[Ab] Resumo:A novel Gram-stain-positive bacterial strain, designated as K13 , was isolated from compost and characterized using a polyphasic approach to determine its taxonomic position. On the basis of 16S rRNA gene sequence analysis, the strain showed highest similarity (93.8 %) to Paenibacillus nanensis MX2-3 . Cells of strain K13 were aerobic, motile rods. The major fatty acids were anteiso C15 : 0 (34.4 %), iso C16 : 0 (17.3 %) and C16 : 0 (10.0 %). The major menaquinone was MK-7, the polar lipid profile included diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylserine and an aminophospholipid. The DNA G+C content was 52.3 %. Based on phenotypic, including chemotaxonomic characteristics and analysis of the 16S rRNA gene sequences, it was concluded that strain K13 represents a novel genus, for which the name Xylanibacillus gen. nov., sp. nov. is proposed. The type species of the genus is Xylanibacillus composti, the type strain of which is strain K13 (=DSM 29793 =NCAIM B.02605 ).

[Ab] Resumo:The gastrointestinal (GI) habitat of ruminant and non-ruminant animals sustains a vast ensemble of microbes that are capable of utilizing lignocellulosic plant biomass. In this study, an indigenous swine (Zovawk) and a domesticated goat (Black Bengal) were investigated to isolate bacteria having plant biomass degrading enzymes. After screening and enzymatic quantification of eighty-one obtained bacterial isolates, Serratia rubidaea strain DBT4 and Aneurinibacillus aneurinilyticus strain DBT87 were revealed as the most potent strains, showing both cellulase and xylanase production. A biomass utilization study showed that submerged fermentation (SmF) of D2 (alkaline pretreated pulpy biomass) using strain DBT4 resulted in the most efficient biomass deconstruction with maximum xylanase (11.98 U/mL) and FPase (0.5 U/mL) activities (55°C, pH 8). The present study demonstrated that bacterial strains residing in the gastrointestinal region of non-ruminant swine are a promising source for lignocellulose degrading microorganisms that could be used for biomass conversion.

[Ab] Resumo:Nonpathogenic soil bacteria can colonize the rhizosphere and induce unique plant phenotypes that may influence plant-insect interactions. However, few studies have considered the influences of bacteria-plant interactions on insect feeding and oviposition. The objective of this study was to determine how rhizobacterial inoculation of bermudagrass affects larval development and ovipositional behaviors of the fall armyworm (Spodoptera frugiperda J.E. Smith). Eight blends of rhizobacteria known to induce root or shoot growth in grasses were applied weekly to hybrid bermudagrass for 5 wk. Oviposition was evaluated in two no-choice trials with bacteria-treated, fertilized, or nontreated grass. Grass blades from these treatments were extracted in polar and nonpolar solvents and assayed for oviposition responses. Another experiment compared the development of fall armyworm larvae on bermudagrass treated with each of the eight rhizobacterial blends for 5 wk to larvae fed nontreated bermudagrass. Females deposited more eggs on nontreated and fertilized grass and ≤34% of eggs on grass treated with rhizobacterial blends. Moths exposed to polar and nonpolar extracts were unable to reproduce these results. Larval and pupal weights at days 10 and 12 and the number of adults to eclose were lower for larvae fed some, but not all, bacteria-treated bermudagrass relative to controls. This is one of the few studies to investigate plant-microbe-insect interactions in an economically important system. Although the effects noted with fall armyworm are limited, induced changes in roots also reported for these bacteria may have greater utility than foliar changes for mediating interactions with biotic or abiotic stresses.

[Ab] Resumo:A thermophilic poly(L-lactide)-degrading Gram-stain-positive filamentous bacterial strain that develops single spores on the aerial mycelium was isolated from forest soil at Srinagarind Dam, Kanchanaburi Province, Thailand. The results of a polyphasic taxonomic study showed that our isolate had characteristics typical of members of the genus Polycladomyces. The isolate grew aerobically at an optimum temperature of 50-55 °C and optimal pH 6-7. Meso-diaminopimelic acid was present as the diagnostic diamino acid in the peptidoglycan but no characteristic sugars are detected. The predominant menaquinone was MK-7. The diagnostic phospholipids were phosphatidylethanolamine, phosphatidylmethylethanolamine diphosphatidylglycerol, phosphatidylglycerol and phosphatidylserine. The predominant cellular fatty acid was iso-C15 : 0. The DNA G+C content of strain KSR 13T was 53.4 mol%. The 16S rRNA gene sequence analysis also indicated that strain KSR 13T belonged to the genus Polycladomyces, being most closely related to Polycladomyces abyssicola JIR-001T (99.2 %). The DNA-DNA relatedness values that distinguished KSR 13T from P. abyssicola JIR-001T were 17.8-32.1 %, which were significantly below the 70 % cutoff value recommended for species delineation. Following an evaluation of phenotypic, chemotaxonomic and genotypic studies, the new isolate is proposed as a novel species and named Polycladomyces subterraneus sp. nov. The type strain is KSR 13T (=BCC 50740T=NBRC 109332T).

[Ab] Resumo:A novel themophilic bacterium, designated YIM 79486T, was isolated from a sediment sample collected from Jinze hot spring in Tengchong county, Yunnan province, south-west China. Phylogenetic analysis based on 16S rRNA gene sequence analysis revealed that strain YIM 79486T should be assigned to the genus Laceyella and formed a monophyletic clade with the type strain Laceyella putida KCTC 3666T (98.7 % similarity). Strain YIM 79486T formed white aerial mycelium and brown substrate mycelium. Abundant endospores were produced on short sporophores. Cell-wall peptidoglycan contained meso-diaminopimelic acid. The predominant menaquinones were MK-9 and MK-8. The genomic DNA G+C content observed for strain YIM 79486T was 47.8 mol%. Based on low DNA-DNA hybridization data, chemotaxonomic characteristics and differential physiological properties, strain YIM 79486T is considered to represent a novel species within the genus Laceyella, for which the name Laceyella thermophila sp. nov. is proposed. The type strain is YIM 79486T (=CCTC AB 2015040T=NBRC 110772T).

[Ab] Resumo:Alicyclobacillus acidoterrestris is a thermotolerant bacterium able to grow in fruit juices and drinks, as the spoilage by Alicyclobacillus in the final product does not product any gas, but leads to a "medicine flavor" due to the formation of guaicol. Also, its detection is a challenge for the quality control departments, because it takes several days to get the results of traditional microbiology methods. This study aimed at developing a more accurate electrical impedance technique for the detection of A. acidoterrestris in concentrated apple juice. Samples of apple juice were inoculated with A. acidoterrestris spores isolated from a peach and grape juice. For the spore germination, several heat-shock treatments were tested (80 °C/10 min, 70 °C/20 min and 60 °C/30 min). Direct and indirect electrical impedance was applied to detect and quantify the microorganism in the inoculated apple juice, using BAT broth and Bimedia 002A (pH 4). The 80 °C/10 min treatment was selected for spore activation. The valid electrical impedance technique was the indirect method in BAT broth, which measured the changes in the impedance through the formation of CO . In addition, a positive correlation (r = 0.98, R = 0.97) was observed between the classical microbiology (BAM agar) and the indirect impedance method.

[Ab] Resumo:A Gram-stain-positive, aerobic, motile, endospore-forming bacterium, designated strain J15A17T, was isolated from sediment of the South China Sea. The strain was oxidase-positive and catalase-negative. Optimal growth occurred at 33 °C, pH 7.5 and in the presence of 3 % (w/v) NaCl. On the basis of 16S rRNA gene sequence analysis, the strain showed closest similarity (92.8 %) to Paenibacillus puldeungensis strain CAU 9324T. Phylogenetic analysis based on 16S rRNA gene sequences showed that the isolate forms a separate branch within the family Paenibacillaceae, with the genus Cohnella as the most closely related genus. The DNA G+C content of strain J15A17T was 37.4 mol%. The strain contained MK-7 as the sole respiratory quinone; anteiso-C15 : 0 and iso-C16 : 0 were the major cellular fatty acids; and its polar lipid pattern consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, glycolipid and four unidentified phospholipids. The strain displayed the peptidoglycan type A4α l-Lys-d-Asp in the cell wall. Phylogenetic, physiological, biochemical and morphological differences between strain J15A17T and its closest relatives in the genera Cohnella, Fontibacillus and Paenibacillus suggest that strain J15A17T (=KCTC 33759T=MCCC 1H00137T) represents the type strain of a novel species in a new genus within the family Paenibacillaceae, Chengkuizengella sediminis gen. nov. sp. nov.

[Ab] Resumo:Two bacterial strains (1-10M-8-7-50T and 1M) were isolated from lagoon sediments. The strains were obligately aerobic, Gram-stain-positive, spore-forming, rod-shaped and motile. Both strains were able to grow at 25-60 ËšC (optimum 40-42 ËšC), at pH 6.0-8.8 (optimum pH 7.0-7.5) and with 0-2.0 % (w/v) (optimum 0-0.8 %) NaCl. The predominant cellular fatty acids were iso-C15 : 0 and iso-C15 : 0 2-OH, and the cell-wall peptidoglycan contained meso-diaminopimelic acid, glutamic acid, glycine and alanine. Phosphatidyl-N-methylethanolamine, phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylglycerol, two unidentified phosphoglycolipids and three unidentified lipids were present as polar lipids. Genomic DNA G+C content was 41.9 mol%. 16S rRNA gene sequence comparisons of both strains indicated that they belong to the genus Aneurinibacillus within the family Paenibacillaceae of the class Bacilli. Both strains had a sequence similarity of 97.6 % with Aneurinibacillus migulanus B0270T, 97.6 % with Aneurinibacillus aneurinilyticus ATCC 12856T, 97.5 % with 'Aneurinibacillus humi' U33 and <97 % with other members of the genus Aneurinibacillus. The DNA-DNA reassociation value between strain 1-10M-8-7-50T and 1M was >90 %, while strains 1-10M-8-7-50T and 1M were only 35.1±1, 29.8±1 and 24.5±1 % related to A. migulanus KACC 18173T, A. aneurinilyticus KACC 18174T and 'A. humi' U33, respectively. Distinct morphological, physiological and genotypic differences from the previously described taxa support the classification of strain 1-10M-8-7-50T as a representative of a novel species in the genus Aneurinibacillus, for which the name Aneurinibacillus sediminis sp. nov. is proposed. The type strain is 1-10M-8-7-50T (=KEMB 563-460T=JCM 31819T).

[Ab] Resumo:A Gram-stain-positive, lactic acid-producing bacterium designed strain MK21-7T, was isolated from tree bark collected from the north east of Thailand. This strain was a facultatively anaerobic spore-forming rod that was catalase-negative. It contained meso-diaminopimelic acid in the cell wall peptidoglycan and had seven isoprene units (MK-7) as the predominant menaquinone. Major fatty acids of MK21-7T were anteiso-C17 : 0, iso-C16 : 0, anteiso-C15 : 0 and C18 : 1ω9c. Polar lipids were phosphatidglycerol, diphosphatidylglycerol, an unknown phospholipid, three unknown glycolipids and an unknown lipid. The results of 16S rRNA gene sequence analysis indicated that it represented a member of the genus Sporolactobacillus. MK21-7T showed the highest 16S rRNA gene sequence similarity to Sporolactobacillus terrae NBRC 101527T with 98.4 % similarity and exhibited 97.6 % similarity with Sporolactobacillus kofuensis NRIC 0334T, 97.5 % with Sporolactobacillus laevolacticus NRIC 0361T, 97.3 % with Sporolactobacillus nakayamaesubsp.nakayamae NRIC 0347T and 97.1 % with Sporolactobacillus nakayamaesubsp.racemicus NBRC 101524T. Analysis of the phylogenetic relationship based on 16S rRNA and gyrB gene sequencing revealed that the position of MK21-7T was clearly separated from all related species of the genus Sporolactobacillus. It had low DNA-DNA relatedness (22.8-57.2 %) with S. terrae NBRC 101527T and related type strains. The DNA G+C content was 43.1 mol%. On the basis of the results of the phenotypic, genotypic and chemotaxonomic studies, MK21-7T should be classified as representing a novel species of the genus Sporolactobacillus for which the name Sporolactobacillus shoreicorticis sp. nov. is proposed. The type strain is MK21-7T (=NBRC 111517T=LMG 29111T=TISTR 2466T).

[Ab] Resumo:Plant-derived protein sources are the most relevant substitutes for fishmeal in aquafeeds. Nevertheless, the effects of plant based diets on the intestinal microbiome especially of juvenile Rainbow trout (Oncorhynchus mykiss) are yet to be fully investigated. The present study demonstrates, based on 16S rDNA bacterial community profiling, that the intestinal microbiome of juvenile Rainbow trout is strongly affected by dietary plant protein inclusion levels. After first feeding of juveniles with either 0%, 50% or 97% of total dietary protein content derived from plants, statistically significant differences of the bacterial gut community for the three diet-types were detected, both at phylum and order level. The microbiome of juvenile fish consisted mainly of the phyla Proteobacteria, Firmicutes, Bacteroidetes, Fusobacteria and Actinobacteria, and thus fits the salmonid core microbiome suggested in previous studies. Dietary plant proteins significantly enhanced the relative abundance of the orders Lactobacillales, Bacillales and Pseudomonadales. Animal proteins in contrast significantly promoted Bacteroidales, Clostridiales, Vibrionales, Fusobacteriales and Alteromonadales. The overall alpha diversity significantly decreased with increasing plant protein inclusion levels and with age of experimental animals. In order to investigate permanent effects of the first feeding diet-type on the early development of the microbiome, a diet change was included in the study after 54 days, but no such effects could be detected. Instead, the microbiome of juvenile trout fry was highly dependent on the actual diet fed at the time of sampling.
