[Ab] Resumo:Triclosan (TRC), chloroxylenol (PCMX) and methylisothiazolinone (MIT) have been commonly used as an antimicrobial in soaps while borax (BRX) is used in household cleaning. After using these chemicals, they are washed down drains and getting into the aquatic ecosystem in which they may affect aquatic living organisms. In the present study, the chronic effects of TRC, PCMX, MIT and BRX on genotoxicity, gene expression and histopathology of rainbow trout (Oncorhynchus mykiss) were evaluated for 40 days under semi static condition. The comet assay results indicated that MIT, TRC and PCMX caused significant DNA damage to erythrocytes of the fish. Transcription of SOD, GPX1, GPX2, GSTA, HSP90BB, HSP90BA, CAT, and HSC70A genes were significantly regulated as a result of TRC, PCMX, MIT, and BRX exposure except PCMX exposed GSTA gene. Histological lesions were detected in gills, spleen liver, and trunk kidney of the fish. Lamellar fusion, hyperplasia and epithelial necrosis in gills, melanomacrophage centers and splenic necrosis in spleen, pyknotic nucleus, fat vacuoles, necrotic hepatocytes in liver, cloudy swelling in the tubules, renal tubule epithelial cells degeneration, glomerular capillaries dilation and glomerulus degeneration in kidney, were observed. Our study demonstrates the chronic toxic effect of TRC, PCMX, MIT, and BRX is high in rainbow trout. Therefore, we should be more careful when using these chemicals for cleaning in order to protect aquatic environment.

[Ab] Resumo:CD capillary electrophoresis methods were developed for complete enantiomeric and diastereoisomeric separations of a series of ten dihydropyridone analogues, of which eight were neutral, one was anionic, and one was cationic. Ten different systems comprising one or two CDs were found to successfully separate the isomers thanks to a screening approach. Among the tested CDs, highly sulfated-γ-CD (HS-γ-CD), either in a single or in a dual system, in a phosphate buffer using capillaries dynamically coated with polyethylene oxide, and SBE-ß-CD, either in a single or in a dual system, in a borate buffer using uncoated capillaries, were the most selective selectors. The effects of different parameters such as the nature and concentration of the CDs, nature and concentration of the buffer, and voltage were examined. The precision and LODs and limits of quantification were evaluated for the optimized methods.

[Ab] Resumo:The Dolichoderus thoracicus (Smith) has seriously infested urban, village, and agricultural environments in Taiwan. To develop optimal bait for the effective control of D. thoracicus, we investigated the feeding preferences of this ant at different sugar and amino acid concentrations in a laboratory setting. The number of feeding workers was significantly higher for 20% sucrose compared with that for other sugars at 10% and 20% concentrations. Furthermore, among all tested concentrations of monosodium glutamate (MSG; 1%, 3%, 5%, and 10%), the highest number of ants were attracted by 1% MSG with 20% sucrose. The consumption of each bait solution was also measured, and the results were similar to those for ant number. Regarding toxicants, the toxicity and repellency of 3% boric acid and borax bait were evaluated individually. Both 3% boric acid and borax demonstrated no repellency against D. thoracicus workers. The LT50 of the 3% boric acid bait was 4.87 d, whereas that of 3% borax bait was only 1.56 d. Thus, 3% boric acid showed further delayed action, facilitating workers spreading the toxicant to their nestmates through trophallaxis. In combination with our unique bait station, we propose an optimal bait formulation comprising 20% sucrose, 1% MSG, and 3% boric acid as a promising candidate for controlling D. thoracicus.

[Ab] Resumo:We report the safety, biodistribution, and internal radiation dosimetry, in humans with thyroid cancer, of F-tetrafluoroborate ( F-TFB), a novel PET radioligand for imaging the human sodium/iodide symporter (hNIS). Serial whole-body PET scans of 5 subjects with recently diagnosed thyroid cancer were acquired before surgery for up to 4 h after injection of 184 ± 15 MBq of F-TFB. Activity was determined in whole blood, plasma, and urine. Mean organ-absorbed doses and effective doses were calculated via quantitative image analysis and using OLINDA/EXM software. Images showed a high uptake of F-TFB in known areas of high hNIS expression (thyroid, salivary glands, and stomach). Excretion was predominantly renal. No adverse effects in relation to safety of the radiopharmaceutical were observed. The effective dose was 0.0326 ± 0.0018 mSv/MBq. The critical tissues/organs receiving the highest mean sex-averaged absorbed doses were the thyroid (0.135 ± 0.079 mSv/MBq), stomach (0.069 ± 0.022 mSv/MBq), and salivary glands (parotids, 0.031 ± 0.011 mSv/MBq; submandibular, 0.061 ± 0.031 mSv/MBq). Other organs of interest were the bladder (0.102 ± 0.046 mSv/MBq) and kidneys (0.029 ± 0.009 mSv/MBq). Imaging using F-TFB imparts a radiation exposure similar in magnitude to many other F-labeled radiotracers. F-TFB shows a biodistribution similar to Tc-pertechnetate, a known nonorganified hNIS tracer, and is pharmacologically and radiobiologically safe in humans. Phase 2 trials for F-TFB as an hNIS imaging agent are warranted.

[Ab] Resumo:The absence of UV chromophores in the structure of most aminoglycosides impedes an easy and straightforward development of analytical methods. It was demonstrated that addition of borate to the mobile phase allowed direct detection of aminoglycosides at the lower end of the UV spectrum. Although the borate ion complexes preferably with compounds possessing vicinal diols, the aim of this study was to develop an assay for tobramycin that does not contain vicinal diols in its structure. Amikacin, an aminoglycoside with a broad spectrum, was added to the study for its high therapeutic importance. An assay for both has been developed and validated. This method was developed on an XBridge C18 column (4.6 mm x 250 mm; 5 µm) and the mobile phase consisting of methanol-disodium tetraborate decahydrate buffer (0.1 M; pH = 9.0)-water (20:20:60) supplemented with 1 g/L sodium octanesulfonate was run isocratically at a column temperature of 40°C. After validation according to International Conference on Harmonization guidelines, this method was tested on two commercially available pharmaceutical formulations, Amukin® and Tobrex®. The developed method was fast, accurate, easy to use and cheap as it did not require sophisticated equipment or tedious derivatization steps.

[Ab] Resumo:The metabolism of carbohydrate polymers drives microbial diversity in the human gut microbiota. It is unclear, however, whether bacterial consortia or single organisms are required to depolymerize highly complex glycans. Here we show that the gut bacterium Bacteroides thetaiotaomicron uses the most structurally complex glycan known: the plant pectic polysaccharide rhamnogalacturonan-II, cleaving all but 1 of its 21 distinct glycosidic linkages. The deconstruction of rhamnogalacturonan-II side chains and backbone are coordinated to overcome steric constraints, and the degradation involves previously undiscovered enzyme families and catalytic activities. The degradation system informs revision of the current structural model of rhamnogalacturonan-II and highlights how individual gut bacteria orchestrate manifold enzymes to metabolize the most challenging glycan in the human diet.

[Ab] Resumo:BACKGROUND: Photoselective vaporization of the prostate is a technique that is widely used for the treatment of benign prostatic hyperplasia (BPH) and has pronounced advantages compared to the traditional transurethral resection of the prostate. Following the recent introduction of end-firing lithium triborate lasers, we have created a new technique called photoselective vaporesection of the prostate (PVRP). This study described our initial experience using the PVRP technique for the treatment of BPH. METHODS: This prospective study included a total of 35 patients with BPH who underwent PVRP from August 2013 to July 2014. The chief clinical parameters were obtained and evaluated during the perioperative period and follow-up, including the International Prostate Symptom Score (IPSS), quality of life (QoL) score, maximum urinary flow rate, and prostate volume. All variables were evaluated for statistically significant differences compared to baseline values using the analysis of variance. RESULTS: The mean subgroup IPSS and QoL scores significantly improved during follow-up; the respective decreases in IPSS storage score, IPSS voiding score, IPSS nocturia score, and QoL score were 75.3%, 83.6%, 51.4%, and 71.7%, respectively (all P < 0.001 compared with baseline). Three patients were diagnosed with prostate cancer based on postoperative pathological examinations. There were no serious perioperative complications. CONCLUSION: The PVRP technique demonstrates satisfactory short-term clinical outcomes and perioperative safety in the treatment of BPH.

[Ab] Resumo:Obesity is a worldwide medical problem resulting in serious morbidity and mortality involving differentiation of pre-adipocytes into mature adipocytes (adipogenesis). Boron treatment has been reported to be associated with weight reduction in experimental animals; however, its effects on pre-adipocyte differentiation and anti-adipogenic molecular mechanisms are unknown. In this study, we demonstrate the inhibitory activities of boric acid (BA) and sodium pentaborate pentahydrate (NaB) on adipogenesis using common cellular models. Boron treatment repressed the expression of adipogenesis-related genes and proteins, including CCAAT-enhancer-binding protein α and peroxisome proliferator-activated receptor γ, by regulating critical growth factors and the ß-catenin, AKT, and extracellular signal-regulated kinase signaling pathways. In addition, although boron treatment did not induce apoptosis in pre-adipocytes, it depressed mitotic clonal expansion by regulation of cell cycle genes. Overall, these data offer promising insights into the prevention/treatment of obesity and associated diseases.

[Ab] Resumo:The effects that an increase of environmental pH has on the triple helix of scleroglucan (Sclg) and on the Sclg/borax hydrogel are reported. Rheological experiments show that the hydrogel is less sensitive to pH increase than Sclg alone, while at pH = 14 a dramatic viscosity decrease takes place for both systems. This effect is evidenced also by the reduced water uptake and anisotropic elongation detected, at pH = 14, by the swelling behaviour of tablets prepared with the Sclg/borax system. On the opposite, a different behaviour was observed with guar gum and locust bean gum tablets, tested as reference polysaccharides. The effect of pH on the structure of Sclg and Sclg/borax was investigated also by means of spectroscopic approaches based on the interaction between Congo red (CR) and the Sclg triple helix. Obtained results indicated that the CR absorbance maximum is shifted as a function of pH and by the presence of borax. Principal component analysis allowed very precise identification of the pH value at which the Sclg helix collapses. Molecular dynamics simulations of the Sclg/borax-CR complex indicated that, at physiological pH, only a few ordered configurations are populated, according to the induced circular dichroism (CD) spectrum evidence.

[Ab] Resumo:With the increasing applications of ionic liquids (ILs), the toxicity of ILs has drawn increasing attention in recent years, especially the influences of different anions and alkyl-chain lengths on the acute toxicity to aquatic organisms. We performed a study on the acute toxicity of 1-alkyl-3-methylimidazolium nitrate ([C mim]NO (n=2, 4, 6, 8, 10, 12)), 1-hexyl-3-methylimidazolium ILs ([C mim]R (R=Cl , Br , BF , PF )) to zebrafish (Danio rerio). We also evaluated the sensibility of the investigated animals and the stability of ILs in water via high performance liquid chromatography (HPLC, Agilent 1260, Agilent Technologies Inc., USA) to prove the reliability of the present study. The results illustrated that the test zebrafish (Danio rerio) were sensitive to the reference toxicant and that the investigated ILs in water were stable. The 50% lethal concentration (LC ) was used to represent the acute toxicity to zebrafish (Danio rerio). The present study showed that the highest toxic IL is [C mim]NO and the lowest toxic IL is [C mim]NO on Danio rerio. The LC s for ILs with different anions had similar values. Accordingly, we believe that ILs with different alkyl-chain lengths cause greater effects than other anions on acute toxicity to aquatic organisms. Furthermore, the present study can also provide scientific methods for future studies to select and assess ILs.
