[Ab] Resumo:BACKGROUND: This study aimed to evaluate the effect of intralipid postconditioning (ILPC) on myocardial damage in patients undergoing valve replacement surgery with concomitant radiofrequency ablation (RFA) for atrial fibrillation (AF). METHODS: Randomized patient and assessor-blind controlled trial conducted in adult patients undergoing valve replacement surgery with concomitant RFA. Sixty-nine patients were randomly assigned to ILPC group (n = 34) or control group (n = 35): ILPC group received an intravenous infusion of 20% intralipid (2 mL/kg) just 10 minutes before aortic cross-unclamping, and control group received an equivalent volume of normal saline. Serum cardiac troponin-T (cTnT) and creatine kinase-MB (CK-MB) was measured before surgery and at 4, 12, 24, 48, and 72 hours after surgery. The primary endpoints were the 72-hour area under the curve (AUC) for cTnT and CK-MB. RESULTS: The total 72-hour AUC of cTnT (P = .33) and CK-MB (P = .52) were comparable between 2 groups. The left ventricle ejection fraction at discharge (P = .011) was higher in the ILPC group than that in the control group, while the AF recurrence did not differ significantly between 2 groups. CONCLUSIONS: There was no observed beneficial effect of ILPC on myocardial injury documented by the cardiac biomarkers in patients undergoing valve replacement surgery with concomitant RFA, and the effect of intralipid against myocardial I/R injury is undetectable within the background of massive biomarker release following ablation owing to localized myocardial necrosis. Besides, there are no other published data about the cardioprotective role of intralipid in patients undergoing this procedure and benefits of this protection need further studies to validate.

[Ab] Resumo:The lipid characterization of storage fat, subcutaneous fat and bone marrow, originated from three different bovine categories, calf, young bull and cow, was carried out in order to develop a re-use of these by-products. After the lipid extraction, the compositions in fatty acids, triacylglycerols and cholesterol were determined by GC-FID. A total of 25 fatty acids were identified in all by-products; the oleic acid was the preponderant component, followed by palmitic, stearic, palmitoleic, linoleic and myristic acid. The long chain triacylglycerols (T48, T50 and T52) were the main products, especially in the cow's by-products. The subcutaneous fat of all animals showed the highest cholesterol content, whereas it was present in low amount in the others. Phospholipids were analysed in bone marrow of all the animals and phosphatidylcholine was the most abundant compound. Because of the high fat content and essential fatty acids and the low cholesterol amount, storage fat and bone marrow could represent a valuable lipid resource in food and pharmaceutical industry.

[Ab] Resumo:Phospholipids (PLs) represent a class of metabolites of interest for evaluating the relationship between diet and the development of several metabolic diseases. Given that PLs are rich in unsaturated fatty acids, they can be oxidized. Because of their structure and reactivity, oxidized phospholipids (PLs-Ox) are increasingly recognized as markers of oxidative stress and of various diseases associated with inflammation. Therefore, there is a growing interest in studying PLs-Ox in lipidomics. Because of their limited commercial availability, very little information is currently available in databases to identify these molecules. The aim of this study is to acquire new knowledge about PLs-Ox in order to propose an analytical strategy for their analyses. For this purpose, a synthesis method of PLs-Ox, in auto-oxidation, has been developed and applied on phosphatidylcholine and phosphatidylethanolamine molecular species with various chain lengths, degree, and position of unsaturations. An analysis method based on mass (MS) and tandem mass spectrometry coupled to electrospray ionization was then developed and enabled the identification of a great diversity of long- and short-chain oxidation products. Formation kinetics of oxidation products was evaluated. Results showed that the formation of oxidized compounds was largely influenced by the degree of unsaturation on fatty acid chains. Oxidation time promotes the formation of some biologically important oxidation products. Coupling the MS method with liquid chromatography in flow injection analysis mode enabled the development of a full analytical strategy. Structural analysis of PLs-Ox allowed the enrichment of databases with important information to identify these molecules in biological matrices.

[Ab] Resumo:Strain SYSU D8010 was isolated from a desert sand sample collected in Saudi Arabia. The taxonomic position of the isolate was investigated by the polyphasic taxonomic approach. The isolate was found to be Gram-positive and aerobic. The strain was able to grow at 14-40 °C, pH 5.0-9.0 and in the presence of up to 22 % (w/v) NaCl. Strain SYSU D8010 contained meso-diaminopimelic acid as cell-wall diamino acid, and arabinose, fucose, galactose, glucose and rhamnose as the whole-cell sugars. The primary polar lipids were diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol, phosphatidylinositol and phosphatidylinositolmannosides. Menaquinone MK-9(H4) was detected as the respiratory quinone; and anteiso-C17 : 0, iso-C16 : 0, iso-C15 : 0 and iso-C17 : 0 as the predominant fatty acids. Pairwise comparison of the 16S rRNA gene sequences indicated that strain SYSU D8010 had a sequence similarity of 97.8 % to Saccharopolyspora halophila YIM 90500 . The genomic DNA G+C content of strain SYSU D8010 was determined to be 69.9 mol%. Based on the analyses of the phenotypic, genotypic and phylogenetic characteristics, strain SYSU D8010 was determined to represent a novel species of the genus Saccharopolyspora, for which the name Saccharopolyspora deserti sp. nov. is proposed. The type strain of the species is SYSU D8010 (=KCTC 39989 =CPCC 204620 ).

[Ab] Resumo:A Gram-negative, motile by gliding, rod-shaped, aerobic bacterium, designated S7-3-19 , was isolated from apple orchard soil in Gyeongsangnam-do Province, Republic of Korea, and characterized taxonomically by using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequencing showed that strain S7-3-19 belonged to the family Cytophagaceae and was most closely related to Spirosoma linguale DSM 74 (96.38 %), Spirosoma fluviale MSd3 (96.38 %), Spirosoma pulveris JSH5-14 (96.35 %) and Spirosoma radiotolerans DG5A (96.24 %). Chemotaxonomic characteristics supported the classification of strain S7-3-19 within the genus Spirosoma. Summed feature 3 (C16 : 1ω7c/C16 : 1ω6c; 46.7 %) and C16 : 1ω5c (23.8 %) were the major fatty acids. Phosphatidylethanolamine, an unidentified aminophospholipid, an unidentified phospholipid and two unidentified lipids were the major polar lipids. Menaquinone with seven isoprene units was the predominant respiratory quinone. The G+C content of the genomic DNA of strain S7-3-19 was 48.6 mol%. On the basis of its phenotypic properties, genotypic distinctiveness and chemotaxonomic features, strain S7-3-19 represents a novel species of the genus Spirosoma, for which the name Spirosomahorti sp. nov. is proposed. The type strain is S7-3-19 (=KCTC 52728 =JCM 32131 ).

[Ab] Resumo:A Gram-stain-variable, aerobic, rod-shaped, motile and spore-forming bacterial strain, designated CJ11 , was isolated from a tidal flat sediment sample from Ganghwa-do, Republic of Korea. Strain CJ11 grew optimally on R2A at 30 °C and pH 7.0. Sequencing results of the 16S rRNA gene revealed that strain CJ11 possesses two copies of the 16S rRNA gene varying at five nucleotide positions. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain CJ11 belonged to the genus Paenibacillus within the family Paenibacillaceae and was most closely related to Paenibacillus lacus KCTC 33691 (99.36-99.15 % similarity). DNA-DNA relatedness levels of strain CJ11 was 41.7 % (reciprocal, 57.8 %) to P. lacus KCTC 33691 . The G+C content of the genomic DNA was 51.0 mol%. Strain CJ11 contained meso-diaminopimelic acid in the cell-wall peptidoglycan. The major isoprenoid quinone was menaquinone-7. The major cellular fatty acids were anteiso-C15 : 0, C16 : 0 and iso-C16 : 0. The predominant polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, two unidentified phospholipids, an unidentified glycolipid and several unidentified lipids. On the basis of the polyphasic taxonomic study, strain CJ11 represents a novel species in the genus Paenibacillus, for which the name Paenibacillustranslucens sp. nov. is proposed. The type strain is CJ11 (=KACC 19304 =JCM 32080 ).

[Ab] Resumo:A Gram-stain-positive, aerobic, non-motile and short-rod-shaped actinobacterium, designated THG-T121 , was isolated from forest soil. Growth occurred at 10-40 °C (optimum 28-30 °C), at pH 6-8 (optimum 7) and at 0-4 % NaCl (optimum 1 %). Based on 16S rRNA gene sequence analysis, the nearest phylogenetic neighbours of strain THG-T121 were identified as Actinotalea ferrariae KCTC 29134 (97.9 %), Actinotalea fermentans KCTC 3251 (97.3 %), Cellulomonas carbonis KCTC 19824 (97.2 %). 16S rRNA gene sequence similarities among strain THG-T121 and other recognized species were lower than 97.0 %. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, two phosphatidylinositol mannosides, one unidentified phospholipid, three unidentified glycolipids and one unidentified lipid. The isoprenoid quinone was menaquinone (MK-10(H4)). The major fatty acids were anteiso-C15 : 0, anteiso-C15 : 1 A, C16 : 0, iso-C16 : 0, anteiso-C17 : 0 and iso-C17 : 0. The whole-cell sugars of strain THG-T121 were rhamnose, ribose, mannose and glucose. The peptidoglycan type of strain THG-T121 is A4ß, containing l-Orn-D-Ser-L-Asp. The DNA G+C content of strain THG-T121 was 72.4 mol%. DNA-DNA hybridization values between strain THG-T121 and A. ferrariae KCTC 29134 , A. fermentans KCTC 3251 and C. carbonis KCTC 19824 were 30.2 % (27.3 %, reciprocal analysis), 28.4 %, (17.3 %) and 16.9 %, (9.3 %), respectively. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA-DNA hybridization data, strain THG-T121 represents a novel species of the genus Actinotalea, for which the name Actinotaleasolisilvae sp. nov. is proposed. The type strain is THG-T121 (=KACC 19191 =CGMCC 4.7389 ).

[Ab] Resumo:A novel actinobacterial strain, designated X5 , was isolated from the sediment of Taihu Lake in China and was subjected to a polyphasic taxonomic characterization. The strain formed orange-red colonies comprising aerobic, Gram-stain-negative, rod-shaped cells on R2A agar. Phylogenetic analysis based on the 16S rRNA gene sequences revealed that the organism was closely related to the genus Sporichthya and consistently formed a distinct clade along with the members of this genus. The closest phylogenetic neighbour was Sporichthya polymorpha NBRC 12702 with 93.7 % 16S rRNA gene sequence similarity. The major fatty acids (>10 %) were iso-C16 : 0 (18.7 %), C18 : 1ω9c (18.6 %) and C17 : 1ω8c (14.0 %). The genomic DNA G+C content was 74.4 mol%. The organism contained menaquinone MK-8(H2), MK-9(H4) and an unidentified menaquinone. Polar lipids were composed of phosphatidylglycerol, an unidentified lipid, two unidentified phospholipids and two unidentified aminolipids. The whole-cell sugars contained ribose, xylose, mannose, glucose and galactose. The cell-wall peptidoglycan contained ll-diaminopimelic acid. Based on the physiological, biochemical and chemotaxonomic data, the organism is proposed to represent a novel genus and species, for which the name Longivirga aurantiaca gen. nov., sp. nov. is proposed. The type strain is X5 (=CGMCC 4.7317 =NBRC 112237 ).

[Ab] Resumo:Strain M1-21 is a Gram-stain-negative, strictly aerobic and short-rod-shaped bacterium, motile by means of a single polar flagellum; it was isolated from freshwater sediment in Korea. It grew at 10-40 °C (optimum 25 °C), pH 6.0-8.0 (optimum pH 7.0) and with 0-0.75 % (w/v) NaCl (optimal growth occurred in the absence of NaCl) on R2A agar, and it accumulated poly-ß-hydroxybutyrate granules inside the cells. According to 16S rRNA gene sequence analysis, strain M1-21 showed highest sequence similarity with Uliginosibacterium gangwonense (94.7 %) and Uliginosibacterium paludis (94.4 %). Phylogenetic analysis of the 16S rRNA gene sequences revealed that strain M1-21 belongs to the genus Uliginosibacterium. The DNA G+C content of strain M1-21 was 61.9 mol%. The predominant respiratory quinone was ubiquinone-8. The major fatty acids (>10 % of the total) were C16 : 0 and summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c), and the major polar lipids were diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. Strain M1-21 showed distinct phenotypic characteristics that differentiated it from species of the genus Uliginosibacterium. Based on these results, strain M1-21 represents a novel species of the genus Uliginosibacterium, for which the name Uliginosibacterium sediminicola sp. nov. is proposed. The type strain is M1-21 (=KACC 19271 =JCM 32000 ).

[Ab] Resumo:A taxonomic study was carried out on strain LW15 , which was isolated from the external lesions of diseased farmed Murray cod (Maccullochella peelii peelii) from an intensive culture pond. Cells of strain LW15 were Gram-negative, facultative-anaerobic, non-motile, and both coccobacillus- and bacillus-shaped. Growth was observed at NaCl concentrations of 0-2 % (w/v) (optimum, 0 %), 4-32 °C (optimum, 25-28 °C) and pH 5.0-9.0 (optimum, 7.0). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain LW15 was affiliated to the genus Acinetobacter, showing the highest similarity to Acinetobacter guillouiae CIP 63.46 (97.7 %) and other Acinetobacter species with validly published names (93.5-97.6 %). Whole-genome sequencing and phylogeny reconstruction based on a core set of 1061 Acinetobacter genes indicated that strain LW15 was most closely related to the clade formed by A. guillouiae CIP 63.46 and Acinetobacter bereziniae CIP 70.12 and distantly related to any of the described species of genus Acinetobacter. Furthermore, strain LW15 could be distinguished from all known Acinetobacter species by its ability to assimilate ß-alanine and l-arginine, but not d-glucose. The principal fatty acids were C18 : 1ω9c, C16 : 0 and C16 : 1ω7c/C16 : 1ω6c. The major respiratory quinone was Q-9. Polar lipids of strain LW15 comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, four phospholipids, aminolipid and two unknown lipids. Based on its phenotypic and genotypic data, strain LW15 represents a novel species of the genus Acinetobacter, for which the name Acinetobacterpiscicola sp. nov. is proposed. The type strain is LW15 (=MCCC 1K03337 =CICC 24241 =KCTC 62134 =JCM 32101 ).
