[Ab] Resumo:Removal of toxic Cr(VI) by microbial reduction is a promising approach to reducing its ecotoxicological impact. To develop bioremediation technologies, many studies have evaluated the application of microorganisms isolated from Cr(VI)-contaminated sites. Nonetheless, little attention has been given to microbes from the environments without a history of Cr(VI) contamination. In this study, we aimed to characterize the Cr(VI) tolerance and removal abilities of a filamentous fungus strain, SL2, isolated from indoor air. Based on phenotypic characterization and rDNA sequence analysis, SL2 was identified as Penicillium oxalicum, a species that has not been extensively studied regarding Cr(VI) tolerance and reduction abilities. SL2 showed high tolerance to Cr(VI) on solid and in liquid media, facilitating its application to Cr(VI)-contaminated environments. Growth curves of SL2 in the presence of 0, 100, 400, or 1000 mg/L Cr(VI) were well simulated by the modified Gompertz model. The relative maximal colony diameter and maximal growth rate decreased as Cr(VI) concentration increased, while the lag time increased. SL2 manifested remarkable efficacy of removing Cr(VI). Mass balance analysis indicated that SL2 removed Cr(VI) by reduction, and incorporated 0.79 mg of Cr per gram of dry biomass. In electroplating wastewater, the initial rate of Cr(VI) removal was affected by the initial contaminant concentration. In conclusion, P. oxalicum SL2 represents a promising new candidate for Cr(VI) removal. Our results significantly expand the knowledge on potential application of this microorganism.

[Ab] Resumo:Bacterial-fungal interactions are widely found in distinct environments and contribute to ecosystem processes. Previous studies of these interactions have mostly been performed in soil, and only limited studies of aerial plant tissues have been conducted. Here we show that a seed-borne plant pathogenic bacterium, Burkholderia glumae (Bg), and an air-borne plant pathogenic fungus, Fusarium graminearum (Fg), interact to promote bacterial survival, bacterial and fungal dispersal, and disease progression on rice plants, despite the production of antifungal toxoflavin by Bg. We perform assays of toxoflavin sensitivity, RNA-seq analyses, lipid staining and measures of triacylglyceride content to show that triacylglycerides containing linolenic acid mediate resistance to reactive oxygen species that are generated in response to toxoflavin in Fg. As a result, Bg is able to physically attach to Fg to achieve rapid and expansive dispersal to enhance disease severity.

[Ab] Resumo:The detection of aerosolized viruses can serve as an important surveillance and control tool in agriculture, human health, and environmental settings. Here, we adapted an anion exchange resin-based method, initially developed to concentrate negatively charged viruses from water, to liquid impingement-based bioaerosol sampling. In this method, aerosolized viruses are collected in a 20ml liquid sample contained within widely used impingers, BioSamplers (SKC Inc., Eighty Four, PA), and further concentrated via adsorption to an anion exchange resin that is suspended within this liquid. Viral nucleic acids are then extracted from the resin to facilitate molecular analyses through a reduction in the effective sample volume. For this study, various quantities of two negatively charged viruses, type A and type B influenza viruses (FluMist Quadrivalent vaccine) and the male-specific (F+) RNA coliphage MS2 (MS2), were nebulized into a custom-built bioaerosolization chamber, and sampled using BioSamplers with and without anion exchange resin. Compared to direct testing of the BioSampler liquid, detection was improved by 6.77× and 3.33× for type A and type B influenza viruses, respectively, by using the anion exchange resin. For MS2, the anion exchange resin method allowed for an average improvement in detection of 8.26×.

[Ab] Resumo:There is limited information on the natural history of building occupants' health in relation to attempts to remediate moisture damage. We examined changes in respiratory and non-respiratory symptoms in 1,175 office building occupants over seven years with multiple remediation attempts. During each of four surveys, we categorized participants using a severity score: 0 = asymptomatic; 1 = mild, symptomatic in the last 12 months, but not frequently in the last 4 weeks; 2 = severe, symptomatic at least once weekly in the last 4 weeks. Building-related symptoms were defined as improving away from the building. We used random intercept models adjusted for demographics, smoking, building tenure, and microbial exposures to estimate temporal changes in the odds of building-related symptoms or severity scores independent of the effect of microbial exposures. Trend analyses of combined mild/severe symptoms showed no changes in the odds of respiratory symptoms but significant improvement in non-respiratory symptoms over time. Separate analyses showed increases in the odds of severe respiratory symptoms (odds ratio/year = 1.15‒1.16, p-values<0.05) and severity scores (0.02/year, p-values<0.05) for wheezing and shortness of breath on exertion, due to worsening of participants in the mild symptom group. For non-respiratory symptoms, we found no changes in the odds of severe symptoms but improvement in severity scores (-0.04‒-0.01/year, p-values<0.05) and the odds for mild fever and chills, excessive fatigue, headache, and throat symptoms (0.65-0.79/year, p-values<0.05). Our study suggests that after the onset of respiratory and severe non-respiratory symptoms associated with dampness/mold, remediation efforts might not be effective in improving occupants' health.

[Ab] Resumo:Demolition can generate fungal spore suspensions in association with various adverse health effects, such as high risk of invasive aspergillosis in immunocompromised patients. One block of Edouard Herriot Hospital was entirely demolished. The aim of the present study was to evaluate Hirst-type spore traps utility in monitoring outdoor Aspergillaceae (Aspergillus spp. + Penicillium spp.) spores in part of Edouard Herriot Hospital (Lyon, France) undergoing major demolition. Three periods were scheduled in 2015: (A) Gutting of building and asbestos removal, (B) Demolition of floors, (C) Excavation and earthwork. Outdoor Aspergillaceae fungal load was monitored by cultivable (Air Ideal®, bioMérieux) and non-cultivable methods (Lanzoni VPPS-2000, Analyzair®, Bologna, Italy). Differences of Aspergillaceae recorded with Hirst-type spore traps were observed between Gerland and Edouard Herriot Hospital. Differences between Aspergillaceae were recorded between day time and night time at Gerland and Edouard Herriot Hospital. Daily paired differences between Aspergillaceae recorded with non-cultivable methodology at Edouard Herriot Hospital and in an area without demolition work were significant in Period A vs Period B (p = 10-4) and Period A vs Period C (p = 10-4). Weak correlation of daily Aspergillaceae recorded by both methods at Edouard Herriot Hospital was significant only for Period C (r = 0.26, p = 0.048, n = 58). Meteorological parameters and type of demolition works were found to heavily influenced Aspergillaceae dispersion. Non-cultivable methodology is a promising tool for outdoor Aspergillaceae scrutiny during major demolition work in hospital, helping infection control staff to rapidly implement control measures.

[Ab] Resumo:Objective The aim of this study was to evaluate the quality of air in surgical centre rooms of a hospital in the South of Brazil. Methods An evaluation of the parameters of comfortability (temperature, luminosity, concentration of carbon dioxide and relative humidity), microbiological analysis and chromatographic of the particulate material present in the filters of air conditioners was carried out. Results Regarding the aspects of comfortability, three surgical rooms did not present temperature in accordance to the current legislation, but were found within the ideal patterns in other environmental factors. In the analysis of particulate material, a larger number of fungal bio-aerosols of viscous colonies was obtained than filamentous. Conclusion It was found that the main aspects related to the quality of air in the hospital being studied are being accomplished; however there is a need of modifying building aspects in the surgical rooms in order to reduce the possibility of contamination by air and decrease the concentration of carbon dioxide in the environment.

Diagnostics and care of the patients must be based on general guidelines, regardless of potential moisture damage in home or work place. No laboratory or other clinical tests exist that can be used to associate potential moisture damage to perceived symptoms of the patient. Buildings affected by moisture damages should be remediated, because it is likely to be beneficial to health of the occupants and other users and for the condition of the building.

[Ab] Resumo:Pollen, fungi, and bacteria are the main microscopic biological entities present in outdoor air, causing allergy symptoms and disease transmission and having a significant role in atmosphere dynamics. Despite their relevance, a method for monitoring simultaneously these biological particles in metropolitan environments has not yet been developed. Here, we assessed the use of the Hirst-type spore trap to characterize the global airborne biota by high-throughput DNA sequencing, selecting regions of the 16S rRNA gene and internal transcribed spacer for the taxonomic assignment. We showed that aerobiological communities are well represented by this approach. The operational taxonomic units (OTUs) of two traps working synchronically compiled >87% of the total relative abundance for bacterial diversity collected in each sampler, >89% for fungi, and >97% for pollen. We found a good correspondence between traditional characterization by microscopy and genetic identification, obtaining more-accurate taxonomic assignments and detecting a greater diversity using the latter. We also demonstrated that DNA sequencing accurately detects differences in biodiversity between samples. We concluded that high-throughput DNA sequencing applied to aerobiological samples obtained with Hirst spore traps provides reliable results and can be easily implemented for monitoring prokaryotic and eukaryotic entities present in the air of urban areas. Detection, monitoring, and characterization of the wide diversity of biological entities present in the air are difficult tasks that require time and expertise in different disciplines. We have evaluated the use of the Hirst spore trap (an instrument broadly employed in aerobiological studies) to detect and identify these organisms by DNA-based analyses. Our results showed a consistent collection of DNA and a good concordance with traditional methods for identification, suggesting that these devices can be used as a tool for continuous monitoring of the airborne biodiversity, improving taxonomic resolution and characterization together. They are also suitable for acquiring novel DNA amplicon-based information in order to gain a better understanding of the biological particles present in a scarcely known environment such as the air.

[Ab] Resumo:Air filtration has been shown to be efficient in reducing pathogen burden in circulating air. We determined at laboratory scale the retention efficiency of different air filter types either composed of a prefilter (EU class G4) and a secondary fiberglass filter (EU class F9) or consisting of a filter mat (EU class M6 and F8-9). Four filter prototypes were tested for their capability to remove aerosol containing equine arteritis virus (EAV), porcine reproductive and respiratory syndrome virus (PRRSV), bovine enterovirus 1 (BEV), Actinobacillus pleuropneumoniae (APP), and Staphylococcus (S.) aureus from air. Depending on the filter prototype and utilisation, the airflow was set at 1,800 m3/h (combination of upstream prefilter and fiberglass filter) or 80 m3/h (filter mat). The pathogens were aerosolized and their concentration was determined in front of and behind the filter by culture or quantitative real-time RT-PCR. Furthermore, survival of the pathogens over time in the filter material was determined. Bacteria were most efficiently filtered with a reduction rate of up to 99.9% depending on the filter used. An approximately 98% reduction was achieved for the viruses tested. Viability or infectivity of APP or PRRSV in the filter material decreased below the detection limit after 4 h and 24 h, respectively, whereas S. aureus was still culturable after 4 weeks. Our results demonstrate that pathogens can efficiently be reduced by air filtration. Consequently, air filtration combined with other strict biosecurity measures markedly reduces the risk of introducing airborne transmitted pathogens to animal facilities. In addition, air filtration might be useful in reducing bioaerosols within a pig barn, hence improving respiratory health of pigs.

[Ab] Resumo:A strictly aerobic Gram-stain-negative bacterium, designated strain KER25-10 , was isolated from a laboratory air conditioning system in South Korea. Cells were yellow-pigmented, non-motile rods showing catalase- and oxidase-positive reactions. The strain grew at pH 4.0-9.0 (optimum, pH 6.0-7.0) and 10-40 °C (optimum, 30 °C) and in the presence of 0-3 % (w/v) NaCl (optimum, 0 %). The G+C content of the genomic DNA was 65.1 mol%. Strain KER25-10 contained ubiquinone-10 (Q-10) as the predominant isoprenoid quinone and C16 : 0, C17 : 1ω6c, summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c) and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c) as the major fatty acids. The major polar lipids were sphingoglycolipid, diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and phosphatidylcholine. Only spermidine was detected as the polyamine. Phylogenetic analysis based on 16S rRNA sequences indicated that strain KER25-10 formed a distinct phylogenetic lineage within the genus Sphingomonas of the family Sphingomonadaceae and the strain was most closely related to Sphingomonas kyeonggiense THG-DT81 with a 96.8 % 16S rRNA gene sequence similarity. On the basis of phenotypic, chemotaxonomic and molecular features, strain KER25-10 clearly represents a novel species of the genus Sphingomonas, for which the name Sphingomonas frigidaeris sp. nov. is proposed. The type strain is KER25-10 (=KACC 19285 =JCM 32053 ).
