[Ab] Resumo:A rapid, portable, and semi-quantitative immunochromatographic strip was developed for rapid and visual detection of alternariol monomethyl ether (AME). For this purpose, the anti-AME monoclonal antibody (mAb) was prepared and identified. AME coupled to bovine serum albumin (BSA) via methyl 4-bromobutanoate was prepared as immunogen. The recoveries of AME in spiked cherry and orange fruits determined by competitive ELISA were 86.1% and 80.7%, respectively. A colloidal gold nanoparticle (CGN) and CGNs-mAb conjugate were synthesized, and on this basis, a competitive colloidal gold immunochromatographic strip was developed and applied to the detection of AME toxin in fruit samples. The intensity of red density of the test line (T line) is inversely proportional to AME concentration in the range 0.1-10 ng/mL. The visual limit of detection (LOD) of AME was found to be about 10 ng/mL. The semi-quantitative detection can be completed in 10 min. Moreover, the immunochromatographic strip has lower cross-reactivity with AME analogues, and it has a good stability performance (following 3 months of storage). Hence, the colloidal gold immunochromatographic strip could be used as a semi-quantitative tool for the on-site, rapid, and visual detection of AME in fruit.

[Ab] Resumo:An immunoaffinity column (IAC) for the selective purification of 3-methyl-quinoxaline-2-carboxylic acid (MQCA) from porcine muscle and the liver as well as the methods for its determination by high-performance liquid chromatography with ultraviolet detection (HPLC-UV) and a colloidal gold-based immunochromatographic assay (GICA) were developed. This is the first study that describes a clean-up method that employs an IAC for the determination of MQCA. Using the HPLC-UV method, the limits of detection and limits of quantification for MQCA in different samples were 1.0-3.0µgkg and 4.0-10.0µgkg , respectively. The average recoveries of MQCA that was spiked into samples at concentrations of 4.0-100µgkg were 80.1-87.7%, with relative standard deviations of less than 8.5%. Using the GICA method, the limits of detection for MQCA in different samples were 10µgkg and 50µgkg . No false negatives or false positives were observed.

[Ab] Resumo:BACKGROUND: Inorganic gold nanoparticles (NPs) have a huge potential in targeted drug delivery. Simple preparation and surface modification procedure with their special physicochemical properties of gold NPs attract their use such as tumor targeting and the detection of cancerous cell. OBJECTIVE: Various studies were reported, however, details of biodistribution profile of gold NPs were not enough evaluated. We have studied biodistribution profile of gold NPs having various particle sizes (20, 50 and 100 nm). METHODS: Gold concentrations in brain, heart, lungs, liver, stomach, pancreas, spleen, kidneys, blood, urine, and feces were measured at 5 minutes, 0.25, 0.5, 1, 2, 3, 6, 12, 18 and 24 hours after administration of gold NPs using inductively coupled plasma atomic emission spectrometry. RESULTS: In lungs and brain, especially 20-nm gold NPs were accumulated after 2-3 hours of dose administration, and they were kept for 24 hours, whereas they showed relatively low accumulation in heart, stomach and pancreas. After 12 hours, 3.3-14.4% of the injected gold were observed in fecal matter and urine. CONCLUSIONS: From this study, the application of gold NPs for targeted delivery to lungs and brain and the excretion route of the gold NPs from the body were suggested.

[Ab] Resumo:BACKGROUND: Schistosomiasis remains a major public health concern in China and an epidemiological survey has revealed that schistosome-infected bovines and goats are the main transmission sources for the disease. Therefore, development of a sensitive technique for the diagnosis of schistosomiasis in domestic animals is necessary. METHOD: A novel colloidal gold immunochromatography assay (GICA) strip was developed for detecting Schistosoma japonicum in domestic animals. The colloidal gold was conjugated with recombinant streptococcal protein G (rSPG). As the test and control lines, the schistosome soluble egg antigen and rSPG, respectively, were blotted on nitrocellulose membrane. RESULTS: The lowest detectable serum dilution was 1∶640 for schistosome-infected buffaloes. The cross-reaction rate of GICA was 14.29% with Paramphistomum sp. in buffaloes, 16.67% with Haemonchus sp. in goats, and 33.33% with Orientobilharzia sp. in goats. These results were slightly lower and similar to those obtained through ELISA. Moreover, the strips for detecting S. japonicum in mice, rabbits, buffaloes, and goats showed high sensitivity (100.00%, 100.00%, 100.00%, and 100.00%, respectively) and specificity (100.00%, 100.00%, 94.23%, and 88.64%, respectively). And the sensitivity or specificity of the GICA strips did not present any significant differences after storage for 12 months at room temperature. When compared with ELISA, the GICA strips exhibited similar sensitivity and specificity in the diagnosis of schistosomiasis in mice, rabbits, buffaloes, and goats. Besides, only 5 µl of serum are required for the test and the detection can be completed within 5 min. CONCLUSION: This study is the first to develop a GICA strip using gold-rSPG conjugate for the diagnosing of schistosomiasis in domestic animals, and preliminary results showed that the developed strip may be suitable for large-scale screening of schistosomiasis in endemic areas.

[Ab] Resumo:The design, development, and biomedical applications of phytochemical-based green synthesis of biocompatible colloidal gold nanoparticles (AuNPs) are becoming an emerging field due to several advantages (safer, eco-friendly, simple, fast, energy efficient, low-cost, and less toxic) over conventional chemical synthetic procedures. Biosynthesized colloidal gold nanoparticles are remarkably attractive in several biomedical applications including cancer theranostics due to small size, unusual physico-chemical properties, facile surface modification, high biocompatibility, and numerous other advantages. Of late, several researchers have investigated the biosynthesis and prospective applications (diagnostics, imaging, drug delivery, and cancer therapeutics) of AuNPs in health care and medicine. However, not a single review article is available in the literature that demonstrates the anti-cancer potential of biosynthesized colloidal AuNPs with detailed mechanistic study. In the present review article, we for the first time discuss the biointerface of colloidal AuNPs, plants, and cancer mainly (i) comprehensive mechanistic aspects of phytochemical-based synthesis of AuNPs; (ii) proposed anti-cancer mechanisms along with biomedical applications in diagnostics, imaging, and drug delivery; and (iii) key challenges for biogenic AuNPs as future cancer nanomedicine.

[Ab] Resumo:Harringtonine (HT) is a natural compound, which is mainly produced by the genus Cephalotaxus, and has been clinically utilized in China for the treatment of acute leukemia and lymphoma. However, the amounts of HT in the Cephalotaxus species are very small; therefore, plant tissue cultures have been focused upon to enhance HT production. Qualitative/quantitative methods for HT detection are required to screen superior cell lines. We developed a one-step indirect competitive immunochromatographic assay (ICA) using colloidal gold nanoparticles conjugated with highly specific monoclonal antibodies against HT (MAb 1D2) for simple, rapid, and sensitive detection of HT in plant samples. This ICA can be completed in 15min after dipping the strip into analytes with a limit of detection of ∼313ng/mL. In developed ICA, fiber pad which is usually used for conventional ICA, was not used to shorten the time for preparing chromatographic strip, resulting in a decrease in the volume of valuable analytes (20µL). Considering simplicity, rapidity, and sensitivity of the developed ICA, this study could be applied to a fieldwork study for finding new natural resources containing HT.

[Ab] Resumo:Colloidal gold and Eu -doped fluorescent microspheres were applied as labels to develop the immunochromatographic strips for detecting melamine in milk. Under the optimized condition, the visual detection limit of colloidal gold-immunochromatographic test strip (ICTS) was 150 µg/L of melamine in phosphate-buffered saline (PBS), although the visual detection limit of fluorescent nanoparticles (FN)-ICTS was 75 µg/L in PBS. As thermal acceleration test, FN-ICTS could be stored at 37 °C for at least 11 d before sample testing, but the color of the lines on colloidal gold-ICTS faded away after 7-d storage. The visual result of FN-ICTS was more stable than that of colloidal gold-ICTS, and the fluorescence intensity of the line on FN-ICTS could be maintained up to 30 d at 22 °C after sample testing. Once the immunochromatographic strips were used to detect melamine in milk, no negative effect of milk components on the performance of FN-ICTS was identified, whereas the performance of colloidal gold-ICTS was significantly influenced by milk matrix.

[Ab] Resumo:Dielectrophoretic motions of Saccharomyces cerevisiae (yeast) cells and colloidal gold are investigated using electrochemically modified electrodes exhibiting fractal topology. Electrodeposition of gold on electrodes generated repeated patterns with a fern-leaf type self-similarity. A particle tracking algorithm is used to extract dielectrophoretic particle velocities using fractal and planar electrodes in two different medium conductivities. The results show increased dielectrophoretic force when using fractal electrodes. Strong negative dielectrophoresis of yeast cells in high-conductivity media (1.5 S/m) is observed using fractal electrodes, while no significant motion is present using planar electrodes. Electrical impedance at the electrode/electrolyte interface is measured using impedance spectroscopy technique. Stronger electrode polarization (EP) effects are reported for planar electrodes. Decreased EP in fractal electrodes is considered as a reason for enhanced dielectrophoretic response.

[Ab] Resumo:To overcome the low bioavailability of siRNA (small interfering RNA) and to improve their transfection efficiency, the use of non-viral delivery carriers is today a feasible approach to transform the discovery of these incredibly potent and versatile drugs into clinical practice. Polymer-modified gold nanoconstructs (AuNCs) are currently viewed as efficient and safe intracellular delivery carriers for siRNA, as they have the possibility to conjugate the ability to stably entrap and deliver siRNAs inside cells with the advantages of gold nanoparticles, which can act as theranostic agents and radiotherapy enhancers through laser-induced hyperthermia. In this study, AuNCs were prepared by coating Gold Nano Stars (GNS) with suitable functionalised polymers, to give new insight on the choice of the coating in order to obtain colloidal stability, satisfying in vitro transfection behaviour and reliability in terms of homogeneous results upon GNS type changing. For this goal, GNS synthesized with three different sizes and shapes were coated with two different polymers: i) α-mercapto-ω-amino polyethylene glycol 3000Da (SH-PEG -NH ), a hydrophilic linear polymer; ii) PHEA-PEG -EDA-LA (PPE-LA), an amphiphilic hydroxyethylaspartamide copolymer containing a PEG moiety. Both polymers contain SH or SS groups for anchoring on gold surface and NH groups, which can be protonated in order to obtain a positive surface for successive siRNA layering. The effect of the features of the coating polymers on siRNA layering, and the extent of intracellular uptake and luciferase gene silencing effect were evaluated for each of the obtained coated GNS. The results highlight that amphiphilic biocompatible polymers with multi-grafting function are more suitable for ensuring the colloidal stability and the effectiveness of these colloidal systems, compared to the coating with linear PEG.

[Ab] Resumo:It is known that the median preoptic nucleus (POMe) sends dense projections to the subfornical organ (SFO). However, the functional significance of these projections have not been well discussed. In this electron microscopic study, we investigated the presence of synapses between POMe-derived axon terminals and SFO neurons that project to the paraventricular hypothalamic nucleus (PVN). After injection of a retrograde tracer, wheat germ agglutinin-conjugated horseradish peroxidase-colloidal gold complex, into the PVN, many labeled neurons were found in the SFO. In contrast, after injection of an anterograde tracer, biotinylated dextran amine, in the POMe, abundant labeled axon varicosities were observed in the SFO. Using electron microscopy, synapses were identified between retrogradely labeled dendrites and cell bodies, and anterogradely labeled axon terminals, indicating that POMe neurons innervate SFO neurons projecting to the PVN. The possibility that POMe neurons play multiple roles in the neuronal circuit responsible for vasopressin release and/or cardiovascular regulation is also discussed.
