[Ab] Resumo:Ion beams present a potential advantage in terms of treatment of lesions with hypoxic regions. In order to use this potential, it is important to accurately model the cell survival of oxic as well as hypoxic cells. In this work, an adaptation of the microdosimetric kinetic (MK) model making it possible to account for cell hypoxia is presented. The adaptation relies on the modification of damage quantity (double strand breaks and more complex lesions) due to the radiation. Model parameters such as domain size and nucleus size are then adapted through a fitting procedure. We applied this approach to two cell lines, HSG and V79 for helium, carbon and neon ions. A similar behaviour of the parameters was found for the two cell lines, namely a reduction of the domain size and an increase in the sensitive nuclear volume of hypoxic cells compared to those of oxic cells. In terms of oxygen enhancement ratio (OER), the experimental data behaviour can be reproduced, including dependence on particle type at the same linear energy transfer (LET). Errors on the cell survival prediction are of the same order of magnitude than for the original MK model. Our adaptation makes it possible to account for hypoxia without modelling the OER as a function of the LET of the particles, but directly accounting for hypoxic cell survival data.

[Ab] Resumo:Exposure to laser irradiation on seeds brings about the changes in agronomical characteristics of the plants. Solanum melongena L. var. Mattu Gulla, a variety of brinjal is of high economic value due to its unique colour and flavour. The aim of the study was to understand the influence of Helium-Neon (He-Ne) laser irradiation on agronomical characters of Solanum melongena L. var. Mattu Gulla in the field conditions. Various growth characteristics including seed germination percentage, survival rate, plant height, number of branches, and flowers and fruits were estimated during different developmental stages of the brinjal. In addition, the chlorogenic acid content of fruits obtained from the laser irradiated seeds were quantified using reversed-phase high-performance liquid chromatography (RP- HPLC). The plants from the seeds irradiated with different doses (20, 25 and 30J/cm ) of He-Ne laser showed significant enhancement on the growth characteristics when compared to the non-irradiated control groups. He-Ne laser irradiation also improved the yield characteristics of the plants significantly in in vivo conditions in comparison with control group. 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed using methanolic extract of matured fruit of Mattu Gulla on HepG2 and fibroblast cell lines. The IC values of fruit extract from laser irradiated groups were found to be similar to non-irradiated control groups. Chlorogenic acid content was found to be higher in 20J/cm and lower in 30J/cm treated fruit tissue. The current study thus elucidates the role of He-Ne laser as a biostimulator on brinjal var. Mattu Gulla not only in the in vitro conditions but also in the in vivo field conditions.

[Ab] Resumo:Excimer lasers, widely used in the semiconductor industry, are crucial for analyzing the purity of premix laser gases for the purpose of controlling stable laser output power. In this study, we designed a system for analyzing impurities in pure neon (Ne) base gas by customized GC. Impurities in pure neon (H2 and He), which cannot be analyzed at the sub-µmol/mol level using commercial GC detectors, were analyzed by a customized pulsed-discharge Ne ionization detector (PDNeD) and a pressurized injection thermal conductivity detector using Ne as the carrier gas (Pres. Inj. Ne-TCD). From the results, trace species in Ne were identified with the following detection limits: H2, 0.378µmol/mol; O2, 0.119µmol/mol; CH4, 0.880µmol/mol; CO, 0.263µmol/mol; CO2, 0.162µmol/mol (PDNeD); and He, 0.190µmol/mol (Pres. Inj. Ne-TCD). This PDNeD and pressurized injection Ne-TCD technique thus developed permit the quantification of trace impurities present in high-purity Ne.

[Ab] Resumo:The article presents the design and development of a neon gas target for the production of (22)Na using a proton beam from the room temperature cyclotron in Variable Energy Cyclotron Centre, Kolkata. The target design is made to handle a beam power of 85 W (17 MeV, 5 µA). The design is based on simulation using the computer code FLUKA for the beam dump and CFD-CFX for target cooling. The target has been successfully used for the production of (22)Na in a 6 day long 17 MeV, 5 µA proton irradiation run.

[Ab] Resumo:Hematoporphyrin monomethyl ether (HMME) combined with He-Ne laser irradiation is a novel and promising photodynamic therapy (PDT)-induced apoptosis that can be applied in vitro on canine breast cancer cells. However, the exact pathway responsible for HMME-PDT in canine breast cancer cells remains unknown. CHMm cells morphology and apoptosis were analyzed using optical microscope, terminal deoxynucleotidyl transferase dUTP nick end labeling fluorescein staining and DNA ladder assays. Apoptotic pathway was further confirmed by Real-time-polymerase chain reaction and Western blotting assays. Our results showed that HMME-PDT induced significant changes in cell morphology, such as formation of cytoplasmic vacuoles and the gradual rounding of cells coupled with decreased size and detachment. DNA fragmentation and cell death was shown to occur in a time-dependent manner. Furthermore, HMME-PDT increased the activities of caspase-9 and caspase-3, and released cytochrome c from mitochondria into the cytoplasm. HMME-PDT also significantly increased both mRNA and protein levels of Bax and decreased P53 gene expression in a time-dependent manner, while the mRNA and protein expression of Bcl-2 were repressed. These alterations suggest that HMME-PDT induced CHMm cell apoptosis via the mitochondrial apoptosis pathway and had anti-canine breast cancer effects in vitro.

[Ab] Resumo:PURPOSE: This work explores a new radiation therapy approach which might trigger a renewed use of neon and heavier ions to treat cancers. These ions were shown to be extremely efficient in radioresistant tumor killing. Unfortunately, the efficient region also extends into the normal tissue in front of the tumor. The strategy the authors propose is to profit from the well-established sparing effect of thin spatially fractionated beams, so that the impact on normal tissues might be minimized while a high tumor control is achieved. The main goal of this work is to provide a proof of concept of this new approach. With that aim, a dosimetric study was carried out as a first step to evaluate the interest of further explorations of this avenue. METHODS: The gate/geant4 v.6.1 Monte Carlo simulation platform was employed to simulate arrays of rectangular minibeams (700 µm × 2 cm) of four ions (Ne, Si, Ar, and Fe). The irradiations were performed with a 2 cm-long spread-out Bragg peak centered at 7 cm-depth. Dose distributions in a water phantom were scored considering two minibeams center-to-center distances: 1400 and 3500 µm. Peak and valley doses, peak-to-valley dose ratios (PVDRs), beam penumbras, and relative contribution of nuclear fragments and electromagnetic processes were assessed as figures of merit. In addition, the type and proportion of the secondary nuclear fragments were evaluated in both peak and valley regions. RESULTS: Extremely high PVDR values (>100) and low valley doses were obtained. The higher the atomic number (Z) of the primary ion is, the lower the valleys and the narrower the penumbras. Although the yield of secondary nuclear products increases with Z, the actual dose being deposited by the secondary nuclear fragments in the valleys starts to be the dominant contribution at deeper points, helping in the sparing of proximal normal tissues. Additionally, a wider center-to-center distance leads to a minimized contribution of heavier secondary fragments in valleys. CONCLUSIONS: The computed dose distributions suggest that a spatial fractionation of the dose combined to the use of submillimetric field sizes might allow profiting from the high efficiency of neon and heavier ions for the treatment of radioresistant tumors, while preserving normal tissues. The authors' results support the further exploration of this avenue. Next steps include the realization of biological experiment to confirm the shifting of normal tissue complication probability curves.

[Ab] Resumo:Histological, cytological and morphometric changes in the thymus of 1 month-old, adult (3-4 months-old) and old (24-30 months-old) rats (24 animals in each group) were studied during muscle regeneration after the alloplasty of the injured area with the muscle tissue from the animal of the same age. Muscles of the donor or recipient were subjected to the course of preliminary irradiation with He-Ne laser (dose: 4.5-5.4 J/cm2 for each extremity; total dose of 9.0-10.8 J/cm2 per animal). It was shown that the exposure of gastrocnemius muscles that were prepared for the operation to He-Ne laser radiation decreased morpho-functional activity of the thymus in young, adult and old recipient rats the before surgery. This was demonstrated by its weaker reaction to the allograft during the early time intervals after surgery. The observed effect was more pronounced with the increasing age of an animal.

[Ab] Resumo:Microwave plasmas excited at electron-cyclotron resonance were studied in the 0.5-15 mTorr pressure range. In contrast with low-limit pressure conditions where the plasma emission highlights a fairly homogeneous spatial structure, a periodic spatial modulation (period ∼6.2 cm) appeared as pressure increased. This feature is ascribed to a local power deposition (related to the electron density) due to the presence of a standing electromagnetic wave created by the feed electromagnetic field (2.45 GHz) in the cavity formed by the reactor walls. Analysis of the electron energy probability function by Langmuir probe and optical emission spectroscopy further revealed the presence of a high-energy tail that showed strong periodic spatial modulation at higher pressure. The spatial evolution of the electron density and of the characteristic temperature of these high-energy electrons coincides with the nodes (maximum) and antinodes (minimum) of the standing wave. These spatially-modulated power deposition and electron heating mechanisms are then discussed.

[Ab] Resumo:The reactions from codeposition of laser-ablated chromium atoms with carbon dioxide in excess neon are studied by infrared absorption spectroscopy. The species formed are identified by the effects of isotopic substitution on their infrared spectra. Density functional calculations are performed to support the spectral assignments and to interpret the geometric and electronic structures of the experimentally observed species. Besides the previously reported insertion products OCrCO and O2Cr(CO)2, the one-to-one Cr(CO2) complex and the one-to-two Cr(CO2)2 complex as well as the CrOCrCO and OCCrCO3 complexes are also formed. The Cr(CO2) complex is characterized to be side-on η(2)-C,O-coordinated. The Cr(CO2)2 complex is identified to involve a side-on η(2)-C,O-coordinated CO2 and an end-on η(1)-O-coordinated CO2. OCCrCO3 is a carbonate carbonyl complex predicted to have a planar structure with a η(2)-O,O-coordinated carbonate ligand. The CrOCrCO complex is predicted to be linear with a high-spin ground state. Besides the neutral molecules, charged species are also produced. The Cr(CO2)(+) and Cr(CO2)2(+) cation complexes are characterized to have linear end-on η(1)-O-coordinated structures with blue-shifted antisymmetric CO2 stretching vibrational frequencies. The OCrCO(-) anion is bent with the Cr-O and CO stretching frequencies red-shifted from those of OCrCO neutral molecule.

[Ab] Resumo:Relatively little is known about the dynamics of electron-transfer reactions at low collision energy. We present a study of Penning ionization of ground-state methyl fluoride molecules by electronically excited neon atoms in the 13 µeV­4.8 meV (150 mK­56 K) collision energy range, using a neutral­neutral merged beam setup. Relative cross sections have been measured for three Ne((3)P2) + CH3F reaction channels by counting the number of CH3F(+), CH2F(+), and CH3(+) product ions as a function of relative velocity between the neon and methyl fluoride molecular beams. Experimental cross sections markedly deviate from the Langevin capture model at collision energies above 20 K. The branching ratios are constant. In other words, the chemical shape of the CH3F molecule, as seen by the Ne((3)P2) atom, appears not to change as the collision energy is varied, in contrast to related Ne((3)PJ) + CH3X (X = Cl and Br) reactions at higher collision energies.
