[Ab] Resumo:Two Gram-staining-positive, strictly aerobic bacilli, designated as strains Ma50-5 and Ma50-6 , were isolated from the hydrothermal sediments of Manus Basin in the western Pacific Ocean. Based on 16S rRNA gene sequence, strains Ma50-5 and Ma50-6 were most closely related to Bacillus alveayuensis (97.0 and 97.2 % identity, respectively). The 16S rRNA gene sequence identity between strains Ma50-5 and Ma50-6 was 97.4 %. The identities between strains Ma50-5 and Ma50-6 and other closely related organisms were below 97.0 %. The G+C contents of the genomic DNA of strains Ma50-5 and Ma50-6 were 43.4 and 47.6 mol%, respectively. The major fatty acids (>10 %) of both strains were iso-C15 : 0 and iso-C17 : 0. The predominant isoprenoid quinone detected in both strains was menaquinone-7. Phylogenetic, physiological, biochemical and morphological analyses suggested that strains Ma50-5 and Ma50-6 represent two novel species of the genus Bacillus, for which the names Bacillus kexueae sp. nov. (type strain Ma50-5 =KCTC 33881 =CCTCC AB 2017020 ) and Bacillus manusensis sp. nov. (type strain Ma50-6 =KCTC 33882 =CCTCC AB 2017019 ), respectively, are proposed.

[Ab] Resumo:Methomyl (S-methyl-1-N-[(methylcarbamoyl)oxy]thioacetimidate) is a carbamate pesticide. It has been noted that in some cases of methomyl poisoning, methomyl is either not detected or detected only in low concentrations in the blood of the victims. However, in such cases, methomyl is detected at higher concentrations in the vitreous humor than in the blood. This indicates that methomyl in the blood is possibly decomposed after death. However, the reasons for this phenomenon have been unclear. We have previously reported that methomyl is decomposed to dimethyl disulfide (DMDS) in the livers and kidneys of pigs but not in their blood. In addition, in the field of forensic toxicology, it is known that some compounds are decomposed or produced by internal bacteria in biological samples after death. This indicates that there is a possibility that methomyl in blood may be decomposed by bacteria after death. The aim of this study was therefore to investigate whether methomyl in blood is decomposed by bacteria isolated from human stool. Our findings demonstrated that methomyl was decomposed in human stool homogenates, resulting in the generation of DMDS. In addition, it was observed that three bacterial species isolated from the stool homogenates, Bacillus cereus, Pseudomonas aeruginosa, and Bacillus sp., showed methomyl-decomposing activity. The results therefore indicated that one reason for the difficulty in detecting methomyl in postmortem blood from methomyl-poisoning victims is the decomposition of methomyl by internal bacteria such as B. cereus, P. aeruginosa, and Bacillus sp.

[Ab] Resumo:Present work describes the in vitro antibacterial evaluation of some new amino acid conjugated antimicrobial drugs. Structural modification was attempted on the three existing antimicrobial pharmaceuticals namely trimethoprim, metronidazole, isoniazid. Twenty one compounds from seven series of conjugates of these drugs were synthesized by coupling with some selected Boc-protected amino acids. The effect of structural features and lipophilicity on the antibacterial activity was investigated. The synthesized compounds were evaluated against five standard American type culture collection (ATCC) i.e. Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi strains of bacteria. Our results identified a close relationship between the lipophilicity and the activity. Triazine skeleton proved beneficial for the increase in hydrophobicity and potency. Compounds with greater hydrophobicity have shown excellent activities against Gram-negative strains of bacteria than Gram-positive. 4-amino unsubstituted trimethoprim-triazine derivative 7b have shown superior activity with MIC = 3.4 µM (2 µg/mL) for S. aureus and 1.1 µM (0.66 µg/mL) for E. coli. The synthesized compounds were also evaluated for their urease inhibition study. Microbial urease from Bacillus pasteurii was chosen for this study. Triazine derivative 7a showed excellent inhibition with IC = 6.23 ±â€¯0.09 µM. Docking studies on the crystal structure of B. pasteurii urease (PDB ID 4UBP) were carried out.

[Ab] Resumo:Two novel (18C and 6C) Gram-stain-positive, rod shaped, motile and endospore-forming bacterial strains were isolated from Lonar soda lake, India. Based on 16S rRNA gene sequence analysis, strains 18C and 6C were identified as belonging to the class Firmibacteria, and were most closely related to Bacillus cohnii KCTC 3572 (99.3 and 99.9%, respectively), Bacillus zhanjiangensis KCTC 13713 (97.4 and 98.0%, respectively), Bacillus halmapalus LMG 17950 (97.0 and 97.6%, respectively) and other members in the genus Bacillus (<97.0%). However, the DNA-DNA relatedness between 18C and 6C and B. cohnii KCTC 3572 (49.6 ± 0.9 and 51.6 ± 0.7, respectively), B. zhanjiangensis KCTC 13713 (42.9 ± 0.8 and 47.1 ± 0.3, respectively) and B. halmapalus LMG 17950 (39.9 ± 0.8 and 40.8 ± 0.3, respectively) indicated that the novel strains were distantly related to these strains. Further, the high 16S rRNA gene sequence similarity (100%) and DNA-DNA relatedness (90 ± 5%) suggested that strains 18C and 6C were members of a genomospecies. The strains grew optimally at a pH of 7.5 with 2-3% (w/v) NaCl and temperature of 37 °C. Strains 18C and 6C were catalase and oxidase negative. The cell wall of strain 18C contained meso-diaminopimelic acid as the diagnostic diamino acid, which was in contrast with its nearest neighbour B. cohnii KCTC 3572 , which contained ornithine and aspartic acid. Polar lipids include diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylethanolamine (PE), an unknown phospholipid (PL) and three unknown lipids (L1-3). The predominant isoprenoid quinone was MK-7. iso-C (32.5%) was the predominant fatty acid and significant proportions of anteiso-C (19.5%), C (11.5%), iso-C (9.5%) and anteiso-C (6.3%) were also detected. The DNA G + C content of strains 18C and 6C were 39.3 and 39.2 mol%, respectively. The results of molecular, biochemical and chemotaxonomic tests showed a clear differentiation of strains 18C and 6C from all other members of the genus Bacillus, for which the name Bacillus catenulatus sp. nov. is proposed. The type strain is 18C (=KCTC 33781 = CGMCC 1.15475 ).

[Ab] Resumo:F -ATPase (F ) is an efficient rotary protein motor, whose reactivity is modulated by the rotary angle to utilize thermal fluctuation. In order to elucidate how its kinetics are affected by the change in the fluctuation, we have extended the reaction-diffusion formalism [R. Watanabe et al., Biophys. J., 2013, 105, 2385] applicable to a wider range of temperatures based on experimental data analysis of F derived from thermophilic Bacillus under high ATP concentration conditions. Our simulation shows that the rotary angle distribution manifests a stronger non-equilibrium feature as the temperature increases, because ATP hydrolysis and P release are more accelerated compared with the timescale of rotary angle relaxation. This effect causes the rate coefficient obtained from dwell time fitting to deviate from the Arrhenius relation in P release, which has been assumed in the previous activation thermodynamic quantities estimation using linear Arrhenius fitting. Larger negative correlation is also found between hydrolysis and P release waiting time in a catalytic dwell with the increase in temperature. This loss of independence between the two successive reactions at the catalytic dwell sheds doubt on the conventional dwell time fitting to obtain rate coefficients with a double exponential function at temperatures higher than 65 °C, which is close to the physiological temperature of the thermophilic Bacillus.

[Ab] Resumo:A pot experiment was conducted to evaluate the potential of two plant growth promoting rhizobacteria (PGPR) viz. Bacillus sp. CIK-516 and Stenotrophomonas sp. CIK-517Y for improving the growth and Ni uptake of radish (Raphanus sativus) in the presence of four different levels of Ni contamination (0, 50, 100, 150 mg Ni kg soil). Plant growth, dry biomass, chlorophyll and nitrogen contents were significantly reduced by the exogenous application of Ni, however, bacterial inoculation diluted the negative impacts of Ni stress on radish by improving these parameters. PGPR strain CIK-516 increased root length (9-27%), shoot length (8-27%), root dry biomass (2-32%), shoot dry biomass (9-51%), root girth (6-48%), total chlorophyll (4-38%) and shoot nitrogen contents (11-15%) in Ni contaminated and non-contaminated soils. Positive regulation of chlorophyll and nitrogen contents by the inoculated plants shows plant tolerance mechanism of Ni stress. Bacterial strain (CIK-516) exhibited indole acetic acid and 1-amino-cyclopropane-1-carboxylate deaminase potentials which would have helped radish plant to stabilize in Ni contaminated soil and thereby increased Ni uptake (24-257 in shoot and 58-609 in root mg kg dry biomass) and facilitated accumulation in radish (bioaccumulation factor = 0.6-1.7) depending upon soil Ni contamination. Based on the findings of this study, it might be suggested that inoculation with bacterial strain CIK-516 could be an efficient tool for enhanced Ni phytoextraction in radish.

[Ab] Resumo:A novel aerobic, Gram-stain-positive, sporogenous, rod-shaped bacterial strain, 7578-1 , was isolated from ripened Pu'er tea. Based on 16S rRNA gene sequence similarity comparisons, strain 7578-1 was grouped into the genus Bacillus and appeared to be closely related to the type strains Bacillus shackletoniiLMG 18435 (98.4 %), Bacillus acidicolaDSM 14745 (97.6 %), Bacillus paralicheniformis KACC 18426 (97.2 %) and Bacillus ginsengihumi KCTC 13944 (96.7 %). The fatty acid profile containing the major fatty acids, iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0 supported the allocation of strain 7578-1 to the genus Bacillus. The strain had a cell-wall type A1γ peptidoglycan with meso-diaminopimelic acid as the diagnostic diamino acid. The major menaquinone was MK-7 (95 %). The predominant polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylethanolamine, one unidentified phospholipid and one unidentified lipid. The average nucleotide identity values between strain 7578-1 and its most closely related species were 67.8-82.4 % by OrthoANIu analysis. The DNA-DNA relatedness value between strain 7578-1 and the type strains of closely related species were 17-39 %, again indicating that strain 7578-1 represented a novel species in the genus Bacillus. The DNA G+C content of strain 7578-1 was 36.0 mol%. On the basis of the presented polyphasic evidence, strain 7578-1 is considered to represent a novel species of the genus Bacillus, for which we propose the name Bacillus camelliae sp. nov. The type strain is 7578-1 (=CGMCC 1.15374 =KCTC 33845 ).

[Ab] Resumo:A Gram-stain-positive, alkaliphilic bacterium, designated EGI 80668 , was isolated from a Tamarix cone soil in Xinjiang, north-west China. Cells were facultatively anaerobic, terminal endospore-forming and motile by means of peritrichous flagella. Colonies were yellowish and the cells showed oxidase-negative and catalase-positive reactions. Strain EGI 80668 grew at pH 8.0-10.0 and with 0-10 % (w/v) NaCl (optimally at pH 9.0 and with 1-2 % NaCl) on marine agar 2216. The predominant menaquinone was MK-7. The major fatty acids were anteiso-C17 : 0 and anteiso-C15 : 0. The cellular polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, four unknown phospholipids and one unknown aminophospholipid. The G+C content of the genomic DNA was 38.3 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain EGI 80668 was affiliated to the genus Bacillus. The highest 16S rRNA gene sequence similarity between strain EGI 80668 and a member of the genus Bacillus was 96.83 % with Bacillus cellulosilyticus JCM 9156 . A polyphasic taxonomic study based on morphological, physiological, biochemical and phylogenetic data indicated that strain EGI 80668 represents a novel species of the genus Bacillus, for which the name Bacillus tamaricis sp. nov. (type strain EGI 80668 =KCTC 33703 =CGMCC 1.15917 ) is proposed.

[Ab] Resumo:Cold shock proteins (Csp) constitute a family of ubiquitous small proteins that act as RNA-chaperones to avoid cold-induced termination of translation. All members contain two subdomains composed of 2 and 3 ß-strands, respectively, which are connected by a hinge loop and fold into a ß-barrel. Bacillus caldolyticus Csp (BcCsp) is one of the most studied members of the family in terms of its folding, function, and structure. This protein has been described as a monomer in solution, although a recent crystal structure showed dimerization via domain swapping (DS). In contrast, other cold shock proteins of the same fold are known to dimerize in a nonswapped arrangement. Hypothesizing that reducing the size of the hinge loop may promote swapping as in several other DS proteins with different folds we deleted two residues from these region (BcCsp∆36-37), leading to a protein in monomer-dimer equilibrium with similar folding stability to that of the wild-type. Strikingly, the crystal structure of BcCsp∆36-37 revealed a nonswapped dimer with its interface located at the nucleic acid-binding surface, showing that the deletion led to structural consequences far from the perturbation site. Concomitantly, circular dichroism experiments on BcCsp∆36-37 demonstrated that binding of the oligonucleotide hexathymidine disrupts the dimer. Additionally, HDXMS shows a protective effect on the protein structure upon dimerization, where the resulting interactions between ligand-binding surfaces in the dimer reduced the extent of exchange throughout the whole protein. Our work provides evidence of the complex interplay between conformational dynamics, deletions, and oligomerization within the Csp protein family. [Ab] Resumo:Bacillus velezensis LM2303 is a biocontrol strain with a broad inhibitory spectrum against plant pathogens, isolated from the dung of wild yak inhabited Qinghai-Tibet plateau, China. Here we present its complete genome sequence, which consists of a single, circular chromosome of 3,989,393bp with a 46.68% G+C content. Genome analysis revealed genes encoding specialized functions for the biosynthesis of antifungal metabolites and antibacterial metabolites, the promotion of plant growth, the alleviation of oxidative stress and nutrient utilization. And the biosynthesis of antimicrobial metabolites in strain LM2303 was confirmed by biochemical analysis, while its plant growth promoting traits were confirmed by inoculation tests. Our results will establish a better foundation for further studies and biocontrol application of B. velezensis LM2303.
