[Ab] Resumo:BACKGROUND: Wheat is the staple food for most of the world's population; however, it is a poor source of zinc. Foliar fertilization of zinc via zinc loaded chitosan nanocarriers (Zn-CNP) post-anthesis has proved to be a promising approach for grain zinc enhancement in durum wheat as evidenced in our earlier study. However, the molecular mechanism of uptake of zinc via Zn-CNP remains unclear. METHODS/PRINCIPLE FINDINGS: Foliar application of Zn-CNP was performed at post anthesis stages in two durum wheat cultivars (MACS 3125 and UC1114, containing the Gpc-B1 gene), and expression levels of several metal-related genes were analyzed during early senescence. Zn-CNP application indeed caused changes in gene expression as revealed by qPCR data on representative genes involved in metal homeostasis, phloem transporters, and leaf senescence. Furthermore, zinc-regulated transporters and iron (Fe)-regulated transporter-like protein (ZIP) family [ZIP1, ZIP7, ZIP15], CA (carbonic anhydrase), and DMAS (2'-deoxymugineic acid synthase) in flag leaves exhibited significant correlation with zinc content in the seeds. The analysis of grain endosperm proteins showed enhancement of gamma gliadins while other gluten subunits decreased. Gene expression within ZIP family members varied with the type of cultivar mostly attributed to the Gpc-B1, concentration of external zinc ions as well as the type of tissue analyzed. Correlation analysis revealed the involvement of the selected genes in zinc enhancement. CONCLUSION: At the molecular level, uptake of zinc via Zn-CNP nanocarrier was comparable to the uptake of zinc via common zinc fertilizers i.e. ZnSO4.

[Ab] Resumo:Soil remediation mitigates hazards from contaminants but could deprive soils of initial biota and enzymes. Historically contaminated acidic soil from Arnoldstein (Austria) and calcareous soil from Meza (Slovenia) were washed with 30 and 100 mmol kg ethylenediaminetetraacetate (EDTA) to remove 78 and 60% of Pb as a main pollutant. Remediation of the Arnoldstein soil decreased urease activity and increased ß-glucosidase activity, measured in a 15-week experiment. The dehydrogenase activity and microbial gene abundances were not significantly impeded compared to the original soil. Conversely, the use of a high dose of EDTA in the Meza soil, necessary for effective remediation of calcareous soils, resulted in pronouncedly decreased enzyme activities (3.2 times on average) and repressed fungal ITS and increased bacterial 16S rRNA gene abundance. Remediation shifted the microbial community composition in both soils. For revitalisation, the remediated soils were amended with compost, inocula of un-contaminated soil and (Arnoldstein soil) biochar enriched with soil extract. Amendments inconsistently affected the Arnoldstein soil: compost increased the dehydrogenase activity and altered the microbial community composition, biochar enhanced the ß-glucosidase activity, and all amendments decreased the microbial abundance (1.6 times on average). In contrast, amendments efficiently revitalised the remediated Meza soil; compost and soil inoculum returned the enzyme activities back to the baseline in the original soil, increased the fungal abundance above that in the original soil and restored the microbial community composition.

[Ab] Resumo:Silkworm silk is an attractive biopolymer for biomedical applications due to its high mechanical strength and biocompatibility; as a result, there is increasing interest in scalable devices to spin silk and recombinant silk so as to improve and customize their properties for diverse biomedical purposes (Vepari and Kaplan 2007 Prog. Polym. Sci. 32 ). While artificial spinning of regenerated silk fibroins adds tunability to properties such as degradation rate and surface functionalization, the resulting fibers do not yet approach the mechanical strength of native silkworm silk. These drawbacks reduce the applicability and attractiveness of artificial silk (Kinahan et al 2011 Biomacromolecules 12 ). Here, we used computational fluid dynamic simulations to incorporate shear in tandem with biomimetic ion gradients by coupling a modular novel glass microfluidic device to our previous co-axial flow device. Fibers spun with this combined apparatus demonstrated a significant increase in mechanical strength compared to fibers spun with the basic apparatus alone, with a three-fold increase in Young's modulus and extensibility and a twelve-fold increase in toughness. These results thus demonstrate the critical importance of ionic milieu and shear stress in spinning strong fibers from solubilized silk fibroin.

[Ab] Resumo:The Mott metal-insulator transition-a manifestation of Coulomb interactions among electrons-is known as a discontinuous transition. Recent theoretical studies, however, suggest that the transition is continuous if the Mott insulator carries a spin liquid with a spinon Fermi surface. Here, we demonstrate the case of a quasi-continuous Mott transition from a Fermi liquid to a spin liquid in an organic triangular-lattice system κ-(ET) Cu (CN) . Transport experiments performed under fine pressure tuning have found that as the Mott transition is approached, the Fermi liquid coherence temperature continuously falls to the scale of kelvins, with a divergent quasi-particle decay rate on the metal side, and the charge gap continuously closes on the insulator side. A Clausius-Clapeyron analysis provides thermodynamic evidence for the extremely weak first-order nature of the transition. These results provide additional support for the existence of a spinon Fermi surface, which becomes an electron Fermi surface when charges are delocalized.

[Ab] Resumo:INTRODUCTION: Percutaneous nephrolithotomy is an efficient approach for treatment of different types of kidney stones. Various types of access techniques have been described like sequential dilatation and one-shot procedure. OBJECTIVE: To determine the differences in time of exposure to X-rays and hemoglobin levels between techniques. METHODS: Controlled clinical trial. Patients older than 18 years with complex/uncomplicated kidney stones, without urine infection were included. They were assigned randomly to one of the two techniques. Response variables were determined before and 24 h after procedures. RESULTS: 59 patients were included: 30 underwent one-shot procedure (study-group) and 29 sequential dilatation (control-group). Baseline characteristics were similar. Study group had a lower postoperative hemoglobin decline than control group (0.81 vs. 2.03 g/dl, respectively; p < 0.001); X-ray exposure time (69.6 vs. 100.62 s; p < 0.001) and postoperative creatinine serum levels (0.93 ± 0.29 vs. 1.13 ± 0.4 mg/dl; p = 0.039). No significant differences in postoperative morbidity were found. CONCLUSION: One-shot technique demonstrated better results compared to sequential dilatation.

[Ab] Resumo:In the supramolecular chemistry field, coordination-driven self-assembly has provided the basis for tremendous growth across many subdisciplines, spanning from fundamental investigations regarding the design and synthesis of new architectures to defining different practical applications. Within this framework, supramolecular coordination complexes (SCCs), defined as large chemical entities formed from smaller precursor building blocks of ionic metal nodes and organic multidentate ligands, resulting in intricate and well-defined supramolecular structures, hold great promise. Notably, interest in the construction of discrete 3D molecular architectures, such as those offered by SCCs, has experienced extraordinary progress because of their potential application as sensors, catalysts, probes, and containers and in basic host-guest chemistry. Despite numerous synthetic efforts and a number of inherent favorable properties, the field of 3D SCCs for biomedical applications is still in its infancy. This Viewpoint focuses on 3D SCCs, specifically metallacages and helicates, first briefly presenting the fundamentals in terms of the synthesis and characterization of their host-guest properties, followed by an overview of the possible biological applications with representative examples. Thus, emphasis will be given in particular to metallacages as drug delivery systems and to chiral helicates as DNA recognition domains. Overall, we will provide an update on the state-of-the-art literature and will define the challenges in this fascinating research area at the interface of different disciplines.

[Ab] Resumo:Dust is a mixture of natural and anthropogenic particles originated from multiple sources, which can represent an hazard for human health. Playgrounds are a site of particularly concern, due to sand dust ingestion by toddlers and inhalation. In this study, 37 sands used in public playgrounds in the city of Barcelona were physico-chemically characterized also in relation to routine maintenance activities such as disinfection and sand renewal. The analyzed sands show a felsic mineralogy dominated by Na-feldspar, quartz, and, to a lesser extent, K-feldspar, with minor amounts of clay minerals, carbonates and hematite. Particle fractions below 10, 2.5 and 1 µm represent, on average, 0.65%, 0.17% and 0.07% of bulk volume, respectively, although, due to the human grinding, these initial fractions increased every year by a 18%, 5% and 2% respectively. Disinfection of sands effectively reduced only the NH concentration, among inorganic species. The average metal content was anthropogenically enriched, with respect to the upper continental crust, only for Sb and As. Both elements show high spatial variation indicating local sources such as road traffic for Sb (contributing mostly to the total concentration), and industry for As (also contributing with highly bioaccessible Sb, Cu and Zn). A clear inverse relationship between total concentrations of some elements and their leachable (Sb) and bioaccessible (Sb and Cr) fractions is observed. The most bioaccessible elements were Ca > Ni > Cu > Sr > Cd > Pb, all above the 25% of the total concentration. Bioaccessibility was higher for the carbonate-bearing particles and for the anthropic emitted metals (>50% of Ba, Cu, K, Pb and Zn).

[Ab] Resumo:An efficient method for rare metal recovery from environmental water and urban mines is in high demand. Toward rapid and high-resolution rare metal ion separation, a novel bis(2-ethylhexyl) phosphate (HDEHP)-impregnated graft-type particle as a filler for a chromatography column is proposed. To achieve rapid and high-resolution separation, a convection-flow-aided elution mode is required. The combination of 35 µm non-porous particles and a polymer-brush-rich particle structure minimizes the distance from metal ion binding sites to the convection flow in the column, resulting in minimized diffusional mass transfer resistance and the convection-flow-aided elution mode. The HDEHP-impregnated graft-type non-porous-particle-packed cartridge developed in this study exhibited a higher separation performance for model rare metals, neodymium (III) and dysprosium (III) ions, and a narrower peak at a higher linear velocity, than those of previous HDEHP-impregnated fiber-packed and commercially available Lewatit VP OC 1026-packed cartridges.

[Ab] Resumo:BACKGROUND: Data regarding the long-term efficacy and safety of everolimus-eluting bioresorbable vascular scaffolds (BVS) compared with everolimus-eluting stents are limited. This meta-analysis aimed to compare the long-term outcomes with both devices. METHODS AND RESULTS: Randomized trials reporting clinical outcomes beyond 1 year and comparing BVS with everolimus-eluting stents were included. Summary estimates risk ratios (RRs) were constructed. The primary efficacy outcome was target lesion failure, defined as cardiac death, target vessel myocardial infarction, and ischemia-driven target lesion revascularization, and the primary safety outcome was definite or probable stent/scaffold thrombosis. Six trials with 5392 patients were included (mean follow-up, 25 months). BVS had a higher rate of target lesion failure (RR, 1.33; 95% confidence interval [CI], 1.11-1.58) driven by the higher rates of target vessel myocardial infarction (RR, 1.65; 95% CI, 1.26-2.17) and target lesion revascularization (RR, 1.39; 95% CI, 1.08-1.78). The risk of definite or probable stent/scaffold thrombosis (RR, 3.22; 95% CI, 1.89-5.49) and very late stent/scaffold thrombosis (>1 year; RR, 4.78; 95% CI, 1.66-13.8) was higher with BVS. The risk of cardiac and all-cause mortality was similar in both groups. CONCLUSIONS: Compared with everolimus-eluting stents, BVS is associated with increased risk of target lesion failure driven by the increased rates of target vessel myocardial infarction and ischemia-driven target lesion revascularization in these studies (mean follow-up, 25 months). The risk of definite or probable stent/scaffold thrombosis and very late stent/scaffold thrombosis seems to be higher with BVS. Further information from randomized trials is critical to evaluate clinical outcomes with BVS on complete resolution of the scaffold.

[Ab] Resumo:The concept of point of darkness has received much attention for biosensing based on phase-sensitive detection and perfect absorption of light. The maximum phase change is possible at the point of darkness where the reflection is almost zero. To date, this has been experimentally realized using different material systems through the concept of topological darkness. However, complex nanopatterning techniques are required to realize topological darkness. Here, we report an approach to realize perfect absorption and extreme phase singularity using a simple metal-dielectric multilayer thin-film stack. The multilayer stack works on the principle of an asymmetric Fabry-Perot cavity and shows an abrupt phase change at the reflectionless point due to the presence of a highly absorbing ultrathin film of germanium in the stack. In the proof-of-concept phase-sensitive biosensing experiments, we functionalize the film surface with an ultrathin layer of biotin-thiol to capture streptavidin at a low concentration of 1 pM.
