[Ab] Resumo:The aim is the study of the circulation, biological activity, persistent potential and genotypic characteristics of opportunistic Enterobacteriaceae of microbial community of the Lena River which is the one of the largest sources of water in Russia and Siberia. Gram-negative component of microbial community in the Lena River was mainly represented by Enterobacteriaceae (80%), including dominated Escherichia (dominant class) and Enterobacter and Klebsiella (subdominant class). In conditions of anthropogenic pollution there is a reorganization of microbial community of the Lena River in the direction of the increase in the proportion of opportunistic pathogens, their species diversity; there is a change of the biological activity of microorganisms, gains of share of strains characterized by antilysozyme activity, hemolytic activity, production DNase, phosphatase. The parameters of system "lysozyme-antilysozyme" are changing, the frequency of the occurrence of bacteria with antilysozyme activity is increasing. This shows the restructurization of the microbial community of water objects. Microorganisms of water objects in territories of anthropogenic pollution are characterized by multiple antibiotic resistance. The proportion of gram-negative opportunistic Enterobacteriaceae in microbial communities of water ecosystems determines the potential danger of water objects and the impact on the level of antibiotic resistance of bacteria. Opportunistic Enterobacteriaceae of microbial community of the Lena River near the city of Yakutsk are characterized by the presence of genetic determinants of pathogenicity (hlyA and sfaG). This is indicative of their potential epidemiological relevance. Microorganisms with high biological activity are markers of their epidemiological danger.

[Ab] Resumo:As a result, of the management of the irrigation system the most part of the runoff headwaters of the river Kuban was transferred to the arid plains of the Stavropol Territory, Rostov Region and Kalmykia Gravity Water via the water tract of the Kuban-Manych. This system was assumed to be supplied by pure mountain water. In fact, 3-4 class contaminated water currently passes to the water intake of the irrigation system (Nevinnomyssky channel). There is a tendency to the further deterioration in the quality of surface waters. It was determined that in the last decades in the catchment area of the upper reaches of the Kuban (Karachaevo-Cherkessia) the population was determined to increase sharply. As a result the discharge of industrial, agricultural, domestic and recreational waste into the river significantly increased. In that in catchment areas there is practically no infrastructure of the acquisition, processing and recycling of waste for the irrigation system. Intensive recreational and transport development of mountainous areas of Karachay-Cherkessia aggravates the situation and may lead to the need for deep water purification for subsequent consumption already in the vast territories of the Central Caucasus. Due to lack of the infrastructure for the water treatment in the upper reaches of the Kuban, it can lead to the serious systemic crisis. It is proposed to start to create in the catchment areas the cost-based system of recycling waste on the base of their processing by pyrolysis.

[Ab] Resumo:Exploration and development of mineral deposits in the Transurals of the Republic of Bashkortostan are accompanied by a comprehensive environmental impact. Ones of the most vulnerable elements of the landscape are the surface water bodies used for fishery purposes, recreation and household needs of the population. Extraction and processing of ores lead to the contamination of aquatic ecosystems by heavy metals (HM), which by passing into a pond, can actively be involved in the cycling of matter and migrate via food chains to the human under the consumption of fish products. Possessing by cumulative properties, HM can express mutagenic, teratogenic and carcinogenic properties. The aim of research was the study of the impact of mining facilities on the content of the priority HM in the components of river ecosystems in the Transurals of the Republic of Bashkortostan and the development of a complex of preventive measures on the improvement of the human environment. There was studied the content of Zn, Cu and Cd in the water, bottom sediments and phytomass of Elodea canadensis Michx. The measurements of mass concentrations of metals were performed by stripping voltammetry with the use of the STA device. The results showed that for the cross-sections there is typical the following descending series of elements in the river components: Zn > Cu > Cd. In water samples there is observed the exceedance of standards for water bodies of potable, cultural and community water use and fishery basins. The concentration of zinc varies in the range of 0.016-5.24 mg/dm; copper - 0.0024-0.095 mg/dm; cadmium - 0.0001-0.019 mg/dm. In bottom sediments, in general, the content of metals in comparison with control plots is increased to the tenfold value. The meaning of mobile forms of zinc in the ground varies in the range of 0.81-9.62 mg/kg; copper - 0.12-18.69 mg/kg; cadmium - 0.00013-0.092 mg/kg. The zinc concentration in above-ground phytomass of Elodea canadensis compared to control increases by 2-3.5 times, copper - 4-8 times, cadmium - 2-7 times. Univariate analysis of variance confirmed the accuracy of the impact of the degree of contamination on the accumulation of metals by Elodea canadensis. The results testify to the potential danger to health of water bodies of the region's population.

[Ab] Resumo:Compound specific stable isotope analysis (CSIA) has been established as a useful tool to evaluate in situ biodegradation. Here, CSIA was used to determine microbial dehalogenation of chloro- and bromobenzenes in microcosms derived from Hackensack River sediments. Gas chromatography-isotope ratio mass spectrometry (GC-IRMS) was used to measure carbon isotope fractionation during reductive dehalogenation of hexachlorobenzene (HCB), pentachlorobenzene (PeCB), 1,2,3,5-tetrachlorobenzene (TeCB), 1,2,3,5-tetrabromobenzene (TeBB), and 1,3,5-tribromobenzene (TriBB). Strong evidence of isotope fractionation coupled to dehalogenation was not observed in the substrate, possibly due to the low solubilities of the highly halogenated benzene substrates and a dilution of the isotope signal. Nonetheless, we could measure a depletion of the δ C value in the dichlorobenzene product during dechlorination of HCB, the sequential depletion and enrichment of δ C value for trichlorobenzene in TeCB dechlorinating cultures, and the enrichment of δ C during debromination of TriBB. This indicates that a measurable isotope fractionation occurred during reductive dehalogenation of highly halogenated chloro- and bromobenzenes in aquatic sediments. Thus, although more quantitative measurements will be needed, the data suggests that CSIA may have application for monitoring in situ microbial reductive dehalogenation of highly halogenated benzenes.

[Ab] Resumo:Water quality criteria (WQC) are considered to be an effective management tool for protecting aquatic environments. To derive site-specific WQC for an area, local data based on local species are essential to improve the applicability of WQC derived. Due to the paucity of local fish data available for the development of site-specific WQC for the Liao River, China, four local and widespread fishes (Pseudorasbora parva, Abbottina liaoningensis, Ctenogobius giurinus, and Misgurnus anguillicaudatus) were chosen to test their sensitivities to ammonia, cadmium and nitrobenzene. These compounds are common and regularly-measured pollutants in Chinese rivers. In addition to the published data for species resident in the Liao River, site-specific WQC for the three chemicals were derived using both a log-logistic species sensitivity distribution (SSD) and the method recommended by the USEPA, in line with current best practice, which were then compared with Chinese national WQC. It was found that A. liaoningensis was the most sensitive, followed, in order, by P. parva, C. giurinus and M. anguillicaudatus was the least sensitive, and this trend was the same to all three chemicals tested. When comparing the SSD derived solely from previously-published data with that including our data on local fish, there were significant differences identified among parameters describing the SSD curves for ammonia and nitrobenzene and significant differences were detected for site-specific WQC derived for all of the three chemicals. Based on the dataset with local fish data taxa, site-specific WQC of Liao River for ammonia, cadmium, and nitrobenzene were derived to be 20.53mg/L (at a pH of 7.0 and temperature of 20°C), 3.76µg/L (at a hardness of 100mg/L CaCO ), and 0.49mg/L, respectively. Using the same deriving method for each chemical, the national Chinese WQC were higher than site-specific WQC derived in this study for ammonia (national WQC of 25.16mg/L) and nitrobenzene (national WQC of 0.57mg/L), while the national WQC for cadmium was lower (national WQC of 1.81µg/L). These results indicated that published data can be helpful for use when deriving site-specific WQC but that there were differences between site-specific and national WQC which may lead to either over- or under-protection depending on the pollutant if national WQC were used as the basis for the water management of specific river systems, like the Liao River.

[Ab] Resumo:An aerobic, Gram-stain-negative, rod-shaped, non-motile bacterium capable of degrading the polycyclic aromatic hydrocarbon pyrene was isolated from sediment of Pearl River and designated PrR001. 16S rRNA gene sequence analysis revealed that this strain was affiliated within the genus Defluviimonas in the family Rhodobacteraceae of the class Alphaproteobacteria and showed great similarity with the type strain Defluviimonas indica 20V17 (96.3 % similarity). The DNA G+C content of strain PrR001 was 68.3 mol%. The major cellular fatty acids comprised summed feature 8 (C18 : 1ω7c/C18 : 1ω6c), C19 : 0 cyclo ω8c, C18 : 0 3OH, and C18 : 0. The sole respiratory lipoquinone was ubiquinone-10. The main polar lipids were diphosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, an unidentified aminolipid, an unidentified aminophospholipid and three unidentified phospholipids. Based on physiological, chemotaxonomic and phylogenetic analysis, strain PrR001 is suggested as a novel species in the genus Defluviimonas, for which the name Defluviimonas pyrenivorans sp. nov. is proposed. The type strain of Defluviimonas pyrenivorans is PrR001 (=CICC 24263 =KCTC 62192 ).

[Ab] Resumo:An aerobic, Gram-negative, motile by means of a single polar flagellum, and ovoid-shaped bacterium, designated D3 , was isolated from shallow stream sediments in Sinan-gun, South Korea. Growth occurred at 15-40 °C (optimum 35 °C), at pH 7.0-8.0 (optimum pH 7.0), and at an optimum NaCl concentration of 0.5 % (w/v). The major cellular fatty acids (>7 % of the total) were C16 : 0, C18 : 0 2-OH, summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and summed feature 8 (C18 : 1ω7c and/or C18 : 1ω6c). The predominant quinone was ubiquinone-10, and the G+C content of the genomic DNA of strain D3 was 73.1 mol%. The major polyamine was spermidine. The major polar lipids of the isolate were diphosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine and phosphatidylglycerol. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain D3 clustered with Roseomonas aquatica TR53 within the genus Roseomonas. The 16S rRNA gene sequence of strain D3 showed the highest sequence similarity to R. aquatica TR53 (95.9 %), followed by Roseomonas rosea 173-96 (95.7 %) and Roseomonas aerilata 5420S-30 (95.0 %). Based on the phenotypic, phylogenetic and chemotaxonomic characterization, strain D3 represents a novel species of the genus Roseomonas, for which the name Roseomonas fluminis sp. nov. is proposed. The type strain is D3 (=KACC 19269 =JCM 31968 ).

[Ab] Resumo:OBJECTIVES: To explore the species, quantity and distribution of diatoms in Ningbo three-river watershed during summer and to provide scientific basis for forensic examination of drowning cases in the waters of Ningbo. METHODS: Water samples were collected in July and August of 2015. Fourteen water sampling points were selected from the Yao River, the Fenghua River and the Yong River. The morphological features of diatom species and dominant diatoms were distinguished by microscope. RESULTS: A total of 16 species of diatoms were detected in the Yao River, the Fenghua River and the Yong River. Melosira was the dominant species in the Yao River, and the quantity and richness were higher than in other rivers. The richness of Cyclotella in the Yong River was higher than in other rivers. The richness of Pinnularia and Licmophora were higher in the Fenghua River than in the Yao River and the Yong River. CONCLUSIONS: The species and proportion of diatom is different in each river. Database of the species and relative composition for the diatoms in corresponding river is established, which may provide data support for forensic examination of drowning cases in Ningbo three-river watershed.

[Ab] Resumo:OBJECTIVES: To explore the species distribution and constituent ratio of diatoms in main sections of urban district rivers where drowning occurs frequently in Chengdu. METHODS: Total 39 water samples from the sampling points of 5 rivers (Jinjiang, Jinniu, Qingyang, Wuhou and Chenghua districts) in October 2014 were collected. The diatoms smear were made and the species distribution and constituent ratio of diatoms from the water samples were analyzed using biological microscope and acquisition system of digital microscope. RESULTS: Total 21 species of diatoms were detected in main sections of urban district rivers in Chengdu. Significant differences in the dominant diatom species and proportions of the different rivers were observed, and there were different species existed in all sampling points of the upstream, midstream and downstream of the rivers. CONCLUSIONS: The database of species map, species distribution and constituent ratio of diatoms in main sections of urban district rivers in Chengdu are preliminarily established, which has special meaning for the analysis and evaluation of falling location inference using diatoms test in case investigation.

[Ab] Resumo:Measurement uncertainties are inherent to assessment of biological indices of water bodies. The effect of these uncertainties on the probability of misclassification of ecological status is the subject of this paper. Four Monte-Carlo (M-C) models were applied to simulate the occurrence of random errors in the measurements of metrics corresponding to four biological elements of surface waters: macrophytes, phytoplankton, phytobenthos, and benthic macroinvertebrates. Long series of error-prone measurement values of these metrics, generated by M-C models, were used to identify cases in which values of any of the four biological indices lay outside of the "true" water body class, i.e., outside the class assigned from the actual physical measurements. Fraction of such cases in the M-C generated series was used to estimate the probability of misclassification. The method is particularly useful for estimating the probability of misclassification of the ecological status of surface water bodies in the case of short sequences of measurements of biological indices. The results of the Monte-Carlo simulations show a relatively high sensitivity of this probability to measurement errors of the river macrophyte index (MIR) and high robustness to measurement errors of the benthic macroinvertebrate index (MMI). The proposed method of using Monte-Carlo models to estimate the probability of misclassification has significant potential for assessing the uncertainty of water body status reported to the EC by the EU member countries according to WFD. The method can be readily applied also in risk assessment of water management decisions before adopting the status dependent corrective actions.
