[Ab] Resumo:A high-throughput small-molecule screen was conducted to identify inhibitors of epithelial-mesenchymal transition (EMT) that could be used as tool compounds to test the importance of EMT signaling in vivo during fibrogenesis. Transforming growth factor (TGF)-ß1-induced fibronectin expression and E-cadherin repression in A549 cells were used as 48-hour endpoints in a cell-based imaging screen. Compounds that directly blocked Smad2/3 phosphorylation were excluded. From 2,100 bioactive compounds, methacycline was identified as an inhibitor of A549 EMT with the half maximal inhibitory concentration (IC50) of roughly 5 µM. In vitro, methacycline inhibited TGF-ß1-induced α-smooth muscle actin, Snail1, and collagen I of primary alveolar epithelial cells . Methacycline inhibited TGF-ß1-induced non-Smad pathways, including c-Jun N-terminal kinase, p38, and Akt activation, but not Smad or ß-catenin transcriptional activity. Methacycline had no effect on baseline c-Jun N-terminal kinase, p38, or Akt activities or lung fibroblast responses to TGF-ß1. In vivo, 100 mg/kg intraperitoneal methacycline delivered daily beginning 10 days after intratracheal bleomycin improved survival at Day 17 (P < 0.01). Bleomycin-induced canonical EMT markers, Snail1, Twist1, collagen I, as well as fibronectin protein and mRNA, were attenuated by methacycline (Day 17). Methacycline did not attenuate inflammatory cell accumulation or alter TGF-ß1-responsive genes in alveolar macrophages. These studies identify a novel inhibitor of EMT as a potent suppressor of fibrogenesis, further supporting the concept that EMT signaling is important to lung fibrosis. The findings also provide support for testing the impact of methacycline or doxycycline, an active analog, on progression of human pulmonary fibrosis.

[Ab] Resumo:This study was designed to examine the interaction of methacyline (METC) with human serum albumin (HSA) by multispectroscopy and a molecular modeling method under simulative physiological conditions. The quenching mechanism was suggested to be static quenching based on fluorescence and ultraviolet-visible (UV-Vis) spectroscopy. According to the Vant' Hoff equation, the values of enthalpy (∆H) and entropy change (∆S) were calculated to be -95.29 kJ/mol and -218.13 J/mol/K, indicating that the main driving force of the interaction between HSA and METC were hydrogen bonds and van der Waals's forces. By performing displacement measurements, the specific binding of METC in the vicinity of Sudlow's site I of HSA was clarified. An apparent distance of 3.05 nm between Trp214 and METC was obtained via the fluorescence resonance energy transfer (FRET) method. Furthermore, the binding details between METC and HSA were further confirmed by molecular docking studies, which revealed that METC was bound at subdomain IIA through multiple interactions, such as hydrophobic effect, polar forces, hydrogen bonding, etc. The results of three-dimensional fluorescence and Fourier transform infrared (FTIR) spectroscopy showed that METC caused conformational and some microenvironmental changes in HSA and reduced the α-helix significantly in the range of 52.3-40.4% in HSA secondary structure. Moreover, the coexistence of metal ions such as Ca(2+), Al(3+), Fe(3+), Zn(2+), Cu(2+), Cr(3+) and Cd(2+) can decrease the binding constants of METC-HSA.

[Ab] Resumo:Mg(2+) -sensitized metacycline fluorescence microscopic imaging technique was applied to detect the raw fresh milk of four cows breeding farms in the Miyun County of Beijing based on the capillary effect of solvent on solid supports. In the presence of NH3-NH4 Cl buffer solution (pH 9.99) and PVA-124, Mg2+ and metacycline can form a strong fluorescence complex of 1 : 1, and Mg(2+) -metacycline complex can form an SOR on the hydrophobic supports with the diameter of 0.93 mm and its ring belt width of 26.2 microm. By measuring the fluorescence intensity of the ring, the quantitative analysis of metacycline was achieved with the detection limit (3sigma) of 8.8 x 10(-14) mol x ring(-1) (1.8 x 10(-7) mol x L(-1)) and linear range of 2.2 x 10(-13) -3.6 x 10(-12) mol x ring(-1) (4.4 x 10(-7) -7.2 x 10(-6) mol x L(-1)) when 0.50 microL droplet was spotted. This method has been satisfactorily applied to the determination of metacycline in the raw fresh milk samples with the recovery of 93.8%-108% and RSD less than 4.3%.

[Ab] Resumo:Tetracycline possesses a great tendency to form complexes with a number of chemical species, particularly with Eu3+ ions. In this work we investigate the europium optical properties of three tetracyclines europium complexes: Metacycline (MTc), Oxytetracycline (OTc), and Chlortetracycline (CTc), in the presence and absence of hydrogen peroxide (HP). The results show that the emission band of EuOTc have enhancement in the presence of hydrogen peroxide. A calibration curve was shown for this complex with the best molar ratio obtained.

[Ab] Resumo:The fluorescence of metacycline enhanced by the synergetic effect of beta-cyclodextrin with anion surfactant sodium dodecylbenzene sulfonate (DBS) was studied. It was found that the fluorescence intensity of metacycline was greatly enhanced in the system of beta-CD-DBS. Based on this a new sensitive method for the determination of metacycline was established. The experiments indicated that MTC-beta-CD-DBS can form 1 : 1 : 1 ternary inclusion complex with apparent stability constant of 1 294 L2 x mol(-2), respectively. The fluorescence intensity was linearly related to the metacycline concentration in the range of 1.7 x 10(-7) to 8.8 x 10(-5) mol x L(-1) with a correlation coefficient of 0.996 6. The detection limit was 7.4 x 10(-9) mol x L(-1) this method was satisfactorily applied to the determination of metacycline in pharmaceutical preparations. The recovery was within the range of 95% to 106%.

[Ab] Resumo:An automated system using on-line solid-phase extraction (SPE) high-performance liquid chromatography (HPLC) with ultraviolet (UV) detection was developed for the determination of tetracyclines (TCs), such as tetracycline (TC), oxytetracycline (OTC), chlortetracycline (CTC), metacycline (MC), and doxycycline (DC) in honey. One milliliter diluted honey sample was injected into a conditioned C18 SPE column and the matrix was washed out with water for 3 min. By rotation of the switching valve, TCs were eluted and transferred to the analytical column by the chromatographic mobile phase. Chromatographic conditions were optimized. TCs were separated in less than 8 min with a gradient elution using a mixture of 0.8% formic acid and acetonitrile. The UV detection was performed at 365 nm. The conditions for on-line SPE, including solvent and total time for loading sample and washing matrix were also optimized. Time for extraction and separation decreased greatly. For the five kinds of TCs, the limits of detection (LODs) at a signal-to-noise of 3 ranged from 5 to 12 ng g(-1). The relative standard deviations (R.S.D.) for the determination of TCs ranged from 3.4 to 7.1% within a day and ranged from 3.2 to 8.9% in 3 days, respectively.

[Ab] Resumo:Eight antibiotics, chlortetracycline, demeclocycline, doxycycline, methacycline, minocycline, oxytetracycline, tetracycline and rolitetracycline, were separated and quantified in Spanish honey extracts of different floral origin using a commercial RP-C18 HPLC column and two different on-line detectors (diode array and electrospray time-of-flight mass spectrometry (ESI-TOF-MS) systems). Operating a linear gradient at a flow of 2 ml min(-1) the HPLC separation of the eight antibiotics was obtained within 10 min with good peak symmetry and an acceptable resolution (2.1) for the critical band pair rolitetracycline and oxytetracycline. Values of the numbers of theoretical plates (N) were comprised between 2328 and 19448 while the limits of detection in honey were within 0.02-1.03 microg kg(-1) in the case of UV detection and 0.05-0.76 microg kg(-1) for ESI-TOF-MS detection (operating in negative mode). A recovery study was carried out by preparing some quality control samples at four levels of concentration (10, 25, 50 and 100 microg kg(-1)) and percentages between 72% and 98% were attained.

[Ab] Resumo:An internal solid contact sensor (ISCS) for the determination of methacycline hydrochloride (MC.Cl), Pt/PPy/PVC(MC-PT), is described, based on the use of conducting poly(pyrrole) (PPy) as solid contact material and MC-phosphotungstate (PT) as the ion exchanger and dibutyl phthalate (DBP) as the plasticizer. A direct potentiometric method for the quantitative analysis of MC.Cl is also described. Under the condition of pH 2.7, the linear concentration range, slope (25 degrees C) and detection limit of the sensor are 6.4 x 1.0(-6) - 3.0 x 1.0(-3) M, 52.4 +/- 0.2 mV/decade and 4.4 x 1.0(-6) M, respectively. The response time is <5 s. The determinations of MC.Cl in tablets were carried out by direct potentiometry. The average recovery and relative standard deviation are 100.1 and 0.7% (n = 4), respectively.

[Ab] Resumo:A new synchronous-derivative fluorimetric method for the determination of metacycline (MTC) in the presence of oxytetracycline (OTC) is proposed. The degradation products of MTC in 0.1 mol x L(-1) NaOH are strongly fluorescent. The addition of cetyltrimethylammonium bromide (CTMAB) enhances the fluorescence intensity 2-fold. The synchronous fluorescence spectra of MTC and OTC degradation products do not interfere with each other under the condition that deltalambda = 100 nm. MTC can be determined in the presence of OTC in the range of OTC+MTC ratio of 1+116-1+1. The recoveries of MTC in the mixture sample are in the range of 94%-102%.

[Ab] Resumo:A CE method for metacycline (MTC) determination was investigated in an inter-laboratory experiment. Many problems were encountered in this study, most of which were related to the transfer of the method to different CE equipment. The reported problems could be classified into different categories: problems related to the precision, to the parameters in the protocol, and to the MTC peak shape. As the peak shape problem was partially responsible for the poor precision, a new CE method was developed in order to obtain a good MTC peak shape on all equipment. The precision of this new method for MTC determination was examined in an intermediate precision study, where the influence of the factors "time" and "equipment" was investigated. Although the new method could be transferred to different instruments, the precision remained poor mainly due to the contributions of the between-replicate and the between-injection variances.
