[Ab] Resumo:A Gram-stain-negative, aerobic, motile, flagellated rod-shaped bacterium, designated ST58-10 , was isolated from an estuarine sediment in the Republic of Korea. The strain was able to degrade benzene. Growth of strain ST58-10 was observed at 4-35 °C (optimum, 20-25 °C), pH 5-9 (optimum, pH 7-8) and 1-8 % NaCl (optimum, 3 %). Phylogenetic analyses based on 16S rRNA gene sequences showed that strain ST58-10 formed a phyletic lineage within the genus Marinobacterium of the family Oceanospirillaceae. Strain ST58-10 was most closely related to Marinobacterium profundum PAMC 27536 (99.6 %) and Marinobacterium rhizophilum CL-YJ9 (98.3 %), and to other members of the genus Marinobacterium(94.5-91.5 %). However, the mean value estimated by using the Genome-to-Genome Distance Calculator was 50.6±7.4 % with M. profundum PAMC 27536 and 30.9±2.8 with M. rhizophilum CL-YJ9 , respectively. An average nucleotide identity value was 89.0 % with M. profundum PAMC 27536 and 85.6 % with M. rhizophilum CL-YJ9 , respectively. The major fatty acids of strain ST58-10 were summed feature 3 (comprising C16 : 1ω7c/C16 : 1ω6c), summed feature 8 (comprising C18 : 1 ω7c/C18 : 1ω6c), C16 : 0 and C10 : 0 3-OH, and contained ubiquinone (Q-8) as the sole isoprenoid quinone. Phosphatidylethanolamine, phosphatidylglycerol, three unidentified aminolipids, an unidentified glycolipid and an unidentified lipid were detected as polar lipids. The DNA G+C content of strain ST58-10 was 58.78 mol%. On the basis of the phenotypic, chemotaxonomic and molecular properties, strain ST58-10 represents a novel species of the genus Marinobacterium, for which the name Marinobacterium aestuarii sp. nov. is proposed. The type strain is ST58-10 (=KCTC 52193 =NBRC 112103 ).

[Ab] Resumo:Strain Eup a-2 , isolated from the torch coral Euphyllia glabrescens, was characterized using a polyphasic taxonomy approach. Cells of strain Eup a-2 were Gram-negative, aerobic and motile by three polar flagella and formed translucent colonies. Optimal growth occurred at 25 °C, pH 6-8 and in the presence of 2-4 % NaCl. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain Eup a-2 belonged to the genus Litoribrevibacter and showed the highest levels of sequence similarity with respect to Litoribrevibacter albus Y32 (97.8 %). Strain Eup a-2 contained summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and C16 : 0 as the predominant fatty acids. The predominant isoprenoid quinone was Q-8. The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol and diphophatidylglycerol. Genomic DNA G+C content of strain Eup a-2 was 49.1 mol%. The DNA-DNA hybridization value for strain Eup a-2 with L. albus Y32 was less than 30 %. Differential phenotypic properties, together with the phylogenetic inference, demonstrate that strain Eup a-2 should be classified as a novel species of the genus Litoribrevibacter, for which the name Litoribrevibactereuphylliae sp. nov. is presented. The type strain is Eup a-2 (=BCRC 81004 =LMG 29725 =KCTC 52438 ).

[Ab] Resumo:A Gram-stain-negative, strictly aerobic, moderately halophilic bacterium, designated A-1 , was isolated from a tidal flat of the Taean coast in South Korea. Cells were motile rods with a single flagellum showing oxidase-negative and catalase-positive activities and contained poly-ß-hydroxyalkanoic acid granules. Growth of strain A-1 was observed at 20-40 °C (optimum, 30 °C), pH 6.0-10.5 (optimum, pH 7.0) and in the presence of 1.0-6.0 % (w/v) NaCl (optimum, 2.0 %). Strain A-1 contained C16 : 0, summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c) and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c) as the major fatty acids. The major polar lipids of strain A-1 were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. The isoprenoid quinones detected were ubiquinone-7 and ubiquinone-8. The G+C content of the genomic DNA was 51.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain A-1 formed a distinct phylogenetic lineage from other genera within the family Oceanospirillaceae. Strain A-1 shared low 16S rRNA gene sequence similarities with other taxa (≤94.9 %). On the basis of phenotypic, chemotaxonomic and molecular properties, it is clear that strain A-1 represents a novel genus and species of the family Oceanospirillaceae, for which the name Pontibacterium granulatum gen. nov., sp. nov. is proposed. The type strain is A-1 (=KACC 18119 =JCM 30136 ).

[Ab] Resumo:A novel Gram-staining-negative, spiral-shaped, pale-yellow, non-sporulating, motile, aerobic bacterium, designated strain AK56T, was isolated from a sediment sample collected at the Coringa Wildlife Sanctuary, India. Colonies on marine agar were circular, pale yellow, shiny, translucent, 1-2 mm in diameter, convex and had an entire margin. The major fatty acids included C16 : 1, C16 : 1ω7c/C16 : 1ω6c and C18 : 1ω7c. Polar lipids included diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, two unidentified aminolipids, one unidentified phospholipid and five unidentified lipids. DNA-DNA hybridization between strain AK56T and Oceanospirillum linum LMG 5214T and 'Oceanospirillum nioense ' NIO-S6 showed relatedness values of 39.91 and 23.62 %, respectively. The DNA G+C content of strain AK56T was found to be 50.3 mol%. A sequence similarity search for the 16S rRNA gene sequence revealed that O. linum and O. nioense were the nearest phylogenetic neighbours, with a pair-wise sequence similarity of 98.9 and 98.2 %, respectively. Phylogenetic analysis also showed the formation of a cluster including strain AK56T with close relative O. linum and O. nioense. Based on the observed phenotypic, chemotaxonomic characteristics and phylogenetic analysis, strain AK56T is described in this study as a novel species in the genus Oceanospirillum, for which the name Oceanospirillum sanctuarii sp. nov. is proposed. The type strain of Oceanospirillumsanctuarii is AK56T (=MTCC 12005T=JCM 19193T=KCTC 52973T).

[Ab] Resumo:Much of the phenotype of a microorganism consists of its repertoire of metabolisms and how and when its proteins are deployed under different growth conditions. Hence, analyses of protein expression could provide important understanding of how bacteria adapt to different environmental settings. To characterize the flexibility of proteomes of marine bacteria, we investigated protein profiles of three important marine bacterial lineages - Oceanospirillaceae (Neptuniibacter caesariensis strain MED92), Roseobacter (Phaeobacter sp. MED193) and Flavobacteria (Dokdonia sp. MED134) - during transition from exponential to stationary phase. As much as 59-80% of each species' total proteome was expressed. Moreover, all three bacteria profoundly altered their expressed proteomes during growth phase transition, from a dominance of proteins involved in translation to more diverse proteomes, with a striking appearance of enzymes involved in different nutrient-scavenging metabolisms. Whereas the three bacteria shared several overarching metabolic strategies, they differed in important details, including distinct expression patterns of membrane transporters and proteins in carbon and phosphorous metabolism and storage compounds. These differences can be seen as signature metabolisms - metabolisms specific for lineages. These findings suggest that quantitative proteomics can inform about the divergent ecological strategies of marine bacteria in adapting to changes in environmental conditions.

[Ab] Resumo:Nine isolates obtained from a great scallop hatchery in Norway were characterized using a polyphasic approach. Strains were Gram-negative, aerobic and motile rods with oxidative metabolism. Phylogenetic analysis based on the sequences of 16S rRNA and rpoB genes showed that these strains formed two different groups associated with members of the genus Neptuniibacter. DNA-DNA hybridization (DDH) and Average Nucleotide Identity (ANI) demonstrated that the isolates constituted two novel species of this genus, which can be phenotypically differentiated from their closest relatives. The names Neptuniibacter marinus sp. nov. and Neptuniibacter pectenicola sp. nov are proposed, with ATR 1.1 (=CECT 8938 =DSM 100783 ) and LFT 1.8 (=CECT 8936 =DSM 100781 ) as respective type strains.

[Ab] Resumo:A Gram-staining-negative, strictly aerobic, transparent and smooth, curved-rod-shaped motile bacterium with a single polar flagellum, designated strain sw100T, was isolated from surface seawater of South Pacific Gyre during Integrated Ocean Drilling Program Expedition 329. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain sw100T was most closely related to the member of the family Oceanospirillaceae and was distinct from the most closely related genera Bacterioplanes, Thalassolituus and Oceanobacter (95.8, 94.5-95.3 and 94.4 % 16S rRNA gene sequence similarities, respectively). Growth occurred in the presence of 0-11 % NaCl (w/v, optimum 7 %), at 10-37 °C (optimum 28 °C) and pH 6.0-9.0 (optimum pH 7.0). The major fatty acids (>10 % of total fatty acids) are C16 : 0, C16 : 0 N alcohol and summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c). The major polar lipids were phosphatidylethanolamine, phosphatidylmonomethylethanolamine, phosphatidylglycerol, diphosphatidylglycerol and two unidentified polar lipids. The DNA G+C content of strain sw100T was 55.3 mol%. The major respiratory quinone was ubiquinone-10 (Q-10). On the basis of polyphasic analyses, strain sw100T is considered to represent a novel species in a novel genus of the family Oceanospirillaceae, for which the name Bacterioplanoides pacificum gen. nov., sp. nov. is proposed. The type strain is sw100T (=JCM 30597T=MCCC 1K00501T=KCTC 42424T).

[Ab] Resumo:Two gram-negative, facultative anaerobic, chemoorganoheterotrophic, motile and rod-shaped bacteria, strains AVMART05(T) and KASP37, were isolated from ascidians (Tunicata, Ascidiaceae) of the genus Ascidiella collected at Gullmarsfjord, Sweden. The strains are the first cultured representatives of an ascidian-specific lineage within the genus Endozoicomonas (Gammaproteobacteria, Oceanospirillales, Hahellaceae). Both strains feature three distinct 16S rRNA gene paralogs, with identities of 98.9-99.1% (AVMART05(T)) and 97.7-98.8% (KASP37) between paralogs. The strains are closely related to Endozoicomonas atrinae and Endozoicomonas elysicola, with which they share 97.3-98.0% 16S rRNA gene sequence identity. Digital DNA-DNA hybridization, average nucleotide identity, and tetra-nucleotide correlation analysis indicate that both strains belong to a single species distinct from their closest relatives. Both strains feature similar DNA G+C contents of 46.70mol% (AVMART05(T)) and 44.64mol% (KASP37). The fatty acid patterns of AVMART05(T) and KASP37 are most similar to those of Endozoicomonas euniceicola and Endozoicomonas gorgoniicola. Based on the polyphasic approach, we propose the species Endozoicomonas ascidiicola sp. nov. to accommodate the newly isolated strains. E. ascidiicola sp. nov. is represented by the type strain AVMART05(T) (=DSM 100913(T)=LMG 29095(T)) and strain KASP37 (=DSM 100914=LMG 29096).

[Ab] Resumo:Two Gram-stain-negative, aerobic, alkaliphilic bacteria (strains MEB087T and MEB142) were isolated from sediment and water samples, respectively, collected from the alkaline Lonar Lake in Maharashtra, India. Strains MEB087T and MEB142 shared 99.8 % 16S rRNA gene sequence similarity and were 85 % related on the basis of DNA-DNA hybridization. The 16S rRNA gene sequences of both strains showed close relationship with the genus Nitrincola, and their closest neighbour was Nitrincola lacisaponensis 4CAT with 97.7 % sequence similarity. MEB087T and MEB142 exhibited only 45 % and 54 % DNA-DNA relatedness, respectively, with Nitrincola lacisaponensis DSM 16316T. Both strains were asporogenous, short, non-motile rods capable of utilizing a limited range of organic acids as sole carbon and energy sources. They were oxidase- and catalase-positive, able to reduce nitrate and nitrite; but unable to degrade DNA, urea, gelatin, casein or starch. They grew optimally at pH 9.5 (tolerating up to pH 11) and could withstand up to 0.6 M NaCl. The predominant cellular fatty acids were summed feature 8 comprising C18 : 1ω7c/C18 : 1ω6c (47-49 %) followed by summed feature 3 comprising C16 : 1ω7c/C16 : 1ω6c (28-32 %). The predominant polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylcholine. The DNA G+C content was 49.3-49.7 mol%. On the basis of the phylogenetic analysis and chemotaxonomic characteristics, strains MEB087T and MEB142 represent a novel species in the genus Nitrincola, for which the name Nitrincola alkalisediminis sp. nov. is proposed. The type strain is MEB087T ( = KCTC 42948T = JCM 19317T) with MEB142 ( = KCTC 42949 = JCM 19318) as an additional strain.

[Ab] Resumo:Neptunomonas antarctica S3-22(T) was isolated from marine sediment of the Nella Fjord, Antarctica. Here we report the draft genome sequence of N. antarctica, which comprises 4,568,828 bp with a mean G+C content of 45.7%. We found numerous genes related to resistance, motility and chemotaxis, nitrogen metabolism, aromatic compound metabolism and stress response. These metabolic features and related genes revealed genetic basis for the adaptation to the marine sediment environment in Antarctica. The genome sequence of N. antarctica S3-22(T) may also provide further insights into the ecological role of the genus Neptunomonas.
