[Ab] Resumo:An ultra-sensitive screen-printed biosensor was successfully developed for phosmet detection in olive oil, based on a genetically-engineered acetylcholinesterase (AChE) immobilized in a azide-unit water-pendant polyvinyl alcohol (PVA-AWP)/Fe-Ni alloy nanocomposite. Fe-Ni not only allowed amplifying the response current but also lowering the applied potential from 80 mV to 30 mV vs Ag/AgCl. The biosensor showed a very good analytical performance for phosmet detection, with a detection limit of 0.1 nM. This detection limit is lower than the allowable concentrations set by international regulations. In addition to the good reproducibility, operational and storage stability, the developed biosensor was successfully used for the determination of phosmet in olive oil samples without any laborious pre-treatment. The phosmet recovery rate was about 96% after a simple liquid-liquid extraction.

[Ab] Resumo:Organophosphorous pesticides (OPPs) are ubiquitous pollutants in the atmospheric environment with adverse effects on human health. In this study, heterogeneous kinetics of particulate phosmet and parathion with NO3 radicals were investigated with a mixed-phase relative rate method. A vacuum ultraviolet photoionization aerosol time-of-flight mass spectrometer (VUV-ATOFMS) and an atmospheric gas analysis mass spectrometer were used to monitor online the decays of particulate OPPs and reference compound, respectively. Reactive uptake coefficients of NO3 radicals on phosmet and parathion particles were (0.12±0.03) and (0.14±0.04), respectively, calculated according to the measured OPPs loss ratios and the average NO3 concentrations. Additionally, the average effective rate constants for heterogeneous reactions of particulate phosmet and parathion with NO3 radicals measured under experimental conditions were (2.80±0.16)×10(-12) and (2.97±0.13)×10(-12) cm(3) molecule(-1) s(-1), respectively. The experimental results of these heterogeneous reactions in the aerosol state provide supplementary knowledge for kinetic behaviors of airborne OPPs particles.

[Ab] Resumo:We use both lyotropic liquid crystals composed of prolate micelles and thermotropic liquid crystals made of rod-like molecules to uniformly disperse and unidirectionally align relatively large gold nanorods and other complex-shaped nanoparticles at high concentrations. We show that some of these ensuing self-assembled orientationally ordered soft matter systems exhibit polarization-dependent plasmonic properties with strongly pronounced molar extinction exceeding that previously achieved in self-assembled composites. The long-range unidirectional alignment of gold nanorods is mediated mainly by anisotropic surface anchoring interactions at the surfaces of gold nanoparticles. Polarization-sensitive absorption, scattering, and extinction are used to characterize orientations of nanorods and other nanoparticles. The experimentally measured unique optical properties of these composites, which stem from the collective plasmonic effect of the gold nanorods with long-range order in a liquid crystal matrix, are reproduced in computer simulations. A simple phenomenological model based on anisotropic surface interaction explains the alignment of gold nanorods dispersed in liquid crystals and the physical underpinnings behind our observations.

[Ab] Resumo:The topical toxicities of five commercial grade pesticides commonly sprayed in apple orchards were estimated on adult worker honey bees, Apis mellifera (L.) (Hymenoptera: Apidae) and Japanese orchard bees, Osmia cornifrons (Radoszkowski) (Hymenoptera: Megachilidae). The pesticides were acetamiprid (Assail 30SG), λ-cyhalothrin (Warrior II), dimethoate (Dimethoate 4EC), phosmet (Imidan 70W), and imidacloprid (Provado 1.6F). At least 5 doses of each chemical, diluted in distilled water, were applied to freshly-eclosed adult bees. Mortality was assessed after 48 hr. Dose-mortality regressions were analyzed by probit analysis to test the hypotheses of parallelism and equality by likelihood ratio tests. For A. mellifera, the decreasing order of toxicity at LD50 was imidacloprid, λ-cyhalothrin, dimethoate, phosmet, and acetamiprid. For O. cornifrons, the decreasing order of toxicity at LD50 was dimethoate, λ-cyhalothrin, imidacloprid, acetamiprid, and phosmet. Interaction of imidacloprid or acetamiprid with the fungicide fenbuconazole (Indar 2F) was also tested in a 1∶1 proportion for each species. Estimates of response parameters for each mixture component applied to each species were compared with dose-response data for each mixture in statistical tests of the hypothesis of independent joint action. For each mixture, the interaction of fenbuconazole (a material non-toxic to both species) was significant and positive along the entire line for the pesticide. Our results clearly show that responses of A. mellifera cannot be extrapolated to responses of O.cornifrons, and that synergism of neonicotinoid insecticides and fungicides occurs using formulated product in mixtures as they are commonly applied in apple orchards.

[Ab] Resumo:BACKGROUND: Pesticides used in agriculture can be taken into worker homes and pose a potential risk for children and other family members. This study focused on identification of potential intervention points at the workplace. METHODS: Workers (N = 46) recruited from two tree fruit orchards in Washington State were administered a 63-item pesticide safety questionnaire. Dust was collected from commuter vehicles and worker homes and analyzed for four organophosphorus (OP) pesticides (azinphosmethyl, phosmet, chlorpyrifos, malathion). RESULTS: Geometric mean azinphosmethyl concentrations in dust for three worker groups (16 pesticide handlers, 15 green fruit thinners, 15 organic orchard workers) ranged from 0.027-1.5 µg/g, with levels in vehicle dust higher than in house dust, and levels in house dust from handlers' homes higher than levels from tree fruit thinners' homes. Vehicle and house dust concentrations of azinphosmethyl were highly associated (R(2) = 0.44, P < 0.001). Significant differences were found across worker groups for availability of laundry facilities, work boot storage, frequency of hand washing, commuter vehicle use, parking location, and safety training. CONCLUSIONS: These findings support a focus on intervention activities to reduce take home pesticide exposure closer to the source of contamination; specifically, the workplace and vehicles used to travel to the workplace.

[Ab] Resumo:A surface plasmon resonance (SPR) immunoassay using a PH-BSA immobilized sensor chip was developed to measure phosmet in food samples. The in-competitive inhibition assay showed highly sensitive and good specificity of the cross-reactivity with analogue structure pesticides. The biosensor exhibited a linear detection range from 8.0 to 60.0 ng/L of phosmet with a lower detection limit of 1.6 ng/L (S/N = 3). The sensitivity obtained with the present SPR affinity biosensor was significantly higher than most of the sensors reported with different measurement methodologies for phosmet. A recovery test of pesticide quantification in peaches, apples, cabbages, and rapes was also studied. Good recoveries (86.4-102.8%) and coefficients of variation (CVs) (5.1-12.6%) were obtained in all cases. The SPR biosensor assay method was compared with cd-ELISA in terms of analysis time, antibody dosage, recoveries, precision, detection limit, pretreatment, and testing costs, and clear advantages could be seen over the traditional ELISA-based detection systems. The developed SPR method was suitable for the rapid quantitative or qualitative determination of phosmet in agricultural products.

[Ab] Resumo:The microscopic structure of fluids of simple spheres is well known. However, the constituents of most real-life fluids are non-spherical, leading to a coupling between the rotational and translational degrees of freedom. The structure of simple dense fluids of spheroids - ellipsoids of revolution - was only recently determined by direct experimental techniques [A. P. Cohen, E. Janai, E. Mogilko, A. B. Schofield, and E. Sloutskin, Phys. Rev. Lett. 107, 238301 (2011)]. Using confocal microscopy, it was demonstrated that the structure of these simple fluids cannot be described by hard particle models based on the widely used Percus-Yevick approximation. In this paper, we describe a new protocol for determining the shape of the experimental spheroids, which allows us to expand our previous microscopy measurements of these fluids. To avoid the approximations in the theoretical approach, we have also used molecular dynamics simulations to reproduce the experimental radial distribution functions g(r) and estimate the contribution of charge effects to the interactions. Accounting for these charge effects within the Percus-Yevick framework leads to similar agreement with the experiment.

[Ab] Resumo:Large-scale field efficacy trials of methoxyfenozide (Intrepid), a reduced-risk molting agonist insecticide, were conducted in 2004 and 2005 in an orchard containing 'Nonpareil' and 'Sonora almonds [Prunus dulcis (Mill.) D.A. Webb] located in Kern County, CA. Methoxyfenozide applied one to three times, the organophosphate phosmet (Imidan) alone or in combination with methoxyfenozide, or the pyrethroid permethrin (Perm-Up) were tested for efficacy against the primary lepidopteran pest navel orangeworm, Amyelois transitella (Walker) (Lepidoptera: Pyralidae), and three other lepidopteran pests of almond: oriental fruit moth, Grapholita molesta (Busck); oblique-banded leafroller, Choristoneura rosaceana (Harris); and peach twig borer, Anarsia lineatella Zeller. Two or three applications of methoxyfenozide (bracketing hull split or spring plus bracketing hull split) were more effective than a single hull split application of phosmet, phosmet combined with permethrin, or methoxyfenozide. In these trials, a spring application followed by a posthull split application was as effective as the applications bracketing hull split. Navel orangeworm accounted for > 60% of the total damage, whereas oriental fruit moth and peach twig borer were the dominant secondary pests. In experiments conducted in 2010 to assess the direct toxicity of methoxyfenozide to navel orangeworm eggs under field conditions, exposure to methoxyfenozide reduced survival by 96-99%. We conclude that this reduced-risk insecticide is effective, although its efficacy is maximized with more than one well-timed application.

[Ab] Resumo:Epidemiological data have associated environmental organophosphate insecticide (OP) exposure during pregnancy with fetal growth deficits. To better understand OP injury that may adversely affect pregnancy, we used the JEG-3 choriocarcinoma cell line, which provide a recognized in vitro model to study placental function. The effects of the OP phosmet (Pm) and chlorpyrifos (Cp) on JEG-3 cells viability, proliferation, cell cycle and inflammatory molecule production were evaluated. Both insecticides affected cellular viability in a concentration- and time-dependent manner, inducing apoptosis and decreasing [(3)H]-thymidine incorporation. However, only Pm reduced DNA synthesis independently of cellular death and decreased the cell percentage at the S-phase. Unlike apoptosis, TNFα production varied with the concentration tested, suggesting that other TNFα independent mechanisms might trigger cell death. No induction of the inflammatory molecule nitric oxide was detected. The mRNA levels of pro-inflammatory IL-6, IL-17 and the anti-inflammatory IL-13 cytokines were differentially modulated. These findings show that Pm and Cp generate a specific toxicity signature, altering cell viability and inducing an inflammatory cytokine profile, suggesting that trophoblasts may represent a possible target for OP adverse effects.

[Ab] Resumo:As an organophosphorous insecticide, phosmet is widely used for plant protection as well as against the ectoparasites on productive livestock. It emits into the atmosphere in both gas and particulate phases via spray drift from treatments and postapplication emissions, but its degradation in the atmosphere is not well-known up to now. In this study, the heterogeneous reaction of phosmet absorbed on the azelaic acid particles with NO(3) radicals is investigated in real-time using a vacuum ultraviolet photoionization aerosol time-of-flight mass spectrometer (VUV-ATOFMS). Two primary products and their nitro-substituted derivates are observed. The apparent reaction rate of phosmet particles with NO(3) radicals is also obtained by a relative rate method with isoprene and 1,4-cyclohexadiene as the reference compounds. The concentrations of the reference substance and phosmet particles are synchronously detected with an atmospheric gas analysis mass spectrometer and the VUV-ATOFMS. Under the experimental conditions, the average apparent reaction rate obtained is (1.92 ± 0.09) × 10(-12) cm(3) molecule(-1) s(-1), and the corresponding atmospheric lifetime toward NO(3) radicals is roughly estimated to be ~0.3 h.
