[Ab] Resumo:The mechanism of the reaction of dichlorvos through hydrolysis reactions and through the reaction with polysulfide (S ) and thiophenolate (PhS ) was investigated by proton nuclear magnetic resonance ( H NMR). The study confirmed product identities of an organophosphorus insecticide reacting with reduced sulfur species using H NMR in oxygen sensitive solutions. The experiments of dichlorvos with polysulfide led to the detection of a previously undetected product. The thiophenolate experiments were further advanced to investigate second-order rate kinetics using an internal standard. The experiments provide new evidence for a nucleophilic attack by the reduced sulfur species at the methoxy carbon of dichlorvos. In addition, the observation of in situ reaction dynamics illustrates the applicability of H NMR spectroscopy toward kinetic investigations in environmental science.

[Ab] Resumo:Degradation models for multilure fruit fly trap dispensers were analyzed to determine their potential for use in large California detection programs. Solid three-component male lure TMR (trimedlure [TML], methyl eugenol [ME], raspberry ketone [RK]) dispensers impregnated with DDVP (2, 2-dichlorovinyl dimethyl phosphate) insecticide placed inside Jackson traps were weathered during summer (8 wk) and winter (12 wk) in five citrus-growing areas. Additionally, TMR wafers without DDVP, but with an insecticidal strip, were compared to TMR dispensers with DDVP. Weathered dispensers were sampled weekly and chemically analyzed. Percent loss of TML, the male lure for Ceratitis capitata (Wiedemann) Mediterranean fruit fly; ME, the male lure for Bactrocera dorsalis (Hendel), oriental fruit fly; RK, the male lure for Bactrocera cucurbitae (Coquillett), melon fly; and DDVP was measured. Based on regression analyses for the male lures, TML degraded the fastest followed by ME. Degradation of the more chemically stable RK was discontinuous, did not fit a regression model, but followed similar seasonal patterns. There were few location differences for all three male lures and DDVP. Dispensers degraded faster during summer than winter. An asymptotic regression model provided a good fit for % loss (ME, TML, and DDVP) for summer data. Degradation of DDVP in TMR dispensers was similar to degradation of DDVP in insecticidal strips. Based on these chemical analyses and prior bioassay results with wild flies, TMR dispensers could potentially be used in place of three individual male lure traps, reducing costs of fruit fly survey programs. Use of an insecticidal tape would not require TMR dispensers without DDVP to be registered with US-EPA.

[Ab] Resumo:Long-lasting insecticide nets (LLINs), which have insecticide incorporated within the fibers, have been widely used for control of malaria and other insect-vectored diseases. Only recently have researchers begun exploring their use for control of agricultural pests. In this study, we evaluated the toxicity of a deltamethrin-incorporated LLIN, ZeroFly (Vestergaard-Frandsen, Washington, DC) for control of the brown marmorated stink bug, Halyomorpha halys (Stål). In the lab, exposure to the ZeroFly net for 10 s resulted in >90% mortality of H. halys nymphs and >40% mortality of H. halys adults. Longer exposure to the net resulted in higher mortality. In another experiment, a 15-cm2 sheet of ZeroFly net placed inside of the stink bug trap provided long-lasting kill of H. halys adults equal to or better than standard dichlorvos kill strip. Potential for the use of ZeroFly nets for H. halys IPM is discussed.

[Ab] Resumo:Current study established a protective action of bacoside A and bromelain against the toxic effects of dichlorvos in kidneys of mice. Experimental design included five groups. The first group was control. Mice of groups II, III and IV were administered doses of dichlorvos, bromelain and bacoside A respectively. In group V, mice were treated with both the antioxidants (bacoside A and bromelain) and dichlorvos. After 21 days of exposure of different doses, levels of acetylcholinesterase (AChE), butyrylcholinesterase (BChE), γ-aminobutyric acid (GABA) and serotonin were measured in renal tissues. Dichlorvos significantly reduced the kidney AChE (p < 0.001), BChE (p < 0.01) and GABA level (p < 0.01) compared to control. A simultaneous significant elevation in the serotonin level (p < 0.01) was recorded after dichlorvos exposure. Concomitant exposure of bacoside A and bromelain followed by dichlorvos treatment in group V not only restored, but increased the renal cholinesterases and GABA level. Meanwhile, a significant decline in serotonin level (p < 0.001) was revealed, compared to dichlorvos exposed mice. Bacoside A and bromelain occupy a tremendous antioxidant action in the mice kidneys and a combination of the same ameliorates the renal toxicity induced by dichlorvos.

[Ab] Resumo:The aim was to investigate whether a human neural stem cell (NSC) line derived from human umbilical cord blood (hUCB) can be used for toxicity study. Toxicity of both neurotoxic environmental xenobiotics, methyl mercury chloride (CH HgCl), lead acetate (CH COOPb), and chlorpyrifos (CP), and non-neurotoxic insecticide, dichlorvos, as well as non-neurotoxic drugs, theophylline and acetaminophen were assessed. Additionally, differentiation of neuronal and glial cell lines derived from hUCB was elucidated. It was observed that CH HgCl was more toxic to human NSCs in comparison to CH COOPb and CP. The minimum inhibitory concentration (MIC) value against NSCs was 3, 10, and 300 mg/L, in each staining process, acridine orange/ethidium bromide (AO/EB) staining, 3-(4,5-dimethylthiazol-2-yl)2,5-diphenyl tetrazolium bromide (MTT) assay, and Hoechst staining, for CH HgCl, CP, and CH COOPb, respectively. CH HgCl had the LC value as 10.0, 14.4, and 12.7 mg/L, by staining method mentioned in succession. CP had the LC value as 21.9, 23.7, and 18.4 mg/L; similarly, CH COOPb had LC values, successively as 616.9, 719.2, and 890.3 mg/L. LC values ranged from 18.2 to 21.7 mg/L for CH HgCl, 56.4 to 60.2 mg/L for CP, and 1000 to 1460.1 for CH COOPb. Theophylline, acetaminophen, and dichlorvos had no impact on the viability of NSCs. This work justified that hUCB-NSC model can be used for toxicity study.

[Ab] Resumo:Dichlorvos is a chemical compound which has been used for decades as a pesticide. Potential inhalational exposure to dichlorvos vapor associated with using commercially-based, dichlorvos-impregnated resin strips in a simulated military guard post was evaluated. A varying number of these pesticide strips, ranging from the manufacturer's guidelines ((3)-16 g strips) up to a full package of strips ((12)-16 g strips), were placed in a small, enclosed space (2.31 m x 2.26 m x 2.44 m, 12.7 m ), which was similar in size to a typical military guard post. Static air sampling was then conducted to simulate personal air sampling, followed by analysis using OSHA Method 62 (GC-ECD). Air sampling was conducted over a range of discrete temperatures (26-38˚C) which approximated average ambient temperatures expected in a variety of deployed environments. Air sampling in this range was conducted to determine the airborne concentration generated at each temperature setting. Airborne concentrations were then compared to established short term military exposure guidelines (MEGs) and the 8-hr OSHA permissible exposure limit (PEL) for dichlorvos (both criteria limits are 0.99 mg/m ). Results from air sampling indicated that exceeding the manufacturer-recommended number of strips for the workspace volume and environmental conditions produced airborne dichlorvos concentrations above established occupational standards (1.77-3.70 mg/m ). Such exposures may potentially lead to adverse effects, such as loss of mental and visual acuity for guard post watch standers who employ more strips within a space than recommended per the manufacturer for the size of the space. However, concentrations of airborne dichlorvos generated when adhering to manufacturer's guidelines based on workspace volume resulted in levels of 0.16-0.39 mg/m for 1-hr and 8-hr timeframes, which were below the established occupational health limits. While dichlorvos-impregnated strips are not currently recommended for use in manned workspaces for periods > 4 hr, findings suggest that prolonged use (8 hr) of similar pesticide strips within manned spaces to repel and/or kill disease-carrying insects may be possible without experiencing adverse health effects.

[Ab] Resumo:The characterization of cholinesterase activity in brain and muscle of gilthead seabream was carried out using four specific substrates and three selective inhibitors. In addition, K and V were calculated from the Michaelis-Menten equation for ASCh and BSCh substrates. Finally, the in vitro sensitivity of brain and muscle cholinesterases to three organophosphates (OPs) was also investigated by estimating inhibition kinetics. The results indicate that AChE is the enzyme present in the brain, whereas in muscle, a typical AChE form is present along with an atypical form of BChE. Very low ChE activity was found in plasma with all substrates used. The inhibitory potency of the studied OPs on brain and muscle AChEs based on bimolecular inhibition constants (k ) was: omethoate < dichlorvos < azinphosmethyl-oxon. Furthermore, muscle BChE was found to be several orders of magnitude (from 2 to 4) more sensitive than brain and muscle AChE inhibition by dichlorvos and omethoate.

[Ab] Resumo:BACKGROUND: This study raises awareness to the use of toxic pesticides and reiterates the well-known danger of these pesticides. The acute toxicity of two pesticides, dichlorvos and lindane against the African air-breathing catfish, Heterobranchus longifilis fingerlings was investigated under static bioassay in the laboratory. Relevant patents relating to acute toxicity of pesticides against fish were reviewed. Invention concerning the detection of biomarkers in material from a living organism relates to a method of testing whether a living organism has been exposed to stress, such as pesticide exposure. METHODS: Range finding bioassays were conducted to get the range of concentrations for the definitive bioassays. The range of concentrations of test media for dichlorvos was 0.2 - 1.5 mg l-1 while that of lindane was 35 - 80 mg l-1. The median lethal concentrations were determined using probit analysis. RESULTS: The test pesticides were found to be differentially toxic to the test species. The 96h LC50 values of 0.8187 and 41.566 mg l-1 for dichlorvos and lindane respectively indicated that dichlorvos was highly toxic while lindane was slightly toxic to the test species. Computed toxicity factor showed that dichlorvos was 50.8 times more toxic than lindane. The unpaired t-test showed that dichlorvos was significantly (0.05) more toxic than lindane. The physical and chemical parameters data showed that over the 96h periods, the test pesticides caused slight increase in temperature, dissolved oxygen and ammonia of the test media, when compared with the untreated control. But they differentially altered pH, conductivity and alkalinity. CONCLUSION: Since H. longifilis is sensitive to the test pesticides, it can therefore be considered as a good test species for the determination of acute toxicity and poisoning resulting from the pesticides and possibly for other compounds with similar intrinsic characteristics.

[Ab] Resumo:Experiments on white non-inbred rats demonstrated that treatment with organophosphorus compound dimethyl dichlorovinyl phosphate (DDVP) decreased T cell-independent antibody production by B cells and blood levels of IL-10 and IL-12; a similar effect was produced by GTS-21, a selective agonist of α7-nicotinic acetylcholine receptor. N-nicotinic receptor antagonist chlorisondamine in combination with DDVP partially prevented suppression of antibody production in comparison with the effect observed during intoxication with DDVP.

[Ab] Resumo:Using the bivalve macrofouler Corbicula fluminea, the suitability of in vitro testing as a stepping stone towards the improvement of control methods based on chemical mixtures was addressed in this study. In vitro cholinesterase (ChE) activity inhibition following single exposure of C. fluminea tissue to four model chemicals (the organophosphates dimethoate and dichlorvos, copper and sodium dodecyl phosphate [SDS]) was first assessed. Consequently, mixtures of dimethoate with copper and dichlorvos with SDS were tested and modelled; mixtures with ChE revealed synergistic interactions for both chemical pairs. These synergic combinations were subsequently validated in vivo and the increased control potential of these selected combinations was verified, with gains of up to 50% in C. fluminea mortality relative to corresponding single chemical treatments. Such consistency supports the suitability of using time- and cost-effective surrogate testing platforms to assist the development of biofouling control strategies incorporating mixtures.
