[Ab] Resumo:The American diet has changed dramatically since 1958, when Congress gave the United States Food and Drug Administration (FDA) the authority to ensure the safety of chemicals in food. Since then, thousands of chemicals have entered the food system. Yet their long-term, chronic effects have been woefully understudied, their health risks inadequately assessed. The FDA has been sluggish in considering scientific knowledge about the impact of exposures-particularly at low levels and during susceptible developmental stages. The agency's failure to adequately account for the risks of perchlorate-a well-characterized endocrine-disrupting chemical-to vulnerable populations is representative of systemic problems plaguing the regulation of chemicals in food. Today, we are faced with a regulatory system that, weakened by decades of limited resources, has fallen short of fully enforcing its mandates. The FDA's inability to effectively manage the safety of hundreds of chemicals is putting our children's health at risk.

[Ab] Resumo:Alantolactone and isoalantolactone isolated from many species of plants are a pair of positional isomers of CC bond. Previously, alantolactone and isoalantolactone have been proved to be good lead compounds for future anticancer agent development. Similarity of their molecular structures increases the separation difficulty for these two isomers on a conventional C column. Silver perchlorate (AgClO ) as mobile phase additives with RP-HPLC for improving the separation was developed for rapid determination of the positional isomers in Inula racemosa Hook.f. The effects of the concentration of silver perchlorate on the separation of the analytes were investigated. The composition of acetonitrile and water containing 5.0% silver perchlorate in a 65:35 (v/v) ratio was used as mobile phase, in which they were well separated within a short period of time on the C column. The method was successfully applied to determine them in an extract of Inula racemosa Hook.f. root. Silver perchlorate in mobile phase can efficiently improve the separation of the positional isomers and could be applied to rapidly determinate their content in this plant.

[Ab] Resumo:A ß-class carbonic anhydrase (CA, EC 4.2.1.1) from the pathogenic bacterium Francisella tularensis (FtußCA) was cloned and purified, and the anion inhibition profile was investigated. Based on the measured kinetic parameters for the enzyme catalyzed CO hydration reaction (k of 9.8×10 s and a k /K of 8.9×10 M s ), FtußCA is a highly effective enzyme. The activity of FtußCA was not inhibited by a range of anions that do not typically coordinate Zn(II) effectively, including perchlorate, tetrafluoroborate, and hexafluorophosphate. Surprisingly, some anions which generally complex well with many cations, including Zn(II), also did not effectively inhibit FtußCA, e.g., fluoride, cyanide, azide, nitrite, bisulphite, sulfate, tellurate, perrhenate, perrhuthenate, and peroxydisulfate. However, the most effective inhibitors were in the range of 90-94µM (sulfamide, sulfamic acid, phenylarsonic and phenylboronic acid). N,N-Diethyldithiocarbamate (K of 0.31mM) was a moderately potent inhibitor. As Francisella tularensis is the causative agent of tularemia, the discovery of compounds that can interfere with the life cycle of this pathogen may result in novel opportunities to fight antibiotic drug resistance.

[Ab] Resumo:Perchlorate (ClO ) contamination is more and more concerned due to the hazards to humans. Based on the common primary bacterium (Helicobacteraceae) of both thiosulfate-acclimated sludge (T-Acc) and sulfur-acclimated sludge (S-Acc) for perchlorate reduction, the rapid start-up of sulfur-based perchlorate reduction reactor (SBPRR) was hypothesized by inoculating T-Acc. Furthermore, the performance of SBPRR, the SO yield, kinetics of ClO reduction and the extracellular polymeric substances (EPS) of biofilm confirmed the hypothesis. The start-up time of R3 (reactor inoculating T-Acc) was 0.18 and 0.21 times that of R1 (control) and R2 (reactor with the influent containing thiosulfate), respectively. The SO yield of R3 was lower than that of R2 and R1 with perchlorate removal rate 166.7mg/(Lh). The kinetic study and EPS demonstrated that inoculating T-Acc was beneficial for the development of biofilm. Consequently, the present study indicated that SBPRR can be rapidly and successfully started-up via inoculation of T-Acc.

[Ab] Resumo:A magnetic amine-crosslinked reed (MACR) was synthesized by an insitu precipitation method and used for perchlorate uptake. The morphological properties of clean MACR, perchlorate-saturated MACR and bio-regenerated MACR samples were determined using scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and zeta potential measurements. The adsorption capacities of perchlorate by clean and bio-regenerated MACRs were determined. The density functional theory (DFT) method was employed to evaluate the binding free energies between various anions and ammonium/hydroxy groups. The maximum adsorption (Q ) of perchlorate by MACR was calculated to be 195.5-232.8 mg/g at 30-50 °C. The theoretical computation of adsorption-free energies indicated that ammonium groups were dominant in the process of perchlorate adsorption; other anions, such as [H PO ] , [NO ] and [SO ] , showed relatively higher binding free energies than [ClO ] , which corresponded to the results of competitive adsorption. The spent MACR was then bio-regenerated in a sealed 250-ml conical flask with perchlorate-reducing bacteria (30 °C, 160 rpm) and reached 81.4% of recovery within 3 days. Some hydrophobic macromolecules of extracellular polymeric substances (EPS) might have attached to the surface of MACR, which was validated by the zeta potential, SEM and excitation emission matrix (EEM) fluorescence spectroscopy results.

[Ab] Resumo:BACKGROUND: At high medicinal doses perchlorate is known to decrease the production of thyroid hormone, a critical factor for fetal development. In a large and uniquely exposed cohort of pregnant women, we recently identified associations between environmental perchlorate exposures and decreased maternal thyroid hormone during pregnancy. Here, we investigate whether perchlorate might be associated with birthweight or preterm birth in the offspring of these women. METHODS: Maternal urinary perchlorate, serum thyroid hormone concentrations, birthweight, gestational age, and urinary nitrate, thiocyanate, and iodide were collected in 1957 mother-infant pairs from San Diego County during 2000-2003, a period when the county's water supply was contaminated with perchlorate. Associations between perchlorate exposure and birth outcomes were examined using linear and logistic regression analyses adjusted for maternal age, weight, race/ethnicity, and other factors. RESULTS: Perchlorate was not associated with birth outcomes in the overall population. However, in analyses confined to male infants, log maternal perchlorate concentrations were associated with increasing birthweight (ß=143.1gm, p=0.01), especially among preterm births (ß=829.1g, p<0.001). Perchlorate was associated with male preterm births ≥2500g (odds ratio=3.03, 95% confidence interval=1.09-8.40, p-trend=0.03). Similar associations were not seen in females. CONCLUSIONS: This is the first study to identify associations between perchlorate and increasing birthweight. Further research is needed to explore the differences we identified related to infant sex, preterm birth, and other factors. Given that perchlorate exposure is ubiquitous, and that long-term impacts can follow altered birth outcomes, future research on perchlorate could have widespread public health importance.

[Ab] Resumo:In the present study we propose a new analytical method for Cl/ Cl analysis in perchlorate by Ion Chromatography(IC) coupled to Multicollector Inductively Coupled Plasma Mass Spectrometry (MC-ICPMS). The accuracy of the analytical method was validated by analysis of international perchlorate standard materials USGS-37 and USGS -38; analytical precision better than ±0.4‰ was achieved. Cl/ Cl isotope ratio analysis in perchlorate during laboratory biodegradation experiment with microbial cultures enriched from the contaminated soil in Israel resulted in isotope enrichment factor ε Cl = -13.3 ± 1‰, which falls in the range reported previously for perchlorate biodegradation by pure microbial cultures. The proposed analytical method may significantly simplify the procedure for isotope analysis of perchlorate which is currently applied in environmental studies.

[Ab] Resumo:Calcined ternary hydrotalcite like compounds had been synthesized by co-precipitation and calcined at 450 °C for 6 h. It had been applied into the nitrate removal from aqueous solutions. Various factors during adsorption process, such as chemical composition of adsorbents, adsorbent dosage, initial concentration of nitrate, initial pH and co-existing anions on nitrate adsorption had been investigated. It was found that 1.5 g/L CHTlcs (Mg/Al/Fe molar ratio = 3:0.1:0.9) could achieve ideal adsorption efficiency at an initial nitrate concentration of 100 mg/L. The value of pH had little effect on nitrate adsorption. The co-existing anions had the inhibitory effect on nitrate adsorption, which was in the order of PO > SO > Cl > ClO . The batch adsorption studies showed that experimental data were well described by the pseudo-second-order kinetic model and Langmuir isotherm model, and the maximum isotherm adsorption capacity was 123.305 mg/g. The adsorption of nitrate was a spontaneous exothermic process with the reduction of entropy. The proposed adsorption mechanism obtained from characterization was that CHTlcs uptake nitrate through reconstruction effect and adsorption on external surface.

[Ab] Resumo:A total of 28 groundwater, paired farmland soil, lettuce, and its rhizosphere soil samples were collected from Chengdu, China to detect perchlorate levels and to evaluate the relationships of perchlorate concentrations among these matrices. The perchlorate concentrations in the groundwater, farmland soil, lettuce, and rhizosphere soil samples ranged from below detection limit to 60.2 µg L , from below detection limit to 249 µg kg dry weight (dw), from 2.07 to 1010 µg kg wet weight, and from below detection limit to 314 µg kg dw, respectively. Significant correlation was found in the perchlorate levels among the farmland soil, lettuce, and rhizosphere soil, suggesting that they have common pollution sources, or perchlorate might transfer from farmland soil-rhizosphere soil-plant. However, there is no significant correlation between groundwater and the other three matrices, indicating that infiltration from perchlorate contaminated farmland soil was not the predominant source for groundwater pollution in Chengdu. The perchlorate concentrations in the farmland soil and lettuce samples were significantly higher than those in the rhizosphere soil, primarily due to uptake of perchlorate through the rhizosphere micro-environment by lettuce, or accelerated degradation by rhizospheric microorganisms, which contributed more needs further investigation.

[Ab] Resumo:Context: Common environmental contaminants can disrupt normal thyroid function, which plays essential but varying roles at different ages. Objective: To evaluate the relationship of perchlorate, thiocyanate, and nitrate, three sodium-iodide symporter (NIS) inhibitors, and thyroid function in different age-sex-stratified populations. Design, Setting, Participants, and Intervention: This was a cross-sectional analysis of data from the 2009 to 2012 National Health and Nutrition Examination Survey evaluating the exposure to perchlorate, thiocyanate, and nitrate in 3151 participants aged 12 to 80. Main Outcome Measure: Blood serum free thyroxine (FT4) as both a continuous and categorical variable. We also assessed blood serum thyroid stimulating hormone. Results: Controlling for serum cotinine, body mass index, total daily energy consumption, race/ethnicity, and poverty-to-income ratio, for each log unit increase in perchlorate, FT4 decreased by 0.03 ng/dL in both the general population (P = 0.004) and in all women (P = 0.005), and by 0.06 ng/dL in adolescent girls (P = 0.029), corresponding to 4% and 8% decreases relative to median FT4, respectively. For each log unit increase thiocyanate, FT4 decreased by 0.07 ng/dL in adolescent boys (P = 0.003), corresponding to a 9% decrease relative to median FT4, respectively. Conclusions: Our results indicate that adolescent boys and girls represent vulnerable subpopulations to the thyroid-blocking effects of NIS symporter inhibitors. These results suggest a valuable screening and intervention opportunity.
