[Ab] Resumo:The green eggshell color produced by ducks is a threshold trait that can be influenced by various factors, such as hereditary, environment and nutrition. The aim of this study was to investigate the genetic regulation of the formation of eggs with green shells in Youxian ducks. We performed integrative analysis of mRNAs and miRNAs expression profiling in the shell gland samples from ducks by RNA-Seq. We found 124 differentially expressed genes that were associated with various pathways, such as the ATP-binding cassette (ABC) transporter and solute carrier supper family pathways. A total of 31 differentially expressed miRNAs were found between ducks laying green eggs and white eggs. KEGG pathway analysis of the predicted miRNA target genes also indicated the functional characteristics of these miRNAs; they were involved in the ABC transporter pathway and the solute carrier (SLC) supper family. Analysis with qRT-PCR was applied to validate the results of global gene expression, which showed a correlation between results obtained by RNA-seq and RT-qPCR. Moreover, a miRNA-mRNA interaction network was established using correlation analysis of differentially expressed mRNA and miRNA. Compared to ducks that lay white eggs, ducks that lay green eggs include six up-regulated miRNAs that had regulatory effects on 35 down-regulated genes, and seven down-regulated miRNAs which influenced 46 up-regulated genes. For example, the ABC transporter pathway could be regulated by expressing gga-miR-144-3p (up-regulated) with ABCG2 (up-regulated) and other miRNAs and genes. This study provides valuable information about mRNA and miRNA regulation in duck shell gland tissues, and provides foundational information for further study on the eggshell color formation and marker-assisted selection for Youxian duck breeding.

[Ab] Resumo: Diphenhydramine (DP), an antihistaminic agent, may become colored and daker or more fluorescent during storage. Herein, we spectroscopically examined the causes of this phenomenon under various DP storage conditions and durations. The infrared vibration-rotation spectrum shows multiple Gauche (G)-type conformers with different intramolecular n→π interaction strengths. The splitting pattern of the dimethylamino group protons in the H-NMR spectrum indicates that DP is mainly in the G-type with a small portion in the Trans (T)-type. The correlation between the red-shifted peak intensity in the UV•VIS absorbance spectrum and the coloring progression indicates a decreased intramolecular n→π interaction of the G-type under elevated temperature during storage. Enhanced fluorescence detected in the Excitation•Fluorescence spectrum demonstrates G-type (quenching) to T-type (fluorescent) conformation conversion, which is due to activated internal rotation of the dimethylamino group under elevated storage temperature and electronic excitation in the phenyl groups under light irradiation during storage. A signal detected in the ESR spectrum corresponds to the G-type charge transfer (CT) structure wherein part of the nonbonding electron pair on the N atom is intramolecularly redistributed to the phenyl groups. The CT structure presents the G-type quenching characteristics, whereas weak CT bonding corresponds to coloring. The results indicate that the quenching G-type is converted to T-type by heat or light to become color faded and bright with enhanced fluorescence and that T-type is reverted to G-type after storage under cool and dark conditions or by vacuum distillation to lose fluorescence.

[Ab] Resumo:PURPOSE: To assess whether the asymmetry in the peripapillary retinal nerve fiber layer (pRNFL) thickness between superior and inferior hemispheres on optical coherence tomography (OCT) is useful for early detection of glaucoma. MATERIALS AND METHODS: The patient population consisted of Training set (a total of 60 subjects with early glaucoma and 59 normal subjects) and Validation set (30 subjects with early glaucoma and 30 normal subjects). Two kinds of ratios were employed to measure the asymmetry between the superior and inferior pRNFL thickness using OCT. One was the ratio of the superior to inferior peak thicknesses (peak pRNFL thickness ratio; PTR), and the other was the ratio of the superior to inferior average thickness (average pRNFL thickness ratio; ATR). The diagnostic abilities of the PTR and ATR were compared to the color code classification in OCT. Using the optimal cut-off values of the PTR and ATR obtained from the Training set, the two ratios were independently validated for diagnostic capability. RESULTS: For the Training set, the sensitivities/specificities of the PTR, ATR, quadrants color code classification, and clock-hour color code classification were 81.7%/93.2%, 71.7%/74.6%, 75.0%/93.2%, and 75.0%/79.7%, respectively. The PTR showed a better diagnostic performance for early glaucoma detection than the ATR and the clock-hour color code classification in terms of areas under the receiver operating characteristic curves (AUCs) (0.898, 0.765, and 0.773, respectively). For the Validation set, the PTR also showed the best sensitivity and AUC. CONCLUSION: The PTR is a simple method with considerable diagnostic ability for early glaucoma detection. It can, therefore, be widely used as a new screening method for early glaucoma.

[Ab] Resumo:Brown eggs are popular in many countries, and consumers regard eggshell brownness as an important indicator of egg quality. Brown eggshell color is controlled by polygene. However, the responsible genes and detailed molecular mechanisms regulating eggshell brownness have not been defined. In the present study, we applied the RNA-seq technology to analyze the transcriptome data of the shell gland epithelium of hens and investigated the candidate genes associated with eggshell brownness. The results indicated that 8461 genes were expressed in the shell gland epithelium, of which 34 genes were differentially expressed in hens laying dark vs. light brown eggs. Functional analysis revealed that two genes, ovotransferrin (TF) and heat-shock protein 70 (HSP70), as well as the oxidative phosphorylation pathway were involved in the synthesis and transport of protoporphyrin â…¨, which might influence the formation of eggshell brownness and result in different shades of brown.

[Ab] Resumo:The Jurassic Yanliao theropods have offered rare glimpses of the early paravian evolution and particularly of bird origins, but, with the exception of the bizarre scansoriopterygids, they have shown similar skeletal and integumentary morphologies. Here we report a distinctive new Yanliao theropod species bearing prominent lacrimal crests, bony ornaments previously known from more basal theropods. It shows longer arm and leg feathers than Anchiornis and tail feathers with asymmetrical vanes forming a tail surface area even larger than that in Archaeopteryx. Nanostructures, interpreted as melanosomes, are morphologically similar to organized, platelet-shaped organelles that produce bright iridescent colours in extant birds. The new species indicates the presence of bony ornaments, feather colour and flight-related features consistent with proposed rapid character evolution and significant diversity in signalling and locomotor strategies near bird origins.

[Ab] Resumo:An optimal design of light-emitting diode (LED) lighting that benefits both the photosynthesis performance for plants and the visional health for human eyes has drawn considerable attention. In the present study, we have developed a multi-color driving algorithm that serves as a liaison between desired spectral power distributions and pulse-width-modulation duty cycles. With the aid of this algorithm, our multi-color plant-growth light sources can optimize correlated-color temperature (CCT) and color rendering index (CRI) such that photosynthetic luminous efficacy of radiation (PLER) is maximized regardless of the number of LEDs and the type of photosynthetic action spectrum (PAS). In order to illustrate the accuracies of the proposed algorithm and the practicalities of our plant-growth light sources, we choose six color LEDs and German PAS for experiments. Finally, our study can help provide a useful guide to improve light qualities in plant factories, in which long-term co-inhabitance of plants and human beings is required.

[Ab] Resumo:The spontaneous occurrence of colour preferences without learning has been demonstrated in several insect species; however, the underlying mechanisms are still not understood. Here, we use a comparative approach to investigate spontaneous and learned colour preferences in foraging bees of two tropical and one temperate species. We hypothesised that tropical bees utilise different sets of plants and therefore might differ in their spontaneous colour preferences. We tested colour-naive bees and foragers from colonies that had been enclosed in large flight cages for a long time. Bees were shortly trained with triplets of neutral, UV-grey stimuli placed randomly at eight locations on a black training disk to induce foraging motivation. During unrewarded tests, the bees' responses to eight colours were video-recorded. Bees explored all colours and displayed an overall preference for colours dominated by long or short wavelengths, rather than a single colour stimulus. Naive Apis cerana and Bombus terrestris showed similar choices. Both inspected long-wavelength stimuli more than short-wavelength stimuli, whilst responses of the tropical stingless bee Tetragonula iridipennis differed, suggesting that resource partitioning could be a determinant of spontaneous colour preferences. Reward on an unsaturated yellow colour shifted the bees' preference curves as predicted, which is in line with previous findings that brief colour experience overrides the expression of spontaneous preferences. We conclude that rather than determining foraging behaviour in inflexible ways, spontaneous colour preferences vary depending on experimental settings and reflect potential biases in mechanisms of learning and decision-making in pollinating insects.

[Ab] Resumo:STATEMENT OF PROBLEM: Three-dimensional (3D) intraoral scanning systems allow for the simultaneous acquisition of 3D information about tooth surfaces and a photorealistic view of the patient's tooth colors. AIM: The goal of this study was the in vivo comparison of a new 3D scanner with a color acquisition mode and conventional visual and digital color measurements. MATERIALS AND METHODS: The colors of 40 teeth of 20 patients were evaluated in seven ways: 1) By dentists using the Vita 3D-Master; 2) By dental technicians using the Vita 3D-Master; 3) With the 3Shape Trios device; 4) With the Vita Easyshade device; 5) With the Vita Easyshade Advance device; 6) With the SpectroShade device; and 7) With the SpectroShade Micro device. Digital measurements of Groups 3 to 7 were repeated three times for each tooth. For all groups, both the CIE Lab values and the Vita 3D-Master values were recorded. The repeatability and relative accuracy of the Vita 3D-Master values were analyzed statistically using Pearson's chi-squared test (α < 0.05). ΔE values were calculated from the CIE Lab values, which served as a basis for performing multidimensional scaling (MDS) and evaluating differences between the groups using the one-way ANOVA with post hoc Tamhane's test (α < 0.05). RESULTS: The results of the ΔE values showed that clinically relevant differences between the evaluation by dentists, dental technicians, and the intraoral scanning device (3Shape) are negligible. The intraoral 3D scanning device (Group 3) and the digital systems (Groups 4 to 7) did not differ significantly in the repeatability of color shade management. The SpectroShade Micro (Group 7) had significantly better relative accuracy than the other devices. CONCLUSIONS: The results demonstrate that intraoral scanning systems can be used to measure both tooth color and tooth surface in 3D. CLINICAL IMPLICATIONS: Intraoral optical scanning devices allow for the acquisition of accurate 3D surface data. Tooth color can be evaluated simultaneously and can be used to determine the color of restorations without requiring additional conventional color-measurement methods.

[Ab] Resumo:As consequences of global warming sea-ice shrinking, permafrost thawing and changes in fresh water and terrestrial material export have already been reported in the Arctic environment. These processes impact light penetration and primary production. To reach a better understanding of the current status and to provide accurate forecasts Arctic biogeochemical and physical parameters need to be extensively monitored. In this sense, bio-optical properties are useful to be measured due to the applicability of optical instrumentation to autonomous platforms, including satellites. This study characterizes the non-water absorbers and their coupling to hydrographic conditions in the poorly sampled surface waters of the central and eastern Arctic Ocean. Over the entire sampled area colored dissolved organic matter (CDOM) dominates the light absorption in surface waters. The distribution of CDOM, phytoplankton and non-algal particles absorption reproduces the hydrographic variability in this region of the Arctic Ocean which suggests a subdivision into five major bio-optical provinces: Laptev Sea Shelf, Laptev Sea, Central Arctic/Transpolar Drift, Beaufort Gyre and Eurasian/Nansen Basin. Evaluating ocean color algorithms commonly applied in the Arctic Ocean shows that global and regionally tuned empirical algorithms provide poor chlorophyll-a (Chl-a) estimates. The semi-analytical algorithms Generalized Inherent Optical Property model (GIOP) and Garver-Siegel-Maritorena (GSM), on the other hand, provide robust estimates of Chl-a and absorption of colored matter. Applying GSM with modifications proposed for the western Arctic Ocean produced reliable information on the absorption by colored matter, and specifically by CDOM. These findings highlight that only semi-analytical ocean color algorithms are able to identify with low uncertainty the distribution of the different optical water constituents in these high CDOM absorbing waters. In addition, a clustering of the Arctic Ocean into bio-optical provinces will help to develop and then select province-specific ocean color algorithms.

[Ab] Resumo:A wide variety of crystallization solutions are screened to establish conditions that promote the growth of a diffraction-quality crystal. Screening these conditions requires the assessment of many crystallization plates for the presence of crystals. Automated systems for screening and imaging are very expensive. A simple approach to imaging trace fluorescently labeled protein crystals in crystallization plates has been devised, and can be implemented at a cost as low as $50. The proteins ß-lactoglobulin B, trypsin and purified concanavalin A (ConA) were trace fluorescently labeled using three different fluorescent probes: Cascade Yellow (CY), Carboxyrhodamine 6G (CR) and Pacific Blue (PB). A crystallization screening plate was set up using ß-lactoglobulin B labeled with CR, trypsin labeled with CY, ConA labeled with each probe, and a mixture consisting of 50% PB-labeled ConA and 50% CR-labeled ConA. The wells of these plates were imaged using a commercially available macro-imaging lens attachment for smart devices that have a camera. Several types of macro lens attachments were tested with smartphones and tablets. Images with the highest quality were obtained with an iPhone 6S and an AUKEY Ora 10× macro lens. Depending upon the fluorescent probe employed and its Stokes shift, a light-emitting diode or a laser diode was used for excitation. An emission filter was used for the imaging of protein crystals labeled with CR and crystals with two-color fluorescence. This approach can also be used with microscopy systems commonly used to observe crystallization plates.
