[Ab] Resumo:A Gram-stain-negative, motile, non-spore-forming, rod-shaped bacterium, designated strain B2T, was isolated from the culture broth of a marine microalga, Picochlorum sp. 122. Phylogenetic analyses based on 16S rRNA gene sequences indicated that strain B2T forms a robust cluster with members of the genus Pelagibacterium, and shares the highest sequence similarity of 96.80 % with Pelagibacterium halotolerans CGMCC 1.7692T. Optimal growth of strain B2T was observed at 33 °C, at pH 8 and in the presence of 1 % (w/v) NaCl. The predominant ubiquinone of strain B2T was Q-10, and the G+C content of the genomic DNA was 58.6 mol%. The major fatty acid profile comprised C18 : 1ω7c/ω6c, C19 : 0 cyclo ω8c and C16 : 0. The major polar lipids of strain B2T were diphosphatidylglycerol, phosphatidylglycerol, two unidentified glycolipids and seven unidentified lipids. Comprehensive analyses based on a polyphasic characterization of strain B2T indicated that it represents a novel species of the genus Pelagibacterium, for which the name Pelagibacterium lentulum sp. nov. is proposed. The type strain is B2T (=MCCC 1K03218T=CGMCC 1.15896T=KCTC 52551T).

[Ab] Resumo:Previously we proposed the reclassification of a thermotolerant phototrophic bacterium, 'Rhodopseudomonas cryptolactis' Stadtwald-Demchick et al. 1990, as 'Rhodoplanes cryptolactis' nom. rev., comb. nov. with strain DSM 9987T (ATCC 49414T) as the type strain. However, while both the names 'Rhodopseudomonas cryptolactis' and 'Rhodoplanes cryptolactis' have not been validated, strain ATCC 49414T is no longer available from the culture collection. This situation indicates that the taxonomic status of the bacterium with both the names to be validated has been lost. In this study, we re-examined the taxonomic characteristics of strain DSM 9987T (TUT3520T as our own collection number) compared with those of six species of the genus Rhodoplanes with validly published names. The results of 16S rRNA gene sequence comparisons indicated that TUT3520T had a 99.0 % level of similarity to the type strains of Rhodoplanes oryzae and Rhodoplanes elegans as its closest relatives and 98.9-96.2 % similarities to other species of the genus Rhodoplanes. Genomic DNA-DNA similarities between TUT3520T and the type strains of the species of the genus Rhodoplanes were less than 50 %. Results of phenotypic testing indicated that TUT3520T could be differentiated from any species of the genus Rhodoplanes by a combination of in vivo absorption spectra, growth temperature, vitamin requirements, carbon nutrition and some other characteristics. Thus, we propose Rhodoplanes tepidamans sp. nov. to accommodate the bacterium previously referred to as 'Rhodoplanes (Rhodopseudomonas) cryptolactis'. The type strain is strain TUT3520T (=DSM 9987T=NBRC 104267T).

[Ab] Resumo:A bacterial isolate, designated strain S37T, was isolated from the rhizosphere of oil palm (Elaeis guineensis). Strain S37T was found to be Gram-stain-negative, aerobic, motile and rod shaped. Based on 16S rRNA gene sequence analysis, strain S37T was most closely related to Devosia albogilva IPL15T (97.3 %), Devosia chinhatensis IPL18T (96.8 %) and Devosia subaequoris HST3-14T (96.5 %). The G+C content of the genomic DNA was 63.0 mol%, and dominant cellular fatty acids were summed feature 8 (C18 : 1ω7c and/or C18 : 1ω6c), 11-methyl C18 : 1ω7c and C16 : 0. The predominant isoprenoid quinone was ubiquinone-10 (Q-10), and the major polar lipids were phosphatidylglycerol, diphosphatidylglycerol, glycolipid and phospholipids. Based on the polyphasic taxonomic data, it is clear that strain S37T represents a novel species of the genus Devosia within the family Hyphomicrobiaceae, for which we propose the name Devosia elaeis sp. nov., with strain S37T (=TBRC 5145T=LMG 29420T) as the type strain.

[Ab] Resumo:A novel arsenic-resistant bacterium, designated 42-50T, was isolated from the high-arsenic sediment of Jianghan Plain, Hubei Province, China. Phylogenetic and biochemical analysis indicated that this bacterium represents the first species of a novel genus belonging to the family Hyphomicrobiaceae. The 16S rRNA gene of strain 42-50T shares 96.3-94.2, 96.3, 96.2 and 94.9-93.8 % sequence identities to those of species from the genera Devosia, Youhaiella, Paradevosia and Pelagibacterium, respectively. The major cellular fatty acids are C16 : 0, C18 : 0, C18 : 1ω7c 11-methyl and summed feature 8 (comprising C18 : 1ω7c and C18 : 1ω6c). The predominant polar lipids are diphosphatidylglycerol, phosphatidylglycerol and two unidentified glycolipids. The predominant respiratory quinone is ubiquinone-10 (Q-10). The DNA G+C content of strain 42-50T is 73.7 mol%. The distinct phylogenetic lineage and unique cellular fatty acids suggest that strain 42-50T represents a novel species of a new genus affiliated with the family Hyphomicrobiaceae, for which the name Arsenicitalea aurantiaca gen. nov., sp. nov. is proposed. The type strain is 42-50T (=CCTCC AB 2014325T=KCTC 42825T).

[Ab] Resumo:A Gram-stain-negative, aerobic, non-flagellated and pleomorphic bacterium, designated HJR-2T, was isolated from a junction between the ocean and a freshwater lake on the East Sea, South Korea. It grew optimally at 30 °C, at pH 7.0-8.0 and in the presence of 0-1.5 % (w/v) NaCl. In the neighbour-joining phylogenetic tree based on 16S rRNA gene sequences, strain HJR-2T clustered with the type strains of Vasilyevaea enhydra and Vasilyevaea mishustinii and this cluster fell within the clade comprising the type strains of Devosia species. Strain HJR-2T exhibited 16S rRNA gene sequence similarity values of 97.2 and 97.0 % to the type strains of V. enhydra and V. mishustinii, respectively, and 93.9-96.2 % to the type strains of Devosia species. Strain HJR-2T contained Q-10 as the predominant ubiquinone and C18 : 1ω7c, 11-methyl C18 : 1ω7c and C16 : 0 as major fatty acids. Major polar lipids of strain HJR-2T were phosphatidylglycerol and two unidentified glycolipids. The DNA G+C content was 66.0 mol% and its mean DNA-DNA relatedness values with the type strains of V. enhydra and V. mishustinii were 11-17 %. Differential phenotypic properties, together with its phylo genetic and genetic distinctiveness, revealed that strain HJR-2T is separated from V. enhydra, V. mishustinii and Devosia species. On the basis of the data presented, strain HJR-2T (=KCTC 52211T=NBRC 112271T) is considered to be the type strain of a novel species of the genus Devosia, for which the name Devosiaconfluentis sp. nov. is proposed. In this study, it is also proposed that V. enhydra and V. mishustinii be reclassified as members of the genus Devosia.

[Ab] Resumo:A Gram-negative, rod shaped and non-spore-forming bacterium, strain H642(T), was isolated from a desert sample collected from Xinjiang province of China, and subjected to a polyphasic taxonomic investigation. Strain H642(T) grew between 20 and 40 °C (optimal 28-37 °C), pH 6.0-10.0 (optimal 7.0-9.0) and at NaCl concentrations between 1.0 and 20.0 % (optimal 2.0-8.0 %). It was positive for catalase, but negative for oxidase. Ubiquinone-10 was present as the major respiratory ubiquinone. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, four unknown glycolipids and one unknown lipid. Phylogenetic analysis of the 16S rRNA gene sequence revealed that strain H642(T) belongs to the genus Pelagibacterium. Strain H642(T) was most closely related to Pelagibacterium halotolerans (with 98.4 % similarity), Pelagibacterium nitratireducens (97.3 %) and Pelagibacterium luteolum (96.2 %). DNA-DNA reassociation values between strain H642(T), P. halotolerans B2(T) and P. nitratireducens JLT2005(T) were 38.0 and 25.3 %, respectively. The genomic DNA G + C content of strain H642(T) was determined to be 44.4 mol%. Based on genotypic, chemotaxonomic and phenotypic data, strain H642(T) represents a novel species of the genus Pelagibacterium, for which the name Pelagibacterium lixinzhangensis sp. nov is proposed. The type strain is H642(T) (=CGMCC 1.10230(T)=JCM 16597(T)).

[Ab] Resumo:In this study, a novel metallo-ß-lactamases fold hydrolase PH-1 was identified from Pelagibacterium halotolerans B2(T). This novel member of the family Hyphomicrobiaceae was isolated from the East China Sea. In silico analysis demonstrated that PH-1 and its relative homologues cluster in a unique branch and constitute a new subgroup among MBLs. PH-1 was cloned and overexpressed in Escherichia coli BL21 in a soluble form. SDS-PAGE, MALDI-TOF/TOF-MS, and size-exclusion chromatography analysis demonstrated that the PH-1 was a monomer with molecular weight of about 29 kDa. Substrate specificity study showed PH-1 preferred penicillin type ß-lactams and exhibited maximum activity toward penicillin-G. Additionally, our experiments also revealed that PH-1 was a halotolerant enzyme since it is active under 4 M NaCl. The enzyme activity of PH-1 was negatively affected by 1 mM Mn(2+) and EDTA. These observations lay a foundation for further study of MBLs from marine bacterium.

[Ab] Resumo:A bacterial isolate, designated strain THG-MM1T, was isolated from soil in Yongin region, Republic of Korea. Strain THG-MM1T was Gram-stain-negative, aerobic, non-motile and rod-shaped. Based on 16S rRNA gene sequence comparisons, strain THG-MM1T was most closely related to Devosia insulae DS-56T (96.5 % 16S rRNA gene sequence similarity), followed by Devosia yakushimensis Yak96BT (95.7 %) and Devosia albogilva IPL15T (95.2 %). The G+C content of the genomic DNA was 63.7 mol%. The major cellular fatty acids were C16 : 0, summed feature 8 (C18 : 1ω7c and/or C18 : 1ω6c) and C18 : 0. The predominant isoprenoid quinone was ubiquinone-10 (Q-10). The major polar lipids were phosphatidylglycerol and an unidentified lipid. The combined phenotypic, chemotaxonomic and phylogenetic data showed that strain THG-MM1T represents a novel species of the genus Devosia, for which the name Devosia humi sp. nov. is proposed, with strain THG-MM1T ( = KACC 18281T = CCTCC AB 2015121T) as the type strain.

[Ab] Resumo:A Gram-stain-negative, aerobic, short rod-shaped and motile bacterial strain, designated M48T, was isolated from a soil sample contaminated with chloroethylene compounds. Strain M48T was able to biodegrade methanol as a sole carbon source. Based on 16S rRNA gene sequences, strain M48T was closely related to Devosia chinhatensis IPL18T, Devosia insulae DS-56T, Vasilyevaea enhydra 9bT and Devosia psychrophila Cr7-05T, showing sequence similarities of 96.0, 95.8, 95.6 and 95.5 %, respectively. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain M48T formed a distinct lineage within the family Hyphomicrobiaceae of the class Alphaproteobacteria. Strain M48T contains Q-10 as the predominant ubiquinone, and C18 : 1ω7c 11-metyl, C16 : 0, C19 : 0 cyclo ω8c and C18 : 1ω7c as the major fatty acids. The polar lipid profile includes diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and dimannosyldiacylglycerol. The DNA G+C content of strain M48T was 60.3 mol%. Based on phenotypic, phylogenetic and chemotaxonomic data, strain M48T represents a novel species of a new genus within the family Hyphomicrobiaceae, for which the name Methyloterrigena soli gen. nov., sp. nov. is proposed. The type strain of the type species is M48T ( = KEMB 224-262T = JCM 30821T).
