[Ab] Resumo:The aim of this study was to compare two irrigation techniques and four devices for endodontic sealer placement into the dentinal tubules. Ninety-nine single-rooted human teeth were instrumented and allocated to either the control (CO) (n=11) or experimental groups according to the irrigation method: syringe and NaveTip needle (NT) (n=44), and EndoActivator (EA) (n=44). These groups were subdivided according to sealer placement into K-File (KF), lentulo spiral (LS), Easy Clean (EC), and EndoActivator (EA) subgroups. Moreover, the distances of 5 mm and 2 mm from the apex were analyzed. The teeth were obturated with AH Plus and GuttaCore X3. Analyses were performed by scanning electron microscopy associated to cathodoluminescence. The percentage and maximum depth of sealer penetration were measured. Data were evaluated by three-way analysis of variance (ANOVA) and Games-Howell test (p<0.05). EA was superior to NT in percentage of sealer penetration. EC was significantly superior to EA (subgroup) for sealer penetration, and both improved the percentage of sealer penetration when compared to LS. Better sealer penetration was observed at the distance of 5 mm from the apex. Sealer penetration into the dentinal tubules was significantly improved by sonic irrigant activation.

[Ab] Resumo:A wide variety of crystallization solutions are screened to establish conditions that promote the growth of a diffraction-quality crystal. Screening these conditions requires the assessment of many crystallization plates for the presence of crystals. Automated systems for screening and imaging are very expensive. A simple approach to imaging trace fluorescently labeled protein crystals in crystallization plates has been devised, and can be implemented at a cost as low as $50. The proteins ß-lactoglobulin B, trypsin and purified concanavalin A (ConA) were trace fluorescently labeled using three different fluorescent probes: Cascade Yellow (CY), Carboxyrhodamine 6G (CR) and Pacific Blue (PB). A crystallization screening plate was set up using ß-lactoglobulin B labeled with CR, trypsin labeled with CY, ConA labeled with each probe, and a mixture consisting of 50% PB-labeled ConA and 50% CR-labeled ConA. The wells of these plates were imaged using a commercially available macro-imaging lens attachment for smart devices that have a camera. Several types of macro lens attachments were tested with smartphones and tablets. Images with the highest quality were obtained with an iPhone 6S and an AUKEY Ora 10× macro lens. Depending upon the fluorescent probe employed and its Stokes shift, a light-emitting diode or a laser diode was used for excitation. An emission filter was used for the imaging of protein crystals labeled with CR and crystals with two-color fluorescence. This approach can also be used with microscopy systems commonly used to observe crystallization plates.

[Ab] Resumo:In this work, a novel core-shell magnetic molecularly imprinted polymers (MMIPs) for the measurement of L-Hydroxyproline (Hyp) in dairy products was prepared. The derivative of Hyp using N-hydroxysuccinimidyl rhodamine B ester (RBS) as derivatization reagent was employed as template to prepare RBS-Hyp-MMIPs (Fe O @MIPs for RBS-Hyp). A new analytical procedure of in situ derivatization with MMIPs (ISD-MMIPs) has been developed for the specific extraction and determination of Hyp in dairy products by ultra high performance liquid chromatography tandem mass spectrometry. The RBS-Hyp-MMIPs was characterized by fourier transform infrared spectrometer and transmission electron microscopy, and evaluated by adsorption experiments. The adsorption process followed Langumuir adsorption isotherm with maximum adsorption capacity of RBS-Hyp on RBS-Hyp-MMIPs at 96 mg/g. In addition, RBS-Hyp-MMIPs showed a short equilibrium time (15.0 min), rapid magnetic separation (5 s) and high stability (retained 95.3% after six cycles). Under the optimized conditions, good linearity was observed with the limits of detection (S/N > 3) and limits of quantification (S/N > 10) at 0.1 and 0.5 ng/mL, respectively. On account of the specific extraction performance of RBS-Hyp-MMIPs, not any interference peak from real sample matrix was observed in the chromatograms of milk powder, liquid milk and milk drink. The proposed procedure was successfully applied for selective determination of Hyp from dairy products with satisfactory validation results, which is of great significance to food safety.

[Ab] Resumo:Ag/AgBr/GdVO composite photocatalysts were designed and synthesized in this paper. The physical and chemical structures, as well as optical properties of the synthesized composite were investigated via XRD, XPS, TEM, and UV-vis. It is found that the composite showed a ternary heterojunction structure of Ag, AgBr and GdVO . Meanwhile, it has a high intensity of light current, indicating its high separation efficiency of electron and hole. Photocatalytic oxidation of rhodamine B (RhB) under visible light irradiation was performed to investigate the activity of the Ag/AgBr/GdVO composite. Result indicates that it shows excellent photocatalytic activity. Under visible light irradiation for 12min, about 80% of RhB (30µmol/L) was degraded. The degradation rate is estimated to be 0.253 min-1, which is three times higher than that of pure AgBr. The high photoactivity can be ascribed to the synergetic effect of AgBr, GdVO , and Ag nanoparticle in separation of electron-hole pairs.

[Ab] Resumo:By using a disulfide-functionalized crosslinker, a pH- and thermo-responsive 2-(dimethylamino) ethyl methacrylate (DMAEMA) monomer and a zwitterionic sulfobetaine methacrylate (SBMA) monomer were conjugated to fabricate a multi-responsive P(DMAEMA-SS-SBMA) copolymeric hydrogel. Apparent UCST and LCST volume transitions were observed in the P(DMAEMA-SS-SBMA) hydrogels with equivalent weight fractions of monomers. Different pore size and response sensitivity of shrunken structures below UCST and above LCST were visualized by SEM images. The hydrogel exhibited a highly swollen state with a swelling ratio of 17.8 and a pore size of 106µm at 45°C, they deswelled unequally at 5°C with a compact surface with pore size of 30µm and a loose bulk with pore size of 83µm, while they deswelled uniformly at 65°C with dense shrunken structure with small pore size of 12µm. The dual-thermoresponsive hydrogel was promising in controlled drug release. The initial drug release was predominantly controlled by diffusion, and the long-term release was influenced by the swelling ratio. Below UCST, the relatively hydrophilic shrunken structure and slow diffusion had a synergistic effect on the sustained release. Above LCST, the fast diffusion and the rapid "off" effect of hydrophobic skin layer resulted in a burst release. Additionally, pH-tunable swelling and redox-sensitive degradation were also observed.

[Ab] Resumo:Histone deacetylase (HDAC) is a validated drug target for various diseases. This study combined indole recognition cap with SAHA, an FDA-approved HDAC inhibitor used to treat cutaneous T-cell lymphoma (CTCL). The structure activity relationship of the resulting compounds that inhibited HDAC was disclosed as well. Some compounds exhibited much stronger inhibitory activities than SAHA. We identified two meta-series compounds 6j and 6k with a two-carbon linker had IC values of 3.9 and 4.5 nM for HDAC1, respectively. In contrast, the same oriented compounds with longer carbon chain linkers showed weaker inhibition. The result suggests that the linker chain length greatly contributed to enzyme inhibitory potency. In addition, comparison of enzyme-inhibiting activity between the compounds and SAHA showed that compounds 6j and 6k displayed higher inhibiting activity for class I (HDAC1, -2, -3 and -8). The molecular docking and structure analysis revealed structural differences with the inhibitor cap and metal-binding regions between the HDAC isozymes that affect interactions with the inhibitors and play a key role for selectivity. Further biological evaluation showed multiple cellular effects associated with compounds 6j- and 6k-induced HDAC inhibitory activity.

[Ab] Resumo:Pd-MnO /TiO nanotube arrays (NTAs) photo-electrodes were successfully fabricated via anodization and electro deposition subsequently; the obtained Pd-MnO /TiO NTAs photo electrodes were analyzed by scanning electron microscopy (SEM), X-ray diffraction (XRD) and characterized accordingly. Moreover, the light harvesting and absorption properties were investigated via ultraviolet-visible diffuse reflectance spectrum (DRS); photo degradation efficiency was investigated via analyzing the photo catalytic degradation of Rhodamine B under visible illumination (xenon light). The performed analyses illustrated that Pd-MnO codoped particles were successfully deposited onto the surface of the TiO nanotube arrays; DRS results showed significant improvement in visible light absorption which was between 400 and 700nm. Finally, the photo catalytic degradation efficiency results of the designated organic pollutant (Rhodamine B) illustrated a superior photocatalytic (PC) efficiency of approximately 95% compared to the bare TiO NTAs, which only exhibited a photo catalytic degradation efficiency of approximately 61%, thus it indicated the significant enhancement of the light absorption properties of fabricated photo electrodes and their yield of OH radicals.

[Ab] Resumo:BACKGROUND: Recent interest in male-based sterile insect technique (SIT) and incompatible insect technique (IIT) to control Aedes aegypti and Aedes albopictus populations has revealed the need for an economical, rapid diagnostic tool for determining dispersion and mating success of sterilized males in the wild. Previous reports from other insects indicated rhodamine B, a thiol-reactive fluorescent dye, administered via sugar-feeding can be used to stain the body tissue and seminal fluid of insects. Here, we report on the adaptation of this technique for male Ae. aegypti to allow for rapid assessment of competitiveness (mating success) during field releases. METHODOLOGY/PRINCIPLE FINDINGS: Marking was achieved by feeding males on 0.1, 0.2, 0.4 or 0.8% rhodamine B (w/v) in 50% honey solutions during free flight. All concentrations produced >95% transfer to females and successful body marking after 4 days of feeding, with 0.4 and 0.8% solutions producing the longest-lasting body marking. Importantly, rhodamine B marking had no effect on male mating competitiveness and proof-of-principle field releases demonstrated successful transfer of marked seminal fluid to females under field conditions and recapture of marked males. CONCLUSIONS/SIGNIFICANCE: These results reveal rhodamine B to be a potentially useful evaluation method for male-based SIT/IIT control strategies as well as a viable body marking technique for male-based mark-release-recapture experiments without the negative side-effects of traditional marking methods. As a standalone method for use in mating competitiveness assays, rhodamine B marking is less expensive than PCR (e.g. paternity analysis) and stable isotope semen labelling methods and less time-consuming than female fertility assays used to assess competitiveness of sterilised males.

[Ab] Resumo:Rhodamine dyes have been banned as food additives due to their potential tumorigenicity. Rhodamine 110 is illegal as a food additive, although its pharmacokinetics have not been characterized, and no accurate bioanalytical methods are available to quantify rhodamine 110. The aim of this study was to develop and validate a fast, stable, and sensitive method to quantify rhodamine 110 using high-performance liquid chromatography coupled to tandem mass spectrometry (HPLC-MS/MS) to assess its pharmacokinetics and organ distribution in awake rats. Rhodamine 110 exhibited linear pharmacokinetics and slow elimination after oral administration. Furthermore, its oral bioavailability was approximately 34-35%. The distribution in the liver and kidney suggests that these organs are primarily responsible for rhodamine 110 metabolism and elimination. Our investigation describes the pharmacokinetics and a quantification method for rhodamine 110, improving our understanding of the food safety of rhodamine dyes.

[Ab] Resumo:Occurrence of Rhodamine B (RhB) contamination in paprika caused by agricultural materials during the vegetation process has been reported. It may transfer during the process of active compounds extraction, and eventually exist in final products. Herein, the re-distribution of RhB during the extraction process was assessed in terms of RhB contents, as well as mass, color value and capsaicinoids yield of each process. Results revealed that natural RhB contamination at 0.55-1.11µg/kg originated from raw paprika fruit then transferred with the extraction proceeded. About 95.5% of RhB was found in red oleoresin. After separation of red oleoresin, 91.6% of RhB was remained in capsicum oleoresin, only 3.7% in paprika red. These results were consistent with total capsaicinoids recovery of each product. The RhB levels in edible capsicum oleoresin in our present study at 0.01-0.34µg/kg did not exceed the legal limits established by the European Union.
