[Ab] Resumo:Hyperglycaemia triggers increased production of methylglyoxal which can cause gross modification in proteins' structure vis-a-vis function though advanced glycation end products (AGEs). The AGEs may initiate vascular and nonvascular pathologies. In this study, we have examined the biochemical and biophysical changes in human IgG under normal and high glucose after introducing methylglyoxal into the assay mixture. This non-enzymatic reaction mainly engaged lysine residues as indicated by TNBS results. The UV results showed hyperchromicity in modified-IgG samples while fluorescence data supported AGEs formation during the course of reaction. Shift in amide I and amide II band position indicated perturbations in secondary structure. Increase carbonyl content and decrease in sulfhydryl suggests that the modification is accompanied by oxidative stress. All modified-IgG samples showed more thermostability than native IgG; the highest Tm was shown by IgG-high glucose-MGO variant. Results of ANS, Congo red and Thioflavin T dyes clearly suggest increase in hydrophobic patches and aggregation, respectively. SEM and TEM images support aggregates generation in modified-IgG samples.

[Ab] Resumo:Targeted photopolymerization is the basis for multiple diagnostic and cell encapsulation technologies. While eosin is used in conjunction with tertiary amines as a water-soluble photoinitiation system, eosin is not widely sold as a conjugate with antibodies and other targeting biomolecules. Here we evaluate the utility of fluorescein-labeled bioconjugates to photopolymerize targeted coatings on live cells. We show that although fluorescein conjugates absorb approximately 50% less light energy than eosin in matched photopolymerization experiments using a 530 nm LED lamp, appreciable polymer thicknesses can still be formed in cell compatible environments with fluorescein photosensitization. At low photoinitiator density, eosin allows more sensitive initiation of gelation. However at higher functionalization densities, the thickness of fluorescein polymer films begins to rival that of eosin. Commercial fluorescein-conjugated antibodies are also capable of generating conformal, protective coatings on mammalian cells with similar viability and encapsulation efficiency as eosin systems.

[Ab] Resumo:Brazilian green propolis (BGP) is noted for its impressive antitumor effects and has been used as a folk medicine in various cultures for many years. It has been demonstrated that BGP could enhance the cytotoxic effect of cytostatic drugs on tumor cells. Photodynamic therapy (PDT) is a therapeutic approach used against malignant cells. To assess the synergistic effect of BGP extract on protoporphyrin IX (PpIX)-mediated photocytotoxicity, MTT assays were performed using A431 and HeLa cells. TUNEL assay and Annexin V-FITC/PI staining were performed to confirm the induction of apoptosis. Western blotting analysis was performed to examine the pro-apoptotic proteins, anti-apoptotic proteins and inflammation related proteins in A431 cells. Intracellular accumulation of PpIX was examined by flow cytometry. The synergistic effect of BGP extract in PpIX-PDT was also evaluated with a xenograft model. Our findings reveal that BGP extract increased PpIX-mediated photocytotoxicity in A431 and HeLa cells. PpIX-PDT with BGP extract treatment resulted in a decrease in Bcl-xL and an increase in NOXA, Bax and caspase-3 cleavage. The protein expression levels of p-IKKα/ß, NF-κB and COX-2 were upregulated by PpIX-PDT but significantly attenuated when in combination with BGP extract. BGP extract was also found to significantly enhance the intracellular accumulation of PpIX in A431 cells. BGP extract increased PpIX-mediated photocytotoxicity in a xenograft model as well. Our findings provide evidence for a synergistic effect of BGP extract in PpIX-PDT both in vitro and in vivo.

[Ab] Resumo:We studied the ability of oligonucleotides C T (n = 2, 5, 7, 9, 12, 25) to form an intermolecular i-motif using circular dichroism, ultra-violet spectroscopy, nuclear magnetic resonance, high-resolution atomic force microscopy, high-performance liquid chromatography, and molecular dynamics simulations. The arrangement of single-stranded oligonucleotides in multimer i-motifs was very unusual: C-tracts of different oligonucleotides followed each other consecutively in order to fold into a closed intermolecular i-motif core with minimal loops (one cytidine in a loop spanning over a minor groove, three cytidines in a loop over a major groove); intact T-tracts protruded from predefined loci allowing visualization of beetle-like nanostructures by atomic force microscopy. The same structures were formed from analogous biotinylated oligonucleotides demonstrating one of the potential applications of such structures as carriers of multiple functional groups. Our findings open up possibilities for the rational design of pH-sensitive DNA aggregates and evaluation of the efficiency of their assembly.

[Ab] Resumo:In the present day, nanotechnology is one of the most promising leading scientific and potentials areas in modern key technology development toward to the humankind. The synthesis of noble metal nanoparticles (NPs) is an expanding research area due to the possible applications for the development of bio-medical applications. Eco-friendly approach for the biosynthesis of gold nanoparticles (AuNPs) using the aqueous extract from Ruellia tuberosa and Phyllanthus acidus (leaf and twig) for the first time. Surface active AuNPs were characterized by UV-Vis spectroscopy, FTIR (Fourier transform infrared) spectroscopy, DSC (differential scanning colorimetry), DLS (dynamic light scattering) and environmental SEM (scanning electron microscope) analysis at room temperature (RT). Enhanced surface plasmon resonance (SPR) absorbance UV visible optical spectra were detected in the range of 552, 548, 558 and 536 nm. SEM and DLS (transmission mode) analysis confirmed the morphology of the nanoparticles to be spherical with the average size in the range of 88.37, 94.31, 82.23 and 81.36 nm. Further they have enhanced the enzyme activity on α-amylase, cellulase, and xylanase. The results suggest that the phyto-fabricated AuNPs from R. tuberosa and P. acidus is simple, less expensive, eco-friendly, green synthesis and also can be exploited for the potential future industrial and bio-medical applications.

[Ab] Resumo:The potential application of any nanoparticles, including silver nanoparticles (AgNPs), strongly depends on their stability against aggregation. In the current study, an aqueous extract of pomegranate peel was used as a stabilizer during synthesis of AgNPs. Nanoparticles have been prepared by the chemical reduction method from an aqueous solution of silver nitrate in the presence of sodium borohydride as a reducing agent. The AgNPs were characterized by dynamic light scattering (DLS), zeta-potential measurements, UV-Vis spectroscopy and transmission electron microscopy (TEM). The antibacterial efficiency of AgNPs against Escherichia coli was investigated. The size, polydispersity index, FWHM, and colloidal stability of nanoparticles in dispersion depends on the extract concentrations. In the presence of pomegranate peel extract, the nanoparticles suspension shows colloidal stability at least for a week. Our studies show that synthesized AgNPs with the above described procedure were stable at pH = 3-12 and in the temperature range of 25-85 °C. Additionally, AgNPs exhibit antibacterial properties, especially at the lowest amount of extract to silver ratio (K ).

[Ab] Resumo:Omeprazole is a proton pump inhibitor used to treat the symptoms of gastro esophageal reflux disease, ulcers, excess stomach acid, infection with Helicobacter pylori, and to control the gastric side effects of various drugs. The approved dosage forms in the U.S. are powder in compounding kits, delayed-release granules for oral suspension, oral delayed-release tablets, and oral delayed-release capsules. An extemporaneously compounded unsweetened oral liquid method, published in the International Journal of Pharmaceutical Compounding, was found to be commonly used by pharmacists. This project investigated the robustness of the compendium omeprazole high-performance liquid chromatographic assay in evaluating an oral liquid made from commercial delayed-release pellets, the potency of extemporaneously compounded solutions having a 1.125% v/v flavored versus unflavored samples stored at controlled cold temperatures at different time points, and examining the absorption spectrum of the flavoring agent. As part of the study, stability-indication testing was also conducted. The studies indicate that the chromatographic area under the plasma concentration-time curve of both study groups remained over 90% of the label claim during the follow-up period. The flavor did not significantly impact the pH of the oral liquid. This study further identified (1) an increase in resilient foam formation in the flavored liquid, potentially hindering dosing accuracy, (2) omeprazole is oxidized easily by 3% hydrogen peroxide, and (3) flavoring agent absorbs in an ultraviolet visible spectroscopy spectral range often used in assay detectors for quantification of drug molecules, and could interfere with assay protocols of the same.

[Ab] Resumo:An inexpensive, rapid method for the determination of octyl methoxycinnamate (OMC) in sunscreen products using ultraviolet-spectrophotometry has been developed and validated according to International Council for Harmonisation and Association of Official Analytical Chemists guidelines. Methanol was the optimal solvent used with a linearity range of 4­12 µg\/ml (r = 0.999) being obtainable. The accuracy of the method is highlighted by the % recovery (98.23­98.50) and relative standard deviation (%RSD, 0.12), and it is widely applicable to prototype products composed of oil in water, and water in oil emulsions. Mineral oils containing low, intermediate, and high OMC levels (1%, 4%, and 7.5%) gave recovery percentages of 99.76­100.76 with %RSD of 0.02­0.28. In addition, this method is repeatable and affords a high degree of precision (%RSD = 0.12 and 0) with 96.08­99.27% recovery. The method is suitable for quality assurance of suncare product formulations, and could be applicable to product development and validation.

[Ab] Resumo:2,4-Dichlorophenoxyacetic acid (2,4-D) is a chlorophenoxy herbicide used worldwide. We describe a high-performance liquid chromatography (HPLC) method with UV detection for the determination of 2,4-D in female and male rat serum. This allows to observe the change of serum 2,4-D concentration in rats with time and its pharmacokinetics characteristics with a simple, rapid, optimized and validated method. The serum samples are pretreated and introduced into the HPLC system. The analytes are separated in a XDB-C18 column with a mobile phase of acetonitrile (solvent A) and 0.02 M ammonium acetate (containing 0.1% formic acid) (solvent B) using a gradient elution at a flow rate of 1.0 mL/min. The wavelength for UV detection was set at 230 nm. Calibration curve for 2,4-D was constructed over a range of 0.1-400 mg/L. The method was successfully applied to study the pharmacokinetics of 2,4-D in rats in this study. After oral administration of 300 mg/kg and 60 mg/kg 2,4-D, the mean Cmax values were 601.9 and 218.4 mg/L, the AUC0→∞ values were 23,722 and 4,127 mg×h/L and the clearance (Cl) were 1.10 and 0.02 L/(h×kg), respectively. The developed method was found to be specific, precise, reproducible and rapid.

[Ab] Resumo:Microbial biosynthesis of metal nanoparticles has been extensively studied for the applications in biomedical sciences and engineering. However, the mechanism for their synthesis through microorganism is not completely understood. In this study, several culture media were investigated for their roles in the microbial biosynthesis of silver nanoparticles (AgNPs). The size and morphology of the synthesized AgNPs were analyzed by UV-vis spectroscopy, Fourier-transform-infrared (FT-IR), transmission electron microscopy (TEM), and dynamic light scattering (DLS). The results demonstrated that nutrient broth (NB) and Mueller-Hinton broth (MHB) among tested media effectively reduced silver ions to form AgNPs with different particle size and shape. Although the involved microorganism enhanced the reduction of silver ions, the size and shape of the particles were shown to mainly depend on the culture media. Our findings suggest that the growth media of bacterial culture play an important role in the synthesis of metallic nanoparticles with regard to their size and shape. We believe our findings would provide useful information for further exploration of microbial biosynthesis of AgNPs and their biomedical applications.
