[Ab] Resumo:The aim of this study was the assessment of levels of pollution of territories of preschool educational institutions (PEI) by components of vehicle emissions, in dependence on their locations relatively to highways with different traffic load and, in this regard, the assessment of children health according to the incidence on the seeking medical advice. To achieve this goal there were solved following tasks: to assess air pollution levels of PEI territories by components of vehicle emissions; to evaluate levels and the structure of morbidity rate according to seeking medical advice by children attending observed PEI. In the article there is presented the evaluation of air pollution levels of the components of vehicle emissions territories 4 PEI of the city of Irkutsk located near to highways with different intensity the road transport load. The research results were obtained with the use of «Methodics for the determination vehicle emissions for summary calculations of ambient air pollution in cities¼, which allows to determine the maximum emissions of a moving vehicle per time unit (g/s) for the main combustion components and with following use of software «Superhighway -city¼ to calculate surface concentrations of considered pollutants in the surface ambient air layer (1 m) in MPC proportions. The highest levels of contamination on the content of nitric oxide and benzo- (a)-pyrene (4 and 6,5 MPC respectively) are registered in the territories adjacent to the PEI motorways with high load tracking. To assess the morbidity rate according to seeking medical advice by children attending PEI with different road transport load there were used records from outpatient medical cards «Medical card of the child¼ (f.112/y). Morbidity rate according to seeking medical advice was evaluated in dynamics throughout five years and calculated per 1000 cases. The total sample size accounted for 670 children. The highest morbidity incidence rate levels according both to seeking medical advice and the class of respiratory diseases were recorded in PEI located in close proximity to the highway with a high load of vehicle transport.

[Ab] Resumo:Routine monitoring of workplace exposure to polycyclic aromatic hydrocarbons (PAHs) is performed mainly via active sampling. However, active samplers have several drawbacks and, in some cases, may even be unusable. Polyurethane foam (PUF) as personal passive air samplers constitute good alternatives for PAH monitoring in occupational air (8 h). However, PUFs must be further tested to reliably yield detectable levels of PAHs in short exposure times (1-3 h) and under extreme occupational conditions. Therefore, we compared the personal exposure monitoring performance of a passive PUF sampler with that of an active air sampler and determined the corresponding uptake rates (Rs). These rates were then used to estimate the occupational exposure of firefighters and police forensic specialists to 32 PAHs. The work environments studied were heavily contaminated by PAHs with (for example) benzo(a)pyrene ranging from 0.2 to 56 ng m , as measured via active sampling. We show that, even after short exposure times, PUF can reliably accumulate both gaseous and particle-bound PAHs. The Rs-values are almost independent of variables such as the concentration and the wind speed. Therefore, by using the Rs-values (2.0-20 m day ), the air concentrations can be estimated within a factor of two for gaseous PAHs and a factor of 10 for particulate PAHs. With very short sampling times (1 h), our method can serve as a (i) simple and user-friendly semi-quantitative screening tool for estimating and tracking point sources of PAH in micro-environments and (ii) complement to the traditional active pumping methods.

[Ab] Resumo:The paper presents information about the content of benzo(a)pyrene in soil samples of petroleum products and the content of heavy metals (iron, copper, lead, arsenic, manganese, chromium, cadmium and mercury) in the soil and hair samples of children. Soil samples and hair were taken in October 2014 in the north-western part of the Yamal-Nenets Autonomous District. The values obtained were compared with the approximately permissible levels and maximum allowable concentrations adopted in the Russian Federation. Levels are determined compounds in the soil in general do not exceed the allowable concentrations. In hair samples there was noted marked exceeding of the recommended values for zinc, copper, chromium and iron 10%, 16.7%, 83.3% and 100% samples, respectively. The cause of the high iron content in the hair samples may be due to the quality of consumed water, which is characterized by low mineralization and high iron content. Increased chromium content of more than 80% of the hair samples can be caused, as in the case of iron, the quality of water consumed, but also a high proportion of local foods in the diet of people, rich by chromium. A conducted study on the state of the environment in the village, away from areas of hydrocarbon production, indicates to the low importance of anthropogenic impact.

[Ab] Resumo:The Love River and Ho-Jin River, two major urban rivers in Kaohsiung City, Taiwan, are moderately to heavily polluted because different types of improperly treated wastewaters are discharged into the rivers. In this study, sediment and river water samples were collected from two rivers to investigate the river water quality and accumulation of polycyclic aromatic hydrocarbons (PAHs) in sediments. The spatial distribution, composition, and source appointment of PAHs of the sediments were examined. The impacts of PAHs on ecological system were assessed using toxic equivalence quotient (TEQ) of potentially carcinogenic PAHs (TEQ ) and sediment quality guidelines. The average PAHs concentrations ranged from 2161 ng/g in Love River sediment to 160 ng/g in Ho-Jin River sediment. This could be due to the fact that Love River Basin had much higher population density and pyrolytic activities. High-ring PAHs (4-6 rings) contributed to 59-90% of the total PAHs concentrations. Benzo(a)pyrene (BaP) had the highest toxic equivalence quotient (up to 188 ng TEQ/g). Moreover, the downstream sediments contained higher TEQ of total TPHs than midstream and upstream sediment samples. The PAHs were adsorbed onto the fine particles with high organic content. Results from diagnostic ratio analyses indicate that the PAHs in two urban river sediments might originate from oil/coal combustion, traffic-related emissions, and waste combustion (pyrogenic activities). Future pollution prevention and management should target the various industries, incinerators, and transportation emission in this region to reduce the PAHs pollution.

[Ab] Resumo:Burning of agricultural biomass generates polycyclic aromatic hydrocarbons (PAHs) including the carcinogen benzo[a]pyrene, of which the catabolism is primarily initiated by a ring-hydroxylating dioxygenase (RHD). This study explores catalytic site accessibility and its role in preferential catabolism of some PAHs over others. The genes flnA1f, flnA2f, flnA3, and flnA4, encoding the oxygenase α and ß subunits, ferredoxin, and ferredoxin reductase, respectively, of the RHD enzyme complex (FlnA) were cloned from Sphingobium sp. FB3 and coexpressed in E. coli BL21. The FlnA effectively transformed fluoranthene but not benzo[a]pyrene. Substitution of the bulky phenylalanine-223 by leucine reduces the steric constraint in the substrate entrance to make the catalytic site of FlnA more accessible to large substrates, as visualized by 3D modeling, and allows the FlnA mutant to efficiently transform benzo[a]pyrene. Accessibility of the catalytic site to PAHs is a mechanism of RHD substrate specificity. The results shed light on why some PAHs are more recalcitrant than others.

[Ab] Resumo:When aromatic hydrocarbons are present in contaminated soils, they often occur in mixtures. The impact of four different (3-ring) nitrogen-containing polycyclic aromatic hydrocarbons (N-PAHs) on C-phenanthrene and C-benzo[a]pyrene (B[a]P) mineralisation in soil was investigated over a 90 d incubation period. The results revealed that both C-phenanthrene and C-benzo[a]pyrene showed no significant mineralisation in soils amended with 10mgkg and 100mgkg N-PAHs (p>0.05). However, increases in lag-phases and decreases in the rates and extents of mineralisation were observed, over time. Among the N-PAHs, benzo[h]quinoline impacted C-phenanthrene mineralisation with extended and diauxic lag phases. Furthermore, C-B[a]P and C-benzo[a]pyrene-nitrogen-containing polycyclic aromatic hydrocarbons ( C-B[a]P-N-PAHs) amended soils showed extensive lag phases (> 21 d); with some C-B[a]P-N-PAH mineralisation recording <1% in both concentrations (10mgkg and 100mgkg ), over time. This study suggests that the presence of N-PAHs in contaminated soil may impact the microbial degradation of polycyclic aromatic hydrocarbons (PAHs) and the impact was most likely the result of limited success in achieving absolute biodegradation of some PAHs in soil.

[Ab] Resumo:The retention of CdS/ZnS QDs on the epidermis has been confirmed to be one of the core procedures during the root uptake process. However, the retention mechanisms of QDs on the epidermis of woody plant were poorly understood for lacking of an appropriate QD quantitative method. In this study, a novel method for in situ determination of CdS/ZnS QDs retained on the root epidermis was established using synchronous nanosecond time-resolved fluorescence spectroscopy. No correlations between K values of oleylamine-CdS/ZnS QDs retained on the epidermal tissues and the surface/bulk composition of mangrove root were observed (p > 0.05) due to the existence of endocytosis mechanisms during the QD uptake processes. Moreover, the difference of the CdS/ZnS QDs in water and further translocated to xylem/phloem of root rather than the combination with cell wall/membranes was the predominant reason that caused the K values to follow the sequence of PEG-COOH-CdS/ZnS QDs < PEG-NH -CdS/ZnS QDs ≪ oleylamine-CdS/ZnS QDs.

[Ab] Resumo:The xeroderma pigmentosum C protein complex (XPC) recognizes a variety of environmentally induced DNA lesions and is the key in initiating their repair by the nucleotide excision repair (NER) pathway. When bound to a lesion, XPC flips two nucleotide pairs that include the lesion out of the DNA duplex, yielding a productively bound complex that can lead to successful lesion excision. Interestingly, the efficiencies of NER vary greatly among different lesions, influencing their toxicity and mutagenicity in cells. Though differences in XPC binding may influence NER efficiency, it is not understood whether XPC utilizes different mechanisms to achieve productive binding with different lesions. Here, we investigated the well-repaired 10R-(+)-cis-anti-benzo[a]pyrene-N -dG (cis-B[a]P-dG) DNA adduct in a duplex containing normal partner C opposite the lesion. This adduct is derived from the environmental pro-carcinogen benzo[a]pyrene and is likely to be encountered by NER in the cell. We have extensively investigated its binding to the yeast XPC orthologue, Rad4, using umbrella sampling with restrained molecular dynamics simulations and free energy calculations. The NMR solution structure of this lesion in duplex DNA has shown that the dC complementary to the adducted dG is flipped out of the DNA duplex in the absence of XPC. However, it is not known whether the "pre-flipped" base would play a role in its recognition by XPC. Our results show that Rad4 first captures the displaced dC, which is followed by a tightly coupled lesion-extruding pathway for productive binding. This binding path differs significantly from the one deduced for the small cis-syn cyclobutane pyrimidine dimer lesion opposite mismatched thymines [ Mu , H. , ( 2015 ) Biochemistry , 54 ( 34 ), 5263 - 7 ]. The possibility of multiple paths that lead to productive binding to XPC is consistent with the versatile lesion recognition by XPC that is required for successful NER.

[Ab] Resumo:Polycyclic aromatic hydrocarbons (PAHs), such as benzo[a]pyrene (B[a]P), are widely distributed environmental contaminants exerting toxic effects such as genotoxicity and carcinogenicity, mainly associated with aryl hydrocarbon receptor (AhR) activation and the subsequent induction of cytochromes P-450 (CYP) 1-metabolizing enzymes. We previously reported an up-regulation of AhR expression and activity in primary cultures of human T lymphocyte by a physiological activation. Despite the suggested link between exposure to PAHs and the risk of lymphoma, the potential of activated human T lymphocytes to metabolize AhR exogenous ligands such as B[a]P and produce DNA damage has not been investigated. In the present study, we characterized the genotoxic response of primary activated T lymphocytes to B[a]P. We demonstrated that, following T lymphocyte activation, B[a]P treatment triggers a marked increase in CYP1 expression and activity generating, upon metabolic activation, DNA adducts and double-strand breaks (DSBs) after a 48-h treatment. At this time point, B[a]P also induces a DNA damage response with ataxia telangiectasia mutated kinase activation, thus producing a p53-dependent response and T lymphocyte survival. B[a]P activates DSB repair by mobilizing homologous recombination machinery but also induces gene mutations in activated human T lymphocytes which could consequently drive a cancer process. In conclusion, primary cultures of activated human T lymphocytes represent a good model for studying genotoxic effects of environmental contaminants such as PAHs, and predicting human health issues.

[Ab] Resumo:This paper analyses sources of sixteen PAHs - polycyclic aromatic hydrocarbons in urbanized areas by using selected diagnostic ratios. Simultaneously, an attempt was made to determine how sewage sludge changes PAHs content in urbanized areas soils. In the experiment three lawns along the main roads in Bialystok with different traffic intensity, three doses of sewage sludge and two years of study were considered. There was no effect of fertilization with sewage sludge on the sum of 16 PAHs in urban soil samples, nevertheless, the sum of 16 PAHs was reduced from 2.6 in 2011 to 2.3 mg/kg in 2012. Among 16 tested PAHs compounds, benzo[a]pyrene was the most dominant compound in samples collected in both years - about 15% of all PAHs. The results suggest that application of sludge into the soil did not influence the concentration of 2-3-ring, 4-ring and 5-6-ring PAHs. For the objects fertilized with a dose 150.0 Mg/ha, of sludge the total sum of potentially carcinogenic PAHs in the urban soil lowered by approximately 68% in comparison with the control plots. PAHs contamination of the urban soil samples resulted from the influence of coal, petroleum and biomass combustion. Moreover, PAHs can enter soil via at mospheric deposition.
