacterial, anti-quorum sensing, anti-diatom, anti-larval and anti-algal properties were investigated for the following algae: a) the invasive brown alga Sargassum muticum from both, its native (Japan) and invasive (Germany) range, b) the two non- or weak invasive species Sargassum fusiforme and Sargassum horneri from Japan, and c) Fucus vesiculosus, a native brown alga from Germany. Crude and surface extracts and lipid fractions of active extracts were tested against common fouling organisms and zygotes of a dominant competing brown alga. Extracts of the native brown alga F. vesiculosus inhibited more bacterial strains (75%) than any of the Sargassum spp. (17 to 29%). However, Sargassum spp. from Japan exhibited the strongest settlement inhibition against the diatom Cylindrotheca closterium, larvae of the bryozoan Bugula neritina and zygotes of the brown alga F. vesiculosus. Overall, extracts of S. muticum from the invasive range were less active compared to those of the native range suggesting an adaptation to lower fouling pressure and competition in the new range resulting in a shift of resource allocation from costly chemical defence to reproduction and growth. Non-invasive Sargassum spp. from Japan was equally defended against fouling and competitors like S. muticum from Japan indicating a necessity to include these species in European monitoring programs. The variable antifouling activity of surface and crude extracts highlights the importance to use both for an initial screening for antifouling activity.

[Ab] Resumo:An aerobic, gliding and yellow-pigmented bacterium, designated strain SFD31T, was isolated from brown alga collected from the South Sea, Republic of Korea. Cells were Gram-stain-negative, and catalase- and oxidase-positive. The neighbour-joining phylogenetic tree based on 16S rRNA gene sequences showed that strain SFD31T forms an independent lineage within the genus Winogradskyella. Strain SFD31T was related distantly to Winogradskyella echinorum KMM 6211T (97.9 %, 16S rRNA gene sequence similarity), Winogradskyellalitoriviva KMM 6491T (97.4 %), Winogradskyellapulchriflava EM106T (97.2 %) and Winogradskyellaeckloniae EC29T (96.9 %). The major fatty acids of strain SFD31T were iso-C15 : 0, iso-C15 : 1 G, summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and unknown 13.565. The only isoprenoid quinone of the isolate was menaquinone 6. The major polar lipids were phosphatidylethanolamine, four unidentified aminolipids and two unidentified lipids. The DNA G+C content of strain SFD31T was 36.0 mol%. Phenotypic characteristics distinguished strain SFD31T from the related species of the genus Winogradskyella. On the basis of the evidence presented in this study, a novel species, Winogradskyella flava sp. nov., is proposed for strain SFD31T (=KCTC 52348T=JCM 31798T).

[Ab] Resumo:Hepatoprotective and reno-potective effect of Sargassum species was investigated in rats against carbon tetrachloride (CCl ) and acetaminophen (AAP) intoxication. The rats were given ethanol extracts of Sargassum ilicifolium, S. lanceolatum and S.swartzii orally at dose of 200mg/kg b.w. (body weight) daily for 14 days. These seaweed treated rats were then intoxicated with single intra-peritoneal dose of CCl or AAP on14th day. The administration of CCl and AAP caused significant (p<0.05) elevation in liver enzymes; alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and other biochemical parameters, bilirubin, glucose, triglyceride and kidney function markers: urea and creatinine. The pre-treatment with ethanol extracts of S. ilicifolium and S. swartzii protected the liver and kidney significantly (p<0.05) by lowering the elevated level of hepatic enzymes, liver and kidney function markers towards normal range. Sargassum species have also showed positive effect on serum glucose, total cholesterol and triglyceride.

[Ab] Resumo:Biosorption of Methyl Blue (MB), Fuchsin Acid (FA), Rhodamine B (RB), Methylene Blue (MEB), Bromocresol purple (BC) and Methyl Orange (MO) onto Sargassum ilicifolium was studied in a batch system. Effect of dye structure on biosorption by Sargassum ilicifolium was studied to define the correlation between chemical structure and biosorption capacity. Different dye groups such as triarylmethane (MB, FA and BC), monoazo (MO), thiazine (MEB) and xanthene (RB) were studied. At optimum experimental conditions for each dye, biosorption capacity was determined and compared. The results indicate that the chemical structure (triarylmethane, monoazo, thiazine, xanthene), number of sulfonic groups, basicity (element of chromophore group: S, N, O) and molecular weight of dye molecules influence their biosorption capacity. Experimental parameters such as biosorbent dose, pH, contact time, and initial dye concentration were optimized for each dye. The biosorption kinetic data were successfully described by the pseudo second-order model. The biosorption results were also analyzed by the Langmuir and Freundlich isotherms. Finally, biosorption capacities obtained using Sargassum ilicifolium were compared with the ones presented in the literature.

[Ab] Resumo:This work focused on the use of waste seaweed Sargassum fluitans (S. fluitans) as carbon source precursor to prepare nitrogen doped carbon dots (NCDs) by hydrothermal method. High resolution transmission electron microscopic (HR-TEM) studies revealed that the synthesized water soluble NCDs are in the size range of 2-8nm and exhibits excellent fluorescent properties with a quantum yield of 18.2%. Elemental nitrogen in NCDs was evidenced by X-ray photoelectron spectroscopy (XPS) and Fourier transformed infrared spectrum (FT-IR). The phytochemical analysis of S. fluitans using H NMR and C NMR revealed the presence of few amino acids which act as nitrogen source in the preparation of NCDs. Application of NCDs as fluorophore for double stranded DNA, single stranded DNA and RNA detection was highlighted in this study. Excellent fluorescent tagging abilities of NCDs with the biological nucleic acids were evidenced using gel electrophoresis. Significant increase in fluorescence was observed upon tagging of NCDs with nucleic acids and this particular phenomenon helps better in visualizing the nucleic acids. All three nucleic acids i.e. double stranded DNA, single stranded DNA and RNA showed similar phenomenon upon tagging with NCDs. Thus synthesized NCDs may be used as an alternate fluorophore for commercial toxic organic staining agents to visualize nucleic acids.

[Ab] Resumo:Pyrolysis of azolla, sargassum tenerrimum and water hyacinth were carried out in a fixed-bed reactor at different temperatures in the range of 300-450°C in the presence of nitrogen (inert atmosphere). The objective of this study is to understand the effect of compositional changes of various aquatic biomass samples on product distribution and nature of products during slow pyrolysis. The maximum liquid product yield of azolla, sargassum tenerrimum and water hyacinth (38.5, 43.4 and 24.6wt.% respectively) obtained at 400, 450 and 400°C. Detailed analysis of the bio-oil and bio-char was investigated using H NMR, FT-IR, and XRD. The characterization of bio-oil showed a high percentage of aliphatic functional groups and presence of phenolic, ketones and nitrogen-containing group. The characterization results showed that the bio-oil obtained from azolla, sargassum tenerrimum and water hyacinth can be potentially valuable as a fuel and chemicals.

[Ab] Resumo:The influence of various solvents (H O, CH OH, and C H OH) on product distribution and nature of products during hydrothermal liquefaction of sargassum tenerrimum algae has been examined. Hydrothermal liquefaction was performed using H O (260, 280 and 300°C) and organic solvents CH OH and C H OH (280°C) for 15min. The use of organic solvents significantly increased the yield of bio-oil. In the case of liquefaction with CH OH and C H OH, the bio-oil yield was 22.8 and 23.8wt.% respectively whereas the bio-oil yield was 16.33wt.% with H O. GC-MS analysis of the liquid products indicated the presence of various organic compounds including aromatics, nitrogenated and oxygenated compounds and higher selectivity amount of ester compounds were observed in the presence of alcoholic solvents. NMR and FT-IR showed that present of solvents have an effect on the decomposition of sargassum tenerrimum algae.

[Ab] Resumo:In this study, we aimed to use different strategies to further uncover the anti-angiogenic molecular mechanism of a fucoidan-like polysaccharide STPC2, isolated from brown alga . A desulfated derivative, STPC2-DeS, was successfully prepared and identified. The native polysaccharide and desulfated product were subjected to evaluate their anti-angiogenic effects. In the tube formation assay, STPC2 showed dose-dependent inhibition. In addition, STPC2 could distinctly inhibit the permeation of HUVEC cells into the lower chamber. Moreover, a significant reduction of microvessel density was observed in chick chorioallantoic membrane assay treated with STPC2. Meanwhile, STPC2 was found to repress the VEGF-induced neovessel formation in the matrigel plug assay in vivo. However, STPC2-DeS failed to suppress the anti-angiogenic activity via these in vitro and in vivo strategies. In addition, we demonstrated that STPC2 could significantly downregulate the phosphorylation of VEGFR2 and its related downstream Src family kinase, focal adhesion kinase, and AKT kinase. Furthermore, surface plasmon resonance assay revealed that STPC2 bound strongly to VEGF to interfere with VEGF-VEGFR2 interaction. Taken together, these results evidently demonstrated that STPC2 exhibited a potent anti-angiogenic activity through binding to VEGF via sulfated groups to impede VEGF-VEGFR2 interaction, thus affected the downstream signaling molecules.

[Ab] Resumo:Phlorotannins are a group of complex polymers of phloroglucinol (1,3,5-trihydroxybenzene), which are unique compounds from marine brown algae. In our present study, a procedure for extraction and enrichment of phlorotannins from with highly antioxidant potentials was established. After comparison of different extraction methods, the optimal extraction conditions were established as follows. The freeze-dried seaweed powder was extracted with 30% ethanol-water solvent with a solid/liquid ratio of 1:5 at temperature of 25 °C for 30 min. After extraction, the phlorotannins were fractioned by different solvents, among which the ethyl acetate fraction exhibited both the highest total phlorotannin content (88.48 ± 0.30 mg PGE/100 mg extract) and the highest antioxidant activities. The extracts obtained from these locations were further purified and characterized using a modified UHPLC-QQQ-MS method. Compounds with 42 different molecular weights were detected and tentatively identified, among which the fuhalol-type phlorotannins were the dominant compounds, followed by phlorethols and fucophlorethols with diverse degree of polymerization. Eckol-type phlorotannins including some newly discovered carmalol derivatives were detected in species for the first time. Our study not only described the complex phlorotannins composition in , but also highlighted the challenges involved in structural elucidation of these compounds.

[Ab] Resumo:Sargassum swartzii, marine macro brown alga, showed a high malachite green (MG) biosorption capacity in batch mode of operation. The analytical evidence from Fourier transform infrared spectra confirmed the involvement of amine group in the biosorption of MG and electrostatic interaction type of mechanism was proposed to occur between the amine group of dye and the cationic MG dye solution. Scanning electron micrograph shows the morphological features and the attachment of dye onto the biosorbent. pH edge experiment shows that biosorption capacity was maximum at pH 10. The effect of biosorbent concentration, pH, temperature, adsorption time was studied for the biosorption of MG using S. swartzii. Langmuir, Freundlich and Temkin models were used to describe the isotherm data, of which Langmuir model described the isotherm data with high coefficient of determination R = 0.999. The maximum dye uptake of 111.1 mg/g was reported at pH 10 based on Langmuir model. Kinetics and temperature profiles were evaluated and reported. Desorption study was carried out with 0.1 M HCl. Efforts were also made to continuously treat MG bearing wastewater using up-flow packed column. Investigations proved that S. swartzii is an excellent biosorbent for the sequestration of MG in aqueous media.
