[Ab] Resumo:In the work there are presented results of studies on the validation of the gas chromatographic (GC) methodfor the determination of chlorothalonil residue amounts in peaches with the use of electron capture detector (ECD). For the analytical control there was selected such stone fruit crop as the peach, referring to the crops, the most contaminated with residue amounts ofpesticides. There was justified the necessity of the inclusion in the procedure of the detection of the method of confirmation, based on mass spectrometry detection (MSD) (the type of ionization - electron impact). The significant source of the obtaining of incorrect data in the identification with the use of ECD of ions are shown to be phthalates, visualized in the chromatogram as intense and/or broad peaks. Mass spectra of compounds of the class ofphthalates are characterized by the dominant peak of the ion with the value of (mass/ charge) 149, just on this peak the detection of low molecular weight phthalates occurs in various matrices, on the spectrum there are also recorded typical ions corresponding to fragments of radical residues. The combination of the use of various types of the detection allows to prove that the revealed response (detector signal) is caused just by the analyte, but not the impurities, and optionally to optimize chromatographic conditions towards to the obtaining reliable results. The lower limit of the quantitation of chlorothalonil in peach fruits accounts for 0.01 mg/kg, determined with a signal/noise ratio of 10. The range of measured concentrations is volatile between 0.01-0.1 mg/kg, recovery rate of chlorothalonil from samples of peaches, established according to results of the analysis of model samples with the introduction of the substance in four points along the detection range, was 84-102%, the SD value of the repeatability varies in the range of 2.0-5.8%.

[Ab] Resumo:A number of poisoning and suicide cases involving formamidine pesticides have been reported, thus developing a rapid and low cost determination method is crucial. In this work, a rapid, sensitive and low-cost method for the simultaneous determination of formamidine pesticides (amitraz, chlordimeform, formetanate) and their main metabolites, N-(2,4-dimethylphenyl)-N-methyl-formamidine, 2,4-dimethylformamidine, 2,4-dimethylaniline, 4-chloro-2-methylaniline and 3-hydroxyacetanilide in human blood by high-performance liquid chromatography with tandem mass spectrometry is developed. The application of columns with core-shell particles significantly reduced the analysis time. Very low LODs (0.01-0.04 µg L ) were obtained for formamidine pesticides and their metabolites. The method was successfully applied to the analysis of human blood samples from a real forensic case. The significantly reduced analysis time, high sensitivity and low cost are the primary advantages of the developed method. This methodology provides important value for sensitive and rapid determination of residue pesticides and metabolites, study of residue pesticides behavior in human body, as well as application in real forensic cases.

[Ab] Resumo:In the present study, a liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) method with a minimal matrix effect (ME) was developed and validated for simultaneous determination of a diverse range of pesticides (49) in kiwifruit. Samples extracted by the quick, easy, cheap, effective, rugged, and safe (QuEChERS) citrate-buffered method were analyzed either without purification or following purification (with primary secondary amine (PSA) or PSA + graphitized carbon black (GCB)). With the addition of a clean-up step, the suppression of the ME decreased, with a higher number of pesticides determined by the application of PSA + GCB. The method exhibited good linearity with coefficients of determination (R ) ≥ 0.9972 and satisfactory recoveries (70-120%) with a relative standard deviations (RSDs) <10%. The limits of quantification (LOQ) were lower than the maximum residue limits (MRLs) set by the Korean Ministry of Food and Drug Safety (MFDS) and the CODEX Alimentarius. The developed method was applied to the real samples and the results indicated that the quantitated levels of all pesticides, except for pyraclostrobin and carbendazim, are lower than the MRLs set by the regulatory authorities. The percentage of the acceptable daily intake was <20%, suggesting that there is no risk associated with the intake of residual pesticides through kiwifruit.

[Ab] Resumo:A method was developed for the confirmatory and quantitative analysis of one pyrethrin and 18 pyrethroid residues in animal fat. Fat was extracted was collected from adipose tissue melted in an oven at 65 °C for 2 h. Fat samples (1 g) were dispersed with deactivated Florisil sorbent and extracted with MeCN. Sample extracts were purified by cold temperature precipitation at -30 °C for 4 h and further purified using dispersive solid-phase extraction (d-SPE) clean-up in tubes containing 500 mg of Z-SEP+ and 125 mg of PSA bonded silica. Purified samples were analysed by ultra-high performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS) detection. Chromatographic separation was carried out on a Acquity C BEH column, using a binary gradient separation comprising of mobile phase A, 5 mM ammonium formate in water:MeOH (80:20, v/v,) and mobile phase B, 5 mM ammonium formate in MeOH. The mass spectrometer was operated in the positive electrospray ionisation mode (ESI(+)). Validation was performed following the 2002/657/EC guidelines. Trueness ranged between 84% and 143% and precision ranged between 3.9% and 29%. The developed method is particularly advantageous because the sample preparation procedure does not require complex sample extraction equipment and uses less solvent compared to other sample preparation protocols.

[Ab] Resumo:A simple method for determining 33 pesticides with a wide polarity range (logK 0.6-4.5) in aquatic products was developed based on LC-MS/MS. The target analytes included three types of widely used pesticides: insecticides, fungicides and herbicides. Based on the optimization of ultrasonic assisted extraction and GPC clean-up procedures, the matrix effect, extraction recoveries and LOD were improved distinctively. LOQ of this method was below 0.5ng/g for all pesticides, which is superior to values in the literature, and the matrix effect was reduced effectively (-14.7% to 7.5%). The method was successfully applied to investigate the pesticide residue levels of twenty-five samples including seven common kinds of fishes from Northeast China. The results showed that all targeted pesticides were present in the fish samples; however, their levels were low, except for atrazine, linuron, ethoprophos, tetrachlorvinphos, acetochlor and fenthion. Atrazine and linuron caught our attention because the concentrations of atrazine in fish samples from Liaoning province were in the range of 0.5-8ng/g (w/w) with mean concentration of 2.3ng/g, which were far above those of other pesticides. The levels of linuron were in the range of 0.6-6ng/g (mean concentration 2.8ng/g), which were the highest among all targeted pesticides in the Inner Mongolia. This is the first systematic investigation on the characteristics and levels of these pesticides in aquatic products from northeast China. Considering their toxicity and bioaccumulation, the potential risk of atrazine and linuron from consuming aquatic products should be paid more attention.

[Ab] Resumo:Over 16.7 million workers in Thailand (42 percent of the working population) are engaged in agriculture, disproportionately from the lower socioeconomic strata of Thai society. Most agricultural workers (over 93 percent) work in the informal sector without the protections of regulations or enforcement of labor or health and safety laws or enrollment in a social security system. Although Thailand's use of herbicides, fungicides, and insecticides is growing, there is little regulation of the sale, use, or application of these potentially toxic chemicals. This paper summarizes the research to date on occupational health and safety for Thai agricultural workers, identifies gaps in pesticide regulations and the current systems for occupational health and safety and social support for Thai agricultural workers, and makes recommendations for future policy and research initiatives to fill the identified gaps.

[Ab] Resumo:Soil aging will influence the chemical speciation of pesticides, thus affecting the uptake route to be bioavailable to the organism. So far, studies on the possible effects of the uptake route on the distribution and elimination of pesticides in the organism that also considers effects of aging are limited. In our study, Eisenia fetida was exposed to 4.5 mg kg lindane aging for 0, 30 and 180 d, and the accumulation, distribution and elimination of lindane in the earthworms were analyzed. The results showed that the 6 h Tenax-extracted fraction exhibited a good linear correlation with the lindane accumulated in the earthworms. With aging time increasing, the bioaccumulation of lindane decreased and the accumulative balance was more easily reached in the earthworms. Lindane distributions were found in the whole earthworm and the proportions of lindane content at sub-organism level and the mass distribution of each fraction were similar for 0 d and 180 d aged groups. The foregut accumulated the highest content of lindane (20%) relative to its low mass distribution proportion (10%). The elimination rate of lindane in the earthworms decreased with aging time extending. Our conclusion was that the 6 h Tenax extraction could be used to assess the bioavailability of aging lindane. Although soil aging decreased the bioavailability of lindane, the soil-bound lindane entered the earthworm through dietary route would take longer to depurate from the organisms than free lindane, which implied the potential ecological risk of bound pesticide residues.

[Ab] Resumo:Due to concerns over negative impacts on insect pollinators, the European Union has implemented a moratorium on the use of three neonicotinoid pesticide seed dressings for mass-flowering crops. We assessed the effectiveness of this policy in reducing the exposure risk to honeybees by collecting 130 samples of honey from bee keepers across the UK before (2014: N = 21) and after the moratorium was in effect (2015: N = 109). Neonicotinoids were present in about half of the honey samples taken before the moratorium, and they were present in over a fifth of honey samples following the moratorium. Clothianidin was the most frequently detected neonicotinoid. Neonicotinoid concentrations declined from May to September in the year following the ban. However, the majority of post-moratorium neonicotinoid residues were from honey harvested early in the year, coinciding with oilseed rape flowering. Neonicotinoid concentrations were correlated with the area of oilseed rape surrounding the hive location. These results suggest mass flowering crops may contain neonicotinoid residues where they have been grown on soils contaminated by previously seed treated crops. This may include winter seed treatments applied to cereals that are currently exempt from EU restrictions. Although concentrations of neonicotinoids were low (<2.0 ng g-1), and posed no risk to human health, they may represent a continued risk to honeybees through long-term chronic exposure.

[Ab] Resumo:A QuEChERS technique-based sample preparation method was optimized and validated in small cardamom to monitor the residues of 154 pesticides by LC with tandem MS. The proposed multiresidue method involved soaking powdered cardamom (2 g) in water (8 mL) for 30 min, followed by extraction with acetonitrile (10 mL). Cleanup by dispersive SPE was performed using primary secondary amine (25 mg/mL), C18 (25 mg/mL), and anhydrous magnesium sulfate (150 mg/mL). The method was validated as per the SANTE/11945/2015 guidelines at 5, 10, 50, and 100 ng/g spiking levels, and most of the analytes showed recoveries between 70 and 120% (with RSDs ≤20%). The LOQ of ≤10 ng/g was achieved for almost 90% of the target pesticides. The measurement uncertainties were evaluated at 100 ng/g, and the global uncertainty values were below 22% for all the analytes.

[Ab] Resumo:The purpose of the present work was the experimental evaluation of pesticides transfer to virgin olive oil during the production step and prediction of their processing factors, which could be eventually used for the calculation of maximum residue limits (MRLs) in olive oil from the MRLs set in olives. A laboratory-scale Abencor system was used for the production of olive oil from olives spiked with the 104 pesticides studied, three different chromatographic methods being used for the analysis of raw olives and the obtained olive oil: (i) gas chromatography-tandem mass spectrometry (GC-MS/MS) for GC-amenable pesticides; (ii) hydrophilic interaction liquid chromatography-tandem mass spectrometry (HILIC-MS/MS) for polar pesticides, and; (iii) reversed-phase liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) for low to medium polarity pesticides. Processing factors experimentally calculated were correlated to their octanol-water partitioning coefficient (logK ), enabling the calculation of the equivalent MRLs in olive oil from the MRLs in olives, considering the percentage of oil extracted (oil yield) and the log K of each pesticide.
