[Ab] Resumo:This study aimed to investigate the fermentation quality and nutritive value of total mixed ration (TMR) silages based on desert wormwood (DW) combined with early stage corn (ESC) as forage and determine an optimum formula. Desert wormwood and ESC were harvested, chopped, and mixed with other ingredients according to a formula, packed into laboratory silos at densities of 500-550 g/L, and stored in the dark for 60 days. The DW proportions in the forage of TMR were 1, 0.75, 0.50, 0.25 and 0, based on fresh weight. As the proportion of DW decreased, the pH also decreased (P < 0.05), while lactic acid, lactic acid/acetic acid, crude protein, starch, and the in vitro digestibility of dry matter and neutral detergent fiber increased (P < 0.05). Ammonia nitrogen/total nitrogen in the TMR silages with DW proportions of 0.75, 0.25 and 0 in the forage was more than 10%. These results indicated that the quality of the TMR silage containing DW alone as forage was poor, TMR silages containing DW proportions of 0.75 and 0.25, and ESC alone, in the forage were not well preserved. The optimum TMR silage formula contained a DW proportion of 0.5 in the forage.

[Ab] Resumo:BACKGROUND: The objective of the present study was to evaluate the effects of four chemicals on the fermentation quality and aerobic stability of alfalfa (Medicago sativa L.) silage. Wilted alfalfa was ensiled without additive (control), or with formic acid (FA), potassium diformate (KDF), sodium diacetate (SDA) or calcium propionate (CAP). RESULTS: After 60 days of ensiling, the pH values in FA, KDF and SDA silages were lower (P < 0.05) compared to that of control and CAP silages, and chemicals (P < 0.05) decreased butyric acid and ammonia N concentrations and populations of aerobic bacteria and yeasts compared to the control. The SDA and CAP silages had a higher (P < 0.05) lactic acid bacteria content compared to the FA and KDF silages. The SDA and CAP silages had higher (P < 0.05) acetic and propionic acid contents compared to the other silages, respectively. The ammonia N concentrations in the FA and KDF silages were lower compared to the other silages during the first 5 days of aerobic exposure, and then increased sharply to 105 and 100 g kg total N, respectively, which was higher (P < 0.05) than that of the SDA and CAP silages on day 9 of aerobic exposure. Yeasts and aerobic bacteria counts in SDA silage slowly increased and remained at lower levels compared to the other silages after 7 days of aerobic exposure. CONCLUSION: Additives prolonged the aerobic stability duration compared to the control, and the SDA and CAP silages remained stable for more than 216 h, followed by the KDF and FA silages (202 and 196 h, respectively). © 2017 Society of Chemical Industry.

[Ab] Resumo:This study evaluated the effects of 3 widely practiced cow feeding systems in the United States, Europe, and Southern Hemisphere regions on the characteristics, quality, and consumer perception of sweet cream butter. Fifty-four multiparous and primiparous Friesian cows were divided into 3 groups (n=18) for an entire lactation. Group 1 was housed indoors and fed a total mixed ration diet (TMR) of grass silage, maize silage, and concentrates; group 2 was maintained outdoors on perennial ryegrass-only pasture (GRS); and group 3 was maintained outdoors on a perennial ryegrass/white clover pasture (CLV). Mid-lactation butter was manufactured in triplicate with milk from each group in June 2015 (137±7d in milk) and was analyzed over a 6-mo storage period at 5°C for textural and thermal properties, fatty acid composition, sensory properties, and volatile compounds. The nutritional value of butters was improved by pasture feeding, and butter from pasture-fed cows had significantly lower thrombogenicity index scores compared with butters from TMR-fed cows. In line with these results, pasture-derived milks (GRS and CLV) produced butter with significantly higher concentrations of conjugated linoleic acid (cis-9,trans-11) and trans-ß-carotene than TMR butter. Alterations in the fatty acid composition of butter contributed to significant differences in textural and thermal properties of the butters. Total mixed ration-derived butters had significantly higher hardness scores at room temperature than those of GRS and CLV. Onset of crystallization for TMR butters also occurred at significantly higher temperatures compared with pasture butters. Volatile analysis of butter by gas chromatography-mass spectrometry identified 25 compounds present in each of the butters, 5 of which differed significantly based on feeding system, including acetone, 2-butanone, 1-pentenol, toluene, and ß-pinene. Toluene was very significantly correlated with pasture-derived butter. Sensory analysis revealed significantly higher scores for GRS-derived butter in several attributes including "liking" of appearance, flavor, and color over those of TMR butter. Partial least square regression plots of fatty acid profiles showed clear separation of butter derived from grazed pasture-based perennial ryegrass or perennial rye/white clover diets from that of a TMR system, offering further insight into the ability of fatty acid profiling to verify such pasture-derived dairy products.

[Ab] Resumo:Brewer's spent grain (BSG) could be tested as an alternative source of polyphenols in animal nutrition. Proper extraction and analytical methods are critical for quantification. Thus, extraction for BSG, corn silage, and brans of rice, corn, and wheat were studied for the highest yield of polyphenols. A method for 18 phenolic monomers by HPLC-DAD was developed, validated, and applied to samples. An aqueous solution of NaOH (0.75% w/v) using integral samples for extraction resulted in the highest values for colorimetric measurements in all analyzed sources. Method by maceration showed the highest phenolic yield when applied in corn silage and BSG. However, for brans the best method was microwave assisted. Results from HPLC-DAD analysis clearly showed that native structures of phenolic compounds were simplified to its monomers allowing quantification and sample discrimination. BSG had the highest concentration of polyphenols and could be a promising and innovative source for animal feed studies.

[Ab] Resumo:Three Gram-stain positive, non-motile, non-spore-forming, catalase-negative and rod-shaped bacterial strains (IWT5T, IWT25T and IWT140), isolated from silage, were investigated by using a polyphasic taxonomic approach. Strains IWT5T and IWT25T grew at 10-37 °C and 30-37 °C, and at pH 4.0-7.5 and 4.0-7.0, respectively. The G+C contents of genomic DNA of strains IWT5T and IWT25T were 43.2 and 44.4 mol%, respectively. Strains IWT5T and IWT25T contained C16 : 0, C18 : 1 ω9c and summed feature 7 (unknown 18.846/C19 : 1 ω6c/C19 : 0cyclo ω10c) as the major fatty acids. Strain IWT5T was most closely related to the type strains of Lactobacillus mixtipabuli (99.9 % 16S rRNA gene sequence similarity) and Lactobacillus silagei (99.5 %). For IWT25T, the 16S rRNA gene sequence similarities with the closely related neighbour type strains L. mixtipabuli and L. silagei were 99.5 and 99.5 %, respectively. The 16S rRNA gene sequence similarities among the three novel isolates were 99.5-99.9 %. The average nucleotide identities of strains IWT5T and IWT25T to other neighbours of the genus Lactobacillus were less than 82 % and the genomes of IWT25T and IWT140 shared 97.3 % average nucleotide identity, demonstrating that the three strains were allocated to two different novel species of the genus Lactobacillus. Together with multilocus sequence analysis, phenotypic and chemotaxonomic characteristics, strains IWT5T (=JCM 31144T=DSM 102973T) and IWT25T (=JCM 31145T=DSM 102974T) are proposed as the type strains of novel species of the genus Lactobacillus, with the names Lactobacillus silagincola sp. nov. and Lactobacillus pentosiphilus sp. nov., respectively.

[Ab] Resumo:The silage quality and anaerobic digestion performance of Pennisetum purpereum with molasses-processed wastewater addition were assessed. The silage samples with molasses-processed wastewater addition obtained higher lactic acid concentration of 15.18-23.38mg/g FM, lower pH value of 3.96-4.45 and lower NH -N content of 0.47-0.64mg/g FM. No obvious difference was observed in the dominant phyla and genus, but the relative abundance of Lactobacillus reached up to 77.39%, increased by 84% compared to the silage samples without molasses-processed wastewater addition. Combined the silage quality and bacterial community, the decreased in pH value of fresh material caused by molasses-processed wastewater addition was the main reason for improving the silage quality. Meanwhile, the increased in COD concentration is beneficial for improving the specific methane yield and the maximum specific methane yield of 259±5.75mL/g VS was obtained with adding 20g/kg molasses-alcoholic wastewater. Molasses-processed wastewater is an alternative additive for silage.

[Ab] Resumo:A newly developed fixed bed disc reactor (FBDR) which combines biofilm formation on biofilm carriers and reactor agitation in one single system was assessed for its applicability to demand-driven biogas production by variable feeding of sugar beet silage. Five different feeding patterns were studied at an organic loading of 4g L d under mesophilic and thermophilic conditions. High methane yields of 449-462L kg were reached. Feeding variable punctual loadings caused immediate response with 1.2- to 3.5-fold increase in biogas production rates within 15min. Although variable feeding did not induce process instability, a temporary decrease in pH-value and methane concentration below 40% occurred. Thermophilic temperature was advantageous as it resulted in a more rapid, higher methane production and less pronounced decrease in methane content after feeding. The FBDR was demonstrated to be well-suited for flexible biogas production, but further research and comparison with traditional reactor systems are required.

[Ab] Resumo:Silage processing has a crucial positive impact on the methane yield of anaerobic treated substrates. Changes in the characteristics of switchgrass after ensiling with different additives and their effects on methane production and microbial community changes during anaerobic digestion were investigated. After ensiling (CK), methane yield was increased by 33.59% relative to that of fresh switchgrass (FS). In comparison with the CK treatment, methane production was improved by 17.41%, 13.08% and 8.72% in response to ensiling with LBr+X, LBr and X, respectively. A modified Gompertz model predicted that the optimum treatment was LBr+X, with a potential cumulative methane yield of 178.31mL/g total solids (TS) and a maximum biogas production rate of 44.39mL/g TS·d. Firmicutes and Bacteroidetes were the predominant bacteria in FS and silage switchgrass; however, the switchgrass treated with LBr+X was rich in Synergistetes, which was crucial for methane production.

[Ab] Resumo:We investigated the effects of different silage storing conditions on the oxygen concentration in the silo and fermentation quality of rice (Oryza sativa L.). Forage rice was ensiled in bottles (with or without space at the bottlemouth, with solid or pinhole cap, and with oxygen scavenger, ethanol transpiration agent, oxygen scavenger and ethanol transpiration agent, or no adjuvant) and stored for 57 days. The oxygen concentration decreased with the addition of the oxygen scavenger and increased with that of the ethanol transpiration agent. The oxygen scavenger facilitated silage fermentation and fungus generation, whereas the ethanol transpiration agent suppressed silage fermentation and fungus generation. However, the combined use of the oxygen scavenger and ethanol transpiration agent facilitated silage fermentation and also suppressed fungus generation. Overall, this study revealed the negative effects of oxygen on the internal silo and the positive effects of the combined use of the oxygen scavenger and ethanol transpiration agent on silage fermentation quality.

[Ab] Resumo:Fatty acids, volatile compounds and sensory attributes of beef from bulls fed concentrates to slaughter (C), grass silage for 120days (GS) followed by C (GSC), or GS followed by 100days at pasture and then C (GSPC), and slaughtered at 3 target carcass weights, were determined. Total intramuscular fat (IMF) was lower for GSPC than for GSC and C. C18:3n-3 concentration and polyunsaturated fatty acid (PUFA) to saturated fatty acid (SFA) ratio were higher and C18:2n-6 and monounsaturated fatty acid concentrations and n-6:n-3 PUFA ratio lower for GSPC than C. C16:0, C18:0 and C18:1c9 increased with carcass weight when expressed quantitatively, but not when expressed proportionately. Hexanal concentration was higher and 2-methyl-1-butanol and toluene lower for C and GSC than for GSPC. Overall liking was negatively correlated with C20:5n-3 and PUFA/SFA ratio, but differences in sensory attributes (tenderness, flavour liking, overall liking) were most strongly correlated with IMF.
