[Ab] Resumo:There is implemented the development of the model of assessment of the risk from working conditions/ This model allows to solve the problem of the safe use of pesticides in agricultural production of the Russian Federation. The importance of the parallel assessment of the risk for the specific factor of the external exposure to operators (SF ex.) and absorbed dose (SF int.) is presented. Studies of more than 500 pesticides have shown the following correlation of the SF ex. and SF int.: in 8.3-37.5% cases SF ex. and SF int. were practically equal, in 44.8-85.7% cases SF ex. was higher up to by 10 times and in 8.3-26.3% cases SF int. is higher up to by 30 times. Introduction of the new evaluation criterion SF int. increases reliability of the risk assessment.

[Ab] Resumo:A selective and sensitive magnetic dispersive solid-phase microextraction (MDSPME) coupled with gas chromatography-mass spectrometry was developed for extraction and determination of organophosphorus pesticides (Sevin, Fenitrothion, Malathion, Parathion, and Diazinon) in fruit juice and real water samples. Zero valent Fe-reduced graphene oxide quantum dots (rGOQDs@ Fe) as a new and effective sorbent were prepared and applied for extraction of organophosphorus pesticides using MDSPME method. In order to study the performance of this new sorbent, the ability of rGOQDs@ Fe was compared with graphene oxide and magnetic graphene oxide nanocomposite by recovery experiments of the organophosphorus pesticides. Several affecting parameters in the microextraction procedure, including pH of donor phase, donor phase volume, stirring rate, extraction time, and desorption conditions such as the type and volume of solvents and desorption time were thoroughly investigated and optimized. Under the optimal conditions, the method showed a wide linear dynamic range with R-square between 0.9959 and 0.9991. The limit of detections, the intraday and interday relative standard deviations (n = 5) were less than 0.07 ngmL , 4.7, and 8.6%, respectively. The method was successfully applied for extraction and determination of organophosphorus pesticides in real water samples (well, river and tap water) and fruit juice samples (apple and grape juice). The obtained relative recoveries were in the range of 82.9%-113.2% with RSD percentages of less than 5.8% for all the real samples.

[Ab] Resumo:To evaluate the effects of indoxacarb on saltwater stress response in Cyprinus carpio, the fish were pre-exposed to indoxacarb (0, 0.75, 1.5 and 3mg/L denoted as CP, 0.75IT, 1.5IT and 3IT, respectively) for 21 days and then released to saltwater. A negative control (CN) group was included (the fish were held in indoxacarb-free water for the entire experiment). The fish were sampled immediately (0h) and 24, 48 and 72h after the salinity exposure for the analysis of plasma cortisol, glucose and sodium, chloride, potassium and calcium levels. All fish pre-exposed to 3mg/L indoxacarb, died after the first day of salinity challenge. CP showed typical cortisol response after the salinity challenge, but, cortisol response of the fish pre-exposed to indoxacarb (0.75IT and 1.5IT) was blocked. Plasma glucose increased significantly in all groups compared to the CN; however, this elevation had no consistent trend in 0.75IT and 1.5IT which indicated interference in glucose response due to indoxacarb exposure. Plasma sodium increased (compared to CN) in all groups after the salinity challenge. However, elevation in plasma chloride and potassium was significantly different among the groups and the indoxacarb-treated fish showed slightly sooner ionic disturbance. The results clearly indicate that indoxacarb impairs stress response of C. carpio and the fish may not be able to respond normally to additional stressors, which threatens their survival.

[Ab] Resumo:In temperate zones, seeds of spring-sown crops may be an attractive food source for breeding farmland birds. We modelled the effects of pesticide seed treatments on the reproductive success of 4 UK arable bird species (Rook, Linnet, Skylark, Yellowhammer) exposed to treated seeds of 3 spring-sown crops (beans, barley and linseed). We ran three types of model, 1) a "broods-at-risk" model looking at the temporal overlap between nesting and seed-sowing dates, and estimating the proportion of those nests that suffered toxicity-exposure ratios < 5; 2) a "seasonal success" Markov chain model estimating the number of chicks successfully raised in the course of a breeding season.; and 3) the potential effects of pesticides on population growth rates. Based on physiology, Rooks, should be less at risk from treated seeds than smaller species because bigger birds eat less as a proportion of their bodyweights. However, in nearly all our scenarios, Rooks were more vulnerable, followed by Skylark and Linnet, with Yellowhammer being least affected. A principal cause is that Rooks are more likely to be breeding at a time when treated seeds are being sown. Furthermore, whereas the other species may make several breeding attempts and early failures from pesticide exposure may be compensated by later successes, Rooks breed only once in a season. The results are also supported by historical evidence of Rook population declines following pesticide seed treatments.

[Ab] Resumo:Industrialization and urbanization have posed serious threats to the environment. Excessive release of heavy metals from industrial effluents and overuse of pesticides in modern agriculture are limiting crop production by polluting environment and deteriorating food quality. Sustaining food quality under heavy metals and pesticide stress is crucial to meet the increasing demands for food. 24-Epibrassinolide (EBL), a ubiquitously occurring plant growth hormone shows great potential to alleviate heavy metals and pesticide stress in plants. This review sums up the potential role of EBL in ameliorating heavy metals and pesticide toxicity in plants extensively. EBL application increases plant's overall growth, biomass accumulation and photosynthetic efficiency by the modulation of numerous biochemical and physiological processes under heavy metals and pesticide stress. In addition, EBL scavenges reactive oxygen species (ROS) by triggering the production of antioxidant enzymes such as SOD, CAT, POX etc. EBL also induces the production of proline and soluble proteins that helps in maintaining osmotic potential and osmo-protection under both heavy metals and pesticide stress. At the end, future needs of research about the application of 24-epibrassinolide have also been discussed.

[Ab] Resumo:A rapid, sensitive, and reliable ultra-fast liquid chromatography combined with electrospray ionization tandem mass spectrometry (UFLC-ESI-MS/MS) method was established and applied to simultaneous determination of 31 triazine herbicides and their metabolites in multiple medicinal parts of traditional Chinese medicines (TCMs). A streamlined pretreatment approach using one-step extraction and dilution was proposed, which provided high-throughput processing, excellent recovery, and negligible interference. Afterwards, multiple-reaction monitoring (MRM) and information-dependent acquisition (IDA) triggered enhanced product ion spectra (EPI) was adopted to identify and quantify the targets in a single analysis. The optimized method was then validated according to the guidelines of the European Commission for the following parameters: Matrix effects, specificity, accuracy, precision, linearity, range, and stability. The LOD and LOQ for the 31 triazine herbicides were 0.1-10 µg kg and 0.5-25 µg kg , respectively. Recoveries at three concentration levels were within 67.9-120.3% with an associated precision RSD <20%. Using the proposed approach, trazines herbicides were determined from 44 commercially available TCMs. The detection rate of triazine herbicides residues was 15.9% of the total samples. Among them, atrazine, simeton, and simetryn were found in the radix, herba, and seed TCMs with values far below the referenced maximum residue limits (MRLs), but no residues were detected in either the flos or fructus. Taken together, this method has the potential to provide a means for triazines screening in extensive matrices, thereby laying the foundation for pesticide registration on TCMs. Moreover, it has the potential to guide further triazine residue control in TCMs.

[Ab] Resumo:Prenatal exposure to organophosphorus pesticides (OPs) has been associated with different neurodevelopmental outcomes across different cohorts. A phenotypic approach may address some of these differences by incorporating information across scales and accounting for the complex correlational structure of neurodevelopmental outcomes. Additionally, Bayesian hierarchical modeling can account for confounding by collinear co-exposures. We use this framework to examine associations between prenatal exposure to OPs and behavior, executive functioning, and IQ assessed at age 6-9 years in a cohort of 404 mother/infant pairs recruited during pregnancy. We derived phenotypes of neurodevelopment with a factor analysis, and estimated associations between OP metabolites and these phenotypes in Bayesian hierarchical models for exposure mixtures. We report seven factors: 1) Impulsivity and Externalizing, 2) Executive Functioning, 3) Internalizing, 4) Perceptual Reasoning, 5) Adaptability, 6) Processing Speed, and 7) Verbal Intelligence. These, along with the Working Memory Index, were standardized and scaled so that positive values reflected positive attributes and negative values represented adverse outcomes. Standardized dimethylphosphate metabolites were negatively associated with Internalizing factor scores (ß^ - 0.13, 95% CI - 0.26, 0.00) but positively associated with Executive Functioning factor scores (ß^ 0.18, 95% CI 0.04, 0.31). Standardized diethylphosphate metabolites were negatively associated with the Working Memory Index (ß^ - 0.17, 95% CI - 0.33, - 0.03). Associations with factor scores were generally stronger and more precise than associations with individual instrument-specific items. Factor analysis of outcomes may provide some advantages in etiological studies of childhood neurodevelopment by incorporating information across scales to reduce dimensionality and improve precision.

[Ab] Resumo:The paper presents experimental data on the pesticides action (molinate (OrdramA), Dinitroortocresolum and chlorophos) at concentrations of 0.01 mg /l to 10 mg/l on sanitary-indicative (TBC, total coliform bacteria, E. coli, E. faecalis), potentially pathogenic ( Ps. aeruginosa) and pathogenic (S. Derby) microflora of the river water. As the test cultures there were used isolated from the reservoir and museum strains of microorganisms. Due to the fact that the rate of decomposition ofpesticides in water increases in direct proportion to the extent of its biological contamination, the experiments were performed with the river water: native and sterilized by autoclaving. There were identified species and strain differences in the microbial response to the action of pesticides. The selectivity of OrdramA action at a concentration of 0.1 to 10 mg/l on Salmonella and of chlorophos at a concentration of 10 mg/l for Salmonella and E. coli is manifested in the stimulation of the reproduction of these bacteria. Dinitroortocresolum in concentrations of 10 mg/l and 1 mg/l gives rise in the reproduction of Pseudomonas aeruginosa and inhibits the growth of total coliform bacteria and E. coli, in a concentration of 0.1 mg/l the preparation stimulates the viability of all the studied microorganisms. When entering the body of water, pesticides were found to cause changes of biocenosis in the river water and disturb bacterial self-cleaning processes. In conditions of pesticide pollution total coliform bacteria, E. coli and E. faecalis unable to maintain their indicative value and therefore the use of only them for the evaluation of the sanitary-epidemiological status of the water source appears to be insufficient. Due to the fact that the epidemic potential of water is directly dependent on quantitative content ofpathogenic and opportunistic pathogenic microorganisms in it, the reproduction Salmonella and Pseudomonas bacteria is a negative moment from the sanitary and epidemiological point of view.

[Ab] Resumo:The article highlights the methodical features of hygienic regulation of pesticide preparation in water bodies, concretizes and supplements (specified) number of positions of the acting current methodical document on hygienic evaluation of new pesticides. From the position of the comprehensive hygienic standardization there are marked principles of the study of MPC for pesticides in water bodies, there is substantiated the expediency of expanding the arsenal of methods for determination of threshold concentrations of pesticides on the impact on organoleptic properties of water and sanitary regime of reservoirs, shows the role and importance of hydrolysis processes of active ingredients ofpesticides in water, which should be considered in the assessment of the impact of drugs on the processes of self-purification of water bodies.

[Ab] Resumo:We developed a high-resolution expression microarray based on 2456 unique transcripts from a cDNA library of the aquatic midge (Chironomus tentans). By using the microarray, we detected that 146, 434 and 243 genes were differentially expressed after C. tentans larvae were exposed to chlorpyrifos (organophosphate insecticide) at 0.1 and 0.5 µg/L, and atrazine (triazine herbicide) at 1000 µg/L, respectively, for 48 h. The number of differentially expressed genes in the larvae exposed to chlorpyrifos at 0.5 µg/L was three times of that in the larvae exposed to chlorpyrifos at 0.1 µg/L. Among the differentially expressed genes in response to chlorpyrifos exposures, 76 genes showed significant Blast hits, and among them 42 were in common between the chlorpyrifos and atrazine exposures. In 19 differentially expressed xenobiotic detoxification genes, 16 were significantly up-regulated in the larvae exposed to chlorpyrifos and/or atrazine. Two cytochrome P450 genes (CtCYP6EV1 and CtCYP4DG2) were specifically up-regulated by chlorpyrifos, whereas three cytochrome P450 genes (CtCYP4DG1, CtCYP6EX3 and CtCYP6EV3) were specifically up-regulated by atrazine. Our results showed that chlorpyrifos exposures even at low concentrations can lead to significant changes in gene expression. The significant transcriptional responses are likely attributed to larval intoxication by the insecticide. These results not only support our previous studies in which candidate gene approaches were used, but also can potentially help develop specific molecular markers for monitoring pesticide exposures in non-target organisms in aquatic systems.
