[Ab] Resumo:Taxonomic studies were carried out on a carotenoid-producing strain, designated WV39 , isolated from the faeces of Antarctic penguins. Cells of strain WV39 were Gram-stain-negative, strictly aerobic, yellow and rod-shaped. 16S rRNA gene sequence analysis revealed that strain WV39 was closely related to Flavobacterium qiangtangense JCM 19739 (96.3 % similarity), Flavobacterium noncentrifugens NBRC 108844 (95.5 %) and Flavobacterium aquatile LMG 4008 (94.9 %). The predominant cellular fatty acids were iso-C15 : 0, iso-C15 : 0 3-OH and summed feature 3 (comprising iso-C15 : 0 2-OH and/or C16 : 1ω7c). Menaquinone-6 was the sole quinone identified, and the major pigment was zeaxanthin. The major polar lipid was phosphatidylethanolamine. DNA-DNA relatedness of strain WV39 with respect to its closest phylogenetic neighbours was 41.8 % for F. qiangtangense JCM 19739 , 25.5 % for F. aquatile LMG 4008 and 25.2 % for F. noncentrifugens NBRC 108844 . The DNA G+C content of strain WV39 was 39.8 mol%. Based on the phenotypic, chemotaxonomic and phylogenetic data, strain WV39 is concluded to represent a novel species of the genus Flavobacterium, for which the name Flavobacteriumkingsejongi sp. nov. is proposed. The type strain is WV39 (=KCTC 42908 =CECT 9085 ).

[Ab] Resumo:A rod-shaped, Gram-staining-negative, strictly aerobic, non-motile bacterium with no flexirubin-type pigment, designated as W201E , was isolated from an intertidal sandy beach in Antarctica. The organism formed faintly yellow, round colonies on marine agar 2216E. The strain required sea salts for growth and grew optimally in the presence of 2 % (w/v) NaCl at pH 7.0, 20 °C. Phylogenetic analysis based on 16S rRNA gene sequence indicated that strain W201E belonged to the genus Flavobacterium and showed the highest sequence similarity to Flavobacterium algicola NBRC 102673 (96.5 %). The major respiratory quinone was menaquinone 6, and the predominant fatty acids were iso-C15 : 1 G, iso-C15 : 0, iso-C15 : 0 3-OH and summed feature 3 (which comprises C16 : 1ω7c and/or C16 : 1ω6c). The polar lipids of strain W201E comprised one phosphatidylethanolamine, two unidentified aminolipids and three unidentified polar lipids. The DNA G+C content of strain W201E was 34.1 mol%. On the basis of the polyphasic analyses, this isolate was considered to represent a novel species in the genus Flavobacterium, for which the name Flavobacterium ovatum sp. nov. is proposed. The type strain is W201E (=KCTC 52693 =MCCC 1K03251 =CGMCC 1.16053 ).

[Ab] Resumo:A Gram-stain-negative, facultatively aerobic, motile-by-gliding, non-flagellated and rod-shaped bacterial strain, designated YSM-43 , was isolated from a tidal flat in Yeosu on the South Sea in the Republic of Korea. Strain YSM-43 grew optimally at 30 °C and in the presence of 1.0-2.0 % (w/v) NaCl. A neighbour-joining phylogenetic tree based on 16S rRNA gene sequences showed that strain YSM-43 fell within the clade comprising type strains of Flavobacterium species, clustering with the type strains of Flavobacterium jejuense and Flavobacterium jumunjinense. It exhibited 16S rRNA gene sequence similarity values of 97.20 and 97.15 % to the type strains of F. jejuense and F. jumunjinense, respectively, and of less than 96.59 % to the type strains of the other Flavobacterium species. Strain YSM-43 contained menaquinone-6 as the predominant menaquinone and iso-C15 : 0, iso-C17 : 0 3-OH, iso-C15 : 1 G and iso-C15 : 0 3-OH as the major fatty acids. The major polar lipids were phosphatidylethanolamine and one unidentified lipid. The DNA G+C content of strain YSM-43 was 29.8 mol% and its DNA-DNA relatedness values with type strains of F. jejuense and F. jumunjinense were 13 and 11 %, respectively. The differential phenotypic properties, together with the phylogenetic and genetic data, revealed that strain YSM-43 is separate from other recognized species of the genus Flavobacterium. On the basis of the data presented, strain YSM-43 is considered to represent a novel species of the genus Flavobacterium, for which the name Flavobacteriumsediminilitoris sp. nov. is proposed. The type strain is YSM-43 (=KACC 19435 =KCTC 62142 =NBRC 113020 ).

[Ab] Resumo:A Gram-staining-negative bacterial strain, designated CJ42 , was isolated from ginseng soil in Anseong, Republic of Korea. Cells were rod-shaped, yellow-pigmented, aerobic and devoid of flagella but showed gliding motility. Strain CJ42 grew optimally at 30 °C and pH 7.0 in the absence of NaCl on tryptic soy agar. Phylogenetic analysis based on the 16S rRNA gene sequence showed that strain CJ42 belonged to the genus Flavobacterium within the family Flavobacteriaceae and was most closely related to Flavobacterium tistrianum KCTC 42679 (97.3 % similarity). Flexirubin-type pigments were produced. The only respiratory quinone was menaquinone 6. The predominant polar lipids were phosphatidylethanolamine, an unknown aminolipid and two unknown lipids. The major fatty acids of strain CJ42 were iso-C15 : 0 and summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c). The G+C content of the genomic DNA was 30.7 mol%. Based on the polyphasic analyses, strain CJ42 represents a novel species in the genus Flavobacterium, for which the name Flavobacteriumfoetidum sp. nov. is proposed. The type strain is CJ42 (=KACC 19302=JCM 32085).

[Ab] Resumo:A Gram-stain-negative, yellow-pigmented, non-flagellated, gliding, rod-shaped, oxidase-negative and catalase-positive bacterium, designated SE14 , was isolated from soil on King George Island, South Shetland Islands, Antarctica. Strain SE14 grew at 4-25 °C (optimum, 20 °C), at pH 6.0-9.0 (optimum, pH 7.0-7.5) and with 0-3.0 % NaCl (optimum, 1.0-1.5 %), and could not produce flexirubin-type pigments. 16S rRNA gene sequence analysis showed the the isolate belonged to the genus Flavobacterium. Strain SE14 had the highest 16S rRNA gene sequence similarity to Flavobacterium antarcticum, F. tegetincola and F. degerlachei with 95.8, 95.5 and 95.2 %, respectively. The strain SE14 consisted of a clade with Flavobacteriumnoncentrifugens (16S rRNA gene sequence similarity 94.9 %) and F. qiangtangense (16S rRNA gene sequence similarity 94.2 %) and simultaneously formed a distinct phyletic lineage in the neighbour-joining phylogenetic tree. Polar lipids of the strain included phosphatidylethanolamine and four unidentified aminolipids. Strain SE14 contained anteiso-C15 : 0, iso-C15 : 0 and a mixture of iso-C15 : 0 2-OH and/or C16 : 1ω7c as the main fatty acids, and the only respiratory quinone was menaquinone-6. The genomic DNA G+C content was 42.3 mol%. The polyphasic taxonomic study revealed that strain SE14 belongs to a novel species within the genus Flavobacterium , and the name Flavobacterium phocarum sp. nov. is proposed. The type strain is SE14 (=CCTCC AB 2017225 =KCTC 52612 ).

[Ab] Resumo:During a study of petroleum hydrocarbon-degrading bacteria, a yellow-coloured, Gram-stain-negative, non-motile and rod-shaped bacterium, designated strain Brt-M , was isolated from oil-contaminated soil of Biratnagar, Morang, Nepal. Strain Brt-M was able to grow at 15-45 °C, pH 5.0-9.0 and 0-1 % (w/v) NaCl concentration. The strain was characterized by multiple taxonomic approaches. Based on 16S rRNA gene sequence analysis, strain Brt-M belonged to the genus Flavobacterium and shared highest sequence similarity with Flavobacterium cloacae wh15 (95.69 %) and Flavobacterium anatoliense MK3 (94.91 %). The only respiratory quinone was MK-6; the major polar lipid was phosphatidylethanolamine; and the predominant fatty acids were iso-C15 : 0, summed feature 9 (iso-C17 : 1ω9c and/or C16 : 010-methyl), iso-C17 : 0 3-OH and summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c). The DNA G+C content was 37.2 mol%. The morphological, physiological, chemotaxonomic and phylogenetic analyses clearly distinguished this strain from its closest phylogenetic neighbours. Thus, strain Brt-M represents a novel species of the genus Flavobacterium, for which the name Flavobacteriumnaphthae sp. nov. is proposed. The type strain is Brt-M (=KEMB 9005-692 =KACC 19393 =JCM 32171 ).

[Ab] Resumo:Emergence of multidrug-resistant bacteria causes an urgent need for new generation of antibiotics, which may have a different mechanism of inhibition or killing action from the existing. Here, we report on the design, synthesis, and biological evaluation of thirty-nine coumarin derivatives in order to solve the antibacterial resistance by targeting at the inhibition of biosynthesis pathway of fatty acids. Their antibacterial activities against Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae, and Flavobacterium cloumnare are tested and action mechanism against the key enzyme in bacterial fatty acid synthesis pathway are studied. The results show that compounds 13 and 18 have potent and broad spectrum antimicrobial activity. In addition, 9, 14 and 19 show eminent antimicrobial efficacy toward S. aureus, S. agalactiae, and F. cloumnare. Mechanistically, coumarin derivatives display the antibacterial activity via the control of FabI and FabK function. The structure-activity relationship analysis indicate that the length of linker and imidazole substitute group could significantly influence the antimicrobial activity, as well as the inhibitory activity against FabI and FabK. The structural optimization analysis of coumarin suggest that derivatives 9, 13, 14, 18 and 19 could be a viable way of preventing and controlling bacteria and considered as promising lead compounds for the development of commercial drugs.

[Ab] Resumo:Antagonistic coevolution of parasite infectivity and host resistance may alter the biological functionality of species, yet these dynamics in nature are still poorly understood. Here we show the molecular details of a long-term phage-bacterium arms race in the environment. Bacteria (Flavobacterium columnare) are generally resistant to phages from the past and susceptible to phages isolated in years after bacterial isolation. Bacterial resistance selects for increased phage infectivity and host range, which is also associated with expansion of phage genome size. We identified two CRISPR loci in the bacterial host: a type II-C locus and a type VI-B locus. While maintaining a core set of conserved spacers, phage-matching spacers appear in the variable ends of both loci over time. The spacers mostly target the terminal end of the phage genomes, which also exhibit the most variation across time, resulting in arms-race-like changes in the protospacers of the coevolving phage population.Arms races between phage and bacteria are well known from lab experiments, but insight from field systems is limited. Here, the authors show changes in the resistance and CRISPR loci of bacteria and the infectivity, host range and genome size of phage over multiple years in an aquaculture environment.

[Ab] Resumo:A Gram-staining-negative, aerobic, rod-shaped bacterium, EA-12 , was isolated from activated sludge in Fujian Province, PR China. The results of phylogenetic analysis of the 16S rRNA gene sequences indicated that it was closely related to Flavobacterium pectinovorum DSM 6368 (97.5 %), Flavobacterium banpakuense 15F3 (97.0 %) and Flavobacterium arsenitoxidans S2-3H (96.9 %). Cells grew at 15-37 °C (optimum, 25 °C), at pH 5.0-9.0 (optimum, pH 7.0) and in the presence of 0-1.0 % (w/v) NaCl. The strain contained MK-6 as the major menaquinone and the major cellular fatty acids were iso-C15 : 0, summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c) and C16 : 0 3-OH. The polar lipids consisted of phosphatidylethanolamine and one unidentified phospholipid. The DNA G+C content was 36.1 mol% (Tm). The DNA-DNA relatedness between strain EA-12 and F. pectinovorum DSM 6368 was 38.6 %. On the basis of phenotypic, chemotaxonomic and phylogenetic comparisons with relatives and DNA-DNA relatedness values, it is concluded that EA-12 represents a novel species within the genus Flavobacterium, for which the name Flavobacterium quisquiliarum sp. nov. is proposed. The type strain is EA-12 (=CGMCC 1.15345 =NBRC 111769 ).

[Ab] Resumo:A Gram-stain-negative, aerobic, yellow-pigmented, non-flagellated, non-gliding, rod-like, oxidase- and catalase-positive bacterium, designated A2-1 , was isolated from soil on Ardley Island, South Shetland Islands, Antarctica. Strain A2-1 grew at 4-22 °C (optimum, 10 °C), at pH 6.0-8.0 (optimum, pH 6.5) and with 0-1.5 % NaCl (optimum, 0.5 %), but could not produce flexirubin-type pigments. 16S rRNA gene sequence analysis showed that the isolates belonged to the genus Flavobacterium. Strain A2-1 had the highest 16S rRNA gene sequence similarity to Flavobacterium cucumis, F. ahnfeltiae and F. cheniae with 95.7, 95.6 and 95.4 %, respectively. The strain A2-1 consisted of a clade with F. cucumis and F. cheniae and simultaneously formed a distinct phyletic lineage in the neighbour-joining phylogenetic tree. Polar lipids of the strain included phosphatidylethanolamine (PE), four unidentified aminolipids and one unidentified lipid. The strain A2-1 contained anteiso-C15 : 0 (20.2 %), iso-C15 : 0 (16.2 %) and C15 : 1 G (11.0 %) as the main fatty acids and the only respiratory quinone was menaquinone MK-6. The genomic DNA G+C content was 34.0 mol%. The polyphasic taxonomic study revealed that the strain A2-1 belongs to a novel species within the genus Flavobacterium and the name Flavobacterium ardleyense sp. nov. is proposed. The type strain is A2-1 (=CCTCC AB 2017157 =KCTC 52644 ).
