[Ab] Resumo:The aim of this study was to elucidate the redox effects of Thiophanate methyl (MT) in the rat liver and kidney. Our results showed, after 3 days of MT injection (700 mg/kg), an increase in malondialdehyde (MDA), hydrogen peroxide and advanced oxidation protein products levels. Glutathione peroxidase and superoxide dismutase activities were also remarkably increased in the liver but decrease in the kidney. Glutathione and vitamin C values were significantly reduced. The changes in biochemical parameters were substantiated by histological and molecular data. A smear without ladder formation on agarose gel was shown, indicating random DNA degradation in the liver and the kidney of MT treated rats. The increase in cyclooxygenase-2 gene expression, marker of inflammation, and an increase in genes expression of glutathione peroxidase and superoxide dismutase in liver and their decrease in the kidney were also occurred after MT exposure. These data confirmed the pro-oxidant and genotoxic effects of this fungicide.

[Ab] Resumo:This research aims at determining residues of thiophanate methyl and propiconazole in grape and mango fruits as an indication for their persistence in this environmental compartment. Fruit extracts were analyzed for thiophanate methyl using High Performance Liquid Chromatography and using Gas Chromatography Electron Capture Detector (GC/ECD), respectively. The results indicated that propiconazole had a less environmental impact since propiconazole had shorter residue half-lives which were 1.24 and 1.19 days in grape and mango fruits, respectively, while thiophanate methyl had half-lives of 2.49 and 2.64 days in mango and grape, respectively. The degradation rates of propiconazole in grape and mango fruits did not change significantly and neither did those of thiophanate methyl. According to the maximum residue level, the pre-harvest intervals of propiconazole were set to be 3 and 7 days for grape and mango fruits, respectively, and the pre-harvest intervals for thiophanate methyl were 15 days for both grape and mango fruits. Propiconazole was generally considered to be less hazardous to humans and will leave the environment less altered because of its faster degradation than that of thiophanate methyl.

[Ab] Resumo:Surface-enhanced Raman scattering (SERS) imaging coupling with multivariate analysis in spectral region of 200 to 1800cm was developed to quantify and visualize thiophanate-methyl (TM) and its metabolite carbendazim residues in red bell pepper (Capsicum annuum L.). Least squares support vector machines (LS-SVM) and support vector machines (SVM) models based on seven optimized characteristic peaks that showed SERS effects of TM and its metabolite carbendazim residues were employed to establish prediction models. SERS spectra with first derivative (1st) and second derivative (2nd) method were subsequently compared and the optimized model of 1st-LS-SVM acquired showed the best performance (RPD=6.08, R =0.986 and RMSEP=0.473). The results demonstrated that SERS imaging with multivariate analysis had the potential for rapid determination and visualization of the trace TM and its metabolite carbendazim residues in complex food matrices.

[Ab] Resumo:Previous research has shown that Botrytis cinerea isolates with resistance to multiple chemical classes of fungicides exist in eastern strawberry fields. In this study, the fungicide resistance profiles of 2,130 isolates from flowers of commercial strawberry fields located in multiple states was determined over four consecutive strawberry production seasons. Producers were asked to alternate single-site fungicides that were considered low risk in their specific location based on resistance monitoring results in their fields. This recommendation led to an increase of chemical class diversity used in the spray programs. Results indicated that simultaneous resistance in individual isolates to two, three, four, five, six, and seven classes of fungicides increased over time. The increase in chemical class resistances within isolates was likely due to a process we termed "selection by association", where fungicide resistance traits were often linked to the trait being selected rather than the selectable trait itself. Data analysis also indicated that the odds were highest for isolates resistant to one chemical class (1CCR) to be resistant to thiophanate-methyl; for 2CCR isolates to be resistant to thiophanate-methyl and pyraclostrobin; and for 3CCR isolates to be resistant to thiophanate-methyl, pyraclostrobin, and either cyprodinil or fenhexamid. We hypothesize that the more chemical classes are used in a spray program, the faster isolates will be selected with increasing numbers of chemical class resistances by virtue of selection by association if such isolates preexist in the population.

[Ab] Resumo:UV-visible irradiation of thiophanate-methyl (TM) led to the formation of nine photoproducts that were characterized by high performance liquid chromatography coupled with high resolution Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS). Although carbendazime has been reported in the literature to be the major metabolite and photoproduct of thiophanate-methyl, it was not detected in this study. However, an isomer of carbendazime referred as PP2, which was unambiguously characterized owing to CID experiments, was found in great abundance. Grape berries and cherry tomatoes treated with aqueous solutions of thiophanate-methyl were submitted to irradiation under laboratory conditions. TM and PP2 were detected in both peel and flesh of berries. The ability of TM and PP2 to pass through the fruit skin has been shown to be highly compound and matrix dependent. In vitro bioassays on Vibrio fischeri bacteria showed that the global ecotoxicity of the TM solution increases significantly with the irradiation time. PP2 should likely contribute to this ecotoxicity enhancement since in silico estimations for Daphnia magna provide a LC50 value seven times lower for PP2 than for the parent molecule.

[Ab] Resumo:A quick, easy, cheap, effective, rugged, and safe QuEChERS (method) was used for the simultaneous detection of four veterinary drug residues, namely naloxone, yohimbine, thiophanate, and altrenogest, in porcine muscle, using liquid chromatography with electrospray ionization triple quadrupole tandem mass spectrometry. Because of the unavailability of a suitable internal standard, matrix-matched calibrations were used for quantification, with determination coefficients ≥ 0.9542. The accuracy (expressed as recovery %) ranged from 60.53 to 83.25%, and the intra- and interday precisions (expressed as relative standard deviations) were <12%. The limits of quantification were 5, 0.5, 2, and 5 ng/g for naloxone, yohimbine, thiophanate, and altrenogest, respectively. Samples purchased from local markets in Seoul, Republic of Korea, revealed no traces of the target analytes. The developed method described herein is sensitive and reliable and can be applied to quantify the tested veterinary drugs in animal tissues.

[Ab] Resumo:BACKGROUND: Cucurbit powdery mildew caused by Podosphaera xanthii limits crop production in Spain, where disease control is largely dependent on fungicides. In previous studies, high levels of resistance to QoI and DMI fungicides were documented in south-central Spain. The aim of this study was to investigate the sensitivity of P. xanthii populations to other fungicides and to provide tools for improved disease management. RESULTS: Using a leaf-disc assay, sensitivity to thiophanate-methyl, bupirimate and quinoxyfen of 50 isolates of P. xanthii was analysed to determine discriminatory concentrations between sensitive and resistant isolates. With the exception of thiophanate-methyl, no clearly different groups of isolates could be identified, and as a result, discriminatory concentrations were established on the basis of the maximum fungicide field application rate. Subsequently, a survey of P. xanthii resistance to these fungicides was carried out by testing a collection of 237 isolates obtained during the 2002-2011 cucurbit growing seasons. This analysis revealed very high levels of resistance to thiophanate-methyl (95%). By contrast, no resistance to bupirimate and quinoxyfen was found. CONCLUSION: Results suggest that thiophanate-methyl has become completely ineffective for controlling cucurbit powdery mildew in Spain. By contrast, bupirimate and quinoxyfen remain as very effective tools for cucurbit powdery mildew management. © 2014 Society of Chemical Industry.

[Ab] Resumo:Methylthiophanate is one of the widely used fungicides to control important fungal diseases of crops. The aim of this study was to elucidate the short-term hematoxicity and genotoxicity effects of methylthiophanate administered by intraperitoneal way at three doses (300, 500 and 700 mg/kg of body weight) after 24, 48 and 72 h. Our results showed, 24 h after methylthiophanate injection, a hematological perturbation such as red blood cells (p < 0.05, p < 0.05 and p < 0.01) and hemoglobin content (p < 0.05), respectively, and a noticeable genotoxic effect in WBC evidenced by a significant increase in the frequency of the micronuclei and a decrease in cell viability. An increase in erythrocyte osmotic fragility was also noted after 24 and 48 h of methylthiophanate treatment at graded doses. A significant increase in hydrogen peroxide, advanced oxidation of protein products and malondialdehyde levels, in erythrocytes of methylthiophanate-treated rats with 300, 500 and 700 mg/kg of body weight, was also observed after 24 h of treatment (p < 0.05, p < 0.01 and p < 0.001, respectively), suggesting the implication of oxidative stress in its toxicity. Antioxidants activities of superoxide dismutase and glutathione peroxidase in erythrocytes significantly increased (p < 0.001) 24 h after the highest dose injected. While all these parameters were improved after 72 h of methylthiophanate injection (300, 500 and 700 mg/kg body weight). In conclusion, these data showed that the exposure of adult rats to methylthiophanate resulted in oxidative stress leading to hematotoxicity and the impairment of defence system, confirming the pro-oxidant and genotoxic effects of this fungicide.

[Ab] Resumo:BACKGROUND: The benzimidazole fungicide thiophanate-methyl is commonly used for the control of brown rot of stone fruits. Low and high levels of resistance to this fungicide have been found in field isolates of the causal pathogen Monilinia fructicola. RESULTS: The minor groove binding (MGB) TaqMan probes specific for alleles E198A and H6Y conferring the high and low levels of resistance in the ß-tubulin gene of M. fructicola were designed. A duplex real-time PCR assay based on these probes was developed for simultaneous quantification of both mutations in a pathogen population. The specificity tests for the primers and probes were conducted using different fungal species of stone and pome fruit pathogens. Similar results were obtained between the duplex real-time (TaqMan) PCR assay and the conventional method to quantify the frequencies of alleles E198A and H6Y of eight samples from different peach orchards. CONCLUSION: The MGB TaqMan probe based duplex real-time PCR provides a useful tool for simultaneous quantification of both alleles, E198A and H6Y, conferring high and low resistance, and has a potential in monitoring the benzimidazole-resistance in M. fructicola populations in stone and pome fruit orchards.

[Ab] Resumo:A method was developed using matrix solid-phase dispersion, together with liquid chromatography with ultraviolet diode array detector for determination of carbofuran, difenoconazole, ß-cyfluthrin, spirodiclofen and thiophanate-methyl in stem of coconut palm. The best results were obtained using 2.0 g of stem, 1.6 g of Florisil as sorbent and cyclohexane:acetone mixture (4:1). The method was validated using stem samples spiked with pesticides at four concentration levels (0.05-2.0 µg/g). Average recoveries ranged from 70 % to 114.3 %, with relative standard deviations between 1.2 % and 19.2 %. Detection and quantification limits were in the ranges 0.02-0.03 and 0.05-0.1 µg/g, respectively.
