[Ab] Resumo:The mechanisms of thrombogenicity in essential thrombocythemia (ET) are complex and not well defined. Our objective was to explore whether phosphatidylserine (PS) exposure on blood cells and endothelial cells (ECs) can account for the increased thrombosis and distinct thrombotic risks among mutational subtypes in ET. Using flow cytometry and confocal microscopy, we found that the levels of PS-exposing erythrocytes, platelets, leukocytes, and serum-cultured ECs were significantly higher in each ET group [JAK2, CALR, and triple-negative (TN) (all P < 0.001)] than those in controls. Among ET patients, those with JAK2 mutations showed higher levels of PS-positive erythrocytes, platelets, neutrophils, and serum-cultured ECs than TN patients or those with CALR mutations, which show similar levels. Coagulation function assays showed that higher levels of PS-positive blood cells and serum-cultured ECs led to markedly shortened coagulation time and dramatically increased levels of FXa, thrombin, and fibrin production. This procoagulant activity could be largely blocked by addition of lactadherin (approx. 70% inhibition). Confocal microscopy showed that the FVa/FXa complex and fibrin fibrils colocalized with PS on ET serum-cultured ECs. Additionally, we found a relationship between D-dimer, prothrombin fragment F1 + 2, and PS exposure. Our study reveals a previously unrecognized link between hypercoagulability and exposed PS on cells, which might also be associated with distinct thrombotic risks among mutational subtypes in ET. Thus, blocking PS-binding sites may represent a new therapeutic target for preventing thrombosis in ET.

[Ab] Resumo:Understanding the sorption mechanisms for organophosphate flame retardants (OPFRs) on impervious surfaces is important to improve our knowledge of the fate and transport of OPFRs in indoor environments. The sorption processes of semivolatile organic compounds (SVOCs) on indoor surfaces are heterogeneous (multilayer sorption) or homogeneous (monolayer sorption). In this study, we adopted simplified Langmuir isotherm and Freundlich isotherm in a dynamic sink model to characterize the sorption dynamics of OPFRs on impervious surfaces such as stainless steel and made comparisons between the two models through a series of empty chamber studies. The tests involve two types of stainless steel chambers (53-L small chambers and 44-mL micro chambers) using tris(2-chloroethyl)phosphate (TCEP) and tris(1-chloro-2-propyl)phosphate (TCPP) as target compounds. Our test results show that the dynamic sink model using Freundlich isotherm can better represent the sorption process in the empty small chamber. Micro chamber test results from this study show that the sink model using both simplified Langmuir isotherm and Freundlich isotherm can well fit the measured gas-phase concentrations of OPFRs. We further applied both models and the parameters obtained to predict the gas phase concentrations of OPFRs in a small chamber with an emission source. Comparisons between model predictions and measurements demonstrate the reliability and applicability of the sorption parameters.

[Ab] Resumo:The study is aimed to investigate the impact of silver nanoparticles on germination of Pisum sativum pea seeds. The influence of synthesized silver nanoparticles on root length and percentage of germinated seeds was revealed. It was found that nanosilver treatment agents do not affect the germination of pea seeds negatively at low concentrations. Also, the treatment of pea seeds with silver nanoparticles provide a significant positive effect on the root length of pea seeds.

[Ab] Resumo:Batch sorption and leaching of arsenic (1-30mgL ) on Fe-sericite composite powder and beads were investigated in this study. Fe-sericite composite powder was made from natural sericite modified with iron, and alginate was used to transform the powder into beads. The maximum sorption capacities of the Fe-sericite composite powder (15.04 and 13.21mgg for As(III) and As(V), respectively) were higher than those of the corresponding beads (9.02 and 7.11mgg for As(III) and As(V), respectively) owing to the higher specific surface area of the powder. In addition, the leaching amounts of As(III) from Fe-sericite composite beads (≤ 15.03%) were higher than those of the corresponding powder (≤ 5.71%). However, acute toxicity of As(III)-sorbed Fe-sericite composite beads toward Daphnia magna was not significantly different from that of the corresponding powder (p > 0.05). Considering higher uptake of the powder particles by the daphnids, Fe-sericite composite beads seem to be a more appropriate and safer sorbent for arsenic removal in practical application. Based on Fe content, Fe-sericite composite beads had similar or higher maximum sorption capacities (71.19 and 56.11mgg Fe for As(III) and As(V), respectively) than those of previously reported sorbents.

[Ab] Resumo:Suppression of plant growth is a common problem in post-mining reclaimed areas, as coarse texture of soils may increase nitrate leaching. Assessing feasibility of using solid waste (precipitated solid matter) produced by water and sewage treatment processes in field conditions is very important in mine soil reclamation. Our work investigated the possibility of plant growth in a degraded site covered with sewage-derived sludge material. A test area (21m × 18m) was established on a mine soil heap. Experimental plant species included Camelina sativa, Helianthus annuus, Festuca rubra, Miscanthus giganteus, Amaranthus cruentus, Brassica napus, Melilotus albus, Beta vulgaris, and Zea mays. ANOVA showed sufficient water content and acceptable physical properties of the soil in each year and layer in a multi-year period, indicating that these species were suitable for phytoremediation purposes. Results of trace elements assays indicated low degree of contamination caused by Carbocrash waste material and low potential ecological risk for all plant species. Detrended correspondence analysis revealed that total porosity and capillary porosity were the most important variables for the biosolids among all water content related properties. Overall, crop plants were found useful on heavily degraded land and the soil benefited from their presence. An addition of Carbocrash substrate to mine soil improved the initial stage of soil reclamation and accelerated plant growth. The use of this substrate in phytoremediation helped to balance the content of nutrients, promoted plant growth, and increased plant tolerance to salinity. Sewage sludge-amended biosolids may be applied directly to agricultural soil, not only in experimental conditions.

[Ab] Resumo:Molecular rotors with an off-center axis and the chiral feature of achiral CuPc molecules on a semi-metallic Bi(111) surface have been investigated by means of a scanning tunneling microscopy (STM) at liquid nitrogen (LN2) temperature. The rotation axis of each CuPc molecular rotor is located at the end of a phthalocyanine group. As molecular coverage increases, the CuPc molecules are self-assembled into various nanoclusters and finally into two-dimensional (2D) domains, in which each CuPc molecule exhibits an apparent chiral feature. Such chiral features of the CuPc molecules can be attributed to the combined effect of asymmetric charge transfer between the CuPc and Bi(111) substrate, and the intermolecular van der Waals interactions.

[Ab] Resumo:Bone regeneration is a complicated process and includes a number of distinct and sequential stages of coordinated cellular actions under the regulation of multiple growth factors. Therefore, bone grafting materials in which growth factors can be incorporated and released in a programmed order in line with the bone tissue healing process may lead to desirable clinical outcomes. In the present study, a double-layered chitosan/gelatin/genipin (d-CSG/G) nanosphere coating is developed by using layer-by-layer electrophoretic deposition and genipin crosslinking. The surface morphology, physicochemical and mechanical properties of the coatings are explored. Cytochrome C is used as a therapeutic model protein and is successfully loaded on the inner and outer layers of the coating. The protein release can be controlled by the loading position, genipin concentration and thickness of the outer layer. Furthermore, the cell response to the coatings was evaluated. Real-time polymerase chain reactions, immunofluorescence staining and extracellular matrix mineralization assay confirmed that the functions of the loaded growth factor are fully preserved after fabrication. Overall, the d-CSG/G nanosphere coating could be a promising growth factor delivery system to promote bone tissue regeneration.

[Ab] Resumo:We report on the fabrication of silicon-reinforced carbon (C:Si) structures by combinatorial pulsed laser deposition to search for the best design for a new generation of multi-functional coated implants. The synthesized films were characterized from the morphological, structural, compositional, mechanical and microbiological points of view. Scanning electron microscopy revealed the presence, on top of the deposited layers, of spheroid particulates with sizes in the micron range. No micro-cracks or delaminations were observed. Energy dispersive x-ray spectroscopy and grazing incidence x-ray diffraction pointed to the existence of a C to Si compositional gradient from one end of the film to the other. Raman investigation revealed a relatively high sp hybridization of up to 80% at 40-48 mm apart from the edge with higher C content. Si addition was demonstrated to significantly increase C:Si film bonding to the substrate, with values above the ISO threshold for coatings to be used in high-loading biomedical applications. Surface energy studies pointed to an increase in the hydrophilic character of the deposited structures along with Si content up to 52 mN m . In certain cases, the Si-reinforced C coatings elicited an antimicrobial biofilm action. The presence of Si was proven to be benign to HEp-2 cells of human origin, without interfering with their cellular cycle. On this basis, reliable C:Si structures with good adherence to the substrate and high efficiency against microbial biofilms can be developed for implant coatings and other advanced medical devices.

[Ab] Resumo:The modification of regenerated cellulose (RC) membranes was carried out by using silane coupling agents presenting primary and secondary amino-groups. The grafting of the amino groups onto the modified cellulose molecule was confirmed by X-ray photoelectron spectroscopies and C nuclear magnetic resonance spectroscopic analyses. The crystallinity of the cellulose membranes (CM) decreased after chemical modification as indicated by the X-ray diffraction results. Moreover, a denser structure was observed at the surface and cross section of the modified membranes by SEM images. The contact angle measurements showed that the silane coupling treatment enhanced the hydrophobicity of the obtained materials. Then the catalytic properties of two types of modified membranes were studied in a batch process by evaluating their catalytic performance in a Knoevenagel condensation. The results indicated that the cellulose membrane grafted with many secondary amines exhibited a better catalytic activity compared to the one grafted only by primary amines. In addition, the compact structure of the modified membranes permitted their application in a pervaporation catalytic membrane reactor. Therefore, functional CM that prepared in this paper represented a promising material in the field of industrial catalysis.

[Ab] Resumo:The present study aimed to evaluate the microtensile bond strength (µTBS) of novel computer-aided design/computer-aided manufacturing (CAD/CAM) restorative materials to dentin using different adhesive strategies. Thirty-two crowns were milled using CAD/CAM materials (Vita Mark II as control, Vita Suprinity, Vita Enamic and Lava Ultimate) and luted to dentin using different resin cements (RelyX ARC, RelyX Unicem 2 and RelyX Ultimate). The specimens were stored in 100% relative humidity at 37°C for 24 h and sectioned. The samples (n = 16) with cross-sectional areas of approximately 0.90 mm2, were submitted to a µTBS test in a universal testing machine with a crosshead speed of 0.5 mm/min. The samples were analyzed with SEM to determinate the failure mode. According to 2-way ANOVA and Tukey's test (α=0.05), the interaction effect (material x luting strategy) was significant (p=0.001). Regardless of the luting strategy, a higher µTBS was obtained with Lava Ultimate and Vita Enamic, which were significantly different from Vita Mark II and Vita Suprinity (p<0.05). For Vita Mark II, Vita Suprinity, Vita Enamic, and Lava Ultimate, the µTBS obtained with RelyX Unicem 2 was not significantly different from that obtained with RelyX ARC or RelyX Ultimate. All groups obtained at least three types of failure. The adhesive strategy with self-adhesive resin cement was comparable to conventional resin cement with total-etch or self-etch adhesive techniques in the bond of novel CAD/CAM materials to dentin.
